Effect of Wild Grape Juice on Lipid Metabolism and Antioxidaive Activity in Rat Fed High Oxidized Lipid (산화된 지방식이를 급여한 흰쥐에서의 머루즙 투여가 지질대사와 항산화에 미치는 영향)

  • This study is to find out the antioxidative effect and serum and liver lipid composition of wild grape juice in vivo. Forty 6-week-old white Sprague Dawley rats were divided into 4 groups such as normal lipid group, normal lipid group with wild grape juice, oxidized lipid (basic diet plus 10% of oxidative lipid) group and oxidized lipid group with wild grape juice, and 2ml juice was provided everyday. After 4 weeks of feeding with experimental diet each groups were examined for the antioxidant enzyme activity in blood and liver microsome and their serum and liver lipid composition. Glutanthione peroxidase activity in blood was significantly higher in oxidized lipid group with wild grape juice than in oxidized lipid group. Glutanthione peroxidase activity showed no difference depending on wild grape juice supplementation. Glutanthione peroxidase activity in liver was significantly higher in the groups with wild grape juice than in the groups supplemented only with oxidized lipid. Glutanthione reductase activity showed no difference depending on the supplementation of wild grape juice. Serum triglyceride level in the group supplemented with oxidized lipid diet and wild grape juice showed similar value to the normal lipid group and the normal lipid group with wild grape juiceoxidized fa6. Liver total lipid in the group supplemented with oxidized lipid and wild grape juice showed similar value to the normal lipid group and the group supplemented with normal lipid and wild grape juice. And it was lower than that of oxidative lipid group without juice. The liver triglyceride level in the group supplemented with normal lipid and wild grape juice was lower than that in the oxidative lipid group, but it was as low as in the group supplemented only with normal lipid.

Effects on Quality Characteristics of Pork Loin Fed with Wild Grape(Vitis amurensis Ruprecht) Wine By-product (머루주 박의 급여가 돈육 등심의 품질에 미치는 영향)

  • This study was carried out to investigate the feeding effect of wild grape wine by-products on pork qualities. The chemical composition, volatile basic nitrogen, thiobarbituric acid reactive substances and pH were not significantly different between control and wild grape pork, but the cholesterol and roast loss of wild grape polk. were lower than those of the control, and 1l1e salt soluble protein extractability of the control was lower than that of the wild grape pork. The Hunter's $a^{\ast}$ and $b^{\ast}$ value of wild grape polk. meat were higher than the control, the $a^{\ast}$ value of wild grape polk. meat was higher than the control, but the $L^{\ast}$ value of meat and fat were not significantly different between control and wild grape pork. The cohesiveness, gumminess and chewiness of control(respectively $66.2\%$ 428kg and 189g) were higher than wild grape polk. (respectively $61.4\%$ 357 kg and 154 g). The total amino acid composition of polk. were not significantly different between control and wild grape wine, The glutamic acid, leucine, arginine and aspartic acid were major amino acids in control and wild grape pork. The arachidonic acid $(C_{20:4})$, EPA $(C_{20:5})$ and DHA $(C_{22:6})$ of wild grape polk. were higher than those of the control. The taste (p<0.001), aroma (p<0.05), flavor (p<0.001), juiciness (p<0.01) and palatability (p<0.01) of wild grape polk. were higher than those of the control, but the texture between control and wild grape polk. were not significantly different.

Antioxidative Activity and Serum Lipid Composition of Wild Grape Seed Powder in Rats fed Hypercholesterolemic Diet (고콜레스테롤혈증 유발식이를 급여한 흰쥐에서 머루씨 분말첨가가 항산화활성과 혈액지질조성에 미치는 효과)

  • This study is to find out the antioxidative effect and serum lipid composition of wild grape seed powder in vivo. 20 white Sprague Dawley rats of six weeks old were divided into 2 groups and AIN-93 basic diet, high fat and cholesterol were provided. And they were examined to know how wild grape seed powder worked for antioxidative effect and serum lipid composition. For the comparing group, wild grape seed powder consisting 5% of the diet weight was provided and the quantity of protein, fat, carbohydrate, and cellulose was controlled following the analysis of the ingredients. The rats were fed for four weeks with experimental diet. Serum lipid and the antioxidant enzyme activity in blood and liver microsome were measured after 4 weeks of experiment. The results are as follows; There was no difference between the experimental groups in the initial body weight, final body weight, weight gain and FER. Food intake was higher in the group wild grape seed powder was provided than in the control group(p<0.05). Serum total cholesterol in the control group was significantly higher than that in the group wild grape seed powder was provided.(p<0.05). There was no difference serum HDL cholesterol and LDL cholesterol between the groups. Serum triglyceride showed no significant difference between the groups. In blood, glutanthione peroxidase activity was higher in the group supplemented with wild grape seed powder than in the control group. The glutathione reductase activity of blood showed no difference between the groups. In liver, the glutanthione peroxidase activity was higher in the group supplemented with wild grape seed powder than in the control group(p<0.05). Glutathione reductase activity in liver showed no difference in accordance with the supplementation of wild grape seed powder.

Quality Characteristics of Yanggeng added with Pesticide Free Wild Grape (Vitis amurensis) Juice (무농약 산머루 즙을 첨가한 양갱의 품질특성)

  • This study was performed to investigate the quality characteristics of Yanggeng added wild grape (Vitis amurensis) juice. The yanggeng was prepared with pesticide-free wild grape juice, agar, sugar and salt. The yanggeng was made with various levels (0, 50, 100, 150, 200 g of wild grape juice in yanggeng, respectively) based on the total weight of water. It was estimated on Hunter's color value, texture profile analysis, and sensory characteristics of the Yanggeng. As the content of wild grape juice increased, The lightness (L) and yellowness (b) decreased and redness (a) increased. In texture profile analysis, hardness was increased; however, springiness, cohesiveness, gumminess and chewiness decreased with increasing levels of wild grape juice. The results of the sensory evaluation indicated that the Yanggeng containing the 150 g level of wild grape juice showed the highest preference scores in terms of color, taste, texture, flavor and overall acceptance. These results show that the yanggeng containing 150 g of wild grape juice is the better.

Optimization of Fermentation Conditions for the Manufacture of Wild Grape Wine (산머루주 제조를 위한 발효조건의 최적화)

  • Yeast with excellent ferment ability was isolated and selected from wild grape to manufacture wild grape wine. Wild grape wine by SMR-3 isolated from wild grape was better than other strains in quality, such as high alcohol content and low acidity, residual sugar, organic acid and fusel oil content. Fermentation condition was optimized to manufacture wild grape wine with response surface methodology using isolated SMR-3 as an alcohol fermentation strain. As a result of culture conditions, 10.61% of alcohol content was expected under the conditions of $21.91^{\circ}C$ fermenting temperature, $21.48^{\circ}brix$ of initial sugar content, and 14.65 day of fermentation time. Residual sugar content showed the lowest value at $24.48^{\circ}C$ fermentation temperature, $12.78^{\circ}brix$ of initial sugar content, and 9.02 day fermentation time. The highest level of sensory evaluation was found at $20.23^{\circ}C$ fermentation temperature, $25.30^{\circ}brix$ of initial sugar content, and 5.94 day fermentation time. Ethyl alcohol was the main alcohol component in wild grape wine and fusel oil in wild grape wine was hardly detected; thus, the quality of wild grape wine was considered excellent. The optimal fermentation conditions of wild grape wine was superimposed by deriving a regression equation for alcohol content, fusel oil, ethyl alcohol content, and overall palatability for each variable of wild grape wine. Hence, the optimal fermentation conditions are estimated to be: fermentation temperature $24{\sim}28^{\circ}C$, initial sugar content $20{\sim}24^{\circ}brix$, and fermenting time $12{\sim}14$ days.

Quality Characteristics and Antioxidant Properties of Rice Chiffon Cakes with Wild Grape Powder (머루 분말로 대체한 쌀 시폰 케이크의 품질과 항산화 활성)

  • In this study, chiffon cakes were prepared with wild grape powder. Different amounts of the wild grape powder were tested (3%, 6%, 9%, and 12%) to determine the optimal amount. Rice chiffon cake was evaluated for quality characteristics, antioxidant properties, and sensory evaluation. The specific gravity of control batter was 0.48 g/mL, and it decreased with increasing amounts of wild grape powder, whereas viscosity showed the reverse effect. The pH of batter and cake decreased with increasing amounts of wild grape powder. The moisture content significantly increased with increasing amounts of wild grape powder, and the water activity of cakes was not significantly different between samples from 0.905~0.908. For color, lightness and yellowness deceased with increasing amounts of wild grape powder, whereas redness showed the reverse effect. For texture, hardness, fracturability, gumminess, and chewiness significantly increased with increasing amounts of wild grape powder. Total polyphenol content, DPPH radical scavenging activity, and ABTS radical scavenging activity significantly increased with increasing amounts of wild grape powder. In the sensory evaluation, the control sample showed the highest scores for color, flavor, softness, and overall acceptability. Based on these results, wild grape powder could be considered as a functional material, and 6% wild grape powder is the optimum level for manufacture of rice chiffon cake.

Effect of Dietary Supplementation of Wild Grape on the Antioxidative Potential of the Breast and Leg Meat of Broilers

  • This study investigated the effect of wild grape (Vitis coignetiae) dietary supplementation on the antioxidative potential and quality of the breast and leg meat of broilers. A total of 36 one-day-old male Cobb broiler chicks were obtained from a commercial hatchery, and randomly assigned to 9 pens with 4 birds per pen. Then, broilers were fed 3 different dietary supplementations, including 0%, 0.25%, or 0.5% wild grape, for 2 wks at the finishing period. After slaughtering, the total phenolic content, ${\alpha}$,${\alpha}$'-diphenyl-${\beta}$-picryl-hydrazyl (DPPH) radical scavenging activity, 2-thiobarbituric acid reactive substances (TBARS), and total cholesterol content of broiler breast and leg meat were measured. Higher total phenolic content was recorded in the leg meat of broilers fed the wild grape when compared with the control, while breast meat did not show any difference. Dietary supplementation of 0.25% and 0.5% wild grape significantly increased DPPH radical scavenging activity of both breast and leg meat. TBARS values of both breast and leg meat were decreased by supplementation of 0.5% wild grape during storage when compared to the control, except for the leg meat at day 7. However, there was no significant difference found in total cholesterol content in both breast and leg meat. The results indicate that the antioxidative potential of broiler meat is improved by supplementing the diet with wild grape.

  • This study investigated the fermentation characteristics of wild grape(Vitis amurensis) wine prepared with reed(Phragmites communis) root. The reed root was added to the fermentation of wild grape wine in the quantities of 0.5%, 1.0% and 2.0%. An addition of reed root during yeast fermentation of wild grape wine decreased the acidity and increased pH of fermentation culture, thus enhancing the consumption of sugar and production of ethanol. Especially, the addition of 2% reed root led to production of about 13% ethanol in 4 day of fermentation time. The sensory evaluation showed that the wild grape wine fermented with 2.0% reed root was most acceptable.

  • To develop a new functional wine, manufacture and quality characteristics of wild grape wine were investigated. Ethanol contents of wild grape wine was 11.0%, and total acidity and volatile acid content were 0.63% and 0.0402%, respectively. Antioxidant activity of the wild grape wine which is related in anti-aging functionality, was 41.6%. From this results, we concluded that the wild grape wine is a high valuable wine.

Physicochemical Analysis and Antioxidative Effects of Wild Grape (Vitis coignetiea) Juice and Its Wine (머루즙과 머루주의 이화학적 분석 및 항산화 효과)

  • Physicochemical analysis of the wild grape juice and its wine extracted with ethanol were assessed. The contents of moisture in wild grape juice and its wine were 78.7${\pm}$0.2% and 74.4${\pm}$O.4%, respectively. Total anthocyanin and flavonoid contents of wild grape juice were higher 4 times and 10 times than those of wild grape wine, respectively. Of the 17 amino acids, the glutamic acid in wild gape juice and threonine in wild grape wine were the highest, which were 43.7${\pm}$1.4mg/100 g and 14.1${\pm}$0.7 mg/100 g, respectively. Electron donating abilities of wild grape juice and its wine at concentration of 1,000 ${\mu}$g/ml were 93.1${\pm}$1.2% and 88.9${\pm}$O.8%, respectively. The nitrite scavenging abilities of wild grape juice and its wine were gradually increased by increasing levels of the extracts to 79.6${\pm}$1.27% and 72.8${\pm}$1.01% at concentration of 1,000 ${\mu}$g/ml under pH 2.5.
