Characterization of Carbohydrate Metabolism during Dark-Induced Senescence

  • To investigate the changes of carbohydrate metabolism in the senescing leaves of Zea mays during dark-induced senescence, the changes in the contents of reducing sugar, sucrose and starch as well as the activities of sucrose synthase, three isozymes of invertase, and ${\alpha}$-amylase were measured. In the senescing leaves, the content of reducing sugars temporarily increased at 4 d and rapidly decreased thereafter, whereas sucrose contents gradually decreased thereafter, whereas sucrose contents gradually decreased until 3 d of senscence and significantly decreased thereafter. The activities of intracellular invertases such as soluble acid and alkaline formed gradually enhanced until 4 d of leaf senescence and significantly declined thereafter. The extracellular invertase activity showed no significant changes during leaf senescence. The deactivation of sucrose synthase was observed within 3 d of leaf senscence. On the other hand, the starch contents gradually declined during 2 d of leaf senescence, and showed a temporary increase at 3 d, which is similar to the pattern of sucrose synthase activity., These results imply that sucrose in the senescing leaves. The major enzymes which correlated to the breakdown of sucrose during dark-induced senescence were soluble acid and alkaline invertases, not sucrose and ABA accelerated leaf senescence by inducing the accumulation of reducing sugar. These result, therefore, that leaf senescence may be mediated by the temporary quantitative changes of reducing sugar induced by the activation of intracellualr inveertases.

  • 본 연구는 독활의 대량증식에 종자를 효과적으로 이용하기 위해, 1990년 11월 7일에 종자를 수확하여 독활종자의 발아특성을 파악하고 발아에 관여하는 물질을 알아보기 위하여 실험하였으며 다음과 같은 결과를 얻었다. 1. 독활종자의 저온습층처리 기간이 길어질수록 종자추출물의 Phenol성 물질은 감소하였다. 2. 보리 무배(無配)종자의 $\alpha-amylase$ 활성실험에 의한 생물검정 결과 활성물질이 존재하고 있는 것 같았으나 저온습층처리 기간이 길어질수록 촉진물질은 감소하였다. 3. 상치종자 발아억제시험에 의한 생물검정 결과 억제물질이 존재하였으나 저온습층처리 기간이 길어질수록 억제물질은 감소되었다.

MCT와 LCT가 혈청(血淸)의 효소활성(酵素活性)에 미치는 영향(影響) (Effect of Medium and Long Chain Triglyceride Diets on the Serum Enzyme Activity in the Rats)

  • To investigate the effect of feeding rats medium-chain triglyceride(MCT), triglyceride containing primarily $C_8$ and $C_10$ fatty acids, it were compared to the effects of feeding triglycerides composed of long-chain triglyceride (LCT), corn oil and lard, on the serum enzyme activity. For 4 weeks rats fed a diet containing 20% MCT or LCT ${\cdot}$ MCT, as compared with LCT, had the following effects: 1) The total lactic dehydrogenase(LDH) activities in the serum of all experimental group were significantly decreased then those of control group. 2) The activities of glutamic oxaloacetic transaminase (GOT) in the serum of all experimental group were decreased than those of control group. 3) The activities of glutamic pyruvic transaminase (GPT) in the serum of all experimental group were decreased, MCT and LCT group were singinficantly decreased than of control group. 4) The activities of ${\alpha}-amylase$ in the serum of all groups were significantly increased than those of control group. 5) According to electrophoresis, LDH of LDH isoezyme activities in MCT and Lard group were increased with those of $LDH_5$ in corn oil group were increased than those of control gourp. It is suggested that the MCT and LCT fed to rats influence on the activity of various serum enzymes.

Chlorella와 Scenedesmus 세포 함유물질이 Lactobacillus delbrukii 와 Bacillus subtilis 에 미치는 성장 촉진효과 (Stimulating effects of Chlorella and Scenedesmus cell upon the growth and fermentation of L. delbrukii and B. subtilis)

  • The accelerate effects on the growth rate and the capacity of fermentation of L. delbrukii and B. subtilis was investigated in the Henneberg's medium added by various amounts of cellular components of Chlorella and Scenedesmus and also was investigated in the media added micronutritional elements such as Mn, Fe and Mo, etc. The results in the comparative experiments are as follow; 1. Various amounts of Chlorella cell components in the media accelerated remarkably the lactic acid formation and growth of L. delbrukii. For example, lactic acid formation in the medium of contained 1 percent Chlorella cell components was promoted more than twice effects compare with control. 2. The formation of .alpha.-amylase by B. subtilis in the medium of 2 percent Chlorella cell contents was also promoted more than nine twice effects compare with control. 3. The formation of lactic acid of L. delbruckii in the medium of Scenedesmus cell contents was a little more than in the medium of Chlorella cell contents. 4. The lactic acid fermented level attained with the addition of 0.2-0.25 percent Chlorella cells was the effect of promoting fermentation attained of saturating level at 100$\mu$g. /ml. of Mn and 0. 1 $\mu$g./ml. of Fe.

  • Neutral protease which was obtained from a genus of Aspergilli as the crystal form were investigated for their purification and properties. The results of biochemical and enzymatic studies for their purification and properties in this enzyme were as follows. 1) On the wheat media containing 70%-water and $CaCo_{3}$, Aspergilus oryzae S.H.W. 131 is satisfactorily grown under the basic optimum conditions temperature $27^{\circ}C$- $30^{\circ}C$at relative humidity 100% for three days. 2) The enzyme solution extracted with water is successively purified through the passing on column of Asmti-177N for decolorization of it. And ion exchanger such as DEAAE Sphadex A-50 or Shepadex G-100 and fraction collector is necessary for the sepearte treatments of this enzyme. After washing it with organic solvents as aceton-EtOH, etc., it should be dried on the vacuum dryer at $40^{\circ}C$) The protease activity is determined by the amounts of amino acids, tyrosine. 4) The optimum pH of neutral protease is 6.0-8.0. 5) In effectively decomposing with this neutral protease, the optimum temperature is $35^{\circ}C$. 6) It is interesting that the amounts of metal ion affects the activity of neutral protease. For examples, if it were treated with manganic ion, its activity would be more effective than any other that.

Evaluation of Immobilization Methods for Cyclodextrin Glucanotransferase and Characterization of its Enzymatic Properties

  • Cyclodextrin glucanotransferase(CGTase) derived from Bacillus macerans was immobilized by (1) covalent linkage on chitosan and chitin with glutaraldehyde, (2) adsorption on DEAE-cellulose and Amberite IRA 900 after succinylation, and (3) entrapment on alginate and polyacrylamide by cross linking. Adsorption on Amberite IRA 900 and covalent linking on chitosan were identified to be the most suitable immobilization methods considering the yield of activity and stability of immobilized CGTase. The enzymatic properties of immobilized CGTase were investigated and compared with those of the soluble CGTase. Thermal stability of CGTase immobilized on chitosan was increased from 50 to $55^{\circ}C$, and the optimum temperature of CGTase immobilized on Amberite IRA 900 was shifted from 55 to $50^{\circ}C$. The effect of molecular size of soluble starch (substrate) on immobilized CGTase investigated using partially liquefied substrates with different dextrose equivalent(DE). Cyclodextrin(CD) conversion yield augmented according to the increase of DE level for immobilized CGTase on Amberite IRA 900. CD conversion yield of partially cyclized starch with soluble CGTase was higher compared with liquefied one with ${\alpha}-amylase$.

Heterologous Production of Pediocin PA-1 in Lactobacillus reuteri

  • The recombinant DNA pLR5cat_PSAB, in which pediocin PA-1 structural and immunity genes (pedAB) fused with the promoter and deduced signal sequence of an ${\alpha}$-amylase gene from a bifidobacterial strain were inserted in Escherichia coli-lactobacilli shuttle vector pLR5cat, was transferred to Lactobacillus reuteri KCTC 3679 and the transformant presented bacteriocin activity. The recombinant L. reuteri KCTC 3679 transformed with the shortened pLR5cat(S)_PSAB, where a nonessential region for the lactobacilli replicon was removed, also showed bacteriocin activity. The molecular mass of the secreted pediocin PA-1 from the recombinant bacteria was the same as that of native pediocin PA-1 (~4.6 kDa) from Pediococcus acidilactici K10 on a sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) gel. In cocultures with Listeria monocytogenes, the recombinant L. reuteri KCTC 3679 effectively reduced the viable cell count of the pathogenic bacterium by a 3 log scale compared with a control where L. monocytogenes was incubated alone.

An Influence of Pretreatment Conditions on Mutagen Binding of Lactobacillus paracasei subsp. tolerans JG22 against MNNG and 2-NF

  • The objectives of this study were to investigate the effect of Lactobacillus paracasei subsp. tolerans JG22 isolated from pepper leaf jangajji on the mutagenic activity of N-methyl, N'-nitro, N-nitrosoguanidine (MNNG) and 2-nitrofluene (2-NF) and to evaluate the effect of physico-chemical pretreatment on the antimutagenic activity of the strain. The viable cells of JG22 strain displayed a significantly high (p <0.05) antimutagenic activity against both mutagens tested. The antimutagenic effect of JG22 strain seems to be positively correlated with the amounts of the cells in the incubation time. This strain produced the antimutagenic activity of the maximum levels after preincubation for 30 min. The binding of this strain against the mutagenic compounds might be mainly present in the cell wall fraction rather than the cytosol fraction. Pretreatment with proteolytic enzymes and simulated gastric and intestinal juices and at different pH values had no significant effect on two mutagens removal by the viable cells. However, the binding activity of the mutagen by the strain seems to be affected by heating, enzymes including $\alpha$-amylase and lysozyme, divalent ions, and sodium metaperiodate. Thus, carbohydrates consisting of the cell walls may be important elements responsible for the binding of MNNG and 2-NF by this strain. In conclusion, the binding of the mutagens to cells of JG 22 strain may play a vital role in suppressing the process of mutagenesis induced by mutagens.

Aspergillus coreanus NR 15-1 과 Aspergillus oryzae NR 2-5의 원형질체 형성의 최적조건 (Optimal Conditions of Protoplast Formation of Aspergillus coreanus NR 15-1 and Aspergilus oryzae NR 2-5)

  • Aspergil-lus coreanus NR-15 and Aspergilus oryzae NR-2-5 from traditional Korean Nuruk were selected as parental strains producing starch hydrolysis enzyme. Xll(Arginine-) mutant from A. coreanus NR 15-1 showed high glu-doamylase activity and total acid productivity. Z6(Adenine-) mutant from A. oryzae NR2-5 showed the highest $\alpha$-amylase activity. Therefore, both XII and Z6 mutants were selected and investigated for the optimal conditions of protoplast formation for protoplast fusion. Mixture of equal amount of cellulase and driselase(10mg/ml each) was the most effective as lytic enzymes. The optimal pH and temperature for protoplast formation were 5.0 and $30^{\circ}C$, respectively. The most effective reaction for protoplast formation time was 4 hours. The maximum of protoplst for- mation of Xll mutant and Z6 mutant were $6.54$\times$10^{7}$ protoplasts/ ml and $3.04$\times$10^{ 7}$ protoplasts/ml, and the regen-eration frequencies of the protoplasts were 11.3% and 11.6%, respectively. The size of the protoplasts from X11 and Z6 mutants were 3~6 $\mu\textrm{m}$ and 4~9$\mu\textrm{m}$, respectively.

Comparison of Digestive Function Among Rabbits, Guinea-Pigs, Rats and Hamsters. II. Digestive Enzymes and Hindgut Fermentation

  • The aim of this trial was to study the response of laboratory animals including omnivores (rats) and herbivores (rabbits, guinea pigs and hamsters) to the same level of dietary fiber on their digestive enzymes and hindgut fermentation. Ten weanling animals of each of four species, rabbits, guinea-pigs, rats and Syrian hamster, were fed a basal diet of 18% crude protein and 10% crude fiber for six weeks. The digesta and tissue of each intestinal segment were collected to measure the activity of digestive enzymes. Rabbits contained the highest secreted pepsin activity in the stomach, whereas rats contained the highest protease and ${\alpha}-amylase$ activity in the small intestine, and lower fibrous hydrolases in the hindgut than rabbits, guinea pigs and hamsters. The total VFA productions in the caecum and colon were highest in rats, followed by hamsters and rabbits, while the guinea pigs contained the lowest VFA and a different pattern of VFA molar ratio from the other laboratory animals. The degree of hindgut fermentation in these laboratory animals was in reverse to the trend for their fiber digestion.
