Antioxidant Activities of Extracts from Different Parts of Sasa borealis (조릿대의 부위별 추출물의 항산화 활성)

  • Objectives : This study was performed to investigate the antioxidant activity of water and ethanol extracts from Sasa borealis leaves, stems and roots. Methods : Sasa borealis leaves, stems and roots extract were prepared using water and ethanol. The total polyphenol and total flavonoid contents were analyzed. 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity, SOD like activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), hydroxyl radical scavenging activity, and Nitrite scavenging activity assays were carried out to determine the antioxidant activities. Results : The antioxidant activities of the Sasa borealis appeared higher in ethanol extract than water extracts. Total polyphenol and total flavonoids contents in ethanol extracts of leaves were $24.6mg/m{\ell}$ and $14.3mg/m{\ell}$, respectively, which were much higher than those of any other parts. SOD like activity was 70% ethanol extract of the leaves was highest with 15.68%. Electron donating ability was 70% ethanol extract of the leaves had the highest 59.07%. It exhibited high electron donating ability than BHT(45.68%). Nitrite scavenging activity of 70% ethanol extract was higher than the water extract at pH 2.5 and pH 4.2. Nitrite scavenging activity was 70% ethanol extract of the leaves was the highest 75.2%. Hydroxyl radical scavenging activity was 70% ethanol extract of the leaves was highest with 16.16%, showed very low activity than BHT(61.56%). Conclusions : These results suggest that 70% ethanol extracts from Sasa borealis leaves, exhibited higher antioxidant activities than those of root and stem, and can be potentially used as proper natural antioxidants.

Selective Solvation and Reasonable Solvation Number of Some Univalent Ions in Water-Ethanol Systems (물-에탄올 混合溶媒에서 몇가지 1가 이온들의 選擇的 溶媒和와 妥當한 溶媒和數)

  • In water-ethanol systems, the limiting equivalent conductances of electrolytes were obtained using conductometric method. Using TATB method, the limiting equivalent ionic conductances of Li+, Na+, K+, Cl-, and Br- ions were also obtained. The effective radii of corresponding ions were determined using Nightingale method. From the volume of the solvation shell, the four solvation numbers were suggested. The reasonable solvation numbers (hH2O+hO) were estimated by comparing the values obtained by from the various suggested methods. The isosolvation point of ion in water-ethanol estimated was found to be larger than that of in water-methanol. This result agree with ET (solvent polarity) values of solvents. From the reasonable solvation numbers of ions in water-ethanol, the selective solvents of corresponding ions in water-ethanol were obtained.

Anti-obesity Effects of Water and Ethanol Extracts of Black Ginseng (흑삼의 열수 및 에탄올 추출물의 항비만 효과)

  • Black ginseng was made by steaming raw white ginseng nine times at $100^{\circ}C$ for 2 h and drying. We then performed pilot experiments using the nine black ginseng extracts for different steaming and drying times to determine their anti-obesity effects. Two ginseng extracts, steaming and drying five times (FSFD) and steaming and drying nine times (NSND), prepared in water or ethanol solution decreased lipid accumulation of 3T3-L1 cells. FSFD in water and ethanol extracts showed higher levels of ginsenosides, in particular, Rh1, Rg2, and Rb1 than NSND, and levels of the three ginsenosides were higher in ethanol extracts than in water extracts. Treatment with FSFD and/or NSND in ethanol extracts significantly regulated $PPAR{\gamma}$, C/$EBP{\alpha}$ and AMPK phosphorylation in 3T3-L1 cells. We verified doubling time of stem cells from both abdominal fat and subcutaneous fat after FSFD and NSND in ethanol and water extracts were added. Although addition of FSFD and NSFD in water extracts had no effects on proliferation, ethanol extracts with FSFD and NSND increased doubling time of stem cells in subcutaneous fat. FSFD and NSND in ethanol extracts more effectively reduced adipogenesis compared to those in water extracts. FSFD in ethanol extracts promoted secretion of anti-inflammatory cytokine such as IL-10 and depressed MCP-1 infiltration in 3T3-L1 preadipocytes co-cultured with RAW264.7 cells. We concluded that FSFD and NSND ethanol extracts may be developed as a functional food for its anti-obesity effect, but anti-inflammatory effect was shown in ethanol extracted FSFD rather than in NSND.

Effect of Ethanol Concentration on Extraction of Vlolatile Components in Cinnamon (에탄올의 농도가 계피가 향기성분 용출에 미치는 영향)

  • In order to select the optimum ethanol concentration for extraction of volatile components in cinnamon, the dried cinnamon was extracted with water and 30∼90% ethanol. The volatile components of cinnamon extracts were isolated by the simultaneous distillation extraction method using Likens and Nickerson's extraction apparatus, and analyzed by GC-MS. In cinnamon bark powder 45 components were detected and 21 components were identified. The major component of cinnamon bark powder was cinnamic aldehyde. In water extract of cinnamon, volatile components were not extracted sufficiently. The volatile components of cinnamon were increased with the increment of ethanol concentraction upto 70%. The volatile component of 70% ethanol extract showed similar pattern and amount to cinnamon bark powder. But in 90% ethanol extracts, the number and amount of volatile component were reduced. The above data suggested that 70% ethanol was the most effective solvent for volatile components extraction of cinnamon.

Stoichiometric Effects. Correlation of the Rates of Solvolysis of Isopropenyl Chloroformate

  • Solvolysis rates of isopropenyl chloroformate (3) in water, $D_2O$, $CH_3OD$ and in aqueous methanol, ethanol, 2,2,2-trifluoroethanol (TFE), acetone, 1,4-dioxane as well as TFE-ethanol at 10 ${^{\circ}C}$ are reported. Additional kinetic data for pure water, pure ethanol and 80%(w/w) 2,2,2-trifuoroethanol (T)-water (W) at various temperatures are also reported. These rates show the phenomena of maximum rates in specific solvents (30% (v/v) methanol-water and 20% (v/v) ethanol-water) and, variations in relative rates are small in aqueous alcohols. The kinetic data are analyzed in terms of GW correlations, steric effect, kinetic solvent isotope effects (KSIE), and a third order model based on general base catalysis (GBC). Solvolyses based on predominately stoichiometric solvation effect relative to medium solvation are proceeding in 3 and the results are remarkably similar to those for p-nitrobenzoyl chloride (4) in mechanism and reactivity.

Biological Activities of Extracts from Cornus kousa Fruit (산딸나무(Cornus kousa) 열매 추출물의 생리활성)

  • The extracted phenolic compounds from Cornus kousa fruit for biological activities as functional resources were examined. The phenolic compounds which were extracted with water and 40% ethanol from Cornus kousa fruit were $7.04{\pm}0.27$ and $4.47{\pm}0.18mg/g$, respectively. The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of water and ethanol extracts were 84% and 86% at $50{\mu}g/mL$phenolics, respectively. The 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization activity of water and ethanol extracts were 84 and 95% at $100{\mu}g/mL$ phenolics, respectively. Antioxidant protection factor in water and ethanol extracts at $50{\mu}g/mL$ phenolics were 1.93 and 1.82 PF, respectively. Thiobarbituric acid reactive substance were 69% in water extracts and 89% in ethanol extracts at $150{\mu}g/mL$ phenolics. The inhibition activity on xanthine oxidase in water and ethanol extracts was 34 and 60%, respectively. The inhibition activity on ${\alpha}$-glucosidase was 29% in water extracts and 87% in ethanol extracts. The tyrosinase inhibitory activity was 19% in ethanol extracts. The collagenase inhibition activity of anti-wrinkle effect showed an excellent wrinkle improvement effect as 53% in water extracts and 77% in ethanol extracts at $200{\mu}g/mL$ phenolics. The hyaluronidase inhibition activity as antiinflammation effect of water extracts was confirmed to 34% of inhibition at $200{\mu}g/mL$ phenolic. The results can be expected extracts from Cornus kousa fruit to use as functional resource for antioxidant, antigout, inhibitor of carbohydrate degradation, antiwrinkle activity and antiinflammation activity.

A Study on the Cooling Characteristics Improvement of TMA-Water Clathrate Compound by Ethanol (에탄올에 의한 TMA-포접화합물의 냉각특성 개선에 대한 연구)

  • This study aims to find out cooling characteristics of TMA(Tri-Methyl-Amine, $(CH_3)_3N$) 25wt%-water clathrate compound with ethanol($CH_3CH_2OH$) such as supercooling, phase change temperature and specific heat. For this purpose, ethanol is added as per weight concentration and cooling experiment is performed at $-6{\sim}-8^{\circ}C$, cooling heat source temperature, and it leads the following result. (1) Phase change temperature is decreased due to freezing point depression phenomenon. Especially, it is minimized as $3.8^{\circ}C$ according to cooling source temperature in case that 0.5wt% of ethanol is added. (2) If 0.5wt% of ethanol is added, average supercooling degree is $0.9^{\circ}C$ and minimum supercooling is 0.8, $0.7^{\circ}C$ according to cooling heat source temperature. The restraint effect of supercooling is shown. (3) Specific heat shows tendency to decrease if ethanol is added. It is $3.013{\sim}3.048\;kcal/kg^{\circ}C$ according to cooling heat source temperature if 0.5wt% of ethanol is added. Phase change temperature higher than that of water and inhibitory effect against supercooling can be confirmed through experimental study on cooling characteristics of TMA 25wt%-water clathrate compound by adding additive, ethanol. This can lead to shorten refrigerator operation time of low temperature latent heat storage system and improve COP of refrigerator and efficiency of overall system. Therefore energy can be saved and efficiency can be improved much more.

Effect of Wongisaengmaek-san Extract on Antioxidative and Anti-Fatigue Activity (원기생맥산(元氣生脈散)의 추출물별 항산화작용과 쥐의 운동피로 회복효과)

  • Objectives : This study aimed to evaluate the effect of Wongisaengmaek-san (extracted with water or 50% ethanol) on antioxidative activity and recovery of fatigue induced by weight-loaded forced swimming exercise. Methods : Antioxidant activity of Wongisaengmaek-san was evaluated in terms of total amount of polyphenol, 1.1-diphenyl-2-picrylhvdroxyl (DPPH) radical scavenging activity and xanthine oxidase inhibition. The anti-fatigue effect of Wongisaengmaek-san was investigated using an acute weight-loaded forced swimming test by monitoring swimming test times and blood biochemical parameters creatinine. BUN, lactic acid dehydrogenase (LDH), free fatty acid (FFA) and lactic acid (LA). Results : 1. 50% EE (ethanol extract) had 2.3 times higher amount of total polyphenol compared to water extract. 2. It was identified that 50% ethanol extract showed enhancement of xanthine oxidase inhibition and DPPH scavenging effect in vitro. 3. According to anti-fatigue effect in the scale of 5% weight loaded mouse swimming test, both water and 50% ethanol extract showed significant improvement of swimming time elongation respectively, and 50% ethanol extract induced a more positive result. 4. After swimming for 1% weight-loaded mouse. water extract and 50% ethanol showed significant anti-fatigue effect with manifestation of blood sampling among both of the intervention: 50% ethanol extract showed a greater result. Conclusions : 50% ethanol extract of Wongisaengmaek-san has more effective antioxidant activity and anti-fatigue than water extract.

Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats (추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. The Effects of Houttuyniae Herba extract on the Activity of Anti-bacteria, Anti-inflammation and Anti-oxidation (어성초(魚腥草) 추출물의 항여드름 효과에 관한 연구)

  • Objective : This experimental study was performed to investigate the effects of Houttuyniae Herba extract on anti-inflammation and anti-oxidation. Methods : The cytotoxicity of Houttuyniae Herba water extract and ethanol extract about viability of Raw 264.7 cell were tested using a colormetric tetrazolium assay(MTT assay). We investigated the inhibitory effects of Houttuyniae Herba water extract and ethanol extract on Propionibactrium acnes using paper disk diffusion method. To investigate the anti-inflammation effects of Houttuyniae Herba water extract and ethanol extract on LPS-induced macrophage Raw 264.7 cell, we used ELISA kit. We evaluated anti-oxidation effects of Houttuyniae Herba water extract and ethanol extract on HaCaT cell by Enzyme recycling method. Results : 1. In Houttuyniae Herba water extract and ethanol extract, cell toxicity depended on the density and wasn't difference between two extracts. 2. Houttuyniae Herba water extract and ethanol extract has not the significant inhibition effect of Propionibactrium acnes. 3. Concentration of 50, $100{\mu}g/m{\ell}$ Houttuyniae Herba water extract inhibited the production of NO in the Raw 264.7 cell stimulated with LPS. 4. All extracts except for $20{\mu}g/m{\ell}$ Houttuyniae Herba water extract showed anti-oxidation effect by decreasing the DPPH radicals. Conclusion : These results indicate that Houttuyniae Herba extract has anti-inflammation and anti-oxidation effects. If further study is performed, the use of Houttuyniae Herba extract will be valuable and benificial in the therapy of Propionibactrium acnes.
