The Experimental Study on the Anti-allergic Effects of Samsoeum and Samsoeumgamibang (蔘蘇飮과 蔘蘇飮加味方의 抗알레르기효과에 관한 實驗的 硏究)

  • Background : Although Samsoeum has been used frequently on allergic rhinitis, but there isn't any experimental research for that. Objective : This study was performed to investigate the anti-allergic effects of Samsoeum and Samsoeumgamibang. Materials and Methods : Katayama's method was used to observe the vascular permeability response induced by serotonin and histamine. Muller's method was used to observe the contact dermatitis response induced by picryl chloride. Miller's method was used to observe the delayed type hypersensitivity response to SRC. Results : 1. In the vascular permeability response to intradermal injection of serotonin, 2,600mg/kg, p.o. group of Samsoeum(蔘蘇飮), 1,300 and 2,600mg/kg, p.o. group of Samsoeumgamibang(蔘蘇飮加味方) showed significant inhibitory effects on the leakage of Evan's blue solution. 2. In the vascular permeability response to intradermal injection of histamine, 1,300 and 2,600mg/kg, p.o. group of Samsoeum, also 1,300 and 2,600mg/kg, p.o. group Samsoeumgamibang showed significant inhibitory effects on the leakage of Evan's blue solution. 3. In the contact dermatitis response induced by picryl chloride, 1,300 and 2,600mg/kg, p.o. group of Samsoeum, 1,300 and 2,600mg/kg, p.o. group of Samsoeumgamibang showed significant inhibitory effects on ear swelling formation. 4. In the delayed type hypersensitivity response to SRC. 2,600mg/kg, p.o. group of Samsoeum and 2,600mg/kg, p.o. group of Samsoeumgamibang showed significant inhibitory effects on foot swelling. Conclusion : This study shows that Samsoeum and Samsoeumgamibang may have anti-allergic effects. So Samsoeum and Samsoeurngamibang can be helpful to treat allergic rhinitis.

Endotoxin Induces Late Increase in the Production of Pulmonary Proinflammatory Cytokines in Murine Lupus-Like Pristane-Primed Modelp

  • Lupus-like syndrome is characterized by multiple organ injuries including lungs and kidneys. Endotoxin induces a transiently intent systemic inflammatory response and indirectly transient acute lung injury in normal condition. However, whether endotoxin may trigger the persistent development of lung injury in chronic, inflammatory lupus-like syndrome compared with normal condition remains unclear. We examined the pulmonary vascular permeability and production of proinflammatory cytokines, such as TNF-${\alpha}$, IL-6, IL-10 and IFN-${\gamma}$, which play prominent roles in the pathogenesis of lupus-like tissue injury, 6 hand 72 h after i.p. lipopolysaccharide (LPS; endotoxin) injection in pristane-primed chronic inflammation ICR mice characterized by a lupus-like syndrome. These results demonstrated that levels of serum IL-6, IL-10 and IFN-${\gamma}$ and bronchoalveolar lavage (BAL) IL-6 and IFN-${\gamma}$ were remarkably increased 6 h in LPS-exposed pristane-primed mice compared with pristane-primed controls, while pulmonary vascular permeability and levels of serum and BAL TNF-${\alpha}$ were not. And levels of BAL TNF-${\alpha}$, IL-6 and IL-10 were significantly enhanced 72 h in LPS-exposed pristane-primed mice compared with pristane-primed controls. Also, LPS significantly induced the increased in vitro production of TNF-${\alpha}$, IL-6 and IL-10 by lung cells obtained from LPS-exposed pristane-primed mice compared with LPS-exposed normal mice. Our findings indicate that LPS may trigger persistent progression of lung injury through late overproduction of BAL TNF-${\alpha}$, IL-6, and IL-10 in lupuslike chronic inflammation syndrome compared with normal condition.

An Experimental Study of Sinichengpaeum(辛荑淸肺飮) on the anti-allergic effect (辛荑淸肺飮의 抗allergy效果에 關한 實驗的 硏究)

  • Sinichengpaeum has been widely used in the oriental medical treatment of nasal diseases. These studies were performed done to investigate the effect of Sinicheng- paeum on the anti-allergic action We studied the vascular permeability response induced by the histamine and serotonin injection(allergy Ⅰ type), homologous PCA provoked by the IgE-like antibody against EWA(allergy Ⅳ type), contact dermatitis induced by picryl chloride(allergy Ⅳ type), delayed type hypersensitivity response to SRBC (allergy Ⅳ type), and the amount of eosinophil and IgE. The results were as follows: 1. The effect of Sinichengpaeum on vascular permeability responses to intradermal histamine and serotonin were significant. 2. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Sinichengpaeum showed a significant effect. 3. In the delayed type hypersensitivity responses to picryl chloride, Sini-chengpaeum provoked a significant effect. 4. After reaction provoked by picryl chloride, Sinichengpaeum showed an insignficant effect on amount of eosinophil, but a significant effect in IgE amount. 5. In the delayed type hypersensitivity responses to SRBC, Sinichengpaeum provoked a significant effect. 6. After reaction provoked by SRBC, Sinichengpaeum showed a significant effect on amount of cosinophil, but an insignificant effect in IgE amount.

Experimental study of Gagam-Cheongsang BangPungTang on the anti-inflammatory effects (가감청상방풍탕(加減淸上防風湯)의 항염증 효과에 대한 실험적 연구)

  • Background and Object : This study was carried out to investigate the effects of GCSBPT (Gagam-Cheongsang BangPungTang) on the in vitro and in vivo anti-inflammatory reactions. Methods : Vascular permeability and Cyclooxygenase inhibition assay are examined in vitro and nitric oxide inhibition assay, radical scavenging activity test, $TNF-{\alpha}$, COX-2 inhibition test are examined in vivo. Results : GCSBPT showed inhibitory effects on vascular permeability and leukocyte migration in animal test. In cyclooxygenase 2 inhibition assay, an ethanol extract of GCSBPT inhibited prostaglandin E2 generation at a concentration of $10{\mu}g/ml$. Among the herbal ingredients of GCSBPT, ethanol extracts of Nepetae Spica exhibited potent inhibitory activities. Ethanol extract of GCSBPT inhibited the release of nitric oxide and the gene expression of inducible nitric oxide synthase in RAW 246.7 cells stimulated by lipopolysaccharide. Ethanol extract of GCSBPT exhibited radical scavenging activity of 54% at $100{\mu}g/ml$. Among the herbal ingredients of GCSBPT. Conclusions : According to the above results, I expected that GCSBPT was a potent anti-inflammatory prescription.

Inhibitory Effect of Saururus chinensis (Lour.) Baill Extracts on Allergy in Mouse Models (Mouse 모델 알레르기 반응에서 삼백초(三白草)가 미치는 영향)

  • We investigated the effect of Saururus chinensis (Lour.) Baill (SCB) on allergy in mice. We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the plasma histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability and anti-DNP IgE-mediated passive cutaneous anaphylaxis. In addition, SCB reduced IL-10 mRNA expression of the lung on ovalbumin-induced allergy. These results indicate that SCB inhibits allergy.

Effects of Okbyungpoongsan Administration on Innate and Specific Immune Response in the Mouse (옥병풍산(玉屛風散)이 생쥐의 선천성 및 특이적 면역반응에 미치는 영향)

  • Okbyungpoongsan(OBPS) has long been known to have anti-allergic effect. In order to evaluate the influence on innate and specific immune response, the effects of OBPS on vascular permeability. hypersensitivities and phagocytic functions were measured. As the results, OBPS increased phagocytic activity of peritoneal macrophages in vitro and in vivo. But OBPS depressed formation of reactive oxygen intermediates(ROI) in vitro and in vivo, while the drug enhanced generation macrophages. Foot pad swelling in the mouse and contact hypersensitivity against dinitroflouorobenzene were decreased. OBPS had no effect on NK cells. But OBPS decreased vascular permeability induced by histamine without statistical significance. These results demonstrate that OBPS suppresses hypersensitivity reactions without affecting phagocytic functions and formation of ROI from macrophages. It also means that OBPS acts as a effective inducer to synthesis of nitric oxide which is effective for the infectious disease while it does damage to tissue less as it suppresses ROI, So we can conclude that OBPS could be used for the treatment of the disease related with immune function.

Antihistaminic Action of Medicinal Plants

  • The antihistaminic action of eighteen herbal medicines was investigated by the radioligand binding and functional assays. The hexane fractions of Trichosanthis radix, Mori cortex radicis and Evodiae fructus dose-dependently inhibited [$^3$H]mepyramine binding to H$_1$ receptor and histamine-induced contraction in guinea-pig brain homogenates and isolated guinea-pig ilea, respectively. Antihistaminic action of the hexane and ethylacetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus was more potent than their antimuscarinic action evaluated from the inhibition of [$^3$H]QNB binding and carbachol response. The ethylacetate and chloroform fractions and six known flavonoids from Scutellariae radix also inhibited histamine-induced contraction, but antihistaminic potencies of these fractions and compounds were almost identical with their antimuscarinic potencies. The hexane fractions of Mori cortex radicis and Evodiae fructus, as shown in ketotifen, inhibited selectively the increase of cutaneous vascular permeability induced by histamine. However, wogonin (SC-1) from Scutellariae radix was a nonselective inhibitor for the effect of histamine and serotonin on the vascular permeability. These results demonstrate that the hexane and ethylacetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus have the selective histamine H$_1$ receptor blocking activities.

  • Experimental studies were done to research the clinical effect of Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang on the Anti-allergie effect in rats and mice. The results obtained as follows: 1. (1) In the effects of Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang on vascular permeability responses to intradermal histamin in rats, Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang revealed significant effect. (2) In the effects of Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang on vascular permeability responses to intradermal serotonin in rats, Woohwangporhyonghwan(minus REALGAR and CINNABARIS) revealed significant effect. 2. In the 48 hour homologous passive cutaneous anaphylaxis in rats provoked by the Ig E-like antibody against egg albumin, Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang revealed significant effect. 3. In the delayed type hypersensitivity responses to picryl chloride in mice, Woohwangporhyonghwan(minus REALGAR and CINNABARIS) revealed significant effect. 4. In the delayed type hypersensitivity responses to SRBC(Sheep Red Blood cell) in mice, Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang revealed significant effect. According to above-stated results, Woohwangporhyonghwan(minus REALGAR and CINNABARIS) and Saengnyosamultang were concluded to be effective as anti-allergic regimen and recommended to be used for treatment of allergic diseases.
