  • Background: The value of 5-aminolevulinic acid (ALA) in fluorescence detection of peritoneal metastases and the underlying mechanisms were evaluated in patients with peritoneal surface malignancies. Materials and Methods: Oral 5-ALA was administered at a concentration of 20 mg/kg body weight with 50 ml of water 2 hours prior to surgery (n=115). The diagnostic value of 5-ALA based fluorescence production was evaluated following white light inspection during prior to cytoreductive surgery and hyperthermic intraperitoneal chemotherapy. Then, peptide transporter PEPT1 (ALA influx transporter) and ATP-binding cassette transporter ABCG2 (porphyrin efflux transporter) gene expression was determined with quantitative real time (qRT)-PCR and pathological diagnoses confirmed for all tissue samples. Results: The 5-ALA based photodynamic detection rate was 17% for appendiceal mucinous neoplasms, 54% for colorectal cancers, 33% for gastric cancers, 67% for diffuse malign peritoneal mesotheliomas, and 89% for epithelial ovarian cancer of peritoneal metastases. 5-ALA was detected in all cases of peritoneal metastases originating from cholangiocarcinomas whereas it was not able to detect any in granulosa cell and gastrointestinal stromal tumor cases. Furthermore, PEPT1 was overexpressed whereas ABCG2 expression was downregulated in tumors detected with fluorescence. Conclusions: 5-ALA provided 100% specificity and high sensitivity to detect peritoneal metastases in subgroups of patients with peritoneal surface mailgnancies. ALA influx transporter PEPT1 and porphyrin efflux transporter ABCG2 genes are important in tumor specific 5-ALA induced fluorescence in vivo. Further studies should clarify diagnostic utility of 5-ALA in peritoneal surface malignancies.

  • In this study, two hypotheses were examined to preliminarily verify for the vegetation recruitment and establishment on riparian bars in unregulated rivers; hydrologic regime change and nutrients influx into streams. In order to preliminarily verify the first hypothesis, precipitation patterns were analyzed during a period from March through to May when reeds, the most common riparian vegetation in Korea, germinate and start to grow in riparian areas. The results show that during the last 35 years, the total precipitation during the three-month period decreased by about 15 %, while the total annual one increased by about 15% in Korea. In order to verify the second hypothesis, a preliminary experiment was conducted with a set of two-vegetative flumes for one year. In this experiment, a stream flow with reeds on the riparian sand bars was simulated with a flume with reeds planted on the sand bed and water with a concentration of 3.5 mg of N flowing in the flume for four hours. For comparison, clean water was circulated in the same way in another flume for simulating a stream flow without N. The experimental results show that the growth rate of reeds in N-mixed flow exceeds that in clean water flow by about 30%. The above two results could explain the phenomenon of change in unregulated rivers from white river to green river in Korea, although they were obtained through limited extents of analysis and experiment.

  • The objective of the present study is to assess the contribution of bulk flow to the regulatory mechanism of amniotic fluid volume and its ionic concentration in the membranes surrounding the amniotic fluid. For quantitative assessment, we prepared 4 kinds of artificial amniotic fIuids (isotonic isovolumetric, hypotonic isovolumetric, isotonic hypervolumetric and hypotonic hypervolumetric ones) by replacing 70% of amniotic fluid of pregnant rabbits with water or normal Tyrode solutions. Isoosmotic saline of 0.5 ml volume containing 0.05% Censored and 15 mM/l LiCl was administered initially into amniotic sacs of all subject animals. Samples of amniotic fluid were collected in after 30 and 90 minute intervals; the concentrations of Censored, $Na^+\;and\;Li^+$ were determined and compared. Followings are the results obtained. 1. from isovolumetric and increased Congcord group, we couldn't find significant change in $Li^+\;and\;Na^+$ concentration in isotonic amniotic fluid. However, $Na^+$ concentration increased significantly as well as a striking increase in Censored concentration in hypotonic amniotic fluid. 2. In isovoIumetric and decreased Censored group, the rate of $[Li^+]$ decrement and the rate of $[Na^+]$ increment were much higher in hypotonic amniotic fluid than in isotonic. 3. In hypervolumetric and increased Censored group, the rate of $Na^+$ efflux increased proportionately with the increment of Censored concentration up to 0.98, which was higher than the rate of $Li^+$ efflux in isotonic amniotic fluid. However, the increment of $Na^+$ concentration was rather related with the initial $Na^+$ concentration in hypotonic amniotic fluid, showing inverse relationship. $Li^+$ concentration increased only when there was a marked increase in Censored concentration and approached near a maximum value or 1. 4. For hypervolumetric and decreased Censored group, the observations were identical to isovolumetric and decreased Censored group. From these results the following conclusions could be made: 1) There is no net movement of water or monovalent cations across the membranes surrounding amniotic fIuid in isotonic isovolumetric condition. In contrast, there is a net efflux of amniotic fluid by osmotic bulk flow, resulting in elevation of $Na^+$ concentration in hypotonic isovolumetric condition. 2) In hypervolumetric conditions, there is a massive efflux of amniotic fluid or solvent drag through the surrounding membranes by fiItrative bulk flow, where the rate of $Na^+$ efflux has a linear relationship with that of water efflux. This is assumed to be carried out through enlarged and newly opened intercellular spaces resulting from increased intraamniotic pressure. 3) Once increasing intraamniotic pressure reaches a point allowing $Li^+$ to pass through during osmotic bulk flow in hypotonic amniotic fIuid, $Na^+$ influx seems to occur by diffusion simultaneously or immediately thereafter, too.

  • Nonylphenol is an endocrine-disrupting chemical that is the degradation product of the nonionic surfactants nonylphenol ethoxylates. To understand the contamination and behavioral characteristics of nonylphenol, we measured the nonylphenol concentrations in surface water in the lower reaches of the Suyeong River, Korea. The results were used to estimate the material balance. The target area was divided into three regions to estimate the material balance of nonylphenol. In region 1, in flux of the dissolved nonylphenol was 282.3 g/day and the nonylphenol influx in particulate suspended solids was 1,582.8 g/day. The dissolved nonylphenol outflow discharged toward region 2 was 192.5 g/day, while the adsorption to particulate suspended solids was 89.8 g/day. Within the particulate suspended solids, the outflow to region 2 was 1,250.0 g/day, while the estimated amount settling in the sediments was 422.7 g/day. The adsorption of dissolved nonylphenol to the particulate suspended solids in regions 1 and 2 was 31.8% and 54.9%, respectively. In region 3, the desorption rate was 8.8%.

  • Road runoff, one of non-point source pollutants, contains various heavy metals, most of which flow into discharge waters without being treated. The mechanism of removing the heavy metals in water is similar to that of removing micro-particles. Therefore, it is considered that it is possible to remove a lot of the heavy metals contained in the road runoff by filtering or absorbing them. In this paper, performed has been a basic study on the characteristics of UNFS (Up Flow Non-Point Source Filtering System) using carbide pellet and zeolite pellet as double-layer filtering mediums to treat the road runoff. The removal rate with filtering and absorption time has been shown as follows: 29.0% for Cr; 27.8% for Cd; 25.7% for Fe; 25.4% for Co; 21.2% for Pb; ]9.6% for Zn; 18.2% for Al; 17.0% for Mn; 11.3% for Ni; 7.5% for Cu. The overall removal rate according to influx change has been shown to be approximately 30%, and the load of heavy metals flowing out in initial precipitation could be reduced by using carbide as a recycling filtering medium. When the removal as coarse particles settle is added up, it is expected that UNFS will result in a higher removal rate.

  • Changwon City first constructed riverbank filtration plants in Book-Myeon and Daesan-Myeon in Korea in the year 2001. This study evaluated hydrogeological characteristics and groundwater flow simulation between the Nakdong River and the fluvial aquifers adjacent to the river in Book-Myeon, Changwon City. The groundwater simulation calculated the influx rate from the Nakdong River and the fluvial aquifers to pumping wells through the riverbank filtration system. The groundwater flow model utilized drilling, grain size analysis, pumping test, groundwater level measurements, river water discharge and rainfall data. Hydraulic heads calculated by the steady-state model closely matched measured heads in pumping and observation wells. According to the transient flow model, using a total pumping amount of 14,000 $m^3$/day, the flux into the pumping wells from the Nakdong River accounts for 8,390 $m^3$/day (60%), 590 $m^3$/day (4%) is from the aquifer in the rectilinea. direction to the Nakdong River, and 5,020 $m^3$/day (36%) is from the aquifer in the parallel direction to the Nakdong River. The particle tracking analysis shows that a particle from the Nakdong River moves toward the pumping wells at a rate of about 1.85 m/day and a particle from the aquifer moves toward the pumping wells at a rate of about 0.75 m/day. This study contributes to surface water/groundwater management modeling, and helps in understanding, how seasonal change affects pumping rates, water quality, and natural recharge.

  • Background: Rapid industrialization has caused various impacts on nature, including heavy metal pollution. However, the impacts of industrialization vary depending on the types of industrializing activity and surrounding environment. South Korea is a proper region because the rapid socio-economical changes have been occurred since the late nineteenth century. Therefore, in this study, we estimate the anthropogenic impacts on an ecosystem from a sediment core of Yonghwasil-mot, an irrigation reservoir on the western coast of Korea, in terms of heavy metal concentrations, nutrient influx, and pollen composition. Results: The sediment accumulation rate (SAR) determined by 210Pb geochronology showed two abrupt peaks in the 1930s and 1950s, presumably because of smelting activity and the Korean War, respectively. The following gradual increase in SAR may reflect the urbanization of recent decades. The average concentrations of arsenic (As), copper (Cu), and lead (Pb) during the twentieth century were > 48% compared to those before the nineteenth century, supporting the influence of smelting activity. However, at the beginning of the twenty-first century, the As, Cu, and Pb concentrations decreased by 19% compared to levels in the twentieth century, which is coincident with the closure of the smelter in 1989 and government policy banning leaded gasoline since 1993. The pollen assemblage and nutrient input records exhibit changes in vegetation cover and water level of the reservoir corresponding to anthropogenic deforestation and reforestation, as well as to land-use alteration. Conclusions: Our results show that the rapid socio-economic development since the twentieth century clearly affected the vegetation cover, land use, and metal pollutions.

  • Aconiti tuber butanol fraction, which is isolated from the chloroform insoluble and water soluble extract of Aconitum volubile, has been recently known to have a potent positive inotropic effect in the isolated cardiac muscle preparations of various animals. The positive inotropic mechanism of Aconiti tuber butanol fraction, in relation with the external calcium, was studied using the isolated cat papillary muscle. The positive inotropic effect was dependent on the calcium concentration in the nutrient medium, and a synergistic relation could be demonstrated between Aconiti tuber butanol fraction and the external calcium. The inotropic effect of $10^{-4}g/ml$ of Aconiti tuber butanol fraction was equivalent to that of 0.06mM of calcium in the medium. After the treatment with a calcium influx inhibitor, Verapamil$(2{\pm}10^{-7}-10^{-6}M)$, the contractile force of the papillary muscle was markedly inhibited. In these preparations, Aconiti tuber butanol fraction restored the decreased contractility in a dose-dependent manner. It was suggested that the positive inotropic effect of Aconiti tuber butanol fraction might be related with the stimulating action on the calcium influx through the slow inward calcium channels in the cardiac cell membrane. In contrast with digitalis cardiac glycoside, Aconiti tuber butanol fraction infused intravenously into the anesthetized rabbit decreased the systemic arterial blood pressure and increased the carotid blood flow, but produced no prominent changes in the heart rate.

  • The most serious problem in oil stockpiles with artificial underground cavern is maintaining the stability of ground water system. In order to understand the ground water system around K-1 site, we determined the regional flow direction and level distribution of groundwater, and investigated the major geologic factors influencing their flow system. Reactivated surface along the contact between granite and gneiss, and fractures and faults along the long acidic dyke may contribute as important pathways for groundwater flow. Within K-1 site, groundwater level fluctuation is closely related to the rainfall events and injection from surface or influx water. In this project, the effect of groundwater pumping from the southern wells was examined. Based on equations relating water level drawdown to pumping rate at those wells, their pumped outflow of groundwater ranged from $80\;m^3$/day to less than $250\;m^3$/day. The modeling results with MODFLOW imply that the previous groundwater pumping at distance of 1.2 km may not affect the groundwater level variations of the K-1 site. However, continuous pumping work at quantity over $250\;m^3$/day in this area will be able to affect the groundwater system of the K-1 site, particularly along the acidic dyke.
