Strawberry Virus Diseases Occurring in Korea, 2007-2008 (우리나라에서 발생하는 딸기 바이러스병(2007-2008))

  • Virus disease surveys of strawberries cultivated and preserved as germplasm resources in Korea was conducted during 2007-2008. Virus detection was conducted by RT-PCR using total RNAs extracted from strawberry samples. We detected the infection with Strawberry mild yellow edge virus (SMYEV), Strawberry mottle virus (SMoV), Strawberry vein banding virus (SVBV) and Strawberry pallidosis associated virus (SPaV) while no infection with Strawberry crinkle virus (SCV), Strawberry necrotic shock virus (SNSV), Strawberry latent ring spot virus (SLRSV) and Arabis mosaic virus (ArMV) was observed. The infection rate of virus disease on 4 cultivars including Seolhyang, Maehyang, Gumhyang, and Dahong, bred in Korea, was 0.1, 1.9, 0, and 0%, respectively. Surprisingly, however, cultivar Red Peal introduced from Japan in 1997 revealed 48.3% virus infection rate. SMYEV, SMoV and SPaV were also identified in strawberries growing in the farm fields of Korea. In the field, however, SMYEV was the most predominant virus (97.4%) among those 3 identified viruses. SVBV was detected only in strawberry kept as a germplasm.

  • There is a good evidence that tabacco necrosis virus, lettuce big vein virus, and tabacoo stunt virus are transmitted by Oplidium brassicae, although absolute proof in aspetic condition is lakcing. Some evidence suggests that polymyxa graminis may be involved in transmission of wheat mosaic virus. One report claims that Synchytrium endobioticum can transmit potato virus X. The cultivated mushroom, Agaricus bisporus, is known to act as a hose of a virus and is apparently involved in the spread of the virus.

  • 참돔 이리도바이러스 질병(red seabream iridovirus disease ; RSIVD)은 1990년 일본 시코쿠지역의 참돔 양식장에서 처음 발병된 후, 매년 발병지역이 확산되고 발병 어종이 다양해지고 있다. 참돔에서 분리된 RSIV는 icosahedral cytoplasmic deoxyribovirus로서 크기가 200∼240nm이며 형태학적 특징에 의해 iridoviridae로. 분류하고 있지만, 어류를 숙주로 하는 iridovirus과 중에서 lymphocystis virus속인 flounder virus(LCDV-1) 및 lymphocystis disease virus(LCDV-2)와 goldfish virus 1-lke virus속인 goldfish virus 1(GFV-1) 및 goldfish virus 2(GFV-2)와는 전혀 다른 바이러스로 알려져 있다.   • A tomato Aspermy Virus (TAV-B) served as a helper virus for multiplication and encapsidation of satellite RNAs which were isolated from two different CMV isolates, D and K. These two satellite RNAs induced renarkable attenuation of TAV symptoms in infected tobacco, which was correlated with a reduction of virus content in the plant. The CMV satellite RNAs also caused lethal necrosis in TAV-infected tomato as in the case of CMV system.

  • Carlaviruses were isolated from naturally infected medicinal plants, and identified by means of test plants and electron microscopy. Mottle symptoms were shown on leaves of Panax ginseng, Aralia cordata, Xanthium strumarium, Taraxacum officinale, Aconitum carmichaeli, and Bupleurum longiradiatum var. breviradiatum. Ring spot on leaves of Abutilon avicennae and ring mosaic or vein clearing on leaves of Sambucus sieboldiana were also shown. These viruses had rather narrow host ranges by mechanical inoculation. The virus particles were scattered or aggregated in cytoplasm of infected host plant leaves. The carlaviruses for which the name panax virus S (PaVS), aralia virus S (ArVS), xanthium mottle virus (XaVS), taraxacum virus S (TaVS), aconite mottle virus (AcMV), bupleurum virus S (BuVS) and abutilon ring spot virus (AbRSV) were proposed, had flexuous particles with width 13 nm and length $620{\sim}720nm$. A reported elder ring mosaic virus was isolated from leaves of Sambucus sieboldiana with ring mosaic or vein-clear symptoms.

A pathological study in subsequent infection of PRRS virus to pigs with porcine pleuropneumoniae (돼지생식기호흡기증후군과 돼지 흉막폐렴의 복합감염에 따른 병리학적 연구)

  • The purpose of this study was to evaluate the effect of a subsequent infection of porcine reproductive and respiratory syndrome(PRRS) virus to pigs with A. pleuropneumonia in pigs. Twenty three 7-weeks-old commercial pigs were infected with PRRS virus and/or A. pleuropneumoniae serotype 5 intratracheally. Feed conversion, clincal signs, gross and histopathological lesions and immunohistochemical findings were examined. 1. Feed conversion ratio in dual-infected pigs with PRRS virus and A. pleuropneumoniae were higher than that of single- infected pigs with PRRS virus or A. pleuropneumoniae. 2. Dual-infected pigs with PRRS virus followed by A. pleuropneumoniae showed more severe clinical signs and gross, histopathological and immunohistochemical pulmonary lesions. The results indicated that dual infections with PRRS virus and A. pleuropneumoniae caused more severe respiratory lesions and growth retardation in pigs than single infection with PRRS virus or A. pleuropneumoniae.

Turnip Mosaic Virus Isolated from Rorippa islandica Borb (속속이풀(Rorippa islandica Borb.)에서 분리한 순무 모자이크 바이러스)

  • Turnip mosaic virus (TuMV) was isolated from Rorippa islandica showing mild mosaic symptom in growing field of Chinese cabbage and radish. Identification of the virus was based on host range, transmission by aphids, electron micrograph, serological reaction and hybridization detection. The virus systemically infected on Chenopodium quinoa, Nicotiana clevelandii, N. glutinosa, Brassica rapa, B. campestris subsp. pekinensis and Raphanus sativus, whereas showed local infection on C. amaranticolor, Gomphrena globosa and Tetragonia tetragonoides. The virus was transmitted by aphid (Myzus persicae). The virus particle was filamentous with 720$\times$12 nm in length, and reacted positively with an antiserum of TuMV in agar gel double duffusion test. In slot-blot hybridization using the digoxigenin(DIG)-labeled RNA probe, TuMV-RNA could be detected in sap of R. islandica infected with the virus. This is the first report of a natural infection of that virus on R. islandica.

Enhanced Virus Removal by Flocculation and Microfiltration

  • In this work we have investigated the feasibility of virus clearance by flocculation and tangential flow microfiltration. Chinese hamster ovary cell feed streams were spiked with minute virus of mice and then flocculated using cationic polyelectrolytes prior to tangential flow microfiltration. Our results indicate that flocculation prior to microfiltration leads to more than 100 fold clearance of minute virus of mice particles in the permeate. Today, validation of virus clearance is a major concern in the manufacture of biopharmaceutical products. Frequently new unit operations are added simply to validate virus clearance thus increasing the manufacturing cost. The results obtained here suggest that virus clearance can be obtained during tangential flow microfiltration. Since tangential flow microfiltration is frequently used for bioreactor harvesting this could be a low cost method to validate virus clearance.

Glial Cell-specific Regulation of the JC virus Early Promoter by Silencer and DNA Methylation (Silencer 및 DNA methylation에 의한 JC virus early promoter의 뇌교세포 특이적인 조절)

  • The human polyomavirus JC virus is the etiologic agent of progressive multifocal leukoencephalopathy (PML). The JC virus early promoter directs cell-specific expression of the viral replication factor large T antigen, thus transcriptional regulation constitutes a major mechanism of glial tropism in PML. Here we found that pentanucleotide sequence immediately upstream of the TATA sequence functions as a cell-specific silencer in the JC virus transcription. In vitro binding studies showed that synthetic oligonucleotides spanning a pentanucleotide sequence, designated "oligo 2", interacts with nuclear proteins from non-glial cells in a cell-specific manner. Furthermore, the sequence preferentially repressed the heterologous thymidine kinase promoter activity in non-glial cells. We also tested whether JC virus transcription is controlled by DNA methylation. Transient transfection of in vitro methylated JC virus promoter abolished transcription in both the glial and non-glial cells. The repression fold was much larger in glial cells than in non-glial cells. Taken together, this finding suggests that glial cell-specific expression of the JC virus is controlled by DNA methylation as well as cell-specific silencers.

Survey and identification of virus diseases on paprika in Jeonnam province

  • Occurrences of virus diseases on paprika ( Capsicum annuum var. grossum) were surveyed in Joennam province from 1999 to 2003 and the collected samples showing virus-like symptoms were tested using ELISA. Virus diseases appeared 4.5%, 17.5%, and 4.9% in 2000, 2002, and 2003, respectively. As the results of investigation of the seasonal incidence with the growing stages of plant, virus was not occurred at seedling stage and was slightly from the planting time to the first harvesting time, but was dramatically increased at the second harvesting time. Virus diseases were more severe on the vinyl house than on the green house. Pepper mild mottle virus (PMMoV) was severely occurred in 2000 but not after that year. Comparing the virus species, Pepper mottle virus (PepMoV) was 35.9%, Broad bean wilt virus (BBWV) was 14.1%, and Cucumber mosaic virus (CMV) was 10.9% in 2002, and 76.0%, 11.1%, and 2.4% in 2003, respectively.
