Development of Quantitative Analytical Method for Isoflavonoid Compounds from Fruits of Cudrania Tricuspidata (꾸지뽕나무 열매 isoflavonoid 화합물에 대한 정량분석법 개발)

  • In this study, an analytical method to evaluate the quality of isoflavonoid compounds purified and isolated from the fruits of Cudrania tricuspidata was developed and validated using Ultra Performance Liquid Chromatography (UPLC). The fruits of C. tricuspidata were extracted with methanol and further fractioned with n-hexane, ethyl acetate and water. The resulting ethyl acetate extract separated into four isoflavonoid compounds by a combination of silica gel and sephadex LH-20 column chromatography. The structures of the compounds were elucidated as alpinumisoflavone, 6,8-diprenyl genistein, 6,8-diprenyl orobol, 4'-O-methylalpinumisoflavone by various techniques such as UV-Vis, ESI-MS, $^1H\;NMR$ and $^{13}C\;NMR$ spectroscopy. Finally, a method to characterize the compounds was developed by using the UPLC equipped with a $C_{18}$ column and a gradient mobile phase system consisting of 2% acetic acid in water (solvent A) and 2% acetic acid in methanol (solvent B). The developed method was validated with the parameters such as selectivity, linearity, limit of detection, limit of quantitation, accuracy, and precision, which are defined by the ICH (International Conference on Harmonization). Using the validated method, the compounds in the fruits harvested in different months were also quantitatively analyzed. We propose this approach this approach can readily be utilized as an efficient evaluation method to quantify the extracts of C. tricuspidata.

  • Coptis chinensis Franch is a valuable traditional oriental medicinal plant used for the treatment of various diseases. The major factors affecting the content of bioactive compounds and the relationship between bioactive compounds and antioxidant capacities of Coptis chinensis Franch were poorly understood. Thus, effects of the solvent, temperature, and extraction time on the extraction yields of bioactive compounds and the antioxidant activity of C. chinensis Franch extracts were investigated in this work. Our cluster analysis indicated that the hydroalcoholic solvent (50% ethanol : 50% water) at $35^{\circ}C$ for 30 min (extract time) was the best extraction condition for a factory use because the highest level of bioactive compounds and antioxidant activities was achieved. Multiple linear regression analysis revealed that total phenolic content (TPC) contributed to the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity, while both total alkaloid content (TAC) and total flavonoid content (TFC) were responsible for ferric reducing antioxidant power (FRAP) activity. These results indicated that extraction conditions controlled the yield of bioactive compounds and the antioxidant activity of C. chinensis Franch, which can provide important information for the industrial extraction.

Effects of Solvent-extracted Fractions from Salicornia herbacea on Anti-oxidative Activity and Lipopolysaccharide-induced NO Production in Murine Macrophage RAW264.7 Cells

  • Salicornia herbacea L. (Chenopodiaceae: S. herbacea) is a salt marsh plant, which has long been prescribed in traditional medicines for the treatment of intestinal ailments, nephropathy, and hepatitis in Oriental countries. In order to elucidate the mechanisms of this herb, we conducted an anti-oxidative activity, the inhibition of nitric oxide (NO) production, and the suppression of the pro-inflammatory cytokine genes, with the solvent-extracts of S. herbacea. We found that both ethyl acetate and n-butanol tractions showed potent anti-oxidative effects in comparison to other fractions using xanthine oxidase assay with $IC_{50}$ values of $66.0{\pm}0.5\;{\mu}g/ml$ and $82.5{\pm}3.8\;{\mu}g/ml$, respectively. In addition, both ethyl acetate and n-butanol fractions showed more electron donating activity (EDA) than other tractions, according to DPPH (2, 2-Diphenyl-lpicrylhydrazyl radical) assay. The EDA of ethyl acetate fraction ($IC_{50}$ values of $117.5{\pm}3.8\;{\mu}g/ml$) is more significant than that of n-butanol fraction ($IC_{50}$ values of $375.0{\pm}12.5\;{\mu}g/ml$). Among potential anti-oxidative tractions, ethyl acetate traction dose-dependently suppressed lipopolysaccharide (LPS, $0.1\;{\mu}g/ml$)-induced nitric oxide (NO) production in RAW264.7 cell, while n-butanol did not. As expected, ethyl acetate fraction suppressed the expression of inducible NO synthase (iNOS) in RAW264.7 cell stimulated by $0.1\;{\mu}g/ml$ of LPS. Moreover, the ethyl acetate traction suppressed the expression of interleukin-1 $(IL)-1{\beta}$ and granulocyte/macrophage colony-stimulating factor (GM-CSF) mRNA in LPS-stimulated RAW264.7 cells. Therefore, these results suggest that S. herbacea may have anti-oxidative and anti-inflammatory activities by modulating radical-induced toxicity and various pro-inflammatory responses.

  • In this study, two species of algae, Ecklonia stolonifera, Sargassum thunbergii and one species of marine plant, Zostera marina(rhizoid and stem) were collected and extamined to determine the extractability of water soluble protein and the influences of various factors including extraction time, temperature, ratio of sample vs solvent and pH upon the extractability were tested. The effects of precipitation treatments for isolation of algal protein from the extracts(TCA treatment, methanol treatment and pH control) were also tested. Amino nitrogen and total nitrogen of purified samples made by obtained optimum conditions were estimated. The effect of the ratio of sample vs solvent on extractability differed from species to species which was enhances at 1:100(w/v) in Sargassum thunbergii and Zostera marina while 1:150(w/v) for Ecklonia stolonifera. The effect of extraction time and temperature was revealed differently in all species which might be considered to be caused by differences in the constitution of algal tissues. But in case of TCA insoluble nitrogen, it was showed the maximum extractability at $40-50^{\circ}C$ for 1 hour extraction. The optimum pH for the ext action of total nitrogen was 9-12 while the optimum pH was 6-7 for TCA insoluble nitrogen. And the pH control appeared to be most effective in the influence of precipitation treatment for isolation of algal protein.

Extraction of Glabridin from Licorice Using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 감초 중의 glabridin 추출)

  • The purpose of this study is to investigate the feasibility of a cosolvent-modified supercritical $CO_2\;(scCO_2)$ extraction technique for the production of licorice extracts with high levels of glabridin. The effects of various parameters such as the type and amount of modifiers, extraction temperature ($40{\sim}80^{\circ}C$) and pressure ($10{\sim}50.0\;MPa$) on the extraction efficiency were examined at a fixed flow rate of 1 mL/min. The organic solvent extraction with pure methanol was also conducted for a quantitative comparison with the $scCO_2$ extraction. The recovery of glabridin from licorice was found to be extremely small for pure $scCO_2$. However, the addition of modifiers such as ethanol and acetone to $scCO_2$ resulted in a significant improvement in the recovery of glabridin. The recovery of glabridin was observed to increase with pressure at a constant temperature. Furthermore, the purity of the glabridin obtained from the $scCO_2$ extraction was higher compared with the organic solvent extraction.

Effect of varying ethanol concentrations on the extraction properties and physiological activity of Artemisia annua L. (에탄올 농도 조건이 개똥쑥 추출 및 생리활성에 미치는 영향)

  • The present study was undertaken to compare the effects exerted by different extraction solvents on the extraction of active components, such as polyphenols and flavonoids, from the dried leaves of Artemisia annua L. Different extracts were prepared using a heating mantle. The extraction solvents used were distilled water, and 20, 40, 60, 80, and 99.5% ethanol solution. It was observed that the 40% ethanol solution yielded the most significant results in the extraction of various phytochemicals with phenol concentration of 154.8±0.28 mg of gallic acid equivalent/g and flavonoid content of 25.28±0.01 mg quercetin equivalent/g. However, based on the extraction solvent used, varying trends were observed in the antioxidant, enzyme inhibition, and bacterial inhibition analyses. It was concluded that the extraction solvent should be selected based on the purpose of use of the dried leaves of A. annua L.

  • In this work, the amounts of Flavone C-glycosides homoorientin, orientin extracted from Black bamboo by various ultrasonic waves frequency (35, 72, 170 KHz, 300 Watt $\pm$1) time (15, 30, 60 min) and temperature ($25^{\circ}C$) were compared using 50% aqueous ethanol solution. And describesanalysis of the antioxidant potential of Black bamboo using an high-performance liquid chromatography (HPLC) on-line $ABTS^+$antioxidant screening method. In conjunction with the analysis of their 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid ($ABTS^+$) radical scavenging ability. The optimum operating conditions were experimentally determined to analyze the Flavone C-glycosides homoorientin, orientin in the pretreatment ultrasonic extracts. From the results, the effect on extraction yield of variations in frequency, solvent composition and extraction time was investigated. The highest yield of Black bamboo was obtained by ultrasonic waves with a frequency of 72 KHz and an extraction time of 60 min, The extraction frequency 35 KHz ($25^{\circ}C$), time 30 min was selected as an optimal antioxidant activity condition.

Concentration of Fucoxanthin from Ecklonia cava Using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용하여 감태로부터 Fucoxanthin 농축)

  • Fucoxanthin has been concentrated efficiently using supercritical carbon dioxide. First, fucoxanthin was extracted from Ecklonia cava using solvents. Three solvents, such as medium chain fatty acid (MCFA), medium chain fatty acid ethyl ester, and fatty acid from coconut oil were tested, and MCFA was selected as a suitable solvent to extract fucoxanthin from Ecklonia cava. The concentration was conducted at various pressures and temperatures. In order to concentrate fucoxanthin from extracts, MCFA was eliminated from the extract using supercritical carbon dioxide, and fucoxanthin was concentrated successfully in residue. Optimal conditions for concentrating the fucoxanthin from fucoxanthin extract were $40^{\circ}C$ at 9.7 MPa, $45^{\circ}C$ at 11.0 MPa, and $50^{\circ}C$ at 12.4 MPa, respectively. The density of carbon dioxide affected the concentration of fucoxanthin from fucoxanthin extract. In this optimal conditions, the density for concentrating the fucoxanthin was 600 g/L.

A comparison of the characteristic properties between soybean (Glycine max [L.] Merrill) seeds with different seed coat colors

  • We profiled the health-promoting bioactive components in nine types of soybean seeds with different seed coat colors (yellow, green, brown, and black) and investigated the effects of different extraction solvents (methanol, ethanol, and water) on their antioxidant activities. The carotenoid and anthocyanin compositions varied greatly by seed color, and the phenolic acids, total phenol, and total flavonoid contents differed by genotype. The carotenoid content was relatively higher in soybean seeds with green and black seed coats than in those with a yellow seed coat while lutein was the most plentiful. The anthocyanin content was considerably higher in the soybean seed with the black seed coat. The results of the DPPH assay showed strong antioxidative activities in the methanol- and water-extracts compared to the ethanol-extract, irrespective of the seed coat colors. Moreover, the soybean seeds with the black seed coat exhibited the highest antioxidant activity among the samples, regardless of the extraction solvent used. Eighteen bioactive compounds were subjected to data-mining processes including principal component analysis and hierarchical clustering analysis. Multivariate analyses showed that brown and black seeds were distinct from the yellow and green seeds in terms of the levels of carotenoids and anthocyanins, respectively. These results help our understanding of the compositional differences in the bioactive components among soybean seeds of various colors, providing valuable information for future breeding programs that seek to enhance the levels of compounds with health benefits.

Comparison of Prunus yedoensis Matsumura bark ethanol extract and solvent fraction antioxidant activities (왕벚나무 껍질의 에탄올 추출물과 용매 분획물의 항산화활성 비교)

  • This study was conducted to investigate the antioxidant compounds and activities of Prunus yedoensis Matsumura bark (PYMB) ethanol extracts (EE) and various other fractions. Among them, the highest total phenol content was 496.80 mg gallic acid equivalent/g in the ethyl acetate fractions (EAF). The flavonoid contents were 7.26-265.52 rutin equivalent mg/g, with the EAF showing the highest levels. The highest proanthocyanidin content was determined to be 326.31 catechin equivalent mg/g in the EAF and the remaining values in descending order were as follows: n-butanol, ethanol, dichloromethane, water, and n-hexane. EAF exhibited the highest DPPH, ABTS+, superoxide- radical scavenging activities, and reducing power, whereas the nitrite scavenging activities were the highest in the case of the EE. The n-hexane and water fraction antioxidant compounds and activities were lower than those of the other fractions. In summary, these results suggest that the PYMB EAF is a potential source of natural antioxidants.
