$^1H$ NMR-Based Metabolomic Approach for Understanding the Fermentation Behaviors of Wine Yeast Strains

  • $^1H$ NMR spectroscopy coupled with multivariate statistical analysis was used for the first time to investigate metabolic changes in musts during alcoholic fermentation and wines during ageing. Three Saccharomyces cerevisiae yeast strains (RC-212, KIV-1116 and KUBY-501) were also evaluated for their impacts on the metabolic changes in must and wine. Pattern recognition (PR) methods, including PCA, PLS-DA and OPLS-DA scores plots, showed clear differences for metabolites among musts or wines for each fermentation stage up to 6 months. Metabolites responsible for the differentiation were identified to valine, 2,3-butanediol (2,3-BD), pyruvate, succinate, proline, citrate, glycerol, malate, tartarate, glucose, N-methylnicotinic acid (NMNA), and polyphenol compounds. PCA scores plots showed continuous movements away from days 1 to 8 in all musts for all yeast strains, indicating continuous and active fermentation. During alcoholic fermentation, highest levels of 2,3-BD, succinate and glycerol were found in musts with the KIV-1116 strain, which showed the fastest fermentation or highest fermentative activity of the 3 strains, whereas the KUBY-501 strain showed the slowest fermentative activity. This study highlights the applicability of NMR-based metabolomics for monitoring wine fermentation and evaluating the fermentative characteristics of yeast strains.

천호지 저질토에서 분리한 철환원세균의 특성 (Characterization of Fe (III)-Reducing Bacteria Isolated from the Sediment of Chunho Reservoir)

  • 금속 이온 환훤 세균에 의한 철(III)환원은 생물지구화학적 물질순환(biogeochemical cycle)에 무척 중요하다. 이는 크롬(Ⅵ)이나 우라늄(Ⅵ)과 같은 독성 중금속 물질의 환원과 유기물질의 산화에 모텔이 되기 때문이다. 총 37균주의 Fe (III)환원 세균을 소양호와 천호지의 저질토에서 각각 분리하였다. 두 정점 중 초기 Fe (II)의 함유량이 가장 높았던 것은 소양호의 저질토였으나 Fe (III)환훤능은 반대로 가장 낮은 Fe (II)함유량을 보여 주었던 천호지가 높게 나타났다. 또한 분리한 균주 중 천호지에서 분리한 균주 C2와 C3가 가장 높은 Fe (III) 환훤능을 보여 주었으며 이 균주를 이용하여 다양한 전자 공여체의 이용 여부를 실험하였다. Glucose, yeast extract, acetate, ethanol, toluene등을 이용하여 실험한 결과 두 균주 모두 glucose와 yeast extract만을 전자 공여체로 이용하였다. 또한 전자 수용체로 토양에 광범위하게 존재하는 humid acid와 nitrate를 이용하였으며 수율이 높은 nitrate reduction에 의해 환원되었던 humic acid가 다시 재 산화되는 것을 관촬할수 있었다. 활성능이 우수한 균주 C2와C3의 165S rRNA유전자 분석 결과에 의하면 Aeromonas hydrophila와 95%의 유사성을 보여주었다.

고추장의 발효미생물에 관한 연구 2 (Microbiological Studles of Red pepper paste Fermentation (II))

  • Among 50 yeasts of red pepper paste isolated and identified in the previous report, 12 strains of yeast which possess osmophilic and non-film characteristics were examined their growth rates and alcohol fermentation ability in red pepper medium. 5 strains of yeast possessing powerful growth rates and excellent alcohol fermentation ability were selected as excellent yeasts in this experiment. And their cultural conditions, salt resistance and red pepper concentration upon the growth were examined in this report. The results obtained were as follows ; 1) Among 12 strains of yeast, Saccharomyces cerevisiae group II(strain $D_1$), Saccharomyces oviformis($D_2$), Saccharomyces steineri($M_3$), Saccharomyces rouxii($D_9$) and Saccharomyces mellis($G_8$) were selected as excellent yeasts. 2) Among 5 strains of yeast selected, the strain $D_1$ and $D_2$showed the best alcohol fermentation ability and the best fermentative flavour. 3) The optimum temperature for growth of the strain $D_1$ and $D_2$ was $25^{\circ}C$$M_3$, $D_9$ and $G_8$ $30^{\circ}C$$D_1$ and $D_2$ was $62^{\circ}C$$M_3$,$D_9$ and $G_8$ ws 60.deg.C with treatment for 10 minutes. 4) The optimum pH for growth of all strains was around 5.0. 5) The strain $D_1$ and $M_3$were grown exceedingly well on the emdia containing 3 percent of NaCl and $D_2$,$D_9$ and $G_8$ on the media containing 6 percent of NaCl. 6) In the case of 5 percent of red pepper liquor added 5 percent of glucose was more nutritional than in the case of 5 percent glucose only. However, the growth rates of yeasts were restrained by the increase of red pepper concentration.

반수체 Saccharomyces diastaticus의 에탄올내성 증진 (Improvement of Ethanol-Tolerance of Haploid Saccharomyces diastaticus)

  • Several mutation procedures have been compared to obtain an ethanol-tolerant Saccha- romyces diastaticus strain secreting glucoamylase. These procedures include spontaneous mutation, EMS treatment, UV irradiation, and combination of EMS treatment and UV irradiation. All these methods were followed by adaptation of the yeast cells to gradually higher ethanol concentration. Among these procedures, the combined method of EMS treatment and UV irradiation gave the promising result, i.e. the ethanol tolerance of the yeast increased from 11.5%(v/v) to 14.0%(v/v). Respiratory deficient petite mutants of industrial and ethanol-tolerant yeast strains have been isola- ted and hybridized with haploid S. diastaticus strains. The resulting hybrids showed increased ethanol tolerance and starch-fermentability.

해국(Aster spathulifolius Maxim.)의 꽃에서 분리한 효모의 분자계통학적 분포해석 및 생물계면활성제 생산 균주의 스크리닝 (Phylogeny of Yeasts Isolated from the Flower of Aster spathulifolius Maxim. and Screening of Biosurfactant Producers)

  • BACKGROUND: Yeast biotechnology finds applications in various industries. Hence, we sought to explore the yeasts associated with the flower of Aster spathulifolius Maxim. This study aimed to isolate yeasts from the flower of the plant and screen for biosurfactant-producing yeasts. METHODS AND RESULTS: We collected flowers of Aster spathulifolius Maxim. and performed pure isolation using four types of media. In total, 117 strains belonging to 4 genera, namely, Cryptococcus (75 strains), Aureobasidium pullulans (30 strains), Candida (11 strains), and Rhodotorula (1 strain), were isolated and identified by ITS sequencing. Upon in-depth analysis, Cryptococcus, the most dominant genus (75 strains) was categorized into the 'Unknown group'. Upon in-depth analysis of A. pullulans, we discovered the 'Unknown group I' (27 strains) and the 'Unknown group II' (2 strains), which have not been reported previously. Two A. pullulans isolates with potent surfactant activity were selected via the screening procedure. CONCLUSION: In this study, a total of 117 strains were isolated from the flower of Aster spathulifolius Maxim. In addition, two biosurfactant-producing yeasts were identified from among the isolated yeasts.

Genomewide Profiling of Rapamycin Sensitivity in Saccharomyces cerevisiae on Synthetic Medium

  • The target of rapamycin (TOR) signaling pathway is a conserved pathway that regulates eukaryotic cell growth in response to environmental cues. Chemical genomic approaches that profile rapamycin sensitivity of yeast deletion strains have given insights into the function of TOR signaling pathway. In the present study, we analyzed the rapamycin sensitivity of yeast deletion library strains on synthetic medium. As a result, we identified 130 strains that are hypersensitive or resistant to rapamycin compared with wild-type cells. Among them, 36 genes are newly identified to be related to rapamycin sensitivity. Moreover, we found 16 strains that show alteration in rapamycin sensitivity between complex and synthetic media. We suggest that these genes may be involved in part of TOR signaling activities that is differentially regulated by media composition.

메탄올 자화효모에 관한 연구 (Studies on Methanol-assimilating Yeasts)

  • The distribution of methanol-assimilating yeasts on three different sources (elm bark, soil and fresh-water mud) and the growth conditions of a new strain of Candidaboidinii (SIO) wereexamines. From 150 samples, 91 methanol yeasts were isolated through enrichment culture ; they were identified as 77 strains of Candida boidinii including four new strains, 5 isolates of Torulopsis pinus, 3 strains of Hansenula polymorpha and one sstrain of Pichia pastoris respectively. The comparison of these yeasts with three sources indicated that decaying bark of elm tree other two, and that Gandida boidinii was most frequently distributed in all three sources. Four new strains of Candida boidinii were freshly isolated and their taxonomical properties were discussed. Of them, SIO strain was selected and characterized for its growth on methanol. This yeast could grow well on less than 1%(v/v) methanol. However, its growth was inhibited at 10% methanol. The cell yield was 3.1g (dry weight) per 1000ml of mineral mediurr, containing 1%(v/v) methanol as well as 01.% yeast extract as additive. The concentration of 0.1% yeast extract appears to be effective for the biomass production. Optimum conditions for growth on methanol was found to be : $28^{\circ}C,\;NH_4^+$ as nitrogen sources, thiamine as vitamin, and pH 4.5 to 6.0. The cell composition was as follows : crude protein and nucleic acids were 54% and 7% respectively. The amino acids were also described.

Flowers in Islands and Mountains of Korea

  • Yeasts were isolated from wild flowers of some islands and mountains such as Jeju-do, Ulleungdo, Yokjido, Seonyudo and Gyejoksan, Oseosan, Beakamsan and Deogyusan in Korea and were identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA or internal transcribed pacer(ITS) 1 and 2 including 5.8S rDNA using BLAST. Seventy two yeast strains of two hundred eighty nine species were isolated from wild flowers in islands and mountains, Korea. Among them, Cryptococcus species were isolated the most dominantly, and Metschnikowia reukaufii were also isolated thirty species, 10.3% of total strains. Twenty-three species including Cryptococcus aureus were overlapped between yeast strains of the islands and mountains. Some physiological functionality of the culture broth and cell-free extracts from two hundred eighty nine yeast strains were determined. The supernatant of Candida sp. 78-J-2 showed antioxidant activity of 22.5%, and supernatant of Metschnilowia reukaufii SY44-6 showed anti-gout xanthine oxidase inhibitory activity of 49.6% and whitening tyrosinase inhibitory activity of 38.4%, respectively.

Killer 효모의 분리 및 특성 (Isolation of the killer yeasts and its characteristics)

  • Ten strains out of about 1,000 yeast strains isolated from byproducts of alcoholic industries, milk products, fruits, greens, food-related industries and soils of nature, revealed the killer activities. Two strains which have excellent killer activities among them were isolated and identified with Saccharomyces cerevisiae B 15-1 and Hansenula anomala Y 33 by investigation of the morphological, cultural and physiological properties. The optimal conditions on these strains for the production of killer toxin were investigated. The strain B 15-1 showed the highest killer toxin activities when it was cultured up to the log phase of 48 hr in YPD medium (pH 4.7) at $25^{\circ}C$. On the other hand, the strain Y33 revealed the highest activities when it was cultured up to the stationary phase of 60 hr in YPD medium (pH 4.0) at $20^{\circ}C$. The sensitive strain Kyokai 7 was found to be killed entirely by the killer toxin produced from the wild killer yeast B 15-1 when B 15-1 was cocultured with the same cell concentration ($10^{6}$ cells/ml) of Kyokai 7 after cultivation of 36 hr, and with large concentration ($9\times 10^{7}$ cells/ml) after 48 hr.

In vitro에서의 Malassezia pachydermatis의 성장에 대한 혈청과 Transferrin의 억제효과 (The Effect of Serum and Tramsferrin on the Growth of Malassezia pachydermatis in vitro)

  • The inhibitory effect of pooled canine serum on the growth of ten strains of Malassezia pachydermatis in vitro was investigated. Studies were also carried out to observe the effect of different concentrations of unsaturated bovine transferrin on Malassezia growth in vitro. Ten strains of Malassezia pachygedrmatis in normal canine serum (11.1%, 44.4%) were found to significantly inhibit the growth (p<0.0005) not only in a dose dependent but also in a time dependent manner. The same strains of yeast treated with 1, 2, 4 and 8 times the normal value of serum transferrin (3.0 mg/ml, 6.0 mg/ml, 12.0 mg/ml, 24.0 mg/fl), were shown to have significantly lower OD readings (p<0.05) when compared to yeast treated in lower concentrations of transferrin (1.5 mg/ml). The optical density (OD) of the ten strains of yeast were significantly lower (p<0.005) when treated with various concentrations of transferrin than with the saline control except at 72 hours post incubation. These results indicate that serum has inhibitory effects on Malassezia pachydermatis growth in vitro, and transfferin is one of the components that contribute towards this inhibitory role. The inhibitions are dose and time dependent.
