The use of isolated wine yeasts in winemaking processes is preferable to spontaneous fermentation. Selection criteria of wine yeast strains depend also on capacity and rate of fermentation and on alcohologenic capabilities. Our studies have described the dynamics of fermentation of wine musts by some isolated wine yeast strains of Saccharomyces genus: strains 6 and 8 of S. cerevisiae var. ellipsoideus (S. ellipsoideus) and strains 5 and 7 of S. bayanus var. oviformis (S. oviformis). All have high technological properties and all are adapted for the specific pedoclimatic conditions of some areas of Sibiu viticultural region. The selected strains were used as inocula to ferment Sauvignon, Muscat Ottonel, Rose Traminer, and Pino Gris musts in controlled laboratory conditions. It was found that higher initial oxygen concentration in must is necessary to accelerate the fermentation of all the wine yeast strains studied. In order to obtain quality wines, strains with considerable fermentative capacity, high alcohologenic capabilities, and a good conversion efficiency are recommended.
In order to investigate the possible application of immobilized yeast cells in sparkling wine production instead of riddling puns by the traditional method, fermentation characteristics were tested during the sparkling wine fermentation in the bottle using immobilized yeast cells with alginate. The rates of sugar consumption and alcohol production were faster with free cells than those with immobilized cells during the fermentation. The higher concentration of yeast cells and the lower concentration of alginate in the cell immobilization resulted in the faster sugar consumption and alcohol production. It also resulted in the increase of yeast cell concentration released from immobilized beads during the fermentation. However, no differences were shown in the contents of alcohol, residual sugar and CO2 pressure after fermentation. In case concentration of yeast cells released from immobilized beads during bottle fermentation, the higher concentration of alginate had and the lower had.
청주가 가지고 있는 향기는 청주의 품질을 결정하는 요인 중의 하나이다. 청주의 향기는 발효과정 중 주로 효모에 의해서 발생하는 것으로 알려지고 있고, ethyl caproate는 향기성분 중의 하나로 esterase가 생성에 관여하고 있는 효소의 하나로 보고되었다. 반면 청주 제조에 사용되는 청주효모와 코지균은 ethyl caproate의 생성과 분해 활성을 가진 esterase를 생산하는 것으로 보고되었다. 본 연구에서는 청주 제조 중 발생하는 ethyl Caproate의 생성에 미치는 청주효모와 코지균이 가지고 있는 esterase들의 영향을 알아보기 위하여 코지균과 청주효모의 생장 중 발생하는 ethyl caproate 합성과 분해에 관련하고 있는 esterase들의 활성 변화에 대하여 알아보았다. 청주효모가 가지고 있는 ethyl caproate 합성 esterase는 caproate에 의해서 유포되고, 분해 esterase는 ethyl caproate에 의해서 유도되지만 caproate에 의해서 저해되는 것으로 나타났다. 코지균의 ethyl caproate합성과 분해 esterase활성은 균체의 성장에 비례하여 증가하였으나 ethyl caproate 생성의 기질이 되는 caproate와 ethanol이 활성에 영향을 미치지 못하는 것으로 나타났다. 또한 코지균의 배양액에서 ethyl caproate 합성 esterase의 활성이 측정됨에도 불구하고 0.01 ppm까지 ethyl caproate를 검출할 수 있는 분석조건 하에서 배지 중의 ethyl caproate는 검출되지 않았다. 본 실험의 결과를 고려한다면 청주의 제조과정 중 생성되는 ethyl caproate는 코지균의 esterase에 의해서는 거의 생성되지 않고 당화과정 후 첨가되는 청주효모가 주로 생성하는 것으로 확인되었다. 또한 청주효모의 증식과정 중에는 효모의 생장과 더불어 ethanol이 생성되어 코지균의 생장을 억제한다. 따라서 청주제조 중 생성되는 ethyl caproate는 청주효모의 esterase에 의해 생산된 것으로 사료된다.
We investigate black raspberry (Rubus occidentalis) wine made using traditional yeast (Saccharomyces cerevisiae A8, B6, GBY2, GBY3) and S. cerevisiae Fermivin (FM), which is widely used in wine manufacturing, and analyze the biogenic amine content and the volatile flavor compounds. Black raspberries were separately inoculated with yeast up to $1{\times}10^9CFU/kg$, followed by incubation at $25^{\circ}C$ for 7 days. FM produced the highest alcohol content, however the final fermentation characteristics of the wine made using four different yeasts were similar. S. cerevisiae A8 had a large biogenic amine (BA) content, specifically tryptamine, thus we excluded this yeast from fermentation. S. cerevisiae GBY3 was selected for black raspberry wine fermentation as a result of sensory evaluation. The volatile flavor compounds of two wines (S. cerevisiae GBY3 and FM) were analyzed by gas chromatography and mass spectrometry. 37 compounds in the samples were separated, and several ester compounds were identified in greater amounts in the wine made with S. cerevisiae GBY3 than in the wine made with FM. A greater amount of the major compound, ethyl benzoate, giving the sweet and fruity flavor, was identified in wine made with S. cerevisiae GBY3 than in the wine made with FM. In conclusion, S. cerevisiae GBY3 was confirmed to produce no major BAs and a better flavored wine. These results give new leads in the production of high quality wine.
참다래로 만든 'Hayward' 와인 제조에 적합한 효모 선발을 위해서 3종의 효모에 의한 발효과정에서의 물리화학적 특성 변화를 조사 분석하였다. 사용된 효모는 Saccharomyces cerevisiae Gervin No.5 strain (GVN), S. bayanus Lavin strain EC1118 (EC1118), 그리고 S. cerevisiae Red star Davis No.796 (No.796)은 와인 발효에 가장 적합하여 상업적으로 이용되고 있는 효모이다. 이 세 효모에 의해 발효된 샘플의 당도 함량은 비슷하게 감소하였으나, 알코올 생산량만 발효 첫 주에 큰 차이가 있었다. 효모중 GVN이 13.8%로 알콜농도가 가장 높았고, 반면에 No.796과 EC1118은 각각13.0%와 12.5%로 상대적으로 알콜함량이 낮았다. 관능평가에서 GVN은 높은 산과 높은 알콜함량을 가지고 있고, 강한 풍미와 쓴맛을 가진 것으로 평가되었다. No.796은 GVN과 유사하지만 낮은 알콜함량과 함께 단맛이 있었다. EC1118은 높은 풍미, 단맛과 낮은 알콜함량으로 부드러운 맛으로 평가되었다. EC1118에 의해 발효된 와인 의 총페놀과 항산화 활성은 다른 효모보다 높았다. 따라서 본 연구에서EC1118이 최적의 효모로 선발되었다. 다른 2종의 효모를 사용한 참다래 발효 와인은 각각 다른 품질 특성을 나타내었다. EC1118효모는 참다래 발효에 가장 바람직한 특성을 가지고 있었다. 이러한 특성은 주로 총페놀과 항산화 활성과 같은 건강기능성과 높은 관능적 기호도를 가지고 있었다.
This study investigated the effects of different commercial yeast strains on the characteristics of peach wine. Peach fruit was inoculated with Fermivin, K1-V1116, EC-1118, D-47 and AR2. Peach wines fermented with the D-47, K1-V1116, and AR2 strains showed faster fermentation than wines fermented with the other strains. Wine fermented with EC-1118 had the lowest titratable acidity and highest pH. The ethanol content of wines fermented with different commercial yeast strains was in the range of 13~14%. In sensory evaluation, the wine made with EC-1118 showed the highest overall scores in color and acceptability.
In order to investigate the possibility of using immobilizing yeast cells with the eliminating purpose of the ridding process in sparkling wine production by the traditional method, the changes in chemical components during and after bottle fermentation by immobilizing yeast cells with alginate were tested. The most volatile compounds, excepting some compounds, were not appreciable different in sparkling wines which obtained from various samples compared. After bottle fermentation, sparkling wine fermented with undergoing riddling process, and tested. The results showed that the taste and aroma of the sparkling wine produced with using immobilized cells were very similar to that produced with using free cells.
This study was performed to investigate the effects of adding a dual starter on the chemical and sensory characteristics of red wine made of Campbell Early grape. The yeast starter, Saccharomyces cerevisiae, and lactic acid bacteria (LAB) starter, Oenococcus oeni, were used for inoculation in the winemaking process for alcoholic fermentation and malolactic fermentation (MLF), respectively. After 200 days of incubation, the chemical compositions of yeast/LAB-added wine (YL-wine) were compared with those of no-starter-added wine (control) and yeast-added wine (Y-wine). The results show that no significant differences were observed in pH, total sugar, and alcohol content among the wine samples, but the malic acid content in YL-wine was significantly reduced, and various esters and higher alcohols were synthesized. The sensory test revealed that the addition of dual starters resulted in improved overall acceptability in wine. This study emphasizes the importance of O. oeni in addition to yeast in making Campbell Early wine.
The objective of this study was to investigate the quality characteristics of Cheongsoo wine using freeze concentration fermented with 5 kinds of yeast strains (Saccharomyces cerevisiae EJ18, EJ30, HK22, HK32 and Fermivin). We compared the characteristics, volatile flavor component and physiological activity of 5 wines. The freeze concentration can increase the sugar concentration in grape juice by reducing its water content and the alcohol content of freeze-cententrated Cheongsoo wines ranged between 15.0~15.8%. The pH of wines ranged from 2.92 to 2.94 and the total acidity and soluble solid of wines ranged from 12.40~13.27. °Brix. The major organic acid in the wines was tartaric acid (4.49~5.11 mg/mL) and malic acid (7.00~7.97 mg/mL). It contains higher functional compounds in wine fermented with yeast (EJ18, EJ30, HK22, HK32) than wine fermented with fermivin. α-glucosidase and tyrosinase inhibitory activity had the highest values (78.87% and 62.25%) in wine fermented with HK22 and EJ18 yeast, respectively. 16 volatile flavor compounds (alcohols, esters, ketones, acids, and others) were detected in the Cheongsoo wines by freeze concentration. These results provide useful information that the quality characteristics of wine developed by the freeze-concentration method using grapes cultivated in Korea.
The diversity and composition of yeast populations may greatly impact wine quality. This study investigated the yeast microbiota in two different types of wine fermentations: direct inoculation of a commercial starter versus pied de cuve method at an industrial scale. The pied de cuve fermentation entailed growth of the commercial inoculum used in the direct inoculation fermentation for further inoculation of additional fermentations. Yeast isolates were collected from different stages of wine fermentation and identified to the species level using Wallersterin Laboratory nutrient (WLN) agar followed by analysis of the 26S rDNA D1/D2 domain. Genetic characteristics of the Saccharomyces cerevisiae strains were assessed by a rapid PCR-based method, relying on the amplification of interdelta sequences. A total of 412 yeast colonies were obtained from all fermentations and eight different WL morphotypes were observed. Non-Saccharomyces yeast mainly appeared in the grape must and at the early stages of wine fermentation. S. cerevisiae was the dominant yeast species using both fermentation techniques. Seven distinguishing interdelta sequence patterns were found among S. cerevisiae strains, and the inoculated commercial starter, AWRI 796, dominated all stages in both direct inoculation and pied de cuve fermentations. This study revealed that S. cerevisiae was the dominant species and an inoculated starter could dominate fermentations with the pied de cuve method under controlled conditions.
