GalhuaHaeJungTang Inhibit Ethanol-induced Gastric Injury in Rats (갈화해정탕이 주상(酒傷)에 미치는 영향 - ethanol-유도 위점막의 산화적 손상을 중심으로 -)

  • This Study was carried out to investigate the protective effect of GalHwaHaJungTang(GHT) on the gastric lesions indused by ethanol in rats. Rats were pretreated with GHT extract 1.7ml/kg, 3.4ml/kg, 5ml/kg and then were given orally 1ml of absolute ethanol 30min later. The animals were killed 1hr after ethanol treatment In morphorogic change, ethanol induced prominent longitudinal hemorrhagic lesions in the stomach. But GHT-pretreatment showed a dose-depentent decrease in the mucosa lesions. GHT extract siginificantly reduced lipid peroxidation and also induced an increase of superoxide dismutase(SOD), catalase and gluthathion(GSH) levels, but showed no influence in the type conversion of xanthine oxidase. These results suggest that the proposed gastroprotective effect may involve activation of antioxidant effects, but independent of the xanthine oxidase system.

Effect of EGF against Oxygen Radical-Induced Neurotoxicity in Cultured Spinal Dorsal Root Ganglion Neurons of Mouse (산소자유기에 의해 저해된 배양 척수감각 신경절 세포에 대한 상피세포성장인자의 영향)

  • In order to elucidate the cytotoxic effect of oxygen radicals on cultured spinal dorsal root ganglion(DRG) neurons derived from mouse. the neurotoxic effect of oxygen radicals w as examined after cultured DRG neurons were exposed to xanthine oxidase(XO) and hypoxanthine(HX)-oxygen radical generating system. In addition. neuroprotective effect of epidermal growth factor(EGF) against oxidant-induced neurotoxicity was also evaluated in these cultures. The results were, as follows: 1. Lethal concentration 50(LC$_{50}$) was 35mU/ml XO and 0.1mM HX in cultured DRG neurons. 2. Oxygen radicals induced the morphological changes such as the decrease of cell number and loss of neurites in these cultures. 3. EGF increased the cell viability and neurofilament in neurons damaged by oxygen radicals. From above the results, it is suggested that oxygen radicals have a cytotoxic effect on cultured DRG neurons of neonatal mouse and selective neurotrophic factors such as EGF are, effective, in blocking the neurotoxicity induced by oxygen radicals in cultured spinal DRG neurons.

Chromatographic Separation of Xanthine Derivatives on Single and Mixed-Template Imprinted Polymers

  • We developed in the present study molecular imprinted polymers (MIPs), using single templates (pentoxifylline, caffeine and theophylline) and mixed-templates (pentoxifylline-caffeine, pentoxifylline-theophylline and caffeine-theophylline). The MIPs were prepared with methacrylic acid (MAA) as the monomer, ethylene glycol dimetharylate (EGDMA) as the crosslinker and 2,2'-azobis(isobutyronitrile) (AIBN) as the initiator. The obtained polymer particles (particle size after grinding was about 25-35 ${\mu}$m) were packed into a HPLC column (3.9 mm i.d. ${\times}$ 150 mm). The selectivity and chromatographic characteristics of the MIPs were studied using acetonitrile as the mobile phase at a flow rate of 0.8 mL/min. UV detector wavelength was set at 270 nm. Different single template MIPs showed different molecular recognitions to the templates and the structurally analogues, according to the rigidity and steric hindrance of the compounds. Recognition was improved on the mixed-template MIPs as a result of the cooperation or sum effect of the templates, whereas on the pentoxifylline-theophylline imprinted polymer, the highest selectivity and affinity were obtained. Separations of the test compounds on different polymers were also investigated.

Inhibitory Effects of H.B.T. on Peroxidation of Lecithin-Liposome and Rat Liver Cell (인공막(人工膜)과 Rat의 간세포(肝細胞)를 이용(利用)한 혈부축어탕(血府逐瘀湯)의 항산화(抗酸化) 작용(作用)에 관(關)한 연구(硏究))

  • Inhibitory effects of Hyeulbuchuckeutang(H.B.T) On the peroxidations of lecithin-liposomes by active oxygens, hydroxyl radical and superoxide anion, derived from hydrogen $peroxidase-Fe^{2+}$ system and xanthine- xanthine oxidase system. These effects were similar to and stronger than those of catalase, mannitol, superoxide dismutase or $dl-{\alpha}-tocopherol$ as a scavenger or an antioxidant. H.B.T. inhibited the peroxidation of lecithin-liposome and active oxygens in concentration-dependent manner. H.B.T. also dose-dependently protected the cell death mduced by tert-butvlhydroperoxide(tBHP) and significantly increased cell viability in the rat normal liver cell (Ac2F). These results suggested that H.B.T might playa protective role in lipid peroxidation by free radicals and tBHP.

Some Physiological Activities Observed from Ethanol Extracts of Germinated Specialty Rough Rice

  • This study analyzed the some of the physiological activities of 70% ethanol extracts of specialty rough rice ('Goami2', 'Keunnunbyeo', and 'Heugkwangbyeo' rough rice cultivars) before and after germination. The physiological activities of the extracts were analyzed for their inhibition of linoleic acid peroxidation, hydrogen peroxide radical scavenging activity, superoxide dismutase-like activity, xanthine oxidase inhibitory activity, and nitrite scavenging effects. The most active rough rice cultivar was 'Heugkwangbyeo' and germinated 'Heugkwangbyeo' rough rice was more effective than ungerminated rough rice, having higher inhibition activity of linoleic acid peroxidation (1.2-fold), hydrogen peroxide radical scavenging activity (1.4-fold), superoxide dismutase-like activity (1.2-fold), and xanthine oxidase inhibitory activity (1.1-fold). The nitrite scavenging effect was increased after germination (2.1-fold) and decreased as pH increased. According to the results obtained in this study, 70% ethanol extracts from germinated 'Heugkwangbyeo' rough rice can be used for industrial applications such as the functional food materials.

Hypoglycemic and Hypolipidemic Effects of Tectorigenin and Kaikasaponin III in the Streptozotocin-Induced Diabetic Rat and their Antioxidant Activity in vitro

  • Tectorigenin and kaikasaponin III from the flowers of Pueraria thunbergiana showed potent hypoglycemic and hypolipidemic effects in the streptozotocin-induced diabetic rats. Intraperitoneal administration of these two compounds with 5 and 10 mg/kg, respectively, for seven days to streptozotocin-induced rats significantly reduced the blood glucose, total cholesterol, LDL- and VLDI-cholesterol and triglyceride levels when compared with those of control group. Glycitein in which 5-OH is unlinked and tectoridin (7-O-glycoside of tectorigenin) isolated from the flowers of P. thunbergiana did not improve hyperglycemia and hyperlipidemia. In addition, tectorigenin showed in vitro antioxidant effects on 1,1-diphenyl-B-pirylhydrazyl (DPPH) radical, xanthine-xanthine oxidase superoxide anion radical, and lipid peroxidation in rat microsomes induced by enzymatic and non-enzymatic methods. We further found that tectorigenin and kaikasaponin III protected the Vero cell line(normal monkey kidney) from injury by hydrogen peroxide. From these findings, it seems likely that the antioxidant action of tectorigenin and kaikasaponin III may alleviate the streptozotocin-induced toxicity and contribute to hypoglycemic and hypolipidemic effects.

Antioxidant and Neuroprotective Effects of Hesperidin and its Aglycone Hesperetin

  • The present study evaluated antioxidant and neuroprotective activities of hesperidin, a flavanone mainly isolated from citrus fruits, and its aglycone hesperetin using cell-free bioassay system and primary cultured rat cortical cells. Both hesperidin and hesperetin exhibited similar patterns of 1,1-diphenyl-2-picrylhydrazyl radical scavenging activities. While hesperidin was inactive, hesperetin was found to be a potent antioxidant, inhibiting lipid peroxidation initiated in rat brain homogenates by $Fe^{2+}$ and L-ascorbic acid. In consistence with these findings, hesperetin protected primary cultured cortical cells against the oxidative neuronal damage induced by $H_2O_2$ or xanthine and xanthine oxidase. In addition, it was shown to attenuate the excitotoxic neuronal damage induced by excess glutamate in the cortical cultures. When the excitotoxicity was induced by the glutamate receptor subtype-selective ligands, only the N-methyl-D-aspartic acid-induced toxicity was selectively and markedly inhibited by hesperetin. Furthermore, hesperetin protected cultured cells against the $A_{{\beta}(25-35)}-induced$ neuronal damage. Hesperidin, however, exerted minimal or no protective effects on the neuronal damage tested in this study. Taken together, these results demonstrate potent antioxidant and neuroprotective effects of hesperetin, implying its potential role in protecting neurons against various types of insults associated with many neurodegenerative diseases.

Inhibitory Effects of Xiaoshuan Zaizao Wan on Excitotoxic and Oxidative Neuronal Damage Induced in Primary Cultured Rat Cortical Cells (일차 배양한 흰쥐 대뇌피질세포의 흥분성 및 산화적 신경세포손상에 대한 소전재조환의 억제효과)

  • Xiaoshuan Zaizao Wan (XZW) has been used in China to improve hemiplegia, deviation of eye and mouth, and dysphasia due to cerebral thrombosis. To characterize pharmacological actions of XZW, we evaluated its effects on neuronal cell damage induced in primary cultured rat cortical cells by various oxidative insults, glutamate or N-methyl-D-aspartate (NMDA), and $\beta$-amyloid fragment ($A_{\beta(25-35)}$). XZW was found to inhibit the oxidative neuronal damage induced by $H_2O_2$, xanthine/xanthine oxidase, or $Fe^{2+}$/ascorbic acid. It also attenuated the excitotoxic damage induced by glutamate or NMDA. The NMDA-induced neurotoxicity was more effectively inhibited than the glutamate-induced toxicity. In addition, we found that XZW protected neurons against the $A_{\beta(25-35)}$-induced toxicity. Moreover; XZW exhibited dramatic inhibition of lipid peroxidation in rat brain homogenates and mild 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Taken together; these results demonstrate that XZW exerts neuroprotective effects against oxidative, excitotoxic, or $A_{\beta(25-35)}$-induced neuronal damage. These findings may provide pharmacological basis for its clinical usage treating the sequelae caused by cerebral thrombosis. Furthermore, XZW may exert beneficial effects on Alzheimer's disease and other oxidative stress-related neurodegenerative disorders.

The role of testosterone on nephrotoxicity of mercuric chloride (Testosterone에 의한 승홍의 신독성 유발에 의한 연구)

  • Mercuric chloride, inorganic compound, is one of the most important drugs that has been used in the field of argriculture, antisyphilitica and anticeptics, but it is not used clinically at present. We have studied the effect of testosterone on the mercuric chloride-induced nephrotoxicity. Renal lipid peroxide concentration of male rat treated with mercuric chloride was significantly increased in comparison with that of the female rat, it showed similar effects on testosterone pretreatment. Changes in renal catalase and gluta- thione peroxidase activities were not siginificantly different in testosterone-treated groups. But, renal xanthine oxidase and aldehyde oxidase activities of testrosterone-treated group given mercuric chloride significantly increased in comparison with that of the testoste- rone-treated alone. Animals treated with testosterone prior to mercuric chloride showed more severe damage on histological observations than those treated with testosterone only. Consequently, we suggest that the mercuric chloride-induced nephrotoxicity might be renal lipid peroxide generating enzyme system by testosterone.

The Effect of Doenjang (Korean Soy Paste) on the Liver Enzyme Activities of the Sarcoma-180 Cell Transplanted Mice

  • Korean traditional fermented soy paste (doenjang) prolonged the life span of Balb/c mice injected with the sarcoma-180 cells. The activities of liver enzymes, such as xanthine oxidase, aminopyrine N-demethylase, aniline hydroxylase, ${\gamma}$-glutamylcysteine synthetase, glutathione reductase and glutathione S-transferase (GST), and the contents of lipid peroxide and glutathione were determined from the sarcoma-180 cell injected mice that were treated with methanol extracts from doenjang, miso and soybean. The content of lipid peroxide and the activity of xanthine oxidase in the liver of Balb/c mice which were increased by the transplantation of the sarcoma-180 cells were decreased by treatment with the methanol extract from doenjang. But the activities of aminopyrine N-dementhylase and aniline hydroxylase were not affected by the treatment of methanol extracts from doenjang to the mice injected with the sarcoma-180 cells. The content of glutathione, the activities of glutamylcysteine synthetase, glutathione reductase and glutathione S-transferase decreased by the injection of the sarcoma-180 were recovered considerably by the treatment of the methanol extract from doenjang.
