RETINOL STABILIZATION BY PSEUDO-LIPOSOME AND LAMELLAR LIQUID CRYSTAL

  • It is well known that all-trans-retinol is not only very unstable in heat, light, air, and water, but also skin-irritant despite a good anti-wrinkle effect. Therefore, it is very difficult to stabilize retinol and make the safe retinol containing cosmetics by using a certain concentration of retinol with real effect. In order to dissolve these problems and apply retinol for skin care cream, firstly retinol is to be encapsulated in the vesicle called Liposphere (pseudo-liposome) which is made by homogenizing under high pressure the mixtures of lecithin, retinol, caprylic/capric triglyceride, and hydroalcoholic solution ; and then this retinol containing Liposphere is to be intercalated in lamellar liquid crystal layer which is prepared by emulsifying in an optimal ratio the mixtures composed of non-ionic emulsifier (cetearyl glucoside, sorbitan stearate & sucrose cocoate etc), cetearyl alcohol, stearic acid, cholesterol, and ceramide. In addition, the stability of the retinol containing oil in water cream by adding the polymeric emulsifier such as acrylate /C10-30 alkyl alkylate crosspolymer is to be ensured even at 55 C. Retinol containing oil in water cream prepared through above procedure could be very stable at 45 C for at least 50 days. The structure identification of lamellar liquid crystal was determined using polarized light microscope and electron microscope Conclusively, we could make the very stable retinol containing oil in water cream by triple procedure, that is, encapsulation of retinol in Liposphere, intercalation of retinol in lamellar liquid crystal layer, and assurance of the high temperature stability of cream even at 55 C.

  • An X-linked skeletal disorder, SEDT (spondyloepiphyseal dysplasia tarda) is a genetic disease characterized by a disproportionately short trunk and short stature caused by mutations in the SEDL gene. This gene is evolutionarily conserved from yeast to human. The yeast SEDL protein ortholog, Trs20p, has been isolated as a member of a large multi-protein complex called the transport protein particle (TRAPP), which is involved in endoplasmic reticulum (ER)-to-Golgi transport. The interaction between SEDL and partner proteins is important in order to understand the molecular mechanism of SEDL functions. We isolated several SEDL-binding proteins derived from rat cells by an immobilized GST-fusion method. Furthermore, the SEDL-homologue proteins were identified using motif based methods. Common motifs between SEDL-binding proteins and SEDL-homologue proteins were classified into seven types and 78 common motifs were revealed. Sequence similarities were contracted to seven types using phylogenetic trees. In general, types I-III and VI were classified as having the function of acetyl-CoA carboxylase, glycogen phosphorylase, isocitrate dehydrogenase, and enolase, respectively, and type IV was found to be functionally related to the GST protein. Types V and VII were found to contribute to TRAPP vesicle trafficking.

Maturation Induction in vitm of Rana dybowskii Oocyte by Lantlianum Ion (Lanthanum 이온에 의한 북방산개구리(Rana dybowskii) 여포난자의 성숙유도)

  • The effect of lanthanum ion (La3 +), which is associated with the mobilization of internal calcium, on the regulation of oocyte maturation was investigated with Rana dybowskii follicles. Follicular oocytes matured (germinal vesicle breakdown, GVBD) dose dependently when they were exposed to La3+ (O.O1-1.O mM) and the maturation occurred in 9-12 hours after the la3+(0.33 mM) stimulation. lanthanum also accelerated the onset of maturation of the lollicular oocytes exhibiting spontaneous maturation. Three hours of exposure to La3+ was enough to induce the maturation. The La3 + -induced maturation was not associated with progesterone production by follicle cells, and the maturation was inhibited by forskolin (9 $\mu$ M), and cyclobeximide (0.01 - 1.0 - $\mu$g/2 ml) in the medium. The La3+ and hormone stimulated maturation showed the same patterns of protein phosphorylation and dephosphorylation during the maturation. The data suggest that the oocyte maturation by La3+ stimulation is very similar to that by progesterone. Thus, it seems that internal mobilization of Ca2+ plays a key role in the initiation of oocyte maturation in amphibia.

The Ultrastructure of the Cutaneous Pigment Cells in Rana nigromaculata coreana Okada (금개구리 피부 색소세포의 미세구조)

  • The authors observed pigment cells in the dorsal skin of Korean frog (Rana nigromaculata coreana Okada) under an electron microscope. The traits of pigment cells were as follows. The three kinds of dermal chromatophores were horizontally expanded; adjacent closely contiguous and partially overlapped with one another and there were the intercellular space between a xanthophore and an iridophore, or an iridophore and a melanophore. 1. Xanthophores: Xanthophores were filled with pterinosomes and carotenoid vesicles, the formers of which were evenly distributed in each cell. Pterinosomes were five kinds of types (type I, type II, type III, type IV and type V pterinosomes). Especially typical type II and type III pterinosomes were well observed in the cytoplasm. 2. Iridophores: Xanthophores were closely contiguous to the xanthophore situated above and they were filled with reflective platelets. Mostly these platelets are arranged regularly, that is, in parallel with the surface of skin, but partly, nonparallel. 3. Melanophores: Melanophores were filled with melanin granules, each of which revealed the same electron density. The pocesses of melanophore entended to bilateral sides of iridophores and melanin granules were dispersed especially in the processes.

Induction and Inhibition of Amphibian(Rana dybowskii) Oocyte Maturation by Proteolytic Enzymes In vitro. (단백질분해효소들의 양서류 난자에 대한 성숙유도와 억제작용에 관하여)

  • Fully grown amphibian oocytes undergo their maturation (germinal vesicle breakdown, GVBD) during in vitro follicle culture when they are stimulated with frog pituitary homogenate (FPH) or progesterone. Present experiments were designed to determine whether proteolytic enzymes are involved in the regulation of the matunation process. Treatment of a $\alpha$ -chymoiyypsin inhibitor, N a -tosyl-L-phenylalanine-chloromethyl-ketone(TP) to the oocytes exhibited a biphasic phenomenon, the induction of the maturation without added hormone at relatively low doses (0.001-1 $\mu$M) and inhibition of the hormone induced oocyte maturation at a high dose (100 $\mu$M). Treatment of a trypsin inhibitor, N a -tosyl-L-lysine-chloromethyl ketone(TLCK) to the oocytes did not induce the maturation, but rather suppressed the hormone induced oocyte maturation in a high dose(100 $\mu$ M). Treatment of exogenous iyypsin to the oocyte induced their maturation without added hormone in a dose dependent manner (0.001-1 $\mu$ M). The data presented here indicate that some proteolytic enzymes play a role in the regulation of the maturation(meiotic arrest or reinitiation) in amphibians.

  • Ultrastmctura changes of multivesicular bodies and mitochondria of oocytes of PseudopotamU- Ia occelata Moore were examined with transmission electron microscope in order to follow the process of yolk formation. Yolk granules begin to form at the previtellogenic stages of 50 $\mu$m diameter from multivesicular bodies. Small vesicles and membranous structures within the multivesicular bodies are fused to form the precursors of core bodies of yolk granules. Some vesicles from cytoplasm are also coalesced into the multivesicular bodies. Mature yolk granules are composed of electron-dense core bodies which are seperated from each other by electronopaque small vesicle-like structures. Structural changes of cristae into vesicular shapes and increase in electron density of matrix in mitochondria strongly suggested that mitochondria are in the process of transformation. The transformed mitochondria appear to be basic structures which later become multivesicular bodies.

Comparison of DNase activities from excretory/secretory productsof Haemonchus contortus fenbendazole-resistantand -susceptible isolates (Fenbendazole에 저항성과 감수성을 지닌 염전위충의 분비배설물에서의 DNase 활성 비료)

  • Change in ${\beta}$-tubulin nucleic acid and protein sequences was the only known difference between Haemonchus contortus fenbendazole (FBZ)-resistant and -susceptible isolates. This change was sufficient to determine the pathologic effect induced by FBZ treatment. This research was initiated to investigate further differences from these two isolates. Since ${\beta}$-tubulin is involved in formation of microtubule, which has functions in secretory vesicle transport, DNase activities from excretory/secretory products (ESP) of the two isolates were compared, based on pH, sensitivity to DNase inhibitors, molecular masses and production of 3'-OH. The most significant difference detected was that a 38.5 kDa DNase activity was identified from ESP of H. contortus FBZ-susceptible isolates but not from those of H. contortus FBZ-resistant isolates. However, it was shown that the 38.5 kDa DNase is expressed with similar level of activity in intestine and whole worm of H. contortus FBZ-resistant and -susceptible isolates. This result demonstrated that the secretory transport pathway of the 38.5 kDa DNase was inhibited by unknown mechanisms, which may be related with ${\beta}$-tubulin sequence change in FBZ-resistant isolates. Other DNases of 34, 36 and 37 kDa were detected from ESP of both H. contortus FBZ-resistant and -susceptible isolates. Overall DNase activities found from ESP of these two isolates were not inhibited by 10 mM EDTA at pH 5.0, but largely inhibited by pH 7.0. In addition, DNase activities in two isolates produced DNA fragments with mixtures of 3'- hydroxyls (OH) and 3'-phosphates (P) at each pH although the 3'-end labeling ratios at pH 5.0 and 7.0 were shown different. Identification of inhibition of the 38.5 kDa DNase secretion in FBZ-resistant isolates suggests existence of further differences, in addition to ${\beta}$-tubulin sequence change, in two isolates. This shows complex effect of FBZ on H. contortus biological mechanisms.

The Effect of n-Alkanols on the Lateral Diffusion of Synaptosomal Plasma Membrane Vesicles Isolated from Bovine Cerebral Cortex (n-Alkanols가 소의 대뇌피질로부터 분리한 Synaptosomal Plasma Membrane Vesicles의 측방확산운동 범위와 속도에 미치는 영향)

  • Intramolecular excimer formation with the fluorescent probe 1,3-di(1-pyrenyl)propane (Py-3-Py) was used to investigate the effects of methanol, ethanol, 1-propanol, 1-butanol, 1-pentanol, 1-hexanol, 1-heptanol, 1-octanol, 1-nonanol and 1-decanol on the lateral diffusion of synaptosomal plasma membrane vesicles isolated from bovine cerebral cortex (SPMV). The n-alkanols increased the excimer to monomer fluorescence intensity ratio (I'/I) of Py-3-Py in the SPMV. In a dose-dependent manner, n-alkanols increased lateral diffusion of hydrocarbon region of bulk (inner+outer monolayers) SPMV lipid bilayers, and the potencies of n-alkanols up to l-nonanol increased with carbon chain length. It appears that the potencies in bilayer fluidization due to the lateral diffusion increase by 1 order of magnitude as the carbon chain length increases by two carbon atoms. The cut-off phenomenon was reached at 1-decanol, where further increase in hydrocarbon length resulted in a decrease in pharmacological activity.

  • The influences of fluorescence, scattering, and flocculation in random media were interpreted for the scattered fluorescence intensity and wavelength, it has been studied the molecular properties by the spectroscopy of laser induced fluorescence(LIF). The effects of optical properties in scattering media have been found by the optical parameters(${\mu}_s$, ${\mu}_a$, ${\mu}_t$). Flocculation is an important step in many solid-liquid separation processes and is widely used in Photodynamic therapy. The interactions of several colloid particles can come into play which have major effect on the flocculation and LIF process. We measured scattering and fluorescence spectra of the sample in vitro as function of distance from lase source to detector. The value of scattering coefficient ${\mu}_s$ is and ${\beta}$-carotene were measured as larger values(I, ${\delta}$) by means of closer distance from source to detector.

In vitro Effects of TBT, TPhT and Aroclor 1254 on Oocyte Maturation and Ovulation in O1ive Flounder (Paralichthys olivaceus) (넙치 (Paralichthys olivaceus)의 in vitro 난모세포 성숙과 배란에 미치는 TBT, TPhT 및 Aroclor 1254의 영향)

  • The effects of tributyltin (TBT), triphenyltin (TPhT) and Aroclor 1254 on germinal vesicle breakdown (GVBD) and ovulation of olive flounder (Paralichthys olivaceus) were investigated in in vitro bioassay. TBT, TPhT and Aroclor 1254 showed the inhibition effects on GVBD and ovulation in response to HCG. The oocyte response appeared to be more sensitive to TBT than Aroclor 1254. TBT was more effective in inhibition GVBD at concentrations of 0.1 and 1 ppm. However, no significant inhibition was obseued in concentrations tested ($0.0001\~1\;ppm$). Significant inhibition of ovulation in response to HCG occurred at TBT (0.01, 0.1, 1 ppm), TPhT (0.01, 0.1, 1 ppm) and Aroclor 1254 (0.01, 1 ppm, except 0,1 ppm), compared to HCG control, The lowest ovulation rate was measured at 1 ppm TBT, These data suggest that TBT (or TPhT) could possibly interfere the actions of progestogens to induce GVBD and ovulation in in vitro bioassay system.
