Effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs

  • Weaning is the most stressful event for nursery pigs because they are moved from familiar to unfamiliar environments. In addition, weaned pigs have immature digestive and immune systems. This situation makes weaned pigs susceptible to diseases and makes the absorption of nutrients from diets difficult. A feed approach, such as dietary enzyme supplementation, can be considered a solution. This study investigated the effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; combination of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Incidence of diarrhea, packed cell volume (PCV), white blood cells (WBC) count, and immunoglobulin content were measured. A significantly lower incidence of diarrhea (p < 0.05) was observed in the Cocktail group as compared with the CON group. The Cocktail group also showed a decreased PCV (p < 0.1) on d 3 after weaning than the CON group. However, no differences were observed for number of WBC and contents of immunoglobulin G, M, and A between the Cocktail and CON groups. Consequently, inclusion of an enzyme cocktail in diets for weaned pigs had a positive influence on gut health by reducing the incidence of diarrhea in the present study.

  • Soybean, one of most widely used swine feed component in the world, contains non-starch polysaccharides (NSP). The digestive system of weaned pigs is not yet fully developed, and thus weaned pigs cannot easily digest diets based on corn and soybean meal. Dietary exogenous enzymes supplementation has been intensively investigated to assist digestion of anti-nutritional factors, such as NSP. This experiment was conducted to investigate the effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; mixture of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Growth performance, morphology of ileum, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter, crude protein, and energy of weaned pigs were measured. No significant differences (p > 0.05) were observed for growth performance for the duration of the experimental period, and morphology of ileum, and nutrient digestibility between CON and Cocktail treatment groups. Therefore, the results from the current study indicated that enzyme cocktail supplementation in diets had no influence on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.

공주 송산리 6호분에서 분리한 진균의 생화학적 특성 분석 및 생장제어 연구 (Biochemical Characteristics and Growth Control for fungi isolated from mural painting of Tomb No.6 at Songsan-ri, Gongju)

  • 공주 송산리 6호분에서 분리한 미생물의 배양조건에 따른 생화학적 특성을 확인하고 천연물 유효성분인 anethole과 eugenol의 항진균력을 평가하여 고분 또는 벽화에 대한 적용 가능성을 확인하였다. 송산리 6호분 내부 공기중 및 벽체에서 포집된 26종의 곰팡이는 자낭균류 15종, 접합균류 2종, 담자균류 1종으로 동정되었다. 분리된 곰팡이의 전분과 젤라틴에 대한 분해도 평가를 실시한 결과 SY-18, SY-23, SY-25 등에서 전분에 대한 분해능이 높게 나타났으며, SY-18, SY-21, SY-23 등에서 높은 젤라틴 분해능을 보였다. 각 기질의 분해능이 우수한 균주 3종을 액체배양하여 생화학적 특성을 확인하였다. 천연물 유효성분인 anethole과 eugenol의 항진균력 평가 결과, anethole과 eugenol의 혼합액(1:2)에서 곰팡이의 생장 억제력이 가장 크게 나타났다. 국내외에 동굴 또는 고분 벽화에 발생 가능한 곰팡이에 대해 강력한 항진균력을 보이는 천연물 유효성분을 이용하여 미생물의 발생을 억제할 수 있을 것으로 기대된다.

비만여성에서 저열량식사와 체중감량제 섭취에 의한 체중 및 체지방 감소 효과 (The Effects of Low Calorie Meal and Weight Control Preparation on the Reduction of Body Weight and Visceral Fat in Obese Females)

  • 저열량 처방을 위한 식사대체제와 식이섬유ㆍ탄수화물 가수분해 억제제ㆍHCA를 주 성분으로 하는 체중감량제의 섭취 효과를 판정하기 위하여 20대∼50대의 BMI 25이상의 여성 자원자를 모집하여 9주간 시험을 실시하였다. 시험기간 동안 1일 1끼 식사는 식사대체제로 대치하고 1일 2회 체중감량제를 섭취하게 한 후 신체계측과 체조성, 배변 상황, 식이섭취 실태, 자각 증상 등의 변화를 조사하여 효과를 판정하였다. 시험 결과는 9주 동안 지속적으로 체중과 체지방, BMI, 허리 및 엉덩이 둘레가 감소한 것으로 나타났으며 체중감소율은 영양학적으로 안전한 범위에 속해 있었다. 혈청 지질에는 유의적인 변화가 없었으며 혈당과 인슐린이나 C-peptide 함량에도 유의적인 차이는 없었으며 BUN이나 ALP는 감소하였고 헤모글로빈은 유의적으로 증가하였으나 모두 정상범위에 있었다. 배변 시간은 단축되었으며 건강에 대한 부정적인 자각증상도 감소되었다. 1일 섭취 열량은 1300 kcal 정도로 유지되었으며 비타민 A, 비타민 $B_2$, 엽산, 칼슘, 철, 아연등의 섭취량은 권장량의 80% 미만으로 나타났다. 그러므로 본 연구의 저열량식사와 체중감량제의 섭취는 체중 및 체지방 감소와 배변에 효과적이었으며 부정수소를 감소시킬 수 있었다.

Varietal Difference in Retrogradation of Cooked Rice and Its Association with Physicochemical Properties of Rice Grain

  • The experiments were carried out to elucidate the varietal variation of retrogradation in aged cooked rice and its association with some physicochemical properties of milled rice. The fifteen rice materials were selected from forty-three low-amylose japonica and Tongil-type rice cultivars based on palatability and retrogradation of cooked rice stratified by preliminary sensory evaluation of warm and cooled cooked rice. One japonica glutinous rice variety was included for comparison of retrogradation of cooked rice. The $\alpha$-amylase-iodine method was adopted for checking the varietal difference in retrogradation of cooked rice. The desirable checking time for evaluating the varietal difference in deterioration of aged cooked rice was four hours after storing in room temperature and two hours after preserving in refrigerator based on the largest coefficients of variations in degree of retrogradation of cooked rice. The rice cultivars revealing the relatively slow retrogradation in aged cooked rice were Ilpumbyeo, Chucheongbyeo, Sasanishiki, Jinbubyeo and Koshihikari. A Tongil-type rice, Taebaegbyeo, and a japonica cultivar, Seomjinbyeo, showed the relatively fast deterioration of cooked rice. The retrogadation index represented by the percentage of retrogradation difference between warm and cooled cooked rice to original estimates of warm cooked rice was significantly affected by the degree of retrogradation of cooled cooked rice. Generally, the better rice cultivars in eating quality of cooked rice showed less retrogradation and much sponginess in cooled cooked rice. Also, the rice varieties exhibiting less retrogradation in cooled cooked rice revealed higher hot viscosity and lower cool viscosity of rice flour in amylogram. The sponginess of cooled cooked rice was closely associated with magnesium content and volume expansion of cooked rice. The hardness-changed ratio of cooked rice by cooling was negatively correlated with solids amount extracted during boiling and volume expansion of cooked rice. The major physicochemical properties of rice grain closely related to the palatability of cooked rice may be directly or indirectly associated with the retrogradation characteristics of cooked nce. The varietal difference in retrogradation of cooked rice can be effectively classified by scatter diagram on the plane of upper two principal components based on some retrogradation properties of cooked rice. The deteriorated structural change in cooled cooked rice by observing through the scanning electron microscope was more conspicuous in the fastly retrograded cooked rice than in the slower one.

막걸리의 제조를 위한 효소제의 개발연구 1 (Studies on improvement of manufacturing method of enzymic source for Maggerley(Korea wine) brewing(I))

  • It is necessry to develop and strengthen the activity of enzymic source which in low applied for maggerley brewing as an amylolytic and proteolytic starter, recently in this country the active and strong enzymic starter is required for the better brewing and to substitute another starch material for the present wheat flour. In this study, manufacturing method the strong enzymic source have been developed and established with use of raw wheat bran plus fungal strains of Rhizopus sp. and Aspergillus usamii the culture of starter. The results on experimental the activities of enzymic sources (stater) are as following ; 1. Method of making the enzymic source (starter) is to cultivate the strains of Asperguillus orzyae, Asp. kawachii, Asp usamii and Rhizopus sp. in the acid treated raw or heatboiled wheat bran. 2. The saccharogenic pwoer (S.P.) of enzymic source which consisted of raw bran plus fungi and cultured in it is generally stronger than those of heat-boiled bran plus fungi, the strongest power was shown in the culture of Rhizopus plus raw bran, and the next other is in mixture of Asp.usamii and Rhizopus on raw wheat bran. 3. The most strong alpha amylase activity was expressed in the plot of Asp.oryzae on heat-boiled wheat bran, the next was in the culture of Rhizopus nad Aspergillus usamii on raw wheatbran. 4. The most vigourous acidic proteinase activity was expressed in the micture of raw bran plus Asp. usamii and Rhizopus those were independentlu cu;tured before mixing for neutral proteinase activity, it was shown in the mixed culture of Asp. usamii and Rhizopus on raw wheat bran, the msot active alkaline proteinase activity of enzymic source was found in the plot of raw bran material. 5. For poly-preptidase activity in pH 6.5 it is found that the culture of Rhizopus and Asp.usamii on raw bran was most active among them of enzymic sources. 6. Generally, it is concluded that culture of fungi on acid treated raw wheat bran is stronger in its activity than those of heat boiled wheat bran, especially the culture of Rhizopus nad Asp.usamii on raw bran exhibited the most vigorous and non-polarized activity for all aspects, so it is considered to be most desirable enzymic stater in Korean Maggerley brewing and this would be able to substitute brewing material for the present wheat flour because of its strong and wide hand activity of amylolytic and proteolytic action.

가금 원료사료의 새로운 엽산 분석방법과 어린 육계의 엽산과 메티오닌의 요구량에 관한 연구 (Studies on the Folic Acid and Methionine Requirements for Young Broiler Chicks Including New Analytical Methods for Folic Acid in Poultry Feedstuffs)

  • In Experiment 1, microbial assays were conducted on 57 feed ingredient samples to determine the content of total folic acid using Lactobacillus casei(ATCC 7469). Folic acid contents of feed samples pretreated with conjugase, ${\alpha}$-amylase, and a mixture of protease(Pronase)were corn, 09${\pm}$1.18($\pi$g${\pm}$SD); fish meal, 23.05${\pm}$1.27; milo, 29.34${\pm}$0.55; bakery meal, 25.80${\pm}$6.93; meat and bone meal, 56.76${\pm}$4.97; wheat middlings, 85.14${\pm}$2.56; and soybean meal, 193.97${\pm}$3.98. Experiments 2 and 3 were conducted to determine the effects of dietary supplemental folic acid and methionine on the performance of starting broiler chicks for 18 days. Four levels of dietary folic acid(0.24. 0.54,1.14 and 2.34mg/kg) and four levels of dietary methionine(0.45, 0.53,0.61, and 0.69%) were fed in a factorial design. The basal diet was based on corn, isolated soybean protein, meat and bone meal, and fish meal. It contained adequate amounts of all nutrients except methionine and folic acid in both experiments. Increased growth rate was observed in chicks fed the basal diet supplemented with either folic acid or methionine. Total dietary folic acid and methionine plus cysteine requirements for optimum growth were estimated to be 1.80 mg/kg and 0.89% in Experiment 2, and 1.47 mg/kg and 0.91% in Experiment 3, respectively. There were interactions between dietary folic acid and methionine on weight gain in both experiments. Chicks fed diets containing 2.34 mg folic acid /kg tended to display slow growth rate in both experiments. There was a significant linear feed conversion response to folic acid in Experiment 2, and a significant quadratic feed conversion resuonse to methionine in Experiment 3. There were both linear and quadratic liver folic acid responses to dietary folic acid in both experiments. There was no indication that dietary methionine had any effect on liver folic acid content. The incidence of tibial dyschondroplasia increased with increasing supplemental methionine, but were no significant differences detected at 5% level.

배지최적화를 통한 재조합 바실러스 서브틸리스에서 바실러스 아밀로리퀴파시엔스 유래 γ-글루타밀펩타이드전달효소의 대량생산 (Overproduction of a γ-glutamyltranspeptidase from Bacillus amyloliquefaciens in Bacillus subtilis through medium optimization)

  • 본 연구를 통해 BAGGT를 재조합 B. subtilis를 이용하여 대량 생산하기 위하여 유전자 클로닝, 발현 시스템 구축 및 배지 최적화를 진행하였다. 이중프로모터 시스템을 이용하여 야생형 균주에 비해 42배 효소 생산성이 향상된 발현 시스템을 구축하였다. 또한 PBD 분석을 통해 당밀과 CSL이 재조합 B. subtilis 시스템에서 BAGGT의 생산성에 큰 영향을 주는 인자임을 확인하였으며, 염류의 첨가에 의한 효소 생산성 증대 효과는 미비하거나 부정적이었다. 탄소원으로 당밀을 선택하고 고가의 질소원인 트립톤을 저가의 CSL로 교체한 후 CCD 분석을 통해서 결정된 최적배지 사용 시 최적화 이전의 LB 배지 대비 4.3배의 생산성 증대를 이루었으며, 이는 LB 배지에서 야생형 균주의 BAGGT 생산성 대비 180배의 효소 생산성 개선에 해당하였다. 본 연구를 통해 식품용 효소로서 BAGGT의 대량생산을 위한 공정을 구축하였으며, 이후 정미성 소재 생산에 활용할 수 있을 것으로 기대한다.

Pilot Scale에서의 Fusant의 Ethanol 발효 (Ethanol Fermentation of Fusant between Heterologous Transformant of Saccharomyces cerevisiae and Candida tropicalis in Pilot Scale)

  • Fusant FSC14-75의 산업적 이용가능성을 검토하기 위한 최종 실험으로서 Working volume이 300 liters인 pilot scale에서의 발효력을 조사하였다. 13,3%의 liquefied sweet potato starch (Total sugar=14.8%)을 ethanol 발효원으로 함유한 fermentation broth 300liters를 0.6% seed로 발효시킨 결과 발효 최종일인 8일만에 6.6%(v/v)의 ethanol을 생성하였고 이때 잔당은 15%이었으므로 발효율은 총당에 대해서는 70%, 소비당에 대해서는 87.5%로 나타났다. 한편 fermentation broth로부터 FSC-14-75 유래의 unicellular cell과 flocculent cell을 분리할 수 있었으며 이를 각각 FSC14-75(S)와 FSC14-75(F)로 명명하였고, 이들 두 균주의 ethanol 발효능을 mini-jar fermentor scale에서 조사한 결과 FSC14-75(F)가 ethanol productivity에 있어서 우수하였으며 15%의 liquefied potato starch로부터 발효 10일만에 8.1%(v/v)의 ethanol을 생성하여 발효율은 총당에 대해 75%였다. FSC-14-73 (F)의 ethanol productivity를 pilot scale에서 조사하기 위해 총당 15.33%의 liquefied sweet potato starch를 함유한 fermentation broth 300 liters를 1% seed로써 발효시킨 결과 ethanol 생성은 배양 8일만에 최대 7.7%(v/v)였으며 이때 잔당은 1.9%였다. 또한 FSC-14-75(F) 균주의 발효과정중 $\alpha$-amylase의 계속적인 작용효과를 검토하기 위하여 300 liters의 fermentation broth를 살균 후1% seed와 동시에 50$m\ell$의 Thermamyl을 재차 첨가하고 ethanol 생성을 조사한 결과 발효율 및 잔당은 첨가하지 않은 경우와 별다른 차이가 없었다.

전자빔 조사에 의한 고구마의 발아관련 효소의 활성과 식품특성 변화 (Change of Sprouting-related Enzymes Activities and Food Quality Characteristics of Sweetpotato Root (Ipomea batatas Lam.) by Electron Beam Irradiation)

  • 고구마의 식품특성에 변화 없이 발아를 억제할 수 있는 물리적 처리법으로 전자빔의 적용가능성을 조사하였다. 전자빔 조사 후 12와 $25^{\circ}C$에 저장된 모든 대조구와 전자빔이 조사된 중간층(7-12 cm)과 하층(12-17 cm)에 위치하는 고구마는 저장 후 6주와 4주에 각각 발아가 시작되었다. 12와 $25^{\circ}C$에 저장된 고구마의 발아율은 저장기간의 경과, 조사선량의 감소, 전자빔과 고구마 간 간격의 증가와 함께 증가하였고 발아율은 12와 $25^{\circ}C$ 저장 후 8주에서 각각 대조구 11.2-12.4와 70.5-74.2%, 전자빔 조사구 8.6-11.3과 42.7-48.7% 이었다. 한편, $4^{\circ}C$에 저장된 모든 고구마와 전자빔 조사 후 12와 $25^{\circ}C$에 저장된 상층(0-7 cm)에 위치한 고구마의 발아는 peroxidase와 indole acetic acid (IAA) oxidase의 활성증가로 인해 완전히 억제되었다. 또한 전자빔이 조사된 모든 시료는 경도, pH, 당도, 중량감소, vitamin C 함량 및 ${\beta}$-carotene 함량에 있어서 대조구와 현저한 차이가 없었다. 따라서 고구마를 박스에 포장하기 전 0.1 kGy 이상의 전자빔 조사 시 $25^{\circ}C$에 저장하여도 식품특성에 변화 없이 발아를 효과적으로 억제할 수 있을 것으로 판단된다.
