The Role of Vitamin C and Vitamin E Supplementation on Iron Contents and Biomarkers of Oxidative Stress in Blood, Liver and Brain of Aging Rats (나이에 따른 흰쥐의 혈액, 간, 뇌조직의 철분함량, 산화 스트레스 지표에 대한 비타민 C 와 비타민 E공급의 역할)

  • The purpose of this study was to investigate the effect of vitamin C and vitamin E supplementation on the iron contents and oxidative stress of the rats. Rats were fed 18g ascorbic acid and 300IU $\alpha$-tocopherol/kg diet, respectively. Rats were sacrificed at 1, 3, 5 and 7 month of age. The blood, liver and brain were selected for the quantitation of iron and malondialdehyde(MDA) contents, glutathione peroxidase(GSHPx), superoxided dismutase(SOD) and catalase(CAT) activity. Iron and MDA contents and GSHPx activities were increased with aging. Vitamin C and Vitamin E supplementation increased iron contents of the plasma. Vitamin C raised iron contents, but vitamin E decreased iron contents of the liver. In the brain vitamin C and vitamin E did not affect the iron level. MDA levels were decreased with vitamin C and vitamin E supplementation in the erythrocyte and liver, and vitamin C supplementation elevated MDA levels in the brain. GSHPx activity was increased with vitamin C and vitamin E supplementation. SOD activities of erythroucyte and brain were not affected with age, but in the liver, SOD activity was raised with age and vitamin C supplementation. Vitamin C and vitamin E supplementation promoted CAT activity of erythroucyte and liver, and CAT activity of brain was eleveated with vitamin addition but was decreaed with vitamin E addition. Vitamin C and vitamin E decreased iron contents of blood plasma, MDA contents of plasma and liver, and CAT activity of erythrocyte. Above results indicated that iron contents and biomarkers of oxidative stress were more affected by age than antioxidant action of vitamin C and vitamin E.

Effects of Vitamin E on Liver Cytochrome $P_{450}$ Content and Xanthine Oxidase Activity in Acute Cadmium-Poisoned Rats (식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 Cytochrome $P_{450}$ 함량과 Xanthine Oxidase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of vitamin E on liver microsomal cytochrome P450 contents and xanthine oxidase activity in acute cadmium poisoned rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(0E Cd group), 40mg vitamin E per kg diet(40E Cd group) or 400mg vitamin E per kg diet(400E Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd2+/kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4 weeks. Body weight, food intake and feed efficiency ratio of cadmium injected animals, were decreased compared with those of normal group. The weights of liver and kidney in cadmium injected groups were not different from those of normal group. Cadmium contents of liver in cadmium groups were 160 fold higher those that of normal group. Accumulation of cadmium poisoned rat liver was reduced by vitamin E supplementation. Contents of blood hemoglobin and hematocrit in 0E Cd groups were decreased to 2~ 14% of those of the other groups. Contents of serum triglyceride in all experimental groups were not significantly different each other. Levels of serum total cholesterol, LDL cholesterol and antherogenic index in 0E Cd and 40E Cd groups were higher than those of normal group, while the contents of HDL cholesterol in 0E Cd and 40E Cd groups were lower than those of normal group. Xanthine oxidase(XOD) activity and cytochrome P450 contents in the liver were significantly increased in cadmium injected groups but these were reduced by vitamin E supplementations. The present results indicate that acute cadmium poisoning in rats causes increasing free radical generation systems in the liver and that leads to liver tissue damage. But these abnormalities can be reduced by dietary vitamin E supplementations.

Suppressive Effects of Vitamin E on the Induction of Placental Glutathione S-transferase(GST-P) Positive foci and Antioxidant Enzyme Activity in Rat Hepatocarcinogenesis (비타민 E가 쥐간 세포의 암화과정에서 태반형 Glutathione S-transferase(GST-P) 양성 병소와 항산화요소계에 미치는 영향)

  • The influences of dietary supplements of vitamin E on hepatocellular chemical carcinogenesis have been studied, Placental glutathione S-transferase(GST-P) positive foci area, antioxidant enzymes(superoxide dismutase(SOD), catalase, glutathione reductase, glutathione peroxidase, glutathione S-transferase(GST)), glucose 6-phosphatase(G6Pase) activities, and lipid peroxidation of mecrosomes(thiobarbituric acid reactive substances(TBARS) contents) were investigated. For is purpose , we used the murine chemical hepatocardinogenic procedure induced by modified Ito model, which consists of 200mg/kg body weight diethylinitrosamine (DEN) injection, 0.01% 2-acethlaminoflurene(2-AAF) feeding for 6 weeks, and partial hepatectomy on week 3. Weanling Sprague-Dawley male rats were fed pulverized Purina rat chow with 15, 000IU/kg diet vitamin E from initiation or promotion stages. We found that vitamin E supplement decreased the area of GST-P positive foci. Catalase, glutathione peroxidase, glutathione reductase. GST activities, and TBARS contents were decreased. On the other hand G6Pase activities were increased by vitamin E supplement. It seemed that vitamin E supplements helped endogenous defense systems against carcinogenesis by decreasing TBARS contents, $H_2O$$_2$ and organic peroxides. So, vitamin E seemed to protect cell from free radical damage in carcinogenesis. Anticarcinogenic effects of vitamin E were more effective at intiation that at promotion stage. These results suggest that vitamin E has suppressive effects on hepatocellular chemical carcinogenesis, probably through antioxidant effects against TBARS contents $H_2O$$_2$ and orgainc peroxides.

Study on Vitamin I Intake of Exclusively Breast-fed Infants (모유 영양아의 비타민 E 섭취에 관한 연구)

  • In order to investigate the vitamin I intake of exclusively breast-fed infants, we examined 33 lactating women and their infants at 0.5, 1, 2, 3, 4 and 5 months of lactation. Vitamin E contents of human milk were determined by HPLC analysis. Vitamin E contents of the milk showed 539, 520, 464, 422, 409 and 351$\mu\textrm{g}$/100m1 during the lactation respectively. Vitamin E contents of the human milk were not affected by energy, lipid and protein consumption of lactating women. Vitamin E intake of breast-fed infants averaged 3375 and 269$\mu\textrm{g}$/day in boys and girls during the first 5 months of lactation. Vitamin E intake per body weight of breast-fed infants appeared 725, 752, 600, 461, 420 and 334$\mu\textrm{g}$/kg/day respectively. We conclude that breast-fed infants most likely receives adequate vitamin E from the human milk compared with recommended dietary allowances for Korean infants.   • Purpose: People have long been interested about antioxidant nutrients such as vitamin A and E to stay in healthy states. This present study provides trustworthy nutritional information and evaluates vitamin A and E contents in emphasized food items. Methods: Vitamin A or vitamin E emphasized foods including beverage, cereal, snack, chocolate product, other cocoa products, and sugar products were analyzed. The contents of vitamin A, E in samples were examined by high performance liquid chromatography. Vitamin A and vitamin E contents were compared with the labeled value indicated on the nutrition label. Results: Vitamin A (n=18) value ranged from 90% to 248% and vitamin E (n=20) value ranged between 96-413% of labeled value. Analyzed samples ranged more than 80% of the labeled value, which complied with food labeling rules. Conclusion: Providing accurate information for nutrition label is essential to confirm nutrient contents in foods. It will help to decide the adequate level of nutrient intake. Therefore, the nutrition labeling system should be supervised continuously with regular monitoring analysis.

Effect of Dietary Polyunsaturated Fatty Acid and $\omega$-Tocopherol on Lipid Peroxidation in Rat Liver (식이 불포화지방산과 Vitamin E 함량이 흰쥐 간장내의 지질과산화에 미치는 영향)

  • To study effect of dietary polyunsaturated fatty acid and $\omega$-tocopherol content on lipid peroxidation in rat liver, rats were fed for 3, 6 and 9 weeks with normal tocopherol diet added 40mg of DL-$\alpha$-tocopherol/kg of diet (PF group), high tocopherol eit 200mg of DL-$\alpha$-tocopherol/kg of diet(PFE group), low tocopherol diet without addition of DL-$\alpha$-tocopherol to diet(PFO group), and control diet added 40mg of DL-$\alpha$-tocopherol/kg of diet(control group). Each diet group supplied 45% of total calorie from corn oil except control group which supplied 12% of total calorie from corn oil. After each feeding period, lipid peroxide and tocopherol contents were measured in the liver as well as activities of glutathione peroxidase and superoxide dismutase. PF group had almost the same contents of liver peroxide, tocopherol contents and activities of glutathione peroxidease and superoxide dimutase as control group, while PFE group had higher tocopherol content and glutathione peroxidase activity and lower lipid peroxide contents and superoxide dismutase activity than control group. On the other hand, changes in the values of PFO group were opposite to those of PFE group. Differences in the values among groups were more pronounced as feeding period became longer.

  • This study was undertatken to investigate the effects of dietary vitamin E on the toxicity of cadmium(Cd) in rats. The two variables were the supplmentary vitamin E(400lU/kg) and the protein amount(10.5% in the low protein diet and 18.0% in the normal protein diet) In cadmium treated rats net weight gain and food intake were decreased but improved by supplementation with vitamin E in the normal protein, hematocrit values reduced by Cd were significantly increased by the addition of vitamin E to normal protein diet in Cd intoxicated rats, The supplementation with vitamin E diminished the effect of Cd on aspartate aminotransf-rase and alanine aminotransferase activities in serum In Cd treated rats fed normal protein diet with vitamin E the contents of triglyceride were decreased and total-cholesterol contents were significantly reduced in serum and both of them in liver were markedly decreased. The activity of alcohol dehydrogenase in liver was decreased by Cd however supplementation with vitamin E reduced the effects of Cd on hepatic alcohol dehydrogenase. the results of this experiment indicated that there was some interaction between vitamin E and protein levels and supplementation with vitamin E had an effect more than protein levels oncd toxicity.

Effects of Vitamin E on Antioxidative Defense System of Liver in Acute Cadmium-Poisoned Rats (식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 항산화계에 미치는 영향)

  • The purpose of this study was to investigate the effects of vitamin E on antioxidative defense system of liver in acute cadmium poisoned rats. Cadmium injected groups were fed vitamin E free diet(OE-Cd group), 40mg vitamin E per kg diet(40E-Cd group) or 400 mg vitamin E per kg diet(400E-Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd$^2$$\^$+//kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4weeks. Activities of superoxide dismutase(SOD), glutathione peroxidase(GSHPx) and glutathione S-transferase(GST) were decreased in cadmium injected groups but those were significantly improved by dietary vitamin I supplementations. Vitamin E contents reduced glutathione(GSH) in the live were decreased in cadmium injected groups, but we., not significantly different among three groups with different levels of vitamin E supplementations. Contents of liver thiobarbituric acid reactive substance (TBARS) of 0E-Cd group were higher than those of 400E-Cd and 400E-Cd groups, but those were markedly alleviated according to vitamin E supplementations. These results indicate that cadmium poisoning in rats causes decreasing antioxidative defense system and increasing peroxidative damage in liver, however can be restored by vitamin E supplements.   • This study was designed to investigate the effect of dietary vitamin E levels on lipid metabolism in caffeine-fed rats. Male Sprague-Dawley rats were fed on diet containing one of three levels of vitamin E(37.5, 750, or 1,500mg/kg diet0 which was with or without 0.3% caffeine. The rats were sacrificed after 5 and 10 weeks of the feed periods. Results obtained from this study were as follows ; Net weight gain, feed intake and FER in the caffeine added groups were significantly decreased as compared to those of only vitamin E fed group. Liver and kidney weights tended to increase in the caffeine added groups, but spleen and heart weights were not affected. Total lipid contents in serum and liver tended to decrease as dietary vitamin E became increasing and caffeine diet adding. Serum total cholesterol content tended to increase in the caffeine added group. But, liver cholesterol content were decreased. And the higher the dietary vitamin E level became, the lower serum and liver cholesterol contents were. Serum and liver triglyceride contents were significantly lower in the caffeine added groups than in the only vitamin E fed groups. The degree of decrease was more evident as dietary vitamin E level became more increasing. Serum aminotransferase activity was not affected in all experimental groups.

  • This experiment is a quantitative experiment where insoluble Vitamins E and K are extracted from the yolk of the spawning which was fed by by-products of tangerine, carrot, or Japanese apricot. Finally the purpose is to raise contents of the insoluble Vitamins E and K inside the yolk of the spawning. The saponification method through ${\alpha}$, ${\alpha}$ - dipyridyl was used to determine the content of Vitamin E. Diethyl dithiocarbamate assay was used to examine the content of Vitamin K. The contents of Vitamin E in egg yolks of chicken fed by by-products of tangerine, carrot, or Japanese apricot increased 2 - 3 times compared to control egg, while the contents of Vitamin K almost no changed.
