  • The transgenic tobacco (Nicotiana tabacum cv. Xanthi) plants containing Bacillus subtilis protoporphyrinogen oxidase gene with cauliflower mosaic virus 35S promoter have recently been generated by using Agrobacterium-mediated gene transformation. The nontransgenic and the transgenic tobacco plants were compared with respect to responses to diphenyl ether herbicide oxyfluorfen and under various environmental conditions. Both cellular leakage and lipid peroxidation caused by oxyfluorfen were found to be less in the transgenic than in the nontransgenic plants. Growth responses of the transgenic plants under various temperature, light, and water conditions were almost the same as those of the nontransgenic plants, although the transgenic plants exhibited slightly more retarded growth under low light or saturated water condition. These results revealed that the transgenic tobacco plants containing B. subtilis protoporphyrinogen oxidase gene under cauliflower mosaic virus 35S promoter were relatively resistant to oxyfluorfen and exhibited normal growth pattern. Possible mechanism of resistance to oxyfluorfen in the transgenic plants is also discussed.

A study on Monitoring the Inner Structure of Dam Body Using High Resolution Seismic Reflection Method (고분해능 탄성파 반사법을 이용한 댐체 내부구조 모니터링 연구)

  • Defects of dam body which can be induced in seepage or leakage procedure can directly affect dam safety. Therefore, a proper inspection method should be carried out in the first place to find out their positions and sizes. After that, some reinforcement works such as grouting and the corresponding assessment could be taken in a proper way. The dam(center core type earth dam) issued in this study has been in need for intensive diagnosis and reinforcement work, because a lot of slumps similar to cracks, seepage and some boggy area have been observed on the downstream slope. High resolution seismic reflection method was performed on the crest profile twice before and after grouting work(Aug. 2001 and Nov. 2004) aimed at the dam inspection and the assessment of grouting efficiency as well. To enhance the data resolution, P-beam energy radiation technique which can reduce the surface waves and hence to reinforce the reflection events was used. Strong reflection events were recognized in the stack section before grouting work, It seems that the events would be caused by e.g. horizontal cracks with a considerable aperture. Meanwhile such strong reflection events were not observed in the section after grouting. That is, the grouting work was dear able to reinforce the defects of dam body. Hence, the section showed an well arranged picture of dam inner structure. In this sense, seismic reflection method will be a desirable technique for dam inspection and for monitoring dam inner structure as well.

THE EFFECT OF LOW-VISCOSITY RESIN SYSTEMS OM MARGINAL LEAKAGE OF COMPOSITE RESIN RESTORATIONS (Low-viscosity Resin Sysem이 복합레진 수복물의 변연누출에 미치는 영향)

  • The purpose of this study was to evaluate and compare the effectiveness of various low-viscosity resin systems used as rebonding agents to prevent microleakage at the margins of class I composite resin restorations. Seventy sound human premolars were selected for experiment. Class I cavities were prepared and each cavity was conditioned with a 37% phosphoric acid for 15 sec, rinsed with water for 15 sec, and dried with compressed air. Bonding agent(Scotchbond Multipurpose, 3M Co.) was applied and a hybrid composite resin (Z-100, 3M Co.) was placed using an incremental technic. The excess cured composite resin was carefully removed with Sof-Lex discs(3M Co.) to expose the original margins of the cavity. The following seven groups were established : group 1 was not rebonded and used as control group ; group 2 was rebonded with a Scotchbond Multipurpose(3M Co.) and finished ; group 3 was rebonded with a Fortify(BISCO) and finished ; group 4 was rebonded with a Concise white sealant(3M Co.) and finished ; group 5 was rebonded with a Concise white sealant(3M Co.) and not finished ; group 6 was rebonded with a P&F sealant(BISCO) and finished; group 7 was rebonded with a P&F sealant(BISCO) and not finished. The specimens were then subjected to 500 thermocycles between 5 & 65 with a 10 see dwell time and immersed in 2% methylene blue dye solution for 24 hours and sectioned with low-speed diamond cutter into two part under water condition. The extent of microleakage at rebonded margins was evaluated microscopically and scored for dye penetration according to the following scale : 0=no dye penetration ; 1=dye penetration to half-way along axial wall between enamel surface and DEJ ; 2=dye penetration beyond halfway along axial wall between enamel surface and DEJ ; 3=dye penetration to the full depth of DEJ or beyond DEJ. Selected samples were prepared for SEM observation to determine the depth of penetration of the rebonding agent into the marginal interface. The obtained results were as follows: 1. In the group 2 and 3, which is rebonded with a Scotchbond Multipupose and Fortify, dye penetration score were decreased significantly than that of group 1 (P<0.05), but group 4 and 6 were not statistically different from group 1(P>0.05). 2. There were significant differences between group 4, 6 and group 5, 7 when compared by dye penetration score (P<0.05). 3. In the SEM observation, Scotchbond Multipurpose and Fortify were penetrated within $30-40{\mu}m$ depth of the outermost surface. However, both sealants were failed to penetrate into the debonded interface.

Research and Development for Decontamination System of Spent Resin in Hanbit Nuclear Power Plant (한빛원전 폐수지 제염공정 개발연구)

  • When reactor coolant leaks occur due to cracks of a steam generator's tube, radioactive materials contained in the primary cooling water in nuclear power plant are forced out toward the secondary systems. At this time the secondary water purification resin in the ion exchange resin tower of the steam generator blowdown system is contaminated by the radioactivity of the leaked radioactive materials, so we pack this in special containers and store temporarily because we could not dispose it by ourselves. If steam generator tube leakage occurs, it produces contaminated spent resins annually about 5,000~7,000 liters. This may increase the amount of nuclear waste productions, a disposal working cost and a unit price of generating electricity in the plant. For this reasons, it is required to develop a decontamination process technique for reducing the radioactive level of these resins enough to handle by the self-disposal method. In this research, First, Investigated the structure and properties of the ion exchange resin used in a steam generator blowdown system. Second, Checked for a occurrence status of contaminated spent resin and a disposal technology. Third, identified the chemical characteristics of the waste radionuclides of the spent resin, and examined ionic bonding and separation mechanism of radioactive nuclear species and a spent resin. Finally, we carried out the decontamination experiment using chemicals, ultrasound, microbubbles, supercritical carbon dioxide to process these spent resin. In the case of the spent resin decontamination method using chemicals, the higher the concentration of the drug decontamination efficiency was higher. In the ultrasound method, foreign matter of the spent resin was removed and was found that the level of radioactivity is below of the MDA. In the microbubbles method, we found that the concentration of the radioactivity decreased after the experiment, so it can be used to the decontamination process of the spent resin. In supercritical carbon dioxide method, we found that it also had a high decontamination efficiency. According to the results of these experiments, almost all decontamination method had a high efficiency, but considering the amounts of the secondary waste productions and work environment of the nuclear power plant, we judged the ultrasound and supercritical carbon dioxide method are suitable for application to the plant and we established the plant applicable decontamination process system on the basis of these two methods.

  • Rainfall infiltration through the unsaturated zone is influenced by a range of factors including topography, geology, soil, rainfall intensity, temperature and vegetation; the actual infiltration varies largely in time and space. The infiltration capacity of soil is a critical factor in identifying groundwater recharge and leakage of surface water. It may differ depending on soil types and geological features of a particular basin or territory as well as on the usage of the land. This study was conducted in forest and farmland region of the mid-mountain area (EL. 50~300 m) of Jeju Island to test soil infiltration capacity of the area where rainfall contributes to groundwater. Results were analyzed using the four soil group classification methods presented by Jeong et al. (1995) and NAS (2007) to discover that the method offered by NAS (2007) is more reliable in the mid-mountain area of Jeju Island. The study compares and reviews the existing classification methods using the results of infiltration capacity tests executed on different soil groups throughout the whole region of the Jeju mid-mountain area. It is expected that this work will serve as a guideline for evaluating surface water recharge and hydraulic characteristics of Jeju Island.

Seasonal Paddy Management Options for the Safe Use of Golden Apple Snails (Pomacea canaliculata) in Eco-friendly Rice Cultivation (친환경 벼 재배지에서 왕우렁이(Pomacea canaliculata) 안전사용을 위한 시기별 논 관리요령)

  • These experiments were conducted for 3 years from 2021 to 2023 to develop a method that can be safely used to prevent the leakage of Golden apple snails (Pomacea canaliculata) from eco-friendly rice plantations. In the southern part of Jeollanam-do, after planting rice, the young golden apple snails placed in the rice field become adults around mid-July and begin spawning. These individuals can overwinter in drains that do not dry out, but individuals hatched after mid-July will not mature enough to overwinter. The size of golden apple snails overwintered in the drainage canal was more than 2.5cm in shell height. Installing a net at the inlet could block 95% of the inflow of snails, and 99% of outflow was blocked by installing an improved water trap and net at the drain. During the mid-drying period and pre-harvest drying period, a water path was created with a power paddy pottery machine. 59.5% of snails were attracted to the waterway, and it took 130 minutes to build the waterway and collect the snails. Based on these results, seasonal paddy management tips for the safe use of giant snails in rice fields were suggested.

Evaluation of the Sealing Capacity of the Supercritical CO2 by the Measurement of Its Injection Pressure into the Tuff and the Mudstone in the Janggi Basin (초임계이산화탄소(scCO2) 주입압력 측정에 의한 장기분지 응회암과 이암의 scCO2 차폐능 평가)

  • The laboratory scale experiment was performed to evaluate the sealing capacity of the capping rock such as tuff and mudstone, measuring the intial supercritical $CO_2$ ($scCO_2$) injection pressure and the $scCO_2$-water-rock reaction for 90 days. The drilling cores sampled from 800 m in depth around the Janggi basin, Korea were used for the experiment. The mineralogical changes of mudstone and tuff were measured to evaluate the geochemical stability during the $scCO_2$-water-rock reaction at $CO_2$ storage condition (100 bar and $50^{\circ}C$). The rock core was fixed in the high pressurized stainless steel cell and was saturated with distilled water at 100 bar of pore water pressure. The effluent of the cell was connected to the large tank filled with 3 L of water and 2 L of $scCO_2$ at 100 bar, simulating the subsurface injection condition. The $scCO_2$ injection pressure, which was higher than 100 bar, was controlled at the influent port of the cell until the $scCO_2$ begin to penetrate into the rock and the initial injection pressure (> 100 bar) of $scCO_2$ into the rock was measured for each rock. The mineralogical compositions of mudstones after 90 days reaction were similar to those before the reaction, suggesting that the mudstone in the Janggi basin has remained relatively stable for the $scCO_2$ involved geochemical reaction. The initial $scCO_2$ injection pressure (${\Delta}P$) of a tuff in the Janggi basin was 15 bar and the continuous $scCO_2$ injection into the tuff core occurred at higher than 20 bar of injection pressure. For the mudstone in the Janggi basin, the initial $scCO_2$ injection pressure was higher than 150 bar (10 times higher than that of the tuff). From the results, the mudstone in Janggi basin was more suitable than the tuff to shield the $scCO_2$ leakage from the reservoir rock at subsurface.

Analysis on the Characteristics of Nonpoint sources during the Precipitation in Residential Area (강우 시 주거지역에서의 비점오염원 유출특성 분석)

  • In this study, divided into small category groups for the residential area it was carried out monitoring for the runoff during precipitation. Based on the results analyzed according to the nonpoint sources Housing leakage characteristics. Analysis of the rainfall runoff and concentration of each type of exclusive detached house with apartments, in the majority of precipitation types runoff concentrations were higher in early. In the case of a difference of two points per runoff rate rainfall it was largely investigation. The average runoff is estimated loadings of BOD $101.1kg/km^2$, SS $232.2kg/km^2$, T-N $18.2kg/km^2$, T-P $2.0kg/km^2$ detached house case, if the apartment was estimated at point BOD $108.82kg/km^2$, SS $329.18kg/km^2$, T-N $57.67kg/km^2$, T-P $4.21kg/km^2$. The average EMCs is BOD BOD 6.6 mg/L, SS 12.8 mg/L, T-N 1.518 mg/L, T-P 0.099 mg/L detached house case, if the apartment was estimated at point BOD 6.3 mg/L, COD 11.2mg/L, SS 14.5 mg/L, T-N 3.1 mg/L, T-P 0.2 mg/L. The initial 30 percentage calculated based on the initial results, the total flow of 30% if the outflow of detached house showed a net percentage difference to T-P 1.04 > T-N 0.97 > BOD 0.90 > SS 0.80. The apartment area showed the percentage difference in the water quality in the order of BOD 1.49 > T-P 1.40 > SS 1.30 > T-N 0.96 per item.

Physiological Responses to Drought Stress of Seven Evergreen Hardwood Species (상록활엽수 7수종의 건조스트레스에 대한 생리적 반응)

  • This research aims to analyze and compare the drought resistance of 7 species of landscape trees commonly grown in Korea. The 7 species are: Camellia japonica, Rhaphiolepis indica, Quercus glauca, Machilus thunbergii, Daphniphyllum macropodum, Dendropanax morbifera and Cinnamomum camphora. In order to analyze their drought resistance, the samples were left without irrigation for 30 days (05/09/2016 ~ 05/10/2016), during which period their respective drought resistor, relative water content, electrolyte elution figures and proline content were measured. As the non-irrigation proceeded, C. camphora was the first to wither, followed by D. morbifera, then D. macropodum, then M. thunbergii, then Q. glauca, then R. indica then finally C. japonica. Of the 7 species, Q. glauca, C. japonica and R. indica can be considered highly drought resistant, since they survived for longer than 3 weeks without irrigation. Relative water content (RWC) plummeted dramatically after the first 15 days of non-irrigation. Whereas RWC readings of C. camphora, D. morbifera, D. macropodum and M. tunbergii dropped by 40% or more, the other 4 species reported a relatively low rate of decrease at 20% or lower. The Camellia japonica, the R. indica and Q. glauca, which were the species with relatively high drought resistance, showed low proline content and electrolyte elution figures, whereas those of C. camphora, D. macropodum, D. morbifera and M. tunbergii were higher. Analysis through the nonlinear regression analysis logistic model showed that non-irrigation proved fatal for the 7 sample species in a range of 22.7 to 37.6 days. The C. japonica, R. indica, Q. glauca and M. tunbergii demonstrated a high drought resistance of 30 days or longer, whereas C. camphora, D. morbifera and D. macropodum had a low resistance of 25 days or less to drought from lack of water. In conclusion, out of the 7 species of broad-leaved evergreen trees tested, C. japonica, R. indica and Q. glauca seem to be suitable for use as landscape trees, owing to their high drought resistance.

Expression of CsRCI2s by NaCl stress reduces water and sodium ion permeation through CsPIP2;1 in Camelina sativa L.

  • Camelina (Camelina sativa L.) is a potential bio-energy crop that has short life cycle about 90 days and contains high amount of unsaturated fatty acid which is adequate to bio-diesel production. Enhancing environmental stress tolerance is a main issue to increase not only crop productivity but also big mass production. CsRCI2s (Rare Cold Inducible 2) are cold and salt stress related protein that localized at plasma membrane (PM) and assume to be membrane potential regulation factor. These proteins can be divide into C-terminal tail (CsRCI2D/E/F/G) or no-tail group (CsRCI2A/B/C/H). However, function of CsRCI2s are less understood. In this study, physiological responses and functional characterization of CsRCI2s of Camelina under salt stress were analyzed. Full-length CsRCI2s (A/B/E/F) and CsPIP2;1 sequences were confirmed from Camelina genome browser. Physiological investigations were carried out using one- or four-week-old Camelina under NaCl stress with dose and time dependent manner. Transcriptional changes of CsRCI2A/B/E/F and CsPIP2;1 were determined using qRT-PCR in one-week-old Camelina seedlings treated with NaCl. Translational changes of CsRCI2E and CsPIP2;1 were confirmed with western-blot using the antibodies. Water transport activity and membrane potential measurement were observed by cRNA injected Xenopus laevis oocyte. As results, root growth rate and physiological parameters such as stomatal conductance, chlorophyll fluorescence, and electrolyte leakage showed significant inhibition in 100 and 150 mM NaCl. Transcriptional level of CsPIP2;1 did not changed but CsRCI2s were significantly increased by NaCl concentration, however, no-tail type CsRCI2A and CsRCI2B increased earlier than tail type CsRCI2E and CsRCI2F. Translational changes of CsPIP2;1 was constitutively maintained under NaCl stress. But, accumulation of CsRCI2E significantly increased by NaCl stress. CsPIP2;1 and CsRCI2A/B/E/F co-expressed Xenopus laevis oocyte showed decreased water transport activity as 61.84, 60.30, 62.91 and 76.51 % at CsRCI2A, CsRCI2B, CsRCI2E and CsRCI2F co-expression when compare with single expression of CsPIP2;1, respectively. Moreover, oocyte membrane potential was significantly hyperpolarized by co-expression of CsRCI2s. However, higher hyperpolarized level was observed in tail-type CsRCI2E and CsRCI2F than others, especially, CsRCI2E showed highest level. It means transport of $Na^+$ ion into cell is negatively regulated by expression of CsRCI2s, and, function of C-terminal tail is might be related with $Na^+$ ion influx. In conclusion, accumulation of NaCl-induced CsRCI2 proteins are related with $Na^+$ ion exclusion and prevent water loss by CsPIP2;1 under NaCl stress.
