효모(Saccharomyces exiguus)의 급여가 육계 생산성, 맹장내 미생물 및 분내 암모니아 가스 발생에 미치는 영향 (Effect of Dietary Yeast (Saccharomyces exiguus) on Growth Performance, Cecal Microflora and Fecal Ammonia Gas in Broiler Chickens)

  • 본 연구는 Saccharomyces exguus의 첨가 급여가 육계 생산성 및 분내 유해가스 발생에 미치는 영향을 구명하기 위하여 수행되었다. 1일령 육계 수평아리(Ross strain) 270수를 공시하여 3처리 3반복 반복당 30수씩 완전 임의배치하였다. 처리구는 대조구인 항생제 첨가구 및 효모 0.5와 1.0% 처리구를 두었다. 본 실험에서 5주 종료 체중 및 증체율이 효모의 첨가 수준에 따라 유의하게 증가하였으며(P<0.05), 사료 요구율은 효모 첨가시 개선되는 경향이 보였으나, 유의성은 인정되지 않았다. 맹장 내 E. coli, Salmonella, Lactobacillus 수는 대조구인 항생제 첨가구와 비교하여 차이가 없었다. 암모니아 가스의 발생량은 대조구에 비해 유의하게 감소하였다(P<0.05). 계분 내 저급 지방산 및 휘발성 유기 화합물은 변화가 없었다. 본 연구 결과 육계에 Saccharomyces exguus급여는 생산성을 향상시키고 계분 내 암모니아 가스 발생량을 감소시켜 항생제 대체 및 환경 개선제로 이용 가능함을 시사하였다.

석유탄화수소를 이용한 단세포단백질의 생산에 관한 연구 -V. 균체의 회수, 정제 및 예비 동물사육 시험- (Production of Single-Cell Protein on Petroleum Hydrocarbon -V. Recovery and Purification of the Yeast Cell and Its Preliminary Animal Feeding Test-)

  • 경유(輕油)를 기질(基質)로 한 석유탄화수소발효액(石油炭化水素醱酵液)으로부터 효과적(效果的)으로 균체(菌體)를 회수(回收), 정제(精製)하기 위한 몇가지 지견(知見)과 예비동물사육시험(飼備動物飼育試驗)결과를 요약(要約)하면 다음과 같다. 1) 발효배양액(醱酵培養液)으로부터 균체(菌體) cream의 정치분리(精置分離)는 30분만에 종료(終了)되며 이때 cream의 균체농도(菌體濃渡)는 5.6%였다. 2) pH 9 이상의 강알칼리성에서 균체(菌體) emulsion은 불안정(不安定)하였으며 열처리(熱處理), 동결(凍結)에 의하여 emulsion으로부터 $60{\sim}80%$의 균체(菌體)를 분리(分離)할 수 있었다. 3) Emulsion 파괴(破壞)를 위한 식용계면활성제(食用界面活性劑)로는 트리오가 가장 우수하였으며 그 적합한 농도(濃渡)는 0.3%였다. 이 외에 Aldo, Monogly 5-7 및 G-4도 유효(有效)하였다. 4) NaCl, KCl 등의 할로겐화알칼리금속염이 균체(菌體) emulsion 파괴(破壞)에우수하였으며 트리오와 같은 계면활성제(界面活性劑)와 혼합(混合)하여 사용하므로서 90% 이상의 균체(菌體)를 분리(分離)할 수 있었다. 5) 건조반제품(乾燥半製品)을 hexane-ethyl alcohol 및 iso-propyl alcohol의 공비혼합물(共沸混合物)로 유출(抽出)하는 것이 가장 기대(期待)되는 균체(菌體)의 정제방법(精製方法)이며 추출온도(抽出溫度)는 $58^{\circ}C$, 추출시간(抽出時間)은 30분(分), 균체(菌體) 대(對) 용매비율(溶媒比率)은 1 : 3이 적합하였다. 6) 균체단백질(菌體蛋白質)의 사양실험결과 어백과 5% 대체(代替)한 수준(水準)에시는 대조구(對照區)보다 3.3%의 더 좋은 증체(增體)를 보였으나 8% 대체구(代替區)는 다른 처리구에 비하여 간(肝)중량이 더 무거운 것이 인정되었다.

효모배양물의 수준별 급여가 육성·비육돈의 생산성, 영양소 소화율, 돈육의 이화학적 특성 및 지방산 조성에 미치는 영향 (Effect of Dietary Supplementation of Yeast Culture on the Performance, Nutrient Digestibility and Physico-Chemical characteristics of the Pork in Growing-Finishing Pigs)

  • 본 연구는 양돈 사료에 효모배양물을 수준별로 급여(0, 0.1, 0.2, 0.4%)하여 총 2회 시행하였다. 실험 1에서는 처리구당 3반복으로 반복당 10두씩 총 120두를 공시하여 이유시부터 출하시까지 112일 동안 사양실험을 통하여 생산성을 조사하였고, 실험 2에서는 실험 종료후 도축된 돈육 등심의 품질을 고찰하였다. 효모배양물을 급여하여 112일 동안 성장 단계별로 조사한 증체량과 사료요구율은 효모배양물 급여구와 대조구간에서 차이를 보이지 않았다. 영양소 소화율은 조단백질과 조섬유 소화율에서 효모배양물 0.1과 0.2% 급여구가 대조구에 비하여 증가하였다. 효모배양물을 급여하여 생산된 돈육 등심의 일반성분 중 조단백질의 함량은 효모배양물 0.1%를 급여한 처리구가 대조구에 비하여 증가하였다. 돈육의 pH, 포장 감량 및 조리 감량은 처리구간에 차이가 없었고, 육색도 대조구와 비교해서 차이가 없는 것으로 나타났다. 돈육의 shearing force는 효모배양물 0.1% 급여구에서 대조구에 비하여 현저하게 낮았으나 cohesiveness, springiness, gumminess 및 brittleness에 있어서는 차이가 없었다. 돈육의 지방산 조성에서 포화지방산, 단가불포화지방산 및 다가불포화지방산은 효모배양물 급여구와 대조구간에 아무런 차이도 없었으며, 콜레스테롤 함량에서도 차이가 없었다.

  • Brownish discoloration develops very rapidly in the storage of dried oyster. This undesirable browning is mainly caused by the series of reactions of sugar-amino condensation, enzymatic oxidation of tyrosine and/or the oxidative rancidity of lipids in the tissue of oyster. Sulfites are commonly used as inhibitors for Maillard type browning reactions in agricultural products. The inhibitory effect of sulfite treatment on canned oysters was also confirmed in some investigations. The results suggested that sulfites not only work on blocking tile amadori rearrangement but also on the reduction of free tyrosine which retards the progress of enzymatic oxidation of tyrosine tyrosinase. In this paper, the effect of sodium sulfite treatment on the reduction of reducing sugar and free tyrosine as a function i)f browning inhibition in oyster was tested and other treatment with glucose-oxidase and yeast were also applied. In preparation of samples, fresh oysters were soaked in sodium sulfite solution by various concentration for different treating times, washed in running water to remove the sulfite residue, and finally dried in the shade. In the result, the treatment of sodium sulfite was certainly effective on the reduction of both free tyrosine and reducing sugars in fresh oyster. The best results were obtained by the treatment of 0.5M sodium sulfite solution for 60 minutes each for soaking and washing. Treatment with, glucose-oxidase and yeast solutions, however, did appear somewhat effective but it required so much time for a certain effect that it seemed not practically applicable.

Effect of Chlorine Dioxide Treatment on Microbial Growth and Qualities of Chicken Breast

  • Chlorine dioxide $(ClO_2)$ treatment was evaluated for microbial growth inhibition and its effects on the quality of vacuum-packaged chicken breasts. Chicken breast samples were treated with 3, 50, and 100 ppm of $ClO_2$ solution, respectively. After $ClO_2$ treatment, chicken breast samples were individually vacuum-packaged and stored at $4^{\circ}C$, a typical storage temperature for meat and meat product, for 7 days. The vacuum-packaged chicken breasts treated with $ClO_2$ had significantly lower total bacteria, yeast and mold, total coliform, and Salmonella spp. were significantly reduced by $ClO_2$ treatment. $D_{10}-values$ of total bacteria count, yeast and mold, total coliform, and Salmonella spp. in vacuum-packaged chicken breasts was 93, 83, 85, and 50 ppm, respectively. The pH of vacuum-packaged chicken breasts decreased with increasing $ClO_2$ concentration. Thiobarbituric acid reacted substance (TBARS) values of vacuum-packaged chicken breasts increased during storage, regardless of $ClO_2$ concentration. $ClO_2$ treatment caused negligible changes in Hunter L, a, and b values in the vacuum-packaged chicken breasts. Sensory evaluation of the vacuum-packaged chicken breasts showed that there were no significant changes among the samples treated with various $ClO_2$ concentration. These results indicate that $ClO_2$ treatment could be useful in improving the microbial safety and quality of meat products.

Yeast Culture(Saccharomyces cerevisiae, Pichia pastoris)가 육계의 생산성, 소장내 미생물 균총 및 혈청 IgG 농도에 미치는 영향 (Effects of Supplementary Yeast Culture(Saccharomyces cerevisiae, Pichia pastoris) on the Performance Small Intestinal Microflora and Serum IgG Concentration in Broiler Chickens)

  • 본 시험은 yeast culture제제들(Saccharomyces cerevisiae, Pichia pastoris)의 첨가가 육계의 성장, 장내 미생물균총 및 혈청 IgG농도에 미치는 영향을 측정하기 위하여 기초사료(control)에 chlorotetracycline 100ppm(CTC), Saccharomyces cerevisiae 0.3%(YC-SC), Pichia pastoris 0.3% (YC-PP), 정제된 Pichia pastoris culture 0.1% (RPPC-0.1) 및 0.3%(RPPC-0.3)를 각각 첨가하였다. Ross 종 육계 1000수(암․수 각각 500수)를 20개의 floor pen에 50수(암 수 각 25수)씩 수용하여 control, CTC, YC-SC와 RPPC-0.1구는 3반복으로 YC-PP와 RPPC-0.3구는 4반복으로 배치하였다. 증체량은 처리간에 유의한 차이가 없었으나 대조구나 항생제첨가구에 비해 효모배양물의 첨가구(YC-SC, RPPC-0.1 및 RPPC- 0.3)에서 증체량이 약 4-5% 개선되는 효과가 있었다. 항생제와 효모배양물은 첨가한 구가 대조구에 비해 사료효율이 개선되며 폐사율이 감소하는 경향이 있었다. 영양소 이용율은 yeast culture 첨가에 의해 유의한 영향을 받지 않았다. 소장하부 내용물의 Lactobacilli의 수는 효모배양물의 첨가구(YC-SC, RPPC-0.1 및 RPPC-0.3)에서 대조구보다 다소 높은 경향을 보였고, Cl. perfringens의 수는 CTC와 정제된 Pichia pastoris culture의 첨가에 의해 뚜렷이 저하되었다. E. coli의 수는 정제된 Pichia pastoris 구들이 낮았는데 특히 RPPC-0.3가 대조구보다 유의하게(P$\prec$0.02) 낮았다. 혈청 IgG 농도는 처간에 유의한 차이는 없었으나 효모배양물의 첨가구들에서 control구나 항생제 첨구에서보다 높은 경향을 나타내었다. 결론적으로 본시험에 사용된 yeast culture제제는 생산성, 소장내 미생물 균총과 혈청 IgG에 있어서 유의한 차이는 없었으나 항생제(CTC)와 같거나 그 이상의 개선효과를 나타내었다.

Functional Screening for Cell Death Suppressors and Development of Multiple Stress-Tolerant Plants

  • Bax, a mammalian pro-apoptotic member of the Bcl-2 family induces cell death when expressed in yeast. To investigate whether Bax expression can induce cell death in plant, we produced transgenic Arabidopsis plants that contained murine Bax cDNA under control of a glucocorticoid-inducible promoter. Transgenic plants treated with dexamethasone, a strong synthetic glucocorticoid, induced Bax accumulation and cell death, suggesting that some elements of cell death mechanism by Bax may be conserved among various organisms. Therefore, we developed novel yeast genetic system, and cloned several Plant Bax Inhibitors (PBIs). Here, we report the function of two PBIs in detail. PBI1 is ascorbate peroxidase (sAPX). Fluorescence method of dihydrorhodamine123 oxidation revealed that expression of Bax in yeast cells generated reactive oxygen species (ROS), and which was greatly reduced by co-expression with sAPX. These results suggest that sAPX inhibits the generation of ROS by Bax, which in turn suppresses Baxinduced cell death in yeast. PBI2 encodes nucleoside diphosphate kinase (NDPK). ROS stress strongly induces the expression of the NDPK2 gene in Arabidopsis thaliana (AtNDPK2). Transgenic plants overexpressing AtNDPK2 have lower levels of ROS than wildtype plants. Mutants lacking AtNDPK2 had higher levels of ROS than wildtype. $H_2O_2$ treatment induced the phosphorylation of two endogenous proteins whose molecular weights suggested they are AtMPK3 and AtMPK6. In the absence of $H_2O_2$ treatment, phosphorylation of these proteins was slightly elevated in plants overexpressing AtNDPK2 but markedly decreased in the AtNDPK2 deletion mutant. Yeast two-hybrid and in vitro protein pull-down assays revealed that AtNDPK2 specifically interacts with AtMPK3 and AtMPK6. Furthermore, AtNDPK2 also enhances the MSP phosphorylation activity of AtMPK3 in vitro. Finally, constitutive overexpression of AtNDPK2 in Arabidopsis plants conferred an enhanced tolerance to multiple environmental stresses that elicit ROS accumulation in situ. Thus, AtNDPK2 appears to playa novel regulatory role in $H_2O_2$-mediated MAPK signaling in plants.

Effect of supplementation of yeast with bacteriocin and Lactobacillus culture on growth performance, cecal fermentation, microbiota composition, and blood characteristics in broiler chickens

  • Objective: The aim of the present study was to investigate the effect of yeast with bacteriocin and Lactobacillus cultures (mixture of Lactobacillus agilis BCRC 10436 and Lactobacillus reuteri BCRC 17476) supplements, alone or in combination, on broiler chicken performance. Methods: A total of 300, 1-d-old healthy broiler chickens were randomly divided into five treatment groups: i) basal diet (control), ii) basal diet+0.25% yeast (Saccharomyces cerevisiae) (YC), iii) basal diet+0.25% yeast with bacteriocin (BA), iv) basal diet+Lactobacillus cultures (LAB), and v) basal diet +0.25% yeast with bacteriocin+Lactobacillus cultures (BA+LAB). Growth performance, cecal microbiota, cecal fermentation products, and blood biochemistry parameters were determined when chickens were 21 and 35 d old. Results: The supplementation of YC, BA, and BA+LAB resulted in a significantly better feed conversion rate (FCR) than that of the control group during 1 to 21 d (p<0.05). The LAB supplementation had a significant effect on the presence of Lactobacillus in the ceca at 35 d. None of the supplements had an effect on relative numbers of L. agilis and L. reuter at 21 d, but the BA supplementation resulted in the decrease of both Lactobacillus strains at 35 d. The BA+LAB supplementation resulted in higher short chain fatty acid (SCFA) in the ceca, but LAB supplementation significantly decreased the SCFA at 35 d (p<0.05). All treatments tended to decrease ammonia concentration in the ceca at 21 d, especially in the LAB treatment group. The BA supplementation alone decreased the triacylglycerol (TG) concentration significantly at 21 d (p<0.05), but the synergistic effect of BA and LAB supplementation was required to reduce the TG concentration at 35 d. The YC supplementation tended to increase the plasma cholesterol at 21 d and 35 d. However, the BA supplementation significantly decreased the cholesterol and low density lipoprotein cholesterol level at 35 d. In conclusion, the BA+LAB supplementation was beneficial to body weight gain and FCR of broiler chickens. Conclusion: The effect of BA and LAB supplementation may be a result of the growth of lactic acid bacteria enhancement and physiological characterization of bacteriocin, and it suggests that the BA and LAB supplementation level or Lactobacillus strain selection should be integrated in future supplementation designs.

Effect of Aqueous Chlorine Dioxide Treatment on the Microbial Growth and Quality of Chicken Legs during Storage

  • The effect of aqueous chlorine dioxide ($ClO_2$) treatment on microbial growth and quality of chicken leg during storage was examined. Chicken leg samples were treated with 0, 50, and 100 ppm of $ClO_2$ solution and stored at $4^{\circ}C$. Aqueous $ClO_2$ treatment significantly decreased the populations of total aerobic bacteria, yeast and mold, and coliforms in chicken leg. One hundred ppm $ClO_2$ treatment reduced the initial populations of total aerobic bacteria, yeast and mold, and coliforms by 0.93, 1.15, and 0.94 log CFU/g, respectively. The pH and volatile basic nitrogen values in the chicken leg decreased with increasing aqueous $ClO_2$ concentration, while concentrations thiobarbituric acid reactive substances (TBARS) increased during storage regardless of aqueous $ClO_2$ concentration. Sensory evaluation results revealed that the quality of the chicken leg treated with aqueous $ClO_2$ during storage was better than that of the control. These results indicate that aqueous $ClO_2$ treatment can be useful for improving the microbial safety of chicken leg during storage.
