Antioxidant activity of partially purified extracts isolated from Acanthopanax sessiliflorum Seeman

Abstract

The antioxidant activity and the qualitative analysis of Acanthopanax sessiliflorum Seeman were studied by partially purified extract using various methods: extraction by using ethanol solutions and temperatures, and absorption to Diaion HP20 column chromatography using 10%, 20%, 40%, 60% ethanol solutions. Major constituents, chlorogenic acid, caffeic acid, eleutheroside E, was determinated by HPLC method in Acanthopanax sessiliflorum S. 10% and 20% ethanol solutions contain chlorogenic acid (3.020$\pm$0.080%, 20.500$\pm$1.150%, respectively). 40% ethanol solution contains caffeic acid and eleutheroside E (12.270$\pm$0.360%, 1.670$\pm$0.140%, respectively). Diaion HP20 fractions (10%, 20%, 40%, 60% ethanol solutions) showed the scavenging activity of radicals and reactive oxygen species with the $IC_{50}$ values of $81.534{\pm}0.992{\mu}g/ml$, $1.748{\pm}0.098{\mu}g/ml$, $11.487{\pm}1.768{\mu}g/ml$, $21.960{\pm}0.547{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazly radical and $1713.548{\pm}34.565{\mu}g/ml$, $131.419{\pm}2.235{\mu}g/ml$, $200.681{\pm}2.444{\mu}g/ml$, $757.897{\pm}6.868{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. Especially, 20% and 40% ethanol fractions showed more antioxidant activity than dl-$\alpha$-tocopherol. These results suggest that Acanthopanax sessiliflorum S. extract and Diaion HP20 fractions may be useful as a potential source of nutraceutical and cosmetic products.

오가피는 건강보조식품, 식품이나 화장품 등의 첨가제로 좋은 약재로 평가되고 있다. 그러나 오가피의 연구의 대부분은 가온 용매추출이나 열수추출을 통한 유효성분의 추출과 생리활성 연구에 치중되어 있다. 따라서 온도 및 추출용매를 조절하여 추출하고 Diaion HP2O 분획을 통한 유효성분의 함량 및 항산화활성에 대하여 연구하였다. 오가피추출물은 열에 불안정하여 갈변현상을 보이므로 상온추출 및 냉온추출하는 것of 좋으며, 에탄올 함량별로 추출시 주 활성성분인 chlorogenic acid의 함량을 보면 100% 정제수 및 100% 에탄올의 단일 용매 추출보다 에탄올수용액을 사용하여 추출시 chlorogenic acid의 함량이 증가함을 알 수 있었다. 에탄올수용액에서의 chlorogenic acid의 함량은 유사하므로 농축시간을 고려하여 70$\sim$85% 에탄올수용액으로 추출하는 것이 바람직할 것으로 사료된다. 최적화된 조건에서 추출된 오가피추출물은 Diaion HP2O column chromatography를 이용하여 10%, 20%, 40%, 60% 에탄올수용액으로 분획 후, 각 분획층의 주 활성성분 합량과 항산화효과를 확인하였다. 10% 및 20% 에탄올 수용액층은 chlorogenic acid가 주 구성성분이었으며, 40% 에탄올 수용액층은 caffeic acid 및 eleutheroside E가 주 구성성분임을 알 수 있었다. Chlorogenic acid와 caffeic acid 함량이 높은 20% 및 40% 에탄올수용액 분획층에서 우수한 DPPH와 superoxide radical에 대한 소거효과를 나타내었다. DPPH radical 소거효과에서 20% 및 40% 에탄올수용액 분획층은 각각 $IC_{50}$ 값이 $1.748{\pm}0.098{\mu}g/ml$, $11.487{\pm}1.768{\mu}g/ml$을 나타내었으며, Superoxide radical 소거효과에서는 $131.491{\pm}2.235{\mu}g/ml$, $200.681{\pm}2.444{\mu}g/ml$로 양성 대조군으로 사용한 ascorbic acid와 동등하거나 dl-$\alpha$-tocopherol보다 우수한 항산화효과를 보였다. 각 분획층의 항산화효과를 미루어 볼 때, 20% 및 40%의 주 구성성분을 조합하여 오가피추출물을 제조하면 더욱 우수한 소재로 개발할 수 있을 것으로 사료된다. 본 기술연구소에서는 최적화된 조건에서 추출된 오가피추출물 및 Diaion HP2O 분획물을 이용하여 B16 melanoma 세포에서의 멜라닌 생성저해 효과를 연구하여 새로운 미백제를 개발 중에 있으며, 건강보조식품, 기능성 향장품소재 등으로의 활용 가치가 높을 것으로 사료된다.
