The purpose of this research was to isolate an active component on immunoytes from 70% MeOH extract of Codonopsis lanceolatae Radix(CLE). CLE was fractionated successively with hexane, methylene chloride, n-butyl alcohol and water, and then the water fraction was separated with molecularporous membrane tubing(m.w. 3,500). Each fraction(50mg/kg) was administered p.o. once a day for 7 days in BALB/c mice respectively. None of these fractions affected the apoptosis and mitochondrial transmembrane potential in thymocyte. Hexane and methylene chloride fractions decreased CD4$^{[-10]}$ CD8$^{+}$ single-positive cells, and the water fraction enhanced CD4$^{+}$ CD8$^{[-10]}$ single-positive cells in thymocyte. The proliferation of thymocytes was decreased by the fraction hexane, but was enhanced by the water fraction. Hexane, methylene chloride and butyl alcohol fractions suppressed the production of nitric oxide, which was not affected by the water fraction. Hexane and butyl alcohol fraction suppressed the phagocytic activity, but water fraction enhanced it. The components(m.w. 3,500 above) separated from the water fraction enhanced the proliferation of thymocyte, the population of CD4$^{+}$ CDB$^{[-10]}$ single-positive cells, and phagocytic activity in macrophage. These results suggest that the stimulative components of proliferation, TH population and phagorytc activity is in the water fraction, and the molecular weight is 3,500 above. The water fraction exhibiting anticancer activity was prepared from 70% methanol extract of Artemisis argyi by stepwise solvent partioning. This water fraction(5 $\mu$g/ml concentration) showed a considerable cytotoxicity against leukemic L1210 cells with a maximal value of 92% for 3 days culture. Contrastingly to such substantial anticancer activities the identical fraction showed far low toxicity against normal lymphocytes than chloroform fraction of Artemisia argyi mitomycine and 5-fluorouracil at every concentration ranging 0.01$\mu$g/ml~10.00$\mu$g/ml. The cytotoxicity displayed against L1210 cells by the water fraction of Artemisia was found to be proportinal to the decrease of viability of L1210 cells. On the other hand, $O_2$ion generation in L1210 cells appeared to be elevated in accordance to cytotoxicity by the water fraction with concurrent increases of superoxide dismuatse (SOD) and glutathione peroxidase (GPx) which are responsible for the conversion of $O_2$ ion and $H_2O$$_2$ respectively These findings taken together indicate that the death of L1210 cells by the water fraction of Auemisia atgyi, may be induced at least in part by the detrimental action of reactive oxygen species (ROS) including $O_2$- in spite of substantial extorts of SOD and GPx to overcome the attack of ROS.
A comprehensive fractionation technique was applied to a set of water samples obtained along drinking water treatment process with ozonation and biological activated carbon (BAC) process to obtain detailed profiles of dissolved organic matter (DOM) and to evaluate the haloacetic acid (HAA) formation potentials of these DOM fractions. The results indicated that coagulation-sedimentation-sand filtration treatment showed limited ability to remove hydrophilic fraction (28%), while removal of hydrophobic and transphilic fraction were 57% and 40%, respectively. And ozonation and BAC treatment showed limited ability to remove hydrophobic fractions (6%), while removal of hydrophilic and transphilic fractions were 25% and 18%. The haloacetic acid formation potential (HAAFP)/dissolved organic carbon (DOC) of hydrophilic fraction was the highest along the treatment train and HAAFP/DOC of hydrophilic fraction was higher than hydrophobic and transphilic fraction as 23%~30%, because of better removal for hydrophobic fraction both in concentration and reactivity.
The present study examined effects of a water-soluble fraction from mulberry leaves (ML water fraction) on the circulatory and autonomic nervous systems, which were compared with those of acetylcholine (ACh) used as a reference drug in order to elucidate its mechanism of action. Intravenous administration of ACh or a ML water fraction produced temporary depressor and tachycardiac responses in a dose-dependent manner in unrestrained, conscious Sprague-Dawley rats. The systemic hemodynamic effects of ACh and a ML water fraction were almost completely blocked by pretreatment with atropine, a muscarinic antagonist. The depressor responses to ACh and a ML water fraction were slightly enhanced and prolonged by pretreatment with neostigmine, an anticholinesterase, whereas the tachycardiac responses were remarkably blocked by pretreatment with pentolinium, a ganglionic blocking agent. In vitro experiments using the ileum isolated from rats showed that ACh and a ML water fraction increased ileal contractility in a dose-dependent manner. The increases in ileal contractility were also completely abolished in the presence of atropine. Finally, the specific binding of [$^3H$]quinuclidinyl benzilate, a muscarinic antagonist, to rat cortical synaptic membranes was inhibited by a ML water fraction in a concentration-dependent manner with an IC$_{50}$ value of 9.5 mg/ml. The results suggest that the effects of a ML water fraction are mediated through direct stimulation of muscarinic cholinergic receptors by unknown cholinomimetic substance(s) contained in that fraction.
Objective ; In order to study the effect of Acanthopanax senticosus(AS) aqua-acupuncture manufactured with water soluble fraction and ether soluble fraction on the streptozotocin induced diabetic rats Methods ; The fractions of AS aqua-acupuncture were carried out on corresponding bilateral loci of Bisu(BL20) everyday for 4 weeks. The experimental animals were divided into control group and AS groups(AS water fraction group and AS ether fraction group). Thereafter the levels of serum glucose, total choleterol, HDL, triglyceride, AST, ALT, creatinine, BUN, urinary albumin excretion, index of kidney hypertrophy, heart rate, mean blood pressure and fibronectin in glomeruli and tubular cells were measured. Results ; The increased serum total cholestrol, triglyceride levels, HDL and urinary albumin excretion, the index of kidney hypertrophy, the mean blood pressure and the amount of fibronectin in glomeruli and tubular cells were significantly decreased in the AS groups, showing more significant decrease in the AS water fraction group as compared with the control group. In the serum ALT, AST, creatinine and BUN levels, there were no significant changes in the AS groups as compared with the control group. Conclusion ; According to the above results, it reveals that Acanthopanax senticosus water soluble and ether soluble fraction have the antidiabetic effect, the antilipidemic effect and the inhibitory effect of renal damage. Also, the results showed that Acanthopanax senticosus water soluble fraction is more effective than ether soluble fraction.
Mugwort was successively fractionated with n-hexane, chloroform, ethyl acetate, n-butanol and water and the fractions were evaluated by their growth-promoting activites for Bifidobacterium sp. in vitro experiments. The growths of Bifidobacterium adolescentis, B. bifidum, B. infantis and B.longum were enhanced with the addition of the water fraction, while the fractions of chloroform and ethylacetate inhibited Clostridium perfringens. When the wate fraction was added to media at a concentration of 0.01-0.5%(w/v), the growhts of Bifidobacterium sp. were increased according to the concentration of water fraction used. The water fraction stimulated also the growth of lactobacillus acidophillus, whereas those of E. coli and Enterococcus faecalis were not affected. The growth-promoting activity of water fraction was stable at the range of pH 2 to pH 10 and kept in thermal treatment at 10$0^{\circ}C$ for 30 minutes.
In this study, we investigated the biological activities of Carthamus tinctorius including antioxidative, fibrinolytic, thrombin inhibitory, ${\alpha}$-glucosidase inhibitory and collagenase inhibitory activities. Carthamus tinctorius, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The hot water extract showed high antioxidative activity and thrombin inhibitory activity at 90.17% and 97.10% respectively. In the fraction activity tests, chloroform fraction showed the highest antioxidative activity at 81.85%. The fibrinolytic activity was strong only in the butanol fraction at 0.70 plasmin units/ml. The thrombin inhibitory activities of hexane, ethyl acetate and butanol fractions were 97.35%, 86.74% and 93.18% respectively. In collagenase inhibitory activity test, hexane fraction showed the highest activity at 87.78%. In conclusion, the hot water extract and solvent fractions of Carthamus tinctorius L can be used as a material for the development of biofunctional tea and foods respectively.
This study was carried out to investigate the changes of yield, total phenolics, saponin content and composition, antimicrobial, and antioxidant activities of various fractions of fine ginseng root (Panax ginseng C.A. Mayer) by maceration method in the order of increasing polarity (hexane, chloroform, ethyl acetate, butanol, and water). Butanol fraction showed the highest total saponin content compare to other fractions. Hexane fraction could harvest significantly high ginsenoside Rg2, Rg1, and Rf (p<0.05). And the contents of ginsenoside Rh1, Rg3, and Rg1 showed relatively higher in the fraction of ethyl acetate than other fractions. The system of hexane-chloroform-ethyl aceate-butanol showed relatively high content of ginsenoside Re, Rd, Rc, Rb3, and Rb1. However, the last fraction of water still remained lots of Rb2 content. The fraction of water was the highest phenolics. The 1,1-diphenyl-2-picryhydrazil, superoxide, and hydroxyl radical scavenging activity of water fraction was higher than the other fractions. In antimicrobial activity, the fraction of hexane showed relatively high antimicrobial activity against Pseudomonas aeruginosa, Salmonella typhimurium, Staphylococcus aureus, Bacillus cereus, and Escherichia coli. And the fractions of the chloroform and ethyl acetate showed higher antimicrobial activities than the other samples in against P. aeruginosa and S. typhimurium.
In this study, S. glauca (Bunge) Bunge extract was testified total polyphenol contents, electron donating ability on DPPH, and scavenging ability of superoxide anion radical and hydrogen peroxide. Total polyphenol contents of S. glauca (Bunge) Bunge extract were 7,053.30±915.93 ㎍ GAE/mL. In the fractions, fraction by ethanol was the highest content of 10,973.30±1,000.24 ㎍ GAE/mL. Fractions of ethyl acetate and water were 2,386.70±166.53 ㎍ GAE/mL, 2,413.30±41.63 ㎍ GAE/mL respectively. It was shown that total content of polyphenol according to solvent was significant relation at p<0.05. In the experiment of the electron donating ability, 70% ethanol extract and methylene chloride fraction were -246.15±24.17%, -254.01± 16.54% respectively. In case of ethyl acetate fraction, it was the highest electron donating ability to DPPH radical, 39.06±0.34%, and then water fraction of 36.71±5.55%, ethanol fraction of 29.77±2.57%, gradually. Electron donating abilities revealed significant difference (p<0.05) between the solvents. The superoxide radical scavenging ability of standard material was 0.029±0.0011; 70% ethanol extract of 0.022±0.00052, methylene chloride fraction of 0.027±0.00031, ethyl acetate fraction of 0.024±0.0011, ethanol fraction of 0.021±0.00024 and water fraction of 0.024±0.00019. Hydrogen peroxide scavenging ability of 70% ethanol extract was -0.0029±0.00040 and the others were as follow; methylene chloride fraction (-0.0042±0.00058), ethyl acetate fraction (-0.003± 0.0041), ethanol fraction (-0.0029±0.0015) and water fraction (-0.0028±0.00090).
The phagocytic activity of murine peritoneal macrophages was determined by lucigenin chemiluminescence with luminometer and engulfment of fluorescein-conjugated E. coli particles. 70% MeOH extract of Lithospermi Radix was fractionated successively with hexane, methylene chloride, n-BuOH and water. The water fraction (m.w. 500 to 1,000) enhanced the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles in murine peritoneal macrophages. The water fraction suppressed the production of nitric oxide in the macrophages. These results suggest that an active fraction of phagocytosis in Lithos-permi Radix is the water fraction and the molecular weight is 500 to 1,000.
