  • In order to understand microbial ecosystem in the fast china sea and Yosu coastal sea horizontal distributions of bacterial flora, Vibrio spp., coliform group, temperature, and salinity in this sea area, were studied 42 sampling stations during 6th-l4th August, 1992. From the results, salinity and temperature were $24.64\~33.78\%_{\circ}$ and $22.53\~29.18^{\circ}C$, respectively. As the open sea far from land goes on, salinity became low while temperature became high. Viable cell counts of bacteral flova, Vibrio spp., and roliform group in Yosu coastal sea water were $1.0\times10^2\~3.0\times10^4/ml,\;0.2\times10\~9.0\times10^3/ml,\;and\;0.3\times10\~3.0\times10^3/ml$ while those of these in the open sea water were $0.4\times10\~2.0\times10^3/ml,\;0.8\times10\~3.0\times10/ml,\;and\;0.9\times10\~1.3\times10/ml$, respectively.

Studies on the Quick Ripening Process for Soysauce (속양(速釀)간장 제조(製造)에 관(關)한 연구(硏究))

  • This experiment was conducted as a preliminary test to invent the quick ripening process for soysauces. The bran koji extract of 2 kinds of mold (Asp. niger and Asp. sojae) was used as enzyme sources in order to improve the rate of enzymatic conversion of raw materials and shorten the ripening period of soysauces. Soybean koji (inoculated Asp. sojae) and steamed soybean were enzymatically decomposed for 8 hours at $40^{\circ}C$, pH 4-6 with stirring at a rate of 1500 rpm. After the enzymatic treatment, soysauces were fermented by 4 kinds of processes with the above-mentioned salt tolerant microorganisms inoculated. After 4 weeks, sensory evolution for these soysauces products and analysis of chemical components were carried out. The results obtained were summarized as follows. 1. Soysauces prepared from decomposed koji by Asp. niger enzyme complex showed higher extractables, such as total nitrogen, amino nitrogen, pure extract and reducing sugar, than any other procedure. Extracted amounts of each components of soysauces prepared from this process during 1 month were similar to those of soysauces made from general procedure (control) for 5 to 6 months. 2. According to sensory evaluation, soysauce prepared from decomposed koji also showed best taste and were similar to that of soysauces prepared by general method for 6 months.

마이크로웨이브 공정에 의한 마늘 올레오레진의 추출조건 설정 및 특성

  • 마늘을 MAE(microwave-assisted extractiona)방법에 의하여 물, 30% 에탄올 및 50% 에탄올로 추출하고 그 특성을 비교하였다. 마늘과 각 용매의 비율은 1:2.5로 하여 마이크로웨이브 60W로 각각 5분 및 20분 동안 추출하였다. 마늘 올레오레진 수율은 50% 에탄올로 20분동안 추출한 추출물이 14.1%로 가장 높았으며 polyphenol함량도 물로 20분동안 추출한 경우가 579.0mg%로 가장 많은 함량을 나타내었다. 전자공여작용 효과 및 피르브산 함량도 물로 20분 동안 추출한 추출물이 각각 32.7% 및 마늘 1g당 13.4$\mu$molus로 가장 높게 나타났다. 각 용매에 대해 20분동안 추출한 추출물들을 37$^{\circ}C$에서 8일동안 저장하면서 피르브산 함량과 전자공여작용의 변화를 조사한 결과, 피르브산 함량은 물 추출물이 에탄올 추출물보다 높은 함량을 나타내었고 저장 시간이 증가할수록 그 함량은 감소하는 경향을 나타내었다. 전자공여작용도 물 추출물이 가장 높은 효과를 나타내었으며 추출시간에 비례하여 증가하다가 저장 6일째부터 감소하는 경향을 나타내었다. 각 용매에 대한 추출물의 색깔을 조사한 결과 물 추출물이 가장 밝고 옅은 갈색을 나타내었으며 30% 에탄올 추출물이 가장 어둡고 푸르스름한 노란색을 나타내었다. 또한 각 추출물들을 37$^{\circ}C$에서 10일간 저장하면서 갈색화 정도를 측정해 본 결과 물 추출물이 에탄올 추출물이 에탄올 추출물보다 갈색화가 많이 진행되었으며 0.1% cysteine을 첨가한 추출물과 에탄올 추출물의 갈색화는 비슷한 경향을 나타내었다. 마늘 추출물을 추출한 후에 0.1% cysteine을 첨가하여 저장한 것 보다 추출하기 전에 첨가하는 경우가 갈색화 억제 효과가 뚜렷하였다.문에 밀가루에 일부를 대용한 wheat flour dough를 사용하고 가정용 제빵기로 구워 최종 단계에까지의 제빵성 결과를 산출했다. amarans folur 5%의 대체에는 빵의 비용적이 비교적 증대했지만 그 이상 amarans flour을 대처하면 확연히 비용적은 감소했다. amarans flour 10% 대체에 hemicellulase 1250U 이상을 첨가하면 비용적은 눈에 띄게 증대했다. farinograph에 있어서 반죽의 안정성은 amarans flour 10% 대용에 현저히 감소했다. 반죽의 점탄성(아축응력, 탄성률, 점성계수)는 amarans flour 10%를 대용한 것이 무첨가한 것보다 많이 단단해졌음을 알 수 있었다. 혼합중의 반죽의 조사형 전자현미경 관찰로 amarans flour로 대체한 gluten이 단단해졌음을 알수 있었다. 유화제 stearly 칼슘, 혹은 hemicellulase를 amarans 10% 대체한 밀가루에 첨가하면 확연히 비용적을 증대시킬 수 있다는 사실을 알 수 있었다. quinoa는 명아주과 Chenopodium에 속하고 페루, 볼리비아 등의 고산지에서 재배 되어지는 것을 시료로 사용하였다. quinoa 분말은 중량의 5-20%을 quinoa를 대체하고 더욱이 분말중량에 대하여 0-200ppm의 lipase를 lipid(밀가루의 2-3배)에 대하여 품질개량제로서 이용했다. 그 결과 quinoa 대량 7.5%에서 비용적, gas cell이 가장 긍정적 결과를 산출했고 반죽의 조직구조가 강화되었다. 또 quinoa 대체에 의해 전분-지질 복합제의 흡열량이 증대된 것으로부터 전분-지질복합제의 형성 촉진이 시사되었다.이것으로 인하여 호화억제에 의한 노화 방지효과가 기대되었지만 실제로 빵의 노화는 현저히 진행되었다. 이것은 quinua 대체량 증가에 따른 반죽의 안정성이 저하되어 버린 것으로 생

  • This study was conducted to investigate inhibitory effects of K-casein, GMP and sialic acid addition on the infection of MA-104 cells by 597(Korean native cattle rotavirus) and JBR(Jeju island bovine rotavirus). MA-104 cells on incomplete Ml99 were infected with domestically separated 597 and ma activated by incubating at 37$\^{C}$ for 6 days, and analyzed for the titer of rotavirus. K-casein, GMP and sialic acid added MA-104 culture infected by activated S97 and nan were incubated for Is hours and stained by the AEC stainning method. The number of infected cells were counted on microscope. The titer of S97 and JBR was 2.5$\times$107 and 2.0$\times$106 PFU/ml, respectively. The inhibition level against cell infection by 597 was 97.4% far 2000UH of K-casein and 97.44% for 2000UM of GMP. The inhibition level against cell infection by JBR was 99.52% for 2000$\mu$M of $\kappa$-casein and 99.78% for 2000$\mu$M of GMP. The inhibition level against cell infection by 597 and JBR was 3.85 and 3.63% for 2000$\mu$M of sialic acid, respectively. The high inhibitory effects (over 97%) of K-casein and CMP against infection of U-1(14 cells with 597 and mR indicated great potentials for the use of K-casein and GMP in the treatment of calf or infant caused by rotavirus.

  • 가토 20마리의 혈액중 혈소판의 수를 수정 전후로 조사한 결과, 수정전과 비교하여 수정후 1일부터 유의하게 감소하였으며 (26.84%, P<0.01), 이러한 감소는 수정후 5일까지 계속되었다. 수정후 5일동안 혈소판의 감소율과 혈소판의 최저치를 수정란의 수와 비교한 바, 이들간의 유의한 상관관계(r=0.5032)가 있었다. 가토수정란 (2세포기) 각각 30∼40개를 5마리의 비장적출수란가토의 난관에 이식한 다음, 혈소판수의 변화를 조사한 결과, 이식전에 비해 이식후 90분 (P<0.025)과 135∼270분 (P<0.025)에 유의한 감소를 확인하였따. 특히 수정란 이식후 180분경에 가장 크게 감소하였다 (P<0.001). 60∼70개의 2세포기 가토수정란을 24시간 배양한 다음, 배양액을 3마리의 비장적출가토에 주사한 후 혈소판의 변화를 조사하였을 때, 주사후 120분경부터 혈소판의 수가 유의하게 감소하기 시작하였다. 이들 배양액을 동결, 해동한 다음 주사한 실험에서도 유사한 결가를 얻었다. 또한 이 배양액을 비장적출생쥐에 주사한 경우에도 가토에서와 마찬가지의 혈소판 촉진현상이 나타났다. 혈소판 촉진인자(PAF)의 길항제인 kadsurenone이 첨가된 배양액에서 24시간동안 가토수정란을 배양한 다음, 이 배양액을 4마리의 비장적출생쥐에 주사한 결과, 혈소판수의 변화가 일어나지 않았다. 또한 kadsurenone이 첨가된 배양액에서는 2세포기 가토수정란의 상실기와 배반포기까지 발달율은 각각 4와 7%로 대조구의 7과 49%보다 유의하게 낮았다. 따라서 본 연구에서 가토의 초기배는 가토나 생쥐의 혈소판수를 감소시키고, 특히 길항제 처리는 이러한 혈소판 감소현상을 억제시킬 뿐만 아니라 가토 초기배발달을 억제한다는 것을 확인하였다. 결론적으로 가토초기배에서는 PAF 또는 수정란 유래의 PAF가 분비된다는 것을 알 수 있었으며, 이러한 인자는 동결처리에서도 그 기능은 전혀 변하지 않는다고 본다. 이후에 있어서 mouse LIF의 첨가는 돼지의 수정란을 배반포 이후의 단계에까지 발달시킬 수 있었다. 있어서 더 적합한 것으로 판단되었다. 5. 개발된 모형은 논 관개의 물리적 측면과 관리목표 모두를 고려한 것으로 계산된 효율은 벼, 생육 각 단계에서의 효율 비교에 양호한 방법임을 알 수 있다.은 Sharpsburg 점질양토에 대한 S.C.S 한계허용치 10ton/ha/year 이내로 나타났다. 비처리구에서의 토양유실량은 평균 2.56ton/ha/year로 높게 나타난 반면 3개의 서로 다른 추리구인 비수구, 초생수로구 및 Bromegrass구에서는 각각 0.152, 0.192 및 0.290ton/ha/year로 낮은 결과를 가져왔다. 6. 평균 침전량에 대한 L.S.D. 검정 걸과 전시험구중 비처리구가 고도의 유의차를 나타낸 반면 비수구, 초생수로구 및 Bromegrass 목초구 간에는 아무런 유의차가 인정되지 않았다. 7. 농지보전 처리구인 배수구와 초생수로구는 비처리구에 비해 낮은 침두 유출량과 낮은 토양유실량을 나타내었다.구보다 14% 절감되는 것으로 나타났다.작용하는 것으로 사료된다.된다.정량 분석한 결과이다. 시편의 조성은 33.6 at% U, 66.4 at% O의 결과를 얻었다. 산화물 핵연료의 표면 관찰 및 정량 분석 시험시 시편 표면을 전도성 물질로 증착시키지 않고, Silver Paint 에 시편을 접착하는 방법으로도 만족한 시험 결과를 얻을 수 있었다.째, 회복기 중에 일어나는 입자들의 유입은 자기폭풍의 지속시간을 연장시키는 경향을 보이며 큰 자기폭풍일수록 현저했다. 주상에서 관측된 이러한 특성은 서브스톰 확장기 활동이 자기폭풍의 발달과 밀접한 관계가 있음을 시사한다.se that were all low in two aspects, named "the Nonsignificant group". Characterization of Biological Chemistry from Over Ripened Kimchi (과숙김치의 생물.화학적 특성)

  • Kimchi is one of the traditional Korean food and a very popular side dish in Korea. To obtain funda mental data on how to prevent over ripening in kimchi after acidity of 0.4% was reached during the lactate fermentation, the physicochemical characteristics such as pH. acidity. organic acids, enzyme activity were measured and the time dependent ecology of microorganism were observed. In the initial stages of fermentation, the pH of kimchi was markedly changed and slowly decreased in 0.5% acidity The acidity was slowly increased and markedly increased in pH 4 by growth of microorganism. HPLC analysis showed oxalic acid, lactic acid, acetic acid, malic acid and succinic acid and this results reconfirmed by GC-MSD. Lactic acid was changed a lot during fermentation period as the time of storage went on, where as malic was decreased. Kimchi A, having acidity of 0.75%, showed the highest acidic Protease and lipase activity. Also, the amylase activity was high in kimchi C, having 0.95% acidity. The total viable bacteria showed 8.1$\times$10$^{5}$ , 4.7$\times$10$^4$, 1.2$\times$10$^3$, 3.2$\times$10$^4$, 4.9$\times$10$^{5}$ cfu/ml in the kimchi A, B, C, D and E, respectively. The numbers of lactic acid bacteria counted 1.0$\times$10$^{5}$ , 1.3$\times$10s, 1.2$\times$10$^3$, 2.3$\times$ 10$^3$, 2.1$\times$10$^4$c1u/m1 in the kimchi A, B, C, D and E, respectively. The numbers of acetobactor were counted 1.8$\times$10$^{5}$ , 9.3$\times$10$^4$, 7.0$\times$10$^1$, 4.5$\times$10$^4$, 5.3$\times$10$^3$cfu/m1 in the kimchi A, B, C, D and E, respectively.

  • Objectives: The aims of this study are 1) to determine if GAST is a better indicator in predicting ovarian response to COH compared with patient's age or basal FSH level and 2) to evaluate its role in detecting abnormal ovarian response. Design: Prospective study in 118 patients undergoing IVF-ET using GnRH-a short protocol during May-September 1995. Materials and Methods: After blood sampling for basal FSH and estradiol $(E_2)$ on cycle day two, 0.5ml (0.525mg) GnRH agonist ($Suprefact^{(r)}$, Hoechst) was injected subcutaneously. Serum $E_2$ was measured 24 hours later. Initial $E_2$ difference $({\Delta}E_2)$ was defined as the change in $E_2$ on day 3 over the baseline day 2 value. Sixteen patients with ovarian cyst or single ovary or incorrect blood collection time were excluded from the analysis. The patients were divided into three groups by ${\Delta}E_2$; group A (n=30):${\Delta}E_2$<40 pg/ml, group B (n=52): 40 pg/ml${\leq}{\Delta}E_2$<100 pg/ml, group C (n=20): ${\Delta}E_2{\leq}100$ pg/ml. COH was done by GnRH agonist/HMG/hCG and IVF-ET was followed. Ratio of $E_2$ on day of hCG injection over the number of ampules of gonadotropins used ($E_2hCGday$/Amp) was regarded as ovarian responsiveness. Poor ovarian response and overstimulation were defined as $E_2$ hCGday less than 600 pg/ml and greater than 5000 pg/ml, respectively. Results: Mean age $({\pm}SEM)$ in group A, B and C were $33.7{\pm}0.8^*,\;31.5{\pm}0.6\;and\;30.6{\pm}0.5^*$, respectively ($^*$: p<0.05). Mean basal FSH level of group $A(11.1{\pm}1.1mlU/ml)$ was significantly higher than those of $B(7.4{\pm}0.2mIU/ml)$ and C $(6.8{\pm}0.4mIU/ml)$ (p<0.001). Mean $E_2hCGday$ of group A was significantly lower than those of group B or C, i.e., $1402.1{\pm}187.7pg/ml,\;3153.2{\pm}240.0pg/ml,\;4078.8{\pm}306.4pg/ml$ respectively (p<0.0001). The number of ampules of gonadotropins used in group A was significantly greater than those in group B or C: $38.6{\pm}2.3,\;24.2{\pm}1.1\;and\;18.5{\pm}1.0$ (p<0.0001). The number of oocytes retrieved in group A was significantly smaller than those in group B or C: $6.4{\pm}1.1,\;15.5{\pm}1.1\;and\;18.6{\pm}1.6$, respectively (p<0.0001). By stepwise multiple regression, only ${\Delta}E_2$ showed a significant correlation (r=0.68, p<0.0001) with $E_2HCGday$/Amp, while age or basal FSH level were not significant. Likewise, only ${\Delta}E_2$ correlated significantly with the number of oocytes retrieved (r=0.57, p<0.001). All four patients whose COH was canceled due to poor ovarian response belonged to group A only (Fisher's exact test, p<0.01). Whereas none of 30 patients in group A (0%) had overstimulation, 14 patients among 72 patients (19.4%) in group B and C had overstimulation (Fisher's exact test, p<0.01). Conclusions: These data suggest that initial $E_2$ difference after GAST may be a better prognostic indicator of ovarian response to COH than age or basal FSH level. Since initial $E_2$ difference demonstrates significant association with abnormal ovarian response such as poor ovarian response necessitating cycle cancellation or overstimulation, GAST may be helpful in monitoring and consultation of patients during COH in IVF-ET cycle.

Characteristics of Marine Algal Communities in Village Fishing Grounds Near Large Wildfires in Uljin-gun (울진군 대형산불 발생 인근 마을어장의 해조류 군집 특성)

  • In this study, we examined the effects of a large wildfire in the coastal area of Uljin-gun. The analysis of water quality and the dominant species, species composition, and community structure of marine algal was conducted quarterly in 2022 at Nagok (F-1), Hujeong (F-2), Bongpyeong (F-3), and Gongse Port (F-C) in Uljin-gun. As a result of water quality analysis, the pH, a factor of wildfire impact was 8.07~8.30 and 8.12~8.48 in surface and bottom layers, respectively, which are normal values in coastal waters of the East Sea, suggesting no direct impact from wildfires. By marine algal species composition, the coastal areas show the following order: Rhodophyta (58.1%) > Ochrophyta (25.8%) > Chlorophyta (14.5%) > Magnoliophyta (1.6%). By season, Undaria pinnatifida was the most dominant at Nagok and Hujeong in March and June, which in September and November, Gelidium elegans and Lithophyllum sp. were the most dominant in Bongpyeong and Gongse Port, respectively. In the cluster analysis, the stations were divided into two groups according to presence and absence of specific marine algal by season. The dominant species were U. pinnatifida, G. elegans and D. divaricata in group A, and Lithophyllum sp. was mainly present in group B. Thus, the species composition and group structure reflected the normal seasonal change pattern with water temperature variation and showed little significant difference from the control site, suggesting no direct effects of the wildfire on algae distribution in Uljin.

Effects of Laminaria Japonica Extract Supplement on Blood Glucose, Serum Lipids and Antioxidant Systems in Type ll Diabetic Patients (다시마추출물이 제2형 당뇨병 환자의 혈당, 지질 및 항산화 체계에 미치는 영향)

  • We performed a randomized double-blind placebo-controlled trial to determine whether Laminaria japonica extract (LJE) supplement modulates blood glucose, serum lipids and antioxidant systems in type II diabetic patients. We also measured critical parameters assessing safety in liver and kidney functions after LJE supplement. A total of 37 patients (18 males and 19 females) were randomized to either LJE group or placebo group. The treatment group received four 350 mg of LJE capsules (1.4 g, total) per day for 12 weeks. The placebo group received the same dose of cellulose capsules. Baseline characteristics regarding general life style and dietary intake pattern were similar between the two groups. There were no significant influences of LJE supplement except for waist circumference on anthropometric parameters. As the whole, 12 weeks of LJE supplement resulted in a little decrease in fasting blood glucose (FBG) and glycated hemoglobin (HbA1c), but a significant decrease was not observed. Total cholesterol, LDL-cholesterol and triglyceride concentrations were significantly (p<0.05) lowered in LJE group. The antioxidant enzymes, glutathion peroxidase (GSH-px) and superoxide dismutase (SOD) levels were elevated in the LJE group (p<0.05) compared to the placebo. The increase of these enzymes was associated significantly with the decrease of MDA concentration (p<0.05). Furthermore, LJE supplement showed no adverse effects on the functions of liver and kidney. Findings from this study suggest that LJE supplement can help improve serum lipid status in type II diabetic subjects without adverse effects.
