Abstract

Stevia rebaudiana is a traditional herb used as a sweetener in Brazil and Paraguay as well as Korea and China. This study investigated the efficacy of Stevia rebaudiana methanol extract (SRE) to protect cells against the mitochondrial dysfunction and apoptosis in hepatocyte. To determine the effects of SRE on oxidative stress, we used the human derived hepatocyte cell line, HepG2 cell. Treatment of arachidonic acid (AA)+iron in HepG2 cells synergistically amplified cytotoxicity, as indicated by the excess reactive oxygen species (ROS) and mitochondrial permeability transition by fluorescence activated cell sorter (FACS) and immunoblot analysis. Treatment with SRE protected hepatocytes from AA+iron-induced cellular toxicity, as shown by alterations in the protein levels related with cell viability such as procaspase-3. SRE also prevented the mitochondrial dysfunction induced by AA+iron, and showed anti-oxidant effects as inhibition of $H_2O_2$ production and GSH depletion. Moreover, we measured the effects of SRE on AMP-activated protein kinase (AMPK), a key regulator in determining cell survival or death. Acetyl-CoA Carboxylase (ACC), a direct downstream target of AMPK. SRE increased phosphorylation of ACC, and prevented the inhibition of ACC phosphorylation by AA+iron. These results indicated that SRE has the ability to protect cells against AA+iron-induced $H_2O_2$ production and mitochondrial impairment, which may be mediated with AMPK-ACC pathway.

서 론

세포는 계속해서 reactive oxygen species (ROS)에 노출되어있고, 산화적 스트레스를 받고 있다. ROS가 과도하게 발생되거나 산화제와 항산화제의 균형이 깨어질 때 세포나 조직이 손상을 받게 되고, 산화적 스트레스가 생성된다1-3). H2O2를 제거하는 catalase, superoxide dismutase 등과 같은 내재적인 항산화 효소와 glutathione (GSH), carotenoids, flavonoids 등과 같은 비효소적 항산화 물질이 ROS로부터 인체를 방어하고, 세포 내 여러부분에서 특이적으로 작용하여 조직의 과산화를 억제한다4-6). 최근, ROS의 불균형 조절을 위하여 항산화제의 복용이 증가되고 있고7,8), 이러한 흐름에 맞추어 질병의 예방 및 치료가 동시에 가능하고 인체에 무해한 천연 항산화제 개발에 대한 연구가 이루어지고 있다9-11).

지방산과 지질의 산화는 ω-6 불포화지방산인 아라키돈 산(Arachidonic acid, AA)의 방출과 염증성 매개인자를 자극하여 인산화의 활성과 cell signal에 해로운 영향을 야기한다11). 여러 종류의 암세포에서 AA와 apoptosis signal과의 관련성이 알려져있다. 세포 내 AA의 증가가 미토콘드리아-매개성 apoptosis와 미토콘드리아 투과성 전이 통로 (mitochondrial permeability transition pore, mPTP)를 개방하여 cytochrome c의 유리를 일으키는 apoptosis를 유도하고, ceramide의 농도를 높임으로써 AA의 세포독성이 일어난다는 사실이 보고되었다12-14). 게다가 iron처리에 앞서, AA를 처리하면 산화를 촉매 하여 세포의 산화적 스트레스를 증가시키고, 미토콘드리아의 기능 장애를 유발시킨다.

Stevia rebaudiana (S. rebaudiana)는 국화과에 속하는 여러해살이풀로, 감미료나 대용 감미료로 시중에서 스테비아로 유통된다15). 주로 그늘에서 말린 경엽(莖葉)이나 어린 잎을 따서 약용부위로 사용하며, 항산화, 항염증, 항비만 등의 효능이 있으나 그 기전은 명확히 밝혀지지 않았다16). AMP-activated protein kinase(AMPK)는 체내 에너지 상태를 감지하는 에너지 센서로 알려져 있는 효소로서, 세포 내 미토콘드리아를 보호하여 산화적 스트레스에 의한 세포 손상을 방어한다고 알려져 있다17-19). 본 연구에서는 이러한 기존 연구결과를 바탕으로 하여 Stevia Rebaudiana methanol extract (SRE)의 산화적 스트레스에 대한 세포 보호 효능을 평가하고, 그 기전으로 AMPK와의 연관성을 제시하고자 한다.

 

재료 및 방법

1. 시약

Anti-procaspase-3, anti-phospho-ACC, 그리고 horseradish peroxidase-conjugated goat anti-mouse IgGs은 Cell Signaling Technology (Beverly, MA, USA)에서 구입하였다. Horseradish peroxidase-conjugated goat anti-rabbit IgGs은 Thermo (Rockford, IL, USA)에서 구입하여 사용하였다. AA, Rhodamine123 (Rh123)은 Calbiochem (San Diego, CA, USA)에서 구입하였고, 2’,7’-dichlorofluorescein diacetate (DCFH-DA), ferric nitrilotriacetic acid (Fe-NTA), anti-β-actin antibody, 그리고 다른 시약들은 Sigma (St. Louis, MO, USA)에서 구입하여 사용하였다.

2. 추출물 제조

Stevia rebaudiana는 이일원 선생님 (자굴산 치유 수목원, 일준 부채 박물관, 의령, 경상남도)께 기증받았으며, 메탄올에 넣고 추출한 후, 추출물을 거즈로 1차 여과 후, 원심분리 하였다. 원심분리 후 상층액만을 취하여 0.22 μm filter (Millipore, USA)로 여과하였다. 이 여과액을 동결건조하여 Stevia rebaudiana 메탄올 추출물 (Stevia rebaudiana methanol extract, SRE)을 얻었으며, 사용 때까지 –20℃에서 보관하였다. in vitro 실험에서 dimethyl sulfoxide (DMSO)에 녹여 사용하였다.

3. 세포배양 및 처리

Human hepatocyte에서 유래된 cell line, HepG2 cell은 ATCC (Rockville, MD, USA)에서 구입하였으며, Dulbecco's modified Eagle's medium (DMEM)에 열처리한 10% fetal bovine serum (FBS), 100 units/ml penicillin and 100 μg/ml streptomycin을 혼합한 배지를 사용하여 37 ℃, 5% CO2 incubator에서 배양하였다. cell은 100mm 배양접시에서 80% confluency에 도달하도록 배양하였고, 일주일에 두 번씩 1:4의 비율로 계대 배양하였다. 12시간 serum-starvation 후, SRE를 0.1 mg/ml의 농도로 한 시간 전 처리, 10 μM AA를 12시간 처리 후 5 μM iron을 첨가하여 1시간 더 배양하였다. SRE는 DMSO에 녹여서 사용하였고, 세포 처리시에는 세포에 무해한 1 μl/ml의 양을 사용하였다.

4. MTT assay

세포 독성을 측정하기 위해, HepG2 cell은 24 well plate에 5×104 cells/well 농도로 0.5 mL/well 분주한 뒤 24시간 동안 배양한 후, SRE를 최종농도 (0, 0.003, 0.01, 0.03, 0.1 mg/ml)가 되도록 한 시간 전 처리, 10μM AA를 12시간 처리 후 5μM iron을 첨가하여 4시간 더 배양하였다. 이 후, 1 mg/ml MTT 용액을 가하고 37℃에서 2시간 더 배양하여 MTT를 환원시켜 생성된 formazon이 떨어져나가지 않도록 배지를 조심스럽게 제거하였다. DMSO를 처리하여 Titertek Multiskan automatic microplate reader (Model MCC/340, Huntsville, AL, USA)로 570 nm에서 흡광도를 측정하였다. 세포 생존율은 control에 대한 백분율로 나타내었다.

5. Immunoblot analysis

HepG2 cell을 PBS로 2회 wash 후, radioimmnoprecipitation buffer (25 mM Tris-HCl pH 7.6, 150 mM NaCl, 1% NP-40, 1% sodium deoxycholate, 0.1% SDS)와 Halt Protease and Phosphatase Inhibitor Cocktail (Sodium Fluoride, Sodium Orthovanadate, β-glycerophosphate, Sodium Pyrophosphate, Aprotinin, Bestatin,Leupeptin, EDTA)을 혼합하여 lysis를 실시하고, 4 ℃ 에서 15,000 g, 30분 간 원심분리하여 단백질을 포함하는 상층액을 취하고, BCA protein assay에 따라 정량 한 후, 순차적으로 일차항체 및 이차항체를 처리하여 enhanced chemiluminescence method (Amersham, Buckinghamshire, UK)로 발광시켜 X-ray필름에 감광하였다. 발색 후 각 단백질의 발현량을 평가하기 위하여 Image J를 이용하여 Densitometric analysis를 실시하였다.

6. Flow cytometric analysis of mitochondrial membrane potential (MMP)

MMP의 변화는 막 삼투 양이온 형광 dye인 Rh123으로 염색하고 난 뒤, fluorescence activated cell sorter (FACS)를 사용하여 flow cytometry에 의해 결정하였다. HepG2 cell을 0.05 μg/ml Rh123으로 30분 간 staining 한 후, trypsin을 이용하여 cell을 취하였다. 1% FBS를 첨가한 PBS에 재부유 시켜, cell의 MMP 변화를 Partec FACS flow cytometer (Goerlitz, DE)로 확인하였다. 각 분석은, 한 샘플 당 20,000개를 기록하였다.

7. 세포 내 H2O2 측정

DCFH-DA는 비극성분자로 용이하게 세포 내로 들어갈 수 있으며, 세포 내의 esterase에 의해 비형광성 극성분자인 DCFH로 분리된 후 세포내의 활성산소에 의해 산화되면 형광성 dichlorofluorescein (DCF)로 전환된다. Cell은 10 μM DCFH-DA로 2시간동안 37 ℃에서 staining하였고, cell의 Fluorescence intensity는 Titertek Multiskan automatic microplate reader(Model MCC/340, Huntsville, AL, USA)를 이용하여 분석하였다.

8. GSH assay

세포에서 환원된 GSH를 정량화하기 위해서 GSH determination kit (Oxis International, Portland, OR, USA)를 사용하였다.

9. 통계처리

각 처리군은 T-test를 실시하여 통계적으로 유의함을 나타내었다. (p<0.05 or p<0.01)

 

결 과

1. SRE의 AA+iron로 유도된 apoptosis 저해 효과.

SRE의 AA+iron으로 유도된 apoptosis 저해능을 알아보기 위해 MTT assay를 진행하였다. 앞서 연구 된 논문에서, HepG2 cell에 AA를 처리하여 (10 μM) 산화스트레스와 apoptosis를 유발하였고, AA로 유도된 독성을 iron (5 μM)의 처리로 증가시켰다20). 따라서 본 실험에서도 10 μM의 AA 와 5 μM의 iron을 처리하였다. SRE를 농도 별로 처리한 결과 0.1 mg/ml 농도 이하에서 세포 독성을 나타내지 않았다 (data not shown). 그러므로 본 실험에서는 SRE 0.003, 0.01, 0.03, 0.1 mg/ml의 농도로 MTT 실험을 진행하였다. 그 결과, AA+iron에 의해 감소한 세포 생존율은 SRE 농도 의존적으로 증가하는 경향을 나타내었으며, 0.1mg/ml의 농도에서 가장 효과적인 세포 보호능을 나타내었다(Fig. 1A). MTT assay에 이어 apoptosis 관련 단백질의 immunoblotting을 진행하였다. HepG2 cell에 AA+iron 처리 시, caspase-3의 cleavage (17kDa)가 증가하여, procaspase-3 단백질 농도가 감소하였고, SRE는 이것을 현저히 억제하였다(Fig. 1B).

2. SRE의 항산화 효과.

세포내의 H2O2는 DCFH-DA를 이용하여 microplate reader로 측정하였다. HepG2 cell에서 AA+iron으로 H2O2의 증가를 유도하고, SRE는 유도된 산화적 스트레스를 억제하였다(Fig. 2A). 또한, GSH는 중요한 endogenous antioxidant로서, 독성으로부터 유발된 ROS의 증가는 GSH의 결핍을 가져온다25). AA+iron과 SRE를 처리한 HepG2 cell에서의 GSH level을 측정함으로써 SRE의 항산화 효과를 평가하였다. AA+iron 처리로 GSH level이 감소하였고, SRE를 처리한 군에서는 GSH level이 회복되었다(Fig. 2B). 또한, SRE 단독 처리 시 control군에 비하여 GSH가 증가하였다.

3. SRE의 mitochondrial dysfunction 저해 효과.

이전 연구에서, AA는 복합체 I 과 III 를 선택적으로 저해함으로써 mitochondria의 호흡기능을 손상시킨다고 보고되었다20). AA+iron은 MMP의 기능 장애를 유발하고, 이는 세포에서 apoptosis를 유도한다21). 본 실험에서 MMP를 측정하기 위하여 막 삼투 양이온 형광 dye인 Rh123으로 cell staining을 실시하였고, FACS를 이용하였다. AA+iron을 처리하였을 때, MMP가 감소, 즉 depolarization 상태가 되어 Rh123의 staining 정도가 떨어지는데22), 이것은 미토콘드리아의 손상과 기능장애가 증가했다는 것을 보여준다 (Fig. 3A and B).

4. SRE에 의한 AMPK-ACC pathway 조절

이전 연구에서 다양한 세포 보호 물질들이 AMPK-ACC pathway를 통하여 AA+iron으로 유도된 미토콘드리아 기능장애 및 산화적 손상을 억제한다고 알려져 있다21). AMPK의 활성화는 ATP를 소비하는 과정에 필요한 효소인 ACC를 인산화 시키므로, AMPK의 downstream인 ACC의 인산화로 AMPK activation을 확인할 수 있다23). SRE 0.1 mg/ml을 HepG2 cell에 10분-6시간 처리하여 관찰한 결과, SRE는 시간 의존적으로 ACC의 인산화를 더욱 증가시켰는데, 1시간에서 가장 많은 활성증가를 나타내었다 (Fig. 4A). 또한, AA+iron은 이러한 ACC의 인산화를 억제하였고, SRE 전처리한 세포에서는 ACC 인산화의 감소를 방어하였다(Fig. 4B).

고 찰

Stevia rebaudiana는 남미의 하천, 습지대 주변에서 약용을 목적으로 재배하여 왔으며, 잎과 줄기에서 추출한 추출물에는 stevioside, rebaudioside 등 여러 종의 폴리페놀이 함유되어 있다. 다양한 허브의 폴리페놀과 항산화 활성을 비교 분석한 결과Stevia rebaudiana에 가장 많은 폴리페놀이 들어있으며, Trolox, 녹차와 비교하여도 더 높은 항산화 결과를 나타내었다16,24). 본 실험에서는 이러한 SRE의 산화 스트레스에 대한 세포 보호효과를 관찰하기 위하여, AA+iron을 사용하여 산화적 스트레스를 유발하였고, 인체 유래 간세포인 HepG2 세포모델을 활용하였다18-21).

Iron은 간에 저장되는데 iron의 과다 축적은 간에서 macrophage를 활성화시켜 산화적 스트레스를 유발한다. 또한 pro-inflammatory cytokine의 생성을 매개하여 phopholipases의 활성을 유도하고, 인지질 막의 변화를 통해 AA의 방출을 촉매한다25,26). AA와 iron의 병용 투여 시, ROS 생성과 cell death가 일어난다. 이러한 이유로 AA+iron 모델은 다양한 항산화 물질들의 세포 보호 효능을 입증하기 위한 도구로 활용되어 왔다18,19). 본 연구에서는 Stevia rebaudiana methanol extract (SRE)의 세포보호 효과를 알아보기 위해 apoptotic marker인 procaspase-3의 발현 변화를 통하여 SRE의 AA+iron으로 유도된 apoptosis의 저해 효과를 확인하였다.

호기성 생물의 건강한 세포는 계속적인 산화과정을 통해 에너지를 만들어야 하므로 생체 내 ROS의 생성이 불가피하며, 세포와 조직에 해로운 독성을 일으켜 여러 가지 질병과 노화를 유발하는 원인이 된다27-29). O2-, H2O2 등의 ROS는 인체 내 대사과정의 부산물로서 지속적으로 생성되며, 불안정하고 반응성이 매우 강하다. 균형이 깨진 활성산소와 만성 산화적 스트레스의 증가는 지질의 산화와 세포막의 파괴, DNA 손상 그리고 단백질 변형을 가져오게 된다30) 본 실험에서 SRE는 AA+iron에 의해 유도된 세포 독성 및 apoptosis를 유의적으로 방어하였고, H2O2 생성 및 GSH의 감소를 효과적으로 억제하였다.

미토콘드리아는 세포의 신호전달, 주기 및 성장조절 등에 관여하고, 전자 전달계에서 생성된 ROS를 스트레스로 감지하여 apoptosis와 세포 생존율을 결정 짓는다31,32). 미토콘드리아의 음이온 이동에 관여하는 이온통로로는 외막에 존재하는 voltage-dependent anion channel (VDAC)과 내막에 존재하는 inner membrane anion channel (IMAC)이다. VDAC은 박테리아의 porin과 유사한 구조를 가지며 전해질과 크기가 작은 비전해질이 통과한다. 과도한 칼슘 유입이 일어나면 미토콘드리아 내막의 용질투과도가 급격히 증가되는데 이를 mitochondrial permeability transition (MPT)이라 한다. MPT를 구성하는 단백질로는 cyclophilin D, adenine nucleotide translocator (ANT) 등이 있다. mPTP의 개방은 대부분 미토콘드리아에서 만들어진 ROS에 의한 막 단백질 thiol의 산화로 일어난다33). 이러한 변화는 미토콘드리아의 산화적 스트레스 및 세포 독성 유발에 증가를 가져왔다20,21). 본 실험에서는 FACS를 통해 MMP의 변화를 측정하였고, 미토콘드리아 손상과 기능장애를 나타내는 RN1 fraction을 통해 SRE의 미토콘드리아 보호효과를 확인하였다. AMPK는 serine/threonine kinase의 일원으로 하나의 catalytic α subunit (α1 또는 α2)과 두 개의 regulatory subunit β (β1 또는 β2)와 γ (γ1, γ2 또는 γ3)로 heterotrimeric complex를 이루고 있다34,35). 그 중, α subunit의 N-terminal 부분에는 catalytic domain을 포함하고 있으며, 이 domain에는 upstream kinase에 의한 AMPK 인산화 및 활성화에 중요한 역할을 하는 threonine residue Thr 172가 존재한다36). AMPK activation은 이화작용을 자극하고, 동화작용을 저해하는 에너지 소비에 반응하여 apoptosis를 조절한다37). 근육 내 AMPK의 활성화는 PGC-1α 발현을 통해 미토콘드리아 관련 유전자들의 발현 증가와 세포 내 NAD(+)를 증가시키고, NAD(+) 의존적 탈아세틸화 효소인 SIRT1의 작용으로 PGC-1α의 탈아세틸화 및 활성화가 이루어지는 것으로 보고되었다18-21,38). 이전 연구에서는 AMPK activator(metformin, AICAR, sauchinone and resveratrol)를 처리함으로써 AA+iron에 의한 세포 독성, ROS 증가, 미토콘드리아 손상에 대한 방어효과가 AMPK의존적이라는 것을 관찰하였다39). 본 연구에서 ACC의 인산화를 통하여 AMPK activation을 관찰한 결과, SRE는 AMPK-ACC pathway를 조절하였다. AA+iron에 의하여 ACC의 인산화가 감소하고, 이는 SRE의 처리에 의해 회복되었다. 이러한 결과를 통하여 SRE의 미토콘드리아 보호효과와 AMPK-ACC pathway와의 관련성을 확인하였다. 또한, 본 연구에서 AA 처리 1시간 전에 SRE를 처리하는데, 사실 SRE는 처리 후 1시간에서 ACC의 인산화가 가장 강하였다. 즉 AA처리 전에 AMPK가 활성화하여 ACC 인산화가 이미 유도된 상황인 것을 알 수 있다. 이러한 결과는 항산화 효과를 가지는 SRE가 AA처리 전에 AMPK활성을 통해 산화스트레스에 대한 내재적 방어시스템을 강화시켰을 수도 있다고 생각된다18).

 

결 론

SRE의 항산화 효과를 검증하기 위하여, AA+iron으로 세포손상을 유발시킨 다음 HepG2 cell에서 cell viability, ROS, mitochondrial dysfunction, AMPK pathway에 미치는 영향을 조사하여 다음과 같은 결론을 얻었다.

SRE (0.1 mg/ml)는 AA+iron에 의한 cell death를 유의하게 보호하고, apoptosis 관련 단백질인 procaspase-3의 발현량 감소를 억제하였다. SRE는 AA+iron으로 증가된 산화적 스트레스를 억제하고, GSH를 회복시켜 항산화 효과를 나타낸다. SRE는 AA+iron으로 유발된 미토콘드리아 기능장애를 억제하여 세포와 미토콘드리아 보호효과를 가진다. SRE는 시간 의존적으로 ACC의 인산화를 증가시키고, AA+iron에 의한 ACC의 인산화 감소를 방어하였다.

일련의 실험결과로 SRE는 HepG2 cell에서 산화적 스트레스를 억제, 미토콘드리아를 보호하는 것으로 확인되었으며, 향후, 다른 세포주를 이용한 SRE의 세포보호 효과, 동물실험과 같은 지속적인 연구가 필요할 것으로 판단된다.
