  • A random study of 574 dairy farms in Gyeongnam area was designed to determine 1) management factors that may be associated with the occurrence of drug residues; 2) the dairy farmer's attitudes and knowledge about residues; 3) how these variables influence the occurrence of residues in dairy cattle. Management factors perceived as having the greatest influence on drug residues in milk were insufficient knowledge about withdrawal periods, errors due to hired help, insufficient identification and record of animals treated for mastitis, dry cow treatment for mastitis, and metritis treatment. Seventy-one percent of farms with residues problem used mixed own feeds compared with twenty-nine percent of farms with premedicated feeds. Factors significantly associated with the occurrence of residues were herd size, increased number of hired persons, increased frequency of use of mixed-own feeds, category of medicated feed, and producer's attitude toward the public health significance of residues. Our findings suggest that residue occurrence was mainly associated with errors due to hired help, insufficient knowledge about withdrawal periods, poor animal identification and records of treatment animals and use of medicated feeds. Any residue avodiance educational program needs to stress how to deal with these factors. This educational program should be directed to dairy farmers and employees, especially temporary employees. In addition, Dairy farmer's attitudes and knowledge about drug residues need to be improved. More evidence on the public healths significance of residues should be available to them. Because belife in importance of public healths concerns was related to successful residue avidance and because 81.3% of the dairy farmers with residue problem thought public healths concerns were less important than economic ones, it would be helpful to provide educational programs specifically directed to this issue. it may be useful to provide programs not only for the dairy farmers but also change of their concerns about on the public healths.

Determination of Flunixin and 5-Hydroxy Flunixin Residues in Livestock and Fishery Products Using Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS)

  • Flunixin is a veterinary nonsteroidal anti-inflammatory agent whose residues have been investigated in their original form within tissues such as muscle and liver. However, flunixin remains in milk as a metabolite, and 5-hydroxy flunixin has been used as the primary marker for its surveillance. This study aimed to develop a quantitative method for detecting flunixin and 5-hydroxy flunixin in milk and to strengthen the monitoring system by applying to other livestock and fishery products. Two different methods were compared, and the target compounds were extracted from milk using an organic solvent, purified with C18, concentrated, and reconstituted using a methanol-based solvent. Following filtering, the final sample was analyzed using liquid chromatography-tandem mass spectrometry. Method 1 is environmentally friendly due to the low use of reagents and is based on a multi-residue, multi-class analysis method approved by the Ministry of Food and Drug Safety. The accuracy and precision of both methods were 84.6%-115% and 0.7%-9.3%, respectively. Owing to the low matrix effect in milk and its convenience, Method 1 was evaluated for other matrices (beef, chicken, egg, flatfish, and shrimp) and its recovery and coefficient of variation are sufficient according to the Codex criteria (CAC/GL 71-2009). The limits of detection and quantification were 2-8 and 5-27 ㎍/kg for flunixin and 2-10 and 6-33 ㎍/kg for 5-hydroxy flunixin, respectively. This study can be used as a monitoring method for a positive list system that regulates veterinary drug residues for all livestock and fisheries products.

LC-MS/MS를 이용한 농산물 중 살균제 Spiroxamine의 시험법 개발 및 검증 (Development and Validation of Analytical Method for Determination of Fungicide Spiroxamine Residue in Agricultural Commodities Using LC-MS/MS)

  • 본 연구는 농산물 중 2017년 기준신설예정농약 spiroxamine에 대한 공정시험법을 개발하고자 수행하였다. Spiroxamine은 모화합물만이 잔류물로 정의되므로 이를 분석하기 위한 시험법을 개발하였고, 확립된 시험법은 실험실 내 및 실험실 간 검증을 통해 공정시험법으로의 유효성을 확인하였다. Spiroxamine의 물리화학적 특성을 고려하여 분석을 위해 LC-MS/MS를 분석기기로 사용하였으며, acetonitrile으로 추출 후 유기용매를 이용하여 액액분배조건 및 florisil cartridge를 이용한 SPE 정제조건을 확립하여 시료의 불순물을 효과적으로 정제하는 시험법을 개발하였다. 개발된 Spiroxamine 시험법의 직선성은 결정계수($r^2$)가 0.99 이상으로 우수하였으며, 검출한계 및 정량한계는 각각 0.0005, 0.001 mg/kg으로 높은 감도를 나타내었다. 대표 농산물 5종(현미, 감자, 대두, 감귤, 고추)에 대한 시험법 검증결과 평균 회수율(5 반복)은 71.9~98.8%였고, 분석오차는 10.0% 이하로 정확성 및 재현성이 우수함을 확인할 수 있었다. 또한 광주지방식약청과의 실험실 간 검증 결과에서 평균 회수율은 70.6~104.6%를 나타내어 모두 Codex 가이드라인 및 식약처 가이드라인의 기준에 부합하는 것으로 확인되었다. 개발된 시험법은 낮은 검출한계 및 정량한계, 우수한 직선성, 회수율 시험을 통한 양호한 정밀성 및 재현성 등이 입증되어 농산물 중 Spiroxamine의 분석을 위한 식품공전 공정시험법으로 활용되기에 적합한 것으로 판단된다.

GC-ECD를 이용한 농산물 중 제초제 aclonifen의 공정분석법 확립 (Development of an Official Analytical Method for Determination of Aclonifen in Agricultural Products Using GC-ECD)

  • 이 연구는 농산물 중 아클로니펜의 안전관리를 위한 공정분석법을 확립하기 위하여 수행하였으며 분석법의 선택성, 검출한계 및 정량한계, 회수율에 대한 검증을 통하여 아클로니펜의 공정시험법으로의 유효성을 확인하였다. 아클로니펜의 이화학적 특징에 따라 분석기기는 GC-ECD를 사용하였고, 아세톤 추출 후 실리카 카트리지를 이용하여 정제하였다. 개발된 분석법의 회수율은 고추에서 92.3-95.0%, 감귤에서 74.3-90.4%, 아몬드에서 76.7-89.8%였으며, 분석오차는 8%이하로 코덱스 가이드라인 (CAC/GL 40)에 적합하였다. 또한 GC-MS를 이용한 재확인 과정을 통해 분석법의 신뢰성을 확보하였다. 따라서 본 분석법의 정확성 및 재현성이 입증되어 농산물 중 아클로니펜의 잔류분석에 적용할 수 있을 것으로 판단된다.

HPLC-UVD를 이용한 농산물 중 Imazapyr의 공정분석법 확립 (Development of an Official Analytical Method for Determination of Imazapyr in Agricultural Commodities using HPLC-UVD)

  • 농산물 중 잔류농약의 안전관리를 위해 2014년 신규 기준설정 농약인 imazapyr의 분석법을 개발하였고 분석법의 정확성, 재현성, 정밀성, 선택성, 직선성에 대한 검증을 통하여 잔류농약 분석을 위한 공정분석법으로서의 유효성을 검증하였다. Imazapyr는 imidazolinone계 화합물로 아미노산의 생성을 억제하여 잡초의 생장을 방제하는 제초제이다. Imazapyr는 CODEX를 포함한 미국, 일본 등에서 잔류허용기준(MRL)이 설정되어 있으며, 우리나라에서도 대두에 대해 잔류허용기준을 설정할 예정이다. 비록 기준은 대두에 우선 설정되지만 농산물 전반에 대한 안전관리를 위해 곡류, 콩류, 채소류, 과일류, 서류에 대한 분석법를 개발하였고, 각 농산물군의 대표작물인 대두, 고추, 감귤, 현미, 감자 시료에 대해 분석법을 개발하였다. 시료 중 imazapyr를 methanol을 이용해 추출하고 dichloromethane으로 액-액 분배하여 극성 간섭물질을 제거하였으며, 현미 및 대두 등 유지시료의 경우 n-hexane/acetonitrile 분배법을 적용하여 유지성분을 추가로 제거하였다. 분석기기로는 Imazapyr의 낮은 증기압과 UV 파장의 빛에 대한 흡광성을 감안하여 HPLC-UVD를 사용하였다. 확립된 분석법에서 검량선의 상관계수($r^2$)는 0.99% 이상으로 우수한 직선성을 보였으며, 5개 대표 농산물에 imazapyr를 3개 농도에서 5회 반복하여 수행한 결과 회수율은 72.1-108.0%이었고 분석오차는 10% 미만으로 잔류농약 분석을 위한 CODEX 가이드라인(CAC/GL 40, 2003)에 적합하였다. 본 연구에서 확립된 분석법은 식품 중 imazapyr의 안전관리를 위한 공정분석법으로 사용될 것이다.

국내 유통 식품의 잔류동물용의약품 모니터링 (Monitoring of Veterinary Drug Residues in Foods Produced in Korea)

  • 알(卵)류 및 축 수산물 321건에 대하여 엔로플록사신, 시프로플록사신, 노르플록사신, 아목시실린, 암피실린, 옥소린산 및 옥시테트라싸이클린, 테트라싸이클린, 클로르테트라싸이클린 및 독시싸이클린에 대한 모니터링을 실시한 결과 총 300건(93.5%)은 조사대상 동물용의약품이 불검출되었으며 검출된 21건(6.5%)의 시료 대부분은 현재 식품공전의 잔류허용기준 이하로 안전한 것으로 나타났다.

  • Great attention form a public health aspect has centered on the safety of tissues residues of veterinary drugs and environmental contaminants, with intensification of animal husbandry, the use of veterinary drugs will become increasingly important. Heavy responsibility is placed on the veterinarian and livestock producer to observe the period for withdrawal of drug prior to slaughter to assure that illegal concentrations of drug residues in meat, milk and egg do not occur. Every nation has their own regulations in relation to the residues and the guidelines on the use of veterinary drugs to fulfil the regulations, and their own national residue programs to monitoring and eliminate illegal products. Good practice of veterinary drug by users and governmental surveillance programs are very important to ensure animal food safety.

Application of ELISA for the Detection of Penicillin Antibiotic Residues in Live Animal

  • Penicillin antibiotics such as penicillin G, ampicillin and amoxicillin have been widely used in the pig industry to control salmonellosis, bacterial pneumonia, and urinary tract infections. Extensive use of antibiotics in veterinary clinics has resulted in tissue residues and bacterial resistance. To prevent unwanted drug residues entering the human food chain, extensive control measures have been established by both government authorities and industries. The demands for reliable, simple, sensitive, rapid and low-cost methods for residue analysis of foods are increasing. In this study, we established a rapid prediction test for the detection of pigs with unacceptable tissue residues of penicillins. The recommended therapeutic doses of three penicillins, penillin G (withdrawal time, 7 days), ampicillin (withdrawal time, 7 days) and amoxicillin (withdrawal time, 14 days), were administered to three groups of 20 pigs each. Blood was sampled before drug administration and during the withdrawal period. The concentration of penicillins in plasma, determined by a semi-quantitative ELISA, were compared to that of internal standard, 4 ppb, which corresponded to the Maximum Residue Limit in milk. The absorbance ratio of internal standard to sample (B/Bs) was employed as an index to determine whether drug residues in pig tissues were negative or positive. That is, a B/Bs ratio less than 1 was considered residue positive, and larger than 1 negative. All 60 plasma samples from pigs were negative to three penicillins at pretreatment. Penicillin G could be detected in the plasma of the treated pigs until day 4 post-treatment and ampicillin until day 2, whereas amoxicillin could be detected until day 10 of its withdrawal period. The present study showed that the semi-quantitative ELISA could be easily adapted to detect residues of penicillin antibiotics (penicillin G, ampicillin and amoxicillin) in live pigs.

Development of Analytical Method and Monitoring of Veterinary Drug Residues in Korean Animal Products

  • This study was conducted to determine the residual amount of veterinary drugs such as meloxicam, flunixin, and tulathromycin in animal products (beef, pork, horsemeat, and milk). Veterinary drugs have been widely used in the rearing of livestock to prevent and treat diseases. A total of 152 samples were purchased from markets located in major Korean cities (Seoul, Busan, Incheon, Daegu, Daejeon, Gwangju, Ulsan and Jeju), including Jeju. Veterinary drugs were analyzed by liquid chromatography-tandem mass spectrometry according to the Korean Food Standards Code. The resulting data, which are located within 70-120% of recovery range and less than 20% of relative standard deviations, are in compliance with the criteria of CODEX. A total of five veterinary drugs were detected in 152 samples, giving a detection rate of approximately 3.3%; and no food source violated the guideline values. Our result indicated that most of the veterinary drug residues in animal products were below the maximum residue limits specified in Korea.

  • The residue of drug in foods of animal origin has increasingly become of interest to the entire livestock industry as growing consumer health concerns. The current overvie of feed additives and veterinary drugs used in Japan and their residual analysis has been reviewed. High performance liquid chromatographic technique(HPLC) with various detectors can be expected to be successfully applied for the routine analysis of residual feed additives and veterinary drugs including anabolic agents in livestock products.
