Contamination of Green Vegetable Juice by E. coli O157:H7 during Storage (E. coli O157:H7에 의한 녹즙 저장 환경에서의 미생물학적 오염도 조사)

  • The market for green vegetable juice (GVJ) is growing owing to the increasing demand for healthy food; however data on the safety and quality of GVJ are poorly reported. The objective of this study was to investigate the change in microbial community in GVJ during storage and its contamination by E. coli O157:H7. The microbial community was analyzed via culturable and non-culturable methods at 5, 10, and $25^{\circ}C$ for different storage times. In the non-culturable method, denaturing gradient gel electrophoresis (DGGE) was used. The initial bacterial concentration was $2.92{\times}10^5CFU/mL$, which exceeded the limit prescribed by the Korean Food Hygiene law. The results of the DGGE analysis indicated that the microbial community during storage was diverse and the spoilage lactic acid bacteria were prevalent at a later stage. Other bacteria such as Rahnella, Citrobacter, Pseudomonas, and Cyanobacteria were identified. Thus, the results strongly emphasize the need to pay attention to GVJ production safety, especially with respect to temperature control, in order to prevent the growth of foodborne pathogens such as E. coli O157:H7 and other spoilage bacteria.

Lupin Seed for Human Consumption (루우핀 콩의 식품이용)

  • The food quality of lupin seed, i.e. soaking, cooking, sprout growing and mold growing for fermentation, was investigated by using the seed of Lupinus angustifolius harvested in Western Australia. A method to produce lupin seed protein concentrate (LPC) was developed, and the wage of LPC in Korean food system was investigated. The water soaking rate of lupin seed was faster than that of soybean, but the cooking rate of lupin seed was much slower compared to soybean. The thermal softening time, $D_{100}$, was 345 min for lupin seed and 84 min for soybean. A two-phase solvent extraction system consisting of haxane-alcohol-water could effectively remove the residual bitter taste, lipid and yellow pigments of lupin seed flour, and the resulting LPC contained over 50% protein and had bland flavor and milky white color. Treatment of LPC with carbohydrate decomposing enzymes resulted in a product of more soluble and higher concentration of protein. Methods to produce lupin seed vegetable milk and lactic beverages from LPC products were discussed.

Effects of Supplementing Russian Comfrey and Kale on the Growth Rate, Nutrients Utilizability and Serum Cholesterol of Chicks (러시안 컴프리와 케일의 급여(給與)가 병아리의 성장률(成長率), 영양소(營養素) 이용률(利用率) 및 혈청(血淸) Cholesterol 함량(含量)에 미치는 영향(影響))

  • This experiment was conducted for 4 weeks to compare the feeding values of dried meal of Russian Comfrey and Kale as green feeds with Acacia leaf meal and Ladino clover meal, and to investigate the effects of feeding these on the total cholesterol in blood serum and liver fat content. Seventy-five male chicks of Single Comb White Leghorn were allotted into 5 treatments. Fifteen birds in each treatment were received Russian Comfrey meal, Kale meal, Acasia leaf meal, Ladino clover meal or no dried meal, respectively. The results obtained are summarized as follows: 1. No significant differences were recognized between the treatments and control in the growth rate, feed consumption, feed efficiency, utilization of nutrients, nitrogen retention and metabolizable energy value. 2. Carotene content in Russian Comfrey and Kale has been decreased as the growing stage advanced, and the leaves of both vegetable contained significantly (p<0.01) more carotene than that in stems. Vitamin C content of leaves was also signifcantly (p<0.01) higher than that in stems, however, vitamin C content has been increased as the growing stage advanced. 3. Toltal cholesterol content in blood serum of the chickens fed the Acacia leaf meal or Russian Comfrey meal was significantly (p<0.05) lower than Ladino clover meal fed group or those received no green feeds. Kale seems to decrease cholesterol content a little. Above results indicated that Acacia leaf and Russian Comfrey might contain certain substances that would depressing the cholesterol content. 4. No significant differences in the liver fat were discovered among the treatment but it was observed that liver fat was decreased as the cholesterol content increased.

Effects of Control Methods on Yields of Oriental Melon in Fields Infested with Meloidogyne arenaria (방제방법이 땅콩뿌리혹선충 밀도와 참외 수량에 미치는 영향)

  • The effect of cultural, physical and chemical control methods on the population density of Meloidogyne arenaria second-stage juveniles (J2) and on fruit yields of oriental melon was investigated at Seongju Fruit Vegetable Experiments Station, Kyungpook province, Korea, for two years from 1999 to 2000. Crops used in a rotation prior to Oriental melon were rice, corn, sesame, and green onion. The physical methods used were either solarization, soil addition or soil drying, and a nematicide, fosthiazate of granular formula, was used as the chemical method, applying at a rate of 300g a. i./10 a. Growing rice in the rotation, solarization, and soil addition controlled the nematode disease most effectively, reducing the number of J2 by 90% and increasing fruit yields two times. However, the effects of these control methods on the J2 population were limited to the early growing season; the J2 population increased later, suggesting that additional control practices may be needed in the following season. The next effective control methods were use of corn in the rotation, the nematicide application, and soil drying. The nematicide application was effective only for the early fruit yield, but neither for the late nor for the total yields. Use of sesame or green onion in the rotation was not effective in controlling the nematode.

  • In indian agricultural , the energy use pattern has played dominant role in influencing the level of mechanization . Besides that the agro-climatic factors as well as the size of holdings do affect the degree of mechanization . Nearly 30 percent of total cultivated area is owned by l76 percent of the small and tiny land holders each owning even less than a hectare. On the other hand, about 2 percent of land owners cultivate land. These variabilitieshave greatly influenced the ownership of power sources on Indian farms. Small farmers, employ human and animal energies with the use of hand tools and animal drawn equipments. Whereases, the use of tractors, power tillers, electric motors, etc. on small farms is on a marginal scale. There are few pockets and also extensive wheat growing regions where mechanical and electrical power sources are extensively used in production agriculture leading to about 185% of cropping intensity . In that region, the animal energy is employed for on the farm transport of fertilizers, fodders and fuel to support milch animals and other household activities . Inspite of high degree of mechanization, the harvesting of crops is done by human labour with few exceptions of harvesting wheat crops by combines in few pockets. In overall assessment of mechanization, the following conclusions are drawn : ⅰ) Farm operation which show a growing trend of mechanization are (a) tillge, (b) seedling (c) Irrigation (d) Plant protection application (e) Threshing and (f) Transport . ⅱ) Crop cultivation system in respect of wheat, maize and sorghum have been greatly mechanized. ⅲ) The least mechanized cropping systems are (a) vegetable production and (b) cultivation of sugarcane, cotton, rice and pulses. ⅳ) Annual production of tractor has touched the figure of 280.000 by 1995 and the total number has crossed 1.5million on Indian farms.

Consideration about Resident Participation Activities for Maintenance and Use OPen Space with Community Development in Hachioji New Town

  • Maintenance open spaces and community development in large scale housing development is one of the most important issues. We(UDC) established a resident participation activity for maintenance and use open space with community development in Hachioji New Town from 1997. The purpose of this study is to establish resident participation activities for maintenance and use open space with community development in Hajichioji New Town, and to clarify the characteristics and to on sider about efficiency of the activity. We established Minamino Shizen-Juku as a methodology for maintenance and use open spaces at the same time Hachioji New Town was opened in 1997. The activity has continued by now in 2001. We conducted questionnaire survey to make sure efficiency and characteristics of the activity past three years. Minamino Shizen-Juku (nature friendly lessons) is a resident participation activity in Hachioji New Town. It was established in 1997 as soon as the new town was opened, this year is fifth since it was started. It has three objects. 1)Maintenance and use woody environment, 2) Community development, 3) Continuation and renewal native culture. And it has general course, three special courses and one extra course were established in the activity. 1) General course is an activity participated all of member. It's included farming experience in the native field and seasonal events. 2) Rice growing and woods maintenance course is rice growing and wild wood maintenance in the park as a series of annual farming activities. 3) Benefaction from farm course is started from planting spring vegetable at Mizukoshi(Leader of Minamino Shezen-Juku)'s field. There is the number of participant limit because of the area of the field. 4) Watching nature course is watching seasonal wild flowers, trees, animals and plants to know about native nature. 5) Extra program is programmed to know about native history and tradition. It's planned including participant's ideas. Not member can participate in this course too.

Comparison of Yield and Workload depending on Stem Training Methods in Oriental Melon Hydroponics (참외 수경재배에서 줄기 유인 방법에 따른 수확량 및 작업 강도 비교)

  • Oriental melon (Cucumis melo L.) is generally cultivated on the ground by creeping culture. A farmer has a higher workload for training stems. This study was conducted to find out a new cultivation of oriental melon to reduce a workload and improve the quality of fruit. There were three treatments for training stem of oriental melon; upward stem growing, downward stem growing, control (creeping stem growing). The results of the plant growth and the net photosynthesis showed higher in upward stem growing. The root activity was higher in downward stem attract. The yield was not significant as 4,055kg/10a in upward stem attract and 3,983kg/10a in downward stem attract. According to the results of the ergonomic agricultural workload evaluation, in the case of the working posture, the working posture of creeping cultivation methods (squatting, bending) showed a higher risk level than the upward and downward cultivation methods. Therefore, it is recommended the upward stem attract of oriental melon is a new cultivation as well as an alternative method for creeping stem attract in terms of improving the plant growth and yield, and reducing the workload.

Relationships between Blossom-End Rot in Pepper and Calcium Fertilization during the First Fruit Setting Period

  • Blossom-end rot is probably the most recognizable Ca deficiency that affects fruit production including pepper. We investigated the induction of blossom-end rot during the period of rapid vegetative growth and fruit setting, and the effect of Ca fertilization on the suppression of blossom-end rot in pepper plants grown in a plastic-film house. During the first fruit setting period, more than 60% of pepper fruits larger than 8 cm showed the symptoms of blossom-end rot. Under the condition of transpiration occurring at a much faster rate, growing leaves of pepper plants could be the greater sinks for Ca than developing fruits. The incidence of blossom-end rot was significantly decreased after two weeks of Ca fertigation. Calcium application during the rapid vegetative growth and fruit setting period could be suggested as a preventive step to overcome the local Ca deficiency inducing blossom-end rot of pepper fruits.

  • The aim of this research is to improve Korean lettuce varieties in terms of Fusarium wilt, bolting under hot weather and nutritional function applying genomics approaches. To find related gene/molecular markers, we selected 96 lettuce varieties which are popular in domestic fresh vegetable markets. To construct frame works of the genomic approaches, we exploited GBS(Genotyping by Sequencing) and found total 61,407 SNPs from lettuce whole genomes (MAF>0.02). We observed that Three SNPs array per 100kb of lettuce genome. Average LD decay is expected to expand up to 3.9M(million)bp. Thus, we concluded that about 104 SNPs exist within a LD, which is sufficient to use GWAS(Genome-wide Association Study) to explore the useful gene/molecular markers. In addition, we optimized mass screening method to evaluate disease resistance levels against Fusarium wilt and are testing the bolting sensitivity during summer growing season for those lettuce allele mining set.

Dietary Patterns and Nutrient Intakes of Individuals with Circulatory Diseases: Ansan-Ansung Cohort Data from the Korean Genome and Epidemiology Study

  • Recently, there is growing interest in studying the dietary patterns that affect the risk of circulatory system diseases (CSDs). We investigated the relationship between CSDs and dietary patterns through a follow-up study in Korea (2001-2016). The participants of this study were obtained from the Korean Genome and Epidemiology Study (KoGES). This study was a large community-based cohort study (the Ansan-Ansung areas) conducted to assess the effects of various factors, especially diet, on the onset of chronic diseases among the Korean population aged 40-69 yrs. Baseline data were collected from 2001 to 2002, and follow-up studies were performed every 2 yrs, with over 7 follow-up studies performed (2015-2016). Three dietary patterns were identified using factor analysis: "vegetable and seafood (men)/soup and stew (women)" pattern, "sweet foods and breads-rice cake" pattern, and "multigrain rice and cooked white rice" pattern. None of the dietary patterns were significantly associated with the risk of CSDs in either men or women. Our follow-up study is meaningful as it investigated whether the dietary patterns of individuals according to sex affects the development of CSDs.
