  • The purpose of this study was to determine if vitamin B-2 and vitamin B-6 deficiency affects hematologic profile in vivo. Rats were fed a vitamin B-2 deficient (-B2) diet or a vitamin B-6 deficient (- B6) diet or a combined vitamin B-2 and vitamin B-6 deficient (-B2-B6) diet or a control diet for 6 weeks. Hematocrit (Ht), hemoglobin (Hb), plasma iron (PI), transferrin saturation (TS) and liver iron concentration (LI) were compared. In -B2 rats vs. control rats, PI, TS and LI were significantly lower but Ht and Hb were not. In - B6 rats vs. control, Ht and Hb were significanty lower but LI was higher. The -B2-B6 rats had intermediate effects in Hb, PI, TS and LI and a decrease in Ht. The repletion with a contro diet for 2 weeks resulted in significant improvements in hematologic indices and LI in both - B2 rats and - B6 rats. This study suggests that the intakes of vitamin B-2 and vitamin B-6 are important for preventive and therap\ulcornereutic approaches to improve the hematologic status in nutritionally anemic groups.

  • The effects of vitamin B6 deficiency on energy utilization during fasting or underfeeding were studied in rats. Fifteen rats were fed a vitamin B6 deficient(-B6) diet and another 15 rats wee fed a control (+B6) diet. These rats were fed for 5 weeks with respective diet, and then subdivided into 3 groups : non-fasted group, fasted group, underfed group. Rats of the fasted group were fasted for 3 days and those of underfed group for 6 days. At the respective time (non-fast, 3 day-fast, 6 day-underfeed at 5 weeks), animals were sacrificed. Feed efficiency ratio of - B6 rats was significantly lower than that of +B6 rats. In - B6 rats, the liver and kidney weights were significantly heavier than those of +B6 rats but spleen and heart weights were not. In non-fasted group, liver protein and triglyceride level of - B6 rats were significantly higher than that of +B6 rats. After - B6 rats were fasted for 3 days, plasma free fatty acid level was significantly lower but liver glycogen level was higher than that of +B6 rats and muscle protein level of +B6 was decreased while that of - B6 was not changed. Vitamin B6 deficiency had little effect on the energy utilization with 6 days underfeeding. These results suggest that vitamin B6 deficiency may impair the utilization of stored fuel during fasting.

  • The purpose of this study was to investigate the effect of vitamin B6 deficiency on the concentration of energy metabolite in streptozotocin-induced diabetic rats. Thirty rats were fed a vitamin B6 deficient diet(-B6) or a control diet(+B6) for 5 weeks and then subdivided into 3 groups respectively ; base group, one day diabetic group and three day diabetic group. Diabetes of rats were induced by streptozotocin injection into the tail vein. Glucose, glycogen, protein, alanine, triglyceride and free fatty acids were compared in plasma, liver skeletal muscle of rats. Also, the total urinary nitrogen and glucose excretion were compared. Compared with +B6 rats, the increase of plasma glucose in -B6 rats due to the diabetes was smaller. After diabetes was induced, the level of plasma alamine was not changed in -B6 rats while increased significantly(p<0.05) in +B6 rats. The increase of urinary nitrogen excretion was smaller and the increase of muscle protein was larger in -B6 rats at the first day diabetes was induced. The levels of plasma free fatty acid and liver triglyceride were significantly (p<0.05) higher in -B6 rats after diabetes was induced. These results suggest that vitamin B6 deficiency may impair the adaptation of animals to the energy metabolism related due to a decrease of the body protein catabolism of fatty acid oxidation in diabetes and aggravate fatty liver which is one of the chronic complications of diabetes.

  • This study investigated the effect of vitamin $B_6$ deficiency on antioxidant enzyme activities and lipid profile in rats with exercise-induced oxidative stress. Forty eight rats were fed either a vitamin $B_6$ deficient diet (B6-) or a control diet (control) for 4 weeks and then subdivided into 3 groups: pre-exercise (PreE); post-exercise (PostE); recess after exercise (recessE). Compared to those of control group, plasma catalase and hepatic cytosol superoxide dismutase (SOD, EC 1.15.1.1) activities of B6- group were lower regardless of exercise. The ratio of reduced glutathione/oxidized glutathione (GSH/GSSG) of B6 - group was lower in PreE and there was no difference between PostE and recessE. The level of malondialdehyde (MDA) of B6- was significantly higher in PreE and PostE. High-density lipoprotein-cholesterol (HDL-C) level of B6- group was lower regardless of exercise. Atherosclerotic index of $B_6$- group was higher in PreE and there was no difference between PostE and recessE. It is suggested that a reduction in antioxidative status caused by vitamin $B_6$ deficiency may be aggravated under exercise-induced oxidative stress.

  • The purpose of this study was to determine whether vitamin B6(B6) deficiency affects fuel utilization and blood cholesterol profile with exercise-training. Twenty-four rats were fed a B6 deficient(-B6) diet or a control (+B6) diet for 5 weeks and either exercised(EX) or nonexercised (NE). EX rats were exercised on treadmill(10$^{\circ}$, 0.5-0.8km/h) for 20 minutes everyday. Glucose(GLU), glycogen (GLY), protein(PRO), trglyceride(TG), free fatty acid(FFA), total cholesterl(TC), HDL-cholesterol(HDL-C) and LDL-choleterol(LDL-C) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. There was a vitamin effect on the level of P-GLU, P-TG, M-TG, L-GLY, L-PRO and an exercise effect on the level of P-PRO, P-FFA, M-PRO, L-GLY, L-TG, P-TC, P-HDL-C, P-LDL-C. Compared to +B6 rats were lower and there were no differences in P-GLU, P-FFA, P-TG. M-GLY, L-TG, P-TC and P-HDL-C. In EX group, the level of P-TG was higher and M-PRO was lower in -B6 rats. There were no differences in M-GLY, L-TG, P-TC and P-HDL-C. These results suggest that a lowered intake of vitamin B6 may impair the adaptation of animals to fuel metabolism related to a decrease of fatty acid oxidation and attenuates the exercise-traning effect on blood lipid profile.

  • BACKGROUND/OBJECTIVES: Recent research regarding vitamin $B_6$ status including biochemical index is limited. Thus, this study estimated intakes and major food sources of vitamin $B_6$; determined plasma pyridoxal 5'-phosphate (PLP); and assessed vitamin $B_6$ status of Korean adults. MATERIALS/METHODS: Three consecutive 24-h diet recalls and fasting blood samples were collected from healthy 20- to 64-year-old adults (n = 254) living in the Seoul metropolitan area, cities of Kwangju and Gumi, Korea. Vitamin $B_6$ intake and plasma PLP were analyzed by gender and by vitamin $B_6$ supplementation. Pearson's correlation coefficient was used to determine associations of vitamin $B_6$ intake and plasma PLP. RESULTS: The mean dietary and total (dietary plus supplemental) vitamin $B_6$ intake was $1.94{\pm}0.64$ and $2.41{\pm}1.45mg/day$, respectively. Median (50th percentile) dietary intake of men and women was 2.062 and 1.706 mg/day. Foods from plant sources provided 70.61% of dietary vitamin $B_6$ intake. Only 6.3% of subjects consumed total vitamin $B_6$ less than Estimated Average Requirements. Plasma PLP concentration of all subjects was $40.03{\pm}23.71nmol/L$. The concentration of users of vitamin $B_6$ supplements was significantly higher than that of nonusers (P < 0.001). Approximately 16% of Korean adults had PLP levels < 20 nmol/L, indicating a biochemical deficiency of vitamin $B_6$, while 19.7% had marginal vitamin $B_6$ status. Plasma PLP concentration showed positive correlation with total vitamin $B_6$ intake (r = 0.40984, P < 0.0001). CONCLUSIONS: In this study, vitamin $B_6$ intake of Korean adults was generally adequate. However, one-third of subjects had vitamin $B_6$ deficiency or marginal status. Therefore, in some adults in Korea, consumption of vitamin $B_6$-rich food sources should be encouraged.

  • The purpose of this study is to evaluate the nutritional status of vitamin and minerals in the patients with alcoholic liver disease and to obtain the materials for nutritional education for alcoholics. The subjects consist of 80 patients with alcoholic liver cirrhosis(ALC) and 12 patients with alcoholic fatty liver(AFL). The control group included 57 alcoholics without liver disease(A), 32 patients with viral liver cirrhosis(VLC) and 194 normal men(NL). Biochemical evaluation of nutritional status was investigated by the analysis of blood samples. The frequency of vitamin B1 deficiency in the ALC, AFL and A groups as indicated by the erythrocyte transketolase activity coefficient were 46.0%, 66.7% and 57.9% respectively. The frequency of vitamin B2 deficiency in the ALC, AFL and A groups as indicated by the erythrocyte glutathione reductase activity coefficient were 9.8%, 8.3% and 38.6% respectively. Vitamin A deficiency was not detected in the alcoholic subjects. The frequency of vi tamin E deficiency in ALC, AFL and A were 96.3%, 66.7% and 86.0% respectively. The levels of plasma lipid peroxidation products were significantly higher in the alcoholic subjects than in the normal subjects. The frequency of subjects below normal range of hemoglobin were 85.0% in ALC, 50.0% in AFL and 31.6% in A. The frequency of copper deficiency in the ALC, AFL and A groups were 48.4%, 16.7% and 17.5% respectively. The frequency of zinc deficiency in the ALC, AFL and A groups were 83.8%, 41.7% and 66.7% respectively. Overall, the vitamin and minerals status of the alcoholic subjects in this study was evaluated to be very poor on the basis of biochemical assessments. The results suggest that alcohol abuse and poor dietary intake could cause malnutrition and may be important risk factors in causing alcoholic liver disease in alcoholics. In addition, vitamin B1, vitamin B2, Cu, Fe and antioxidant supplementation may be effective in nutritional therapy for chronic alcoholics.

  • The purpose of this study was to investigate the effect of vitamin {TEX}$B_{6}${/TEX} deficiency on the utilization and recuperation of stored fuel in exercising rats. Thirty six rats were fed either a vitamin {TEX}$B_{6}${/TEX} deficient diet(-{TEX}$B_{6}${/TEX}) or a control diet(+{TEX}$B_{6}${/TEX}) for 5 weeks, then subdivided into 3 group: non-exercise group(NE), exercise group(EX), exercise and recuperation group(EX). EX group were exercised in treadmill({TEX}$10^{o}${/TEX}, 0.5~0.8km/h)for 2 hours and ER group were recuperated three days with the respective diet after exercise. Glucose(GLU), glycogen(GLY), protein(PRO), trigyceride(TG) and free fatty acid(FFA) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. Compared to +{TEX}$B_{6}${/TEX} rats, in NE group, the level of L-GLY of -{TEX}$B_{6}${/TEX} rats was higher, M-TG, L-PRO of -{TEX}$B_{6}${/TEX} rats were lower and there were no differences in P-PRO, P-FFA, P-TG, M-GLY, M-PRO and L-TG. In EX group, the levels of P-FFA, L-PRO of -{TEX}$B_{6}${/TEX} rats were higher. P-TG, L-TG of -{TEX}$B_{6}${/TEX} rats were lower and there was no difference in L-GLY. In EG group, the levels of P-GLU, P-PRO, P-RG, L-PRO of -{TEX}$B_{6}${/TEX} rats were lower and there were no differences in L-GLY, L-TG, M-TG and M-GLY. these results suggest that a lowered intake of vitamin {TEX}$B_{6}${/TEX} may impair the recuperation of aminals after exercise related to exercise fuel stores although there is a compensation among stored fuel utilization during exercise.

  • Purpose: The most common metabolic defect appearing following a gastrectomy is anemia. Two types have been identified: One is related to a deficiency in iron and the other is related to an impairment in vitamin B12 metabolism. The purpose of this study is to evaluate the incidence and treatment of vitamin B12 deficiency after a total gastrectomy. Materials and Methods: Serum vitamin B12 concentrations were measured in 128 patients who had undergone a total gastrectomy. The group with a serum concentration under 200 pg/ml was supplemented at one-month intervals with Actinamide and five-six months later, serum concentrations of the vitamin B12 were rechecked. Results: The group with vitamin B12 under 200 pg/ml was 61 (47.6%) of the 128 patients who had undergone a total gastrectomy. In this group, the cumulative incidences of vitamin B12 deficiency were 7.0, 23.4, 33.6, 39.1, 41.4, and 47.7% at 6 months, 1, 2, 3, 4, and 5 or more years, respectively. The treated group with vitamin B12 under 200 pg/ml had 36 (28.17%) of the 128 patients. The 16 of those cases with vitamin B12 levels of $100{\sim}150\;pg/ml$ were supplemented 5.21 times and the vitamine B12 was elevated to above 650 pg/ml. The other 20 cases with an average of vitamin B12 levels of $150{\sim}200pg/ml$ were supplemented an average of 4.75 times, and the vitamin B12 was elevated to above 780 pg/ml. Conclusion: It is necessary to supplement vitamin B12 even 1 year later after a total gastrectomy. The group with vitamin B12 under 200 pg/ml was supplemented $5{\sim}6$ times at one-month intervals with Actinamide $1,000\;{\mu}g$ IM injections and reached normal levels. • The purpose of this study was to determine whether vitamin {TEX}$B_{6}${/TEX} deficiency and age affect the blood cho-lesterol profile in exercising rats. Fifty four rats were fed either a viramin {TEX}$B_{6}${/TEX} deficient dief(-{TEX}$B_{6}${/TEX}) of a control diet(+{TEX}$B_{6}${/TEX}) for 6 weeks, then subdivided into 3 groups:non-exercise group(NE), exercise and sacrifice group(ES), exercise and recuperation group(ER). ES group was exercised in treadmill({TEX}$10^{o}${/TEX}, 0.5~0.8km/h) for 2 hours and sacrifice. ER group was recuperated three days with respective diet after exercise. At week 3 and 6, and level of plasma total cholesterol(TC), high density lipoprotein cholesterol(HDL_C) and low density lipoprotein cholesterol(LDL_C) were compared. In NE group, there was no difference in the levels of TC, HDL_C and LDL_C between +{TEX}$B_{6}${/TEX} rats and -{TEX}$B_{6}${/TEX} rats. The plasma levels of TC and LDL_C of 6 weeks were higher than those of 3 weeks and on difference in HDL_V between 3 weeks rats and 6 weeks rats. In ES group, there was also no difference in the levels of TC, HDL_C and LDL_C between +{TEX}$B_{6}${/TEX} rats and -{TEX}$B_{6}${/TEX} rats and there was no difference in TC, LDL-cholesterol between 3 weeks rats and 6 weeks rats. The level of HDL_C pf 6 weeks was lower than that of 3 weeks rats. In ER group, there was no difference in the levels of TC and LDL_C not only between +{TEX}$B_{6}${/TEX} rats and-{TEX}$B_{6}${/TEX} rats but also between 3 weeks rats and 6 weeks rats. The level of HDL_C was lower in -{TEX}$B_{6}${/TEX} rats than in +{TEX}$B_{6}${/TEX} rats and higher in 6 weeks rats than in 3 weeks rats. These results suggest that vitamin {TEX}$B_{6}${/TEX} deficiency may affect the HDL_C during exercise and after recuperation. The desirable effect of exercise on plasma Cholesterol profile is strengthened in adult age than young age.
