Detection of Wound-inducible Trans-Cinnamic Acid-4-Hydroxylase in Avocado, Persea americana, Roots

  • Trans-cinnamic acid-4-hydroxylase(tC4H) is the first cytochrome P450-dependent monooxygenase of the phenylpropanoid pathway. The roots of avocado seedlings were wounded and examined to determine whether the tC4H would be activated in response to wounding and/or whether tC4H activity be modulated by the application of exogenous p-coumarate. At the specified length of times, the wounded and treated roots were either frozen in liquid nitrogen or used immediately to extract microsomal proteins. The microsomal proteins were subjected to immunoblot analysis using polyclonal antibodies against CYP73 of tC4H gene. In this study, tC4H was induced in wounded roots sealed in bags within 6 hours, and in low level({TEX}$10^{-8}${/TEX}M) of p-coumarate solution within 24 hours, whereas the olution without p-coumarate and high levels of p-coumarate solution repressed tC4H induction in wounded roots. These results indicate that tC4H is induced by wounding in the root of avocado, and is inhibited by the application of exogenous p-coumarate.

Anthracnose of Safflower Caused by Colletotrichum acutatum

  • Anthracnose occurred severely on safflower plants grown in Euiseong and Jecheon areas of Korea in 1997 and 1998. The disease incidence was up to 100% in some fields, and symptoms developed on seedlings, leaves, stems, roots and hulls of the plants. Coletotrichum sp. was consistently present on the diseased plant parts, and all the isolates from the lesions were identified as Colletotrichum acutatum based on the morphological and cultural characteristics. Similar symptoms were produced on the host plants by artificial inoculation with isolates of the fungus. The fungus was reisolated from lesions on the plants inoculated. This is the first report that c. acutatum causes anthracnose of safflower.

Immunocytolocalization of Cell Wall Peroxidase and Other Wall Antigens from Maize Seedlings

  • Immunocytochemistry utilizes the specificity of the antigen-antibody reaction to localize specific antigens in cells or cellular organelles. Here we report the use of monoclonal antibodies, in conjunction with gold-labeled second antibodies to study the ultrastructural localization and tissue distribution of the Mr 98, 000 anionic peroxidase and other wall antigens. The antibody specific for this wall peroxidase, mWP3, labeled mainly the cell wall area. At the tissue level, the Mr 98, 000 peroxidase is located predominantly in the leaf mesophyll, internal coleoptile and sieve elements, but not in the root, as assayed with these procedures. The coleoptile walls were less heavily stained than the walls of leaf mesophyll cells. At the subcellular level, it is localized mainly in intercellular regions of the cell walls. A similar staining pattern was revealed by mWP19, one of anti-$\beta$ glucosidase antibody, though it looked less heavily stained than one with mWP3. In order to serve as a control wall staining using IgM monoclonal antibodies, mWP18 was used. Most of the label is localized over wall regions of cells of the young leaf mesophyll and coleoptile.

Trends of Several Air Pollutants and the Effects of Ozone on the Plant Antioxidant system in Platanus occidentalis in Korea

  • This study investigated concentrations of the several air pollutants and compared antioxidative enzyme activities on Platanus occidentalis because this tree species is one of the widespread street trees in Korea. This species has been emerging the ambient air pollutants during its growing periods. The purpose of this study was to identify the relationship between air pollution on the tree species and antioxidant enzyme activities on the trees. $O_3$, $NO_2$, CO and $SO_2$ concentrations of several cities in Korea were compared for last decades. Among the air pollutants, $O_3$ and $NO_2$ concentrations in six big cities in Korea showed similar increasing trends during this period. In contrast, $SO_2$ and CO concentrations in the same cities dramatically decreased between 1994 and 2005. Platanus occidentalis trees were controlled to investigate, ascorbate peroxidase (APX) and glutathione reductase (GR) activity. Ozone exposure generally increased APX and GR activities of tree seedlings. It is a typical compensatory strategy of stressed trees.

The Effects of Waste Leachate on the Eco-Physiological Characteristics of Populus euramericana

  • Populus euramericana has been identified as a possible species for use for phytoremediation of landfills. To identify the effects of waste leachate on the growth and physiological characteristics of Populus euramericana. four different treatments were applied to Populus euramericana seedlings: leachate solution (100% leachate). 25% dilution (75% leachate: 25% water. v/v), 50% dilution (50% leachate: 50% water. v/v) and control (100% tab water) were applied to Populus euramericana. Treatment with waste leachate significantly stimulated Populus euramericana height. diameter at root collar and biomass production relative to the water control. Chlorophyll contents. photosynthesis and transpiration of leachate irrigated-trees were significantly higher than those of water control. These results suggested that poplar could be a suitable species for phytoremediation in landfills because these species showed good growth performance and were capable of taking up waste leachate.

Development and Ultrastructure of Interfascicular Cambium in Stem of Ginkgo biloba Seedling (은행나무 유직물의 줄기에서 유관동문 형성층의 발생과 미세구조)

  • The progressive differentiation of interfascicular cambium from residual meristem in the first internode of Ginkgo biloba seedlings was elucidated by light and electron microscopy. The cells of residual meristem were small and homogeneous and heterogeneous in their arrangement but those of the adjacent cortex and pith were large and homogeneous. Some interprocambial residual meristem progressively became elongated and vacuolated during the process of the differentiation. In tangential section, residual meristem composed of long and short cells. The eventual interfascicular cambium had long fusiform initials and short ray initials. Storage materials in the cells progressively disappeared from the interprocambial residual meristem and were absent in early interfascicular cambium. Both the radial and tangential walls of cells of the interprocambial residual meristem were almost the same, but the radial wall became progressively thicker than the tangential wall during differentiation of interfascicular cambium. From these results, it is clear that interfascicular cambium is gradually differentiated from residual meristem.

Effect of Plant Age on Infection of Soybean by Calonectria ilicicola (Calonectria ilicicola의 감염에 대한 콩 식물체 나이가 미치는 영향)

  • A series of greenhouse test was conducted to evaluate infection of Calonectria ilicicola on soybean plants of different ages at time of inoculation. Lesion length and number of perithecia were determined on cultivars Braxton, Deltapine 726, and Riverside 699 that were 10∼40 days old and 4-10 days old at time of inoculation. Quadratic and linear relationships were described between plant age at inoculation and lesion length or perithecia production in greenhouse studies. Soybean seedlings exhibited low susceptibility to C. ilicicola regardless of cultivar susceptibility. On 8- or 1-0-days-old Braxton, lesion lenght and perithecia numbers were reduced. Lesion lengths were longest on plants 30 days old whereas perithecia production was greatest on plants 20∼30 days old at time of inoculation. Differences in lesion length and perithecia production that were observed on young plants (4∼10 days old) were similar to relative levels of susceptibility in soybean cultivars in greenhouse and field tests, suggesting that reaction to C. ilicicola in soybean cultivars may be determined early in plant development.

NPR1 is Instrumental in Priming for the Enhanced flg22-induced MPK3 and MPK6 Activation

  • Pathogen-associated molecular patterns (PAMPs) activate mitogen-activated protein kinases (MAPKs), essential components of plant defense signaling. Salicylic acid (SA) is also central to plant resistance responses, but its specific role in regulation of MAPK activation is not completely defined. We have investigated the role of SA in PAMP-triggered MAPKs pathways in Arabidopsis SA-related mutants, specifically in the flg22-triggered activation of MPK3 and MPK6. cim6, sid2, and npr1 mutants exhibited wild-type-like flg22-triggered MAPKs activation, suggesting that impairment of SA signaling has no effect on the flg22-triggered MAPKs activation. Pretreatment with low concentrations of SA enhanced flg22-induced MPK3 and MPK6 activation in all seedlings except npr1, indicating that NPR1 is involved in SA-mediated priming that enhanced flg22-induced MAPKs activation.

Prevalence and Transmission of Seed-Borne Fungi of Maize Grown in a Farm of Korea

  • Seed-borne fungi of some maize cultivars/lines grown during the months from May to September of 2001, collected from Dongguk University farm, Go Young City, IL Sang Gu, Korea were detected by blotter method. In all six fungi namely Alternaria alternata(Fr.) Keissler, Aspergillus niger Van Tiegh, Fusarium moniliforme Sheldon, Fusarium sp., Penicillium sp. and Ustilago zeae Unger. were found to associated with maize seeds. Prevalence of seed-borne fungi also varied. The highest percentages of seed-borne fungi were recorded with Fusarium moniliforme and the lowest in Penicillium sp. Transmission of all seed-borne pathogens from seeds to seedlings were also detected by test tube seedling symptom test. Among the seed-borne fungi, Alternaria alternata, Fusarium moniliforme and Fusarium sp. produced distinct seed rot and seedling infection symptoms. All the transmitted seed-borne fungi might be caused primary source of infection to the maize crop.
