Biochemical Properties and Localization of the β-Expansin OsEXPB3 in Rice (Oryza sativa L.)

  • ${\alpha}$-Expansins are bound to the cell wall of plants and can be solubilized with an extraction buffer containing 1 M NaCl. Localization of ${\alpha}$-expansins in the cell wall was confirmed by immunogold labeling and electron microscopy. The subcellular localization of vegetative ${\beta}$-expansins has not yet been studied. Using antibodies specific for OsEXPB3, a vegetative ${\beta}$-expansin of rice (Oryza sativa L.), we found that OsEXPB3 is tightly bound to the cell wall and, unlike ${\alpha}$-expansins, cannot be solubilized with extraction buffer containing 1 M NaCl. OsEXPB3 protein could only be extracted with buffer containing SDS. The subcellular localization of the OsEXPB3 protein was confirmed by immunogold labeling and electron microscopy. Gold particles were mainly distributed over the primary cell walls. Immunohistochemistry showed that OsEXPB3 is present in all regions of the coleoptile and root tissues tested.

  • D10 values obtained for radiation alone in Bacillus subtilis and Clostridium perfrigenes were 0.35-0.48 kGy in vegetative cells, and 2~2.08 kGy in spores, respectively. Irradiation dose of 24 kGy completely inhibited spores. In the case of heat treatment, D50, 60 values ranged from 10 to 14 minutes in vegetative cells, and D70, 80, 90 values ranged from 10 to 140 minutes in spores. In the case of combined treatment with heat and radiation, D10 values ranged form 1 to 1.25 kGy in vegetative cells, and from 3.42 to 3.61 kGy in spores. Thus, resistance of cells to gamma radiation did not seem to be influences by pre-heating.

Helicobactericidal Activity of Cissus quadrangularis L. Variant I

  • Cissus quadrangularis L. variant I (Family: Vitaceae), the common variant with square stem is widely used for peptic ulcer disorders (PUD) in traditional medicine. Aerial parts were collected during flowering and vegetative seasons and analysed. Aqueous (hot and cold) and solvent extracts (acetone, chloroform and ethanol) were screened for their anti-Helicobacter pylori (Hp) activities. Among them chloroform extract was observed to recover bioactive principles markedly with low minimal inhibitory concentration (MIC) and minimal lethal concentration (MLC). MIC was $30\;{\mu}g$ in both samples and MLC was $35\;{\mu}g$ for vegetative and $30\;{\mu}g$ for flowering seasons, respectively. Extracts from samples collected during flowering season were better than thse of vegetative season.

  • Polysiphonia senticulosa Harvey (Ceramiales: Rhodophyta) was recorded for the first time in Korea, and its vegetative and tetrasporangial morphologies are described. This species is characterized by the combined features of having a vegetative axial segment with 4 pericentral cells, a main axis without cortication, pseudodichotomous branching, branches with sharply pointed apical cells, flexuous branches hooked backward below, the presence of cicatrigenous branches, axillary endogenous stichidia, and the production of 1-3 tetrasporangia per stichidium. Based on this study, it is distinguished from Polysiphonia morrowii Harvey by its flexuous branches and the number of tetrasporangia produced per stichidium.

Unusual Rhizoidal Development in Bangia (Bangiales, Rhodophyta) -Another Form of Vegetative Reproduction?

  • The gametophytic filaments of two genetically distinct taxa of Bangia from New Zealand showed unusual rhizoidal development in comparative culture experiments. In the past Bangia has been reported to possess simple, colourless rhizoids that extend from the basal cells of the unbranched filaments, whereas in this study the rhizoids observed became pigmented and multicellular. A reversal of growth direction occurred and filamentous extensions developed from the rhizoids under some culture conditions. These extensions were either prostrate or resembled new gametophytic filaments. This is the first report for filamentous members of the Bangiales of the development of such stolon-like rhizoids, apparently serving as a form of vegetative reproduction.

Listeria monocytogenes의 열저항성에 미치는 pH와 Nisin의 효과 (Effect of pH and Nisin on Heat Resistance of Listeria monocytogenes Scott A)

  • The effect of pH (7, 5 and 4) and nisin (100 and 200IU/ml) on heat resistance of Listeria monocytogenes Scott A were determined using citrate-phosphate buffer system. Heat resistance of vegetative and starved cell was decreased as pH value was lower at 65 and 72C. Starved L. monocytogenes was more resistant than vegetative cell at both temperature. Heat resistance of vegetative and starved cell was decreased significantly with treatment of nisin. The effect of nisin was increased significantly at low pH(5, 4). Adherent microcolony was more resistant to heat and nisin than planktonic cell. Contamination of L. monocytogenes may be prevent by using nisin in food and food processing environments.

Antimicrobial Activity of Hemidesmus indicus var.indicus R.Br. Against Human Isolates of Helicobacter pylori

  • Hemidesmus indicus var. indicus belonging to the family Asclepiadaceae was screened for its activity against Helicobacter pylori(Hp) human isolates. Flowering and vegetative period samples were analysed. Aqueous (hot and cold) and solvent extracts (acetone, chloroform and methanol) were screened. Among them chloroform extract was observed to recover bioactive principles with low MIC and MLC. MIC was $75\;{\mu}g$ in both seasons and MLC was $100\;{\mu}g$ for vegetative and $75\;{\mu}g$ for flowering periods respectively. Extracts from samples collected during flowering period was better than that of vegetative period.

  • Single ascospore isolates of Cordyceps bassiana were observed for their colony pigmentation on Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates and were inoculated in a brown rice medium for production of fruiting bodies. Colony pigmentation did not show any relationship with perithecial stromata formation. The isolates were also grown on opposite sides of SDAY agar plates and were observed for vegetative compatibility. Neither vegetative compatibility nor perithecial stromata could be found to be related to each other. It was concluded that fertile fruiting body production was independent of colony pigmentation and vegetative compatibility. Synnemata formation was found to be more common than perithecial stromata formation. This might be due to its highly conidiogenous anamorphic stage, i.e., Beauveria bassiana.

Vegetative endocarditis associated with Arcanobacterium haemolyticum in a Holstein cow: a case report

  • A 4-year-old Holstein cow with progressive atrophy and ataxia was submitted for diagnosis. The postmortem examination revealed multifocal yellowish nodules in the pulmonary parenchyma and vegetative masses in the mitral and tricuspid valve of the heart. Both kidneys were severely enlarged, with multiple yellow nodules on the parenchyma. Histopathologically, pulmonary abscesses, vegetative endocarditis, suppurative glomerulonephritis, and fibrino-purulent arthritis were observed. The tiny β-hemolytic bacterial colonies were isolated from the lesions and identified as Arcanobacterium haemolyticum by the VITEK 2 system (bioMérieux, USA). This is the first documented report of an A. haemolyticum infection in a Holstein cow in Korea.

하천식생 이입현상의 원인 별 유형화 및 연구 방향 (Cause-based Categorization of the Riparian Vegetative Recruitment and Corresponding Research Direction)

  • 본 연구는 충적하천의 식생이입 현상, 즉 경관생태적으로 하천이 화이트리버에서 그린리버로 변하는 현상을 원인 별로 구분하고, 구분된 각각의 유형을 설명하고, 앞으로의 연구방향을 제시하는 것이다. 문헌연구와 과거 필자들이 제한된 범위에서 시행한 관련 국내 연구성과에 의하면 사주 상 하천식생 이입과 퇴행의 필요/충분 조건은 기계적 교란 (하상소류력), 토양습윤(지하수위, 지형, 하상재료 구성, 강수 등), 침수기간, 극단적 기후, 그리고 영양염류의 유입 등이다. 본 연구에서 사주 상 하천식생 이입과 퇴행의 원인 별 유형으로서 부분적으로 입증된 가설인 봄철 강우양상 변화로 인한 비조절하천에서 홍수 저감과 하천에 영양물질 유입증가 유형을 추가하였다. 하천식생 이입과 퇴행 현상을 과학적으로 더 규명하고 실제 이용가능한 하천식생모형의 개발을 위해서는 화이트리버가 그린리버로 바뀌는 현상의 전국적인 현황조사, 가설의 검증과 세분화를 통한 원인 별 유형 정립, 동적 하천식생 모델링 기법 개발, 변화에 따른 적응관리 등에 대해 연구가 필요하다.
