Toxoplasma gondii Induces Apoptosis via Endoplasmic Reticulum Stress-Derived Mitochondrial Pathway in Human Small Intestinal Epithelial Cell-Line

  • Toxoplasma gondii, an intracellular protozoan parasite that infects one-third of the world's population, has been reported to hijack host cell apoptotic machinery and promote either an anti- or proapoptotic program depending on the parasite virulence and load and the host cell type. However, little is known about the regulation of human FHs 74 small intestinal epithelial cell viability in response to T. gondii infection. Here we show that T. gondii RH strain tachyzoite infection or ESP treatment of FHs 74 Int cells induced apoptosis, mitochondrial dysfunction and ER stress in host cells. Pretreatment with 4-PBA inhibited the expression or activation of key molecules involved in ER stress. In addition, both T. gondii and ESP challenge-induced mitochondrial dysfunction and cell death were dramatically suppressed in 4-PBA pretreated cells. Our study indicates that T. gondii infection induced ER stress in FHs 74 Int cells, which induced mitochondrial dysfunction followed by apoptosis. This may constitute a potential molecular mechanism responsible for the foodborne parasitic disease caused by T. gondii.

Effects of Gossypetin from Hibiscus sabdariffa on Interleukin-6 Production in Porphyromonas gingivalis Lipopolysaccharide-Stimulated Human Gingival Fibroblasts

  • Background: Periodontal disease is a major cause of tooth loss in adults and is a representative oral disease commonly suffered by most people around the world. Mainly the proliferation of Gram-negative bacteria and secreted virulence factors cause an inflammatory response and destroy periodontal tissue. Gossypetin, isolated from Hibiscus sabdariffa L, is known to have various pharmacological effects, including antibacterial and anticancer activities. We aimed to confirm the anti-inflammatory effect of gossypetin through interleukin-6 (IL-6) regulation in human gingival fibroblasts (HGFs) stimulated with lipopolysaccharide (LPS) of Porphyromonas gingivalis, a major cause of adult periodontitis. Methods: CCK-8 assay was performed to confirm the concentration-dependent cytotoxicity of gossypetin against HGFs. The secretion level and mRNA expression of IL-6, an inflammation-related cytokine, and the effect of gossypetin on these in HGFs stimulated with P. gingivalis LPS were confirmed by ELISA and qRT-PCR analysis, respectively. Results: Up to a concentration of 100 µM gossypetin with or without P. gingivalis LPS, the survival rate for HGFs was maintained at over 95% and showed no toxicity. ELISA and qRT-PCR analysis results showed that P. gingivalis LPS increased IL-6 secretion and mRNA levels in HGFs compared to the control group. However, this increase in IL-6 was significantly down-regulated by gossypetin treatment in a dose-dependent manner. In particular, 80 µM gossypetin inhibited IL-6 production to the level of the control group. Conclusion: These results indicated that gossypetin attenuated IL-6 production in HGFs stimulated by P. gingivalis LPS, which may ultimately suppress the inflammatory response in periodontal tissue. Therefore, gossypetin may have potential as a natural ingredient for the prevention and treatment of periodontal disease.

Magnaporthe oryzae Effector AVR-Pii Helps to Establish Compatibility by Inhibition of the Rice NADP-Malic Enzyme Resulting in Disruption of Oxidative Burst and Host Innate Immunity

  • Plant disease resistance occurs as a hypersensitive response (HR) at the site of attempted pathogen invasion. This specific event is initiated in response to recognition of pathogen-associated molecular pattern (PAMP) and subsequent PAMP-triggered immunity (PTI) and effector-triggered immunity (ETI). Both PTI and ETI mechanisms are tightly connected with reactive oxygen species (ROS) production and disease resistance that involves distinct biphasic ROS production as one of its pivotal plant immune responses. This unique oxidative burst is strongly dependent on the resistant cultivars because a monophasic ROS burst is a hallmark of the susceptible cultivars. However, the cause of the differential ROS burst remains unknown. In the study here, we revealed the plausible underlying mechanism of the differential ROS burst through functional understanding of the Magnaporthe oryzae (M. oryzae) AVR effector, AVR-Pii. We performed yeast two-hybrid (Y2H) screening using AVR-Pii as bait and isolated rice NADP-malic enzyme2 (Os-NADP-ME2) as the rice target protein. To our surprise, deletion of the rice Os-NADP-ME2 gene in a resistant rice cultivar disrupted innate immunity against the rice blast fungus. Malic enzyme activity and inhibition studies demonstrated that AVR-Pii proteins specifically inhibit in vitro NADP-ME activity. Overall, we demonstrate that rice blast fungus, M. oryzae attenuates the host ROS burst via AVR-Pii-mediated inhibition of Os-NADP-ME2, which is indispensable in ROS metabolism for the innate immunity of rice. This characterization of the regulation of the host oxidative burst will help to elucidate how the products of AVR genes function associated with virulence of the pathogen.

  • 시장에는 프로바이오틱스를 포함한 무수히 많은 제품이 판매되고 있는데, 우리에게 어떤 이로운 점을 줄 수 있는지 궁금해 한 적이 있었을 것이다. 프로바이오틱스는 기본적인 영양 측면을 배제하고, 우리가 유효한 양을 섭취하였을 때 건강상의 이점을 주는 살아있는 미생물로 정의된다. 프로바이오틱스는 유용하고 건강에 좋은 미생물로도 불려지고 있으며, 다음과 같은 다섯 가지 측면에서 건강 기능성이 있는 것으로 강조되어 왔는데, 1) 대장암 및 IBS와 같은 다른 대장(결장)관련 질병의 발병율 감소, 2) 면역 시스템의 촉진, 3) 항고혈압 및 항콜레스테롤 작용, 4) 장내세균에 작용하는 항생제의 효과를 경감시키고, 5) 위장관 감염을 예방시키는 점이다. 그러나 이러한 건강기능 작용들에 대한 과학적인 근거가 충분히 구축되어 있지는 않아, 유럽식품 위생위원회(Euroupe Food Safety Authority)는 항생제 투여에 따른 설사증상의 완화와 같은 건강기능 표시를 금지하고 있어, 프로바이오틱스의 작용기전에 대한 연구가 필요한 시점이라 하겠다.

MoJMJD6, a Nuclear Protein, Regulates Conidial Germination and Appressorium Formation at the Early Stage of Pathogenesis in Magnaporthe oryzae

  • In plant-pathogen interactions, Magnaporthe oryzae causes blast disease on more than 50 species of 14 monocot plants, including important crops such as rice, millet, and most 15 recently wheat. M. oryzae is a model fungus for studying plant-microbe interaction, and the main source for fungal pathogenesis in the field. Here we report that MoJMJD6 is required for conidium germination and appressorium formation in M. oryzae. We obtained MoJMJD6 mutants (ΔMojmjd6) using a target gene replacement strategy. The MoJMD6 deletion mutants were delayed for conidium germination, glycogen, and lipid droplets utilization and consequently had decreased virulence. In the ΔMojmjd6 null mutants, global histone methyltransferase modifications (H3K4me3, H3K9me3, H3K27me3, and H3K36me2/3) of the genome were unaffected. Taken together, our results indicated that MoJMJD6 function as a nuclear protein which plays an important role in conidium germination and appressorium formation in the M. oryzae. Our work provides insights into MoJMJD6-mediated regulation in the early stage of pathogenesis in plant fungi.

녹농균(Pseudomonas aeruginosa)의 쿼럼 센싱 수용체인 QscR의 활성에 영향을 미치는 아미노산 잔기 분석 (Analysis of Amino Acid Residues Affecting the Activity of QscR, a Quorum Sensing Receptor of Pseudomonas aeruginosa)

  • 그람 음성균인 녹농균(Pseudomonas aeruginosa)은 다양한 환경에 존재하는 기회감염성 병원균으로, 병원성의 발현에 쿼럼센싱(QS) 기전이 중요한 역할을 담당한다. 녹농균의 여러 QS 신호물질 수용체들 중 하나인 QscR은 다른 QS 수용체들과는 구분되는 특별한 특성들을 가진다. 본 연구에서는 이러한 특성들 중 특히 넓은 신호물질 특이성을 QscR에 부여해 주는 아미노산 잔기가 무엇인지 알아보기 위해, QscR의 72번째 threonine, 132번째 arginine, 140번째 threonine 잔기가 각각 isoleucine, methionine, isoleucine 잔기로 치환된 돌연변이 QscR들($QscR_{T72I}$, $QscR_{R132M}$, $QscR_{T140I}$)을 제조하였다. 이들의 활성을 측정해 보았을 때 $QscR_{R132M}$은 N-3-oxododecanoyl homoserine lactone (3OC12-HSL)에 대한 반응성이 사라졌고, $QscR_{T72I}$$QscR_{T140I}$는 민감성이 많이 감소하기는 하였으나 여전히 3OC12-HSL에 대한 반응성을 가지고 있었다. 이들 돌연변이 QscR들에 다양한 구조의 acyl-HSL을 처리해 보았을 때, $QscR_{T72I}$$QscR_{T140I}$는 야생형 QscR처럼 자기 자신의 신호물질인 3OC12-HSL 보다 N-decanoyl HSL (C10-HSL)이나 N-dodecanoyl HSL (C12-HSL)처럼 10개 혹은 12개의 탄소 사슬을 가지면서 3번째 탄소에 oxo-moiety가 없는 acyl-HSL에 대해 더 높은 반응성을 보였으며, $QscR_{R132M}$은 3OC12-HSL 뿐만 아니라 본 연구에서 사용된 어떤 acyl-HSL에도 반응성을 보이지 않았다. 또한 $QscR_{T72I}$$QscR_{T140I}$는 QscR 억제제인 5f에 의해 야생형 QscR과 비슷한 수준으로 활성이 억제되었다. 이러한 결과들은 130번째 arginine의 경우 QscR의 활성과 acyl-HSL들과의 결합에 중요한 역할을 하는 반면, 72번째와 140번째 threonine들의 경우 QscR의 활성에는 중요하지만, 다른 구조의 acyl-HSL들에 대한 선택적 결합이나, 경쟁적 억제자들의 결합 간섭에는 영향을 주지 않음을 시사하는 것이다.
