  • To study some informations on the morphogenesis and developmental process of the coronal suture in rats, the author performed daily oral administrations of demethylchlorotetracycline, a kind of tetracycline group, in the amount of 30mg/kg of body weight to the female rats from the 7th day of pregnancy to the time of delivery. Microscopic evaluation was undertaken on the fetal rats in the experimental group. The subject of this experiment were defined to the fetal rats of each group at 1st, 3rd, 7th and 14th day after birth. All these fetal rats were sacrificed and the heads were removed. All the tissue sections were fixed with $10\%$ formalin, Bouin' and Carnoy' solution and then stained by Hematoxylin-Van Gieson stain, or Feulgen and Rossenbeck, Periodic acid Schiff, and prepared for alcian blue reaction. The results were as follows; 1. The directions of osteogenic fibers were arranged irregulary during first 3 days, but after the 7th day they tended to change radial directions like control group. 2. The density of deep stained cells by Feulgen-Rossenbeck reaction were shown leu in the experimental group than that in the control group in first 3 days, but there was shown no significant difference between both groups after the 7th day. 3. PAS reaction in early stage was generally negative in the experimental group unlike as in the control group, but diffuse reaction was observed in the loose middle zone like as in the control group after 14th day. 4. Alcian blue reaction was negative in cambial zone, and slightly positive in uniting zone compared with control group in early stage. After 14th day, however, there was observed a tendency of moderately positive reaction.

  • Introduction. The human cutaneous scars manifest themselves many ways in different types according to the factors such as the age, sex, race of the patient as well as the location,. kind and heal ing process of the wound. Among the scars it is quiet difficult to verify the clinical course of the hypertrophic or keloidal scars from the true keloids. However, clinical observations indicate that stress, either mechanical or in the forms of chronic infections, can induce a functional change in the fibroblasts causing an excessive production of collagenous matrix. In this study, we preliminary attempt to justify any difference of the cellular structure between keloids and hypertrophic scars by using electron microscope. Material and Methods. A total of 23 cases: 2 scars, 2 hypertrophic scars and 19 keloids are examined. Immediately, the biopsy tissue was fixed in 10% neutral formalin and 4% glutaraldehyde solution in phosphate buffer for 4 hours, post fixed in 1 % osmium tetraoxide for two hours, dehydrated with graded alcohol, and embedded in Epon 812. Thick sections were stained with hematoxylin eosin, periodic acid-Schiff(PAS) and Van Gieson stain. Thin sections were cut and uranyle acetate, lead citratestain and examined with the electron microscope. Result. The morphologic features of keloid showed thick, homogenously eosinophilic bands of collagen and numberous large active fibroblasts. The hypertrophic scar and soft scar are more cellular than keloid and composed thinner collagenous fiber. For this paper in the etiology of keloids can not as be defined, but and interesting keloidal tissue fibroblast showed irregular nucleus with irregular shape dense bodies and fibril materials contained in to the cytoplasm.

  • It has been studied that aminoacetonitrile was associated with the inhibition of collagen fiber, argyrophilic fiber and oxytalan fiber synthesis. This experiment was performed, by the basic knowledge of above mentioned study, to study on the biological effect of aminoacetonitrile to the developing periodontal ligament in Sprague Dawley rat. twenty two of female rats weighing about 200gm were gestated. In 7 days after gestation, the experimental rats were injected aminoacetonitrile 7 times intraperitoneally. After parturition, delivered fJtuses were divided into 4 groups and each group was sacrificed to 1 day, 7 days, 14 days, and 21 days after delivery, schematically. All the fetuses were observed on their periodontal ligament by histological and histo chemical methods. To study on the components of periodontal ligament fiber in these experimental study van Gieson, Masson's trichrom, argyrophilic fiber, oxytalan fiber, methyl green pyronin and periodic acid-Shiff staining were performed. Results were as follows; 1) Retardation of functional orientation in periodontal ligament collagen fiber was observed in 1 day fetuses hut this appearance was diminished gradually and recovered in normal condition in 7 days fetuses. 2) Distribution of argyrophilic fiber in 1 day fetuses was oriented delicately and loosely but volume of this fiber was gradually thickened and distributed densely. 3) Oxytalan fiber was oriented dendritic ally and contradictorily in 14 days fetuses but their orientation was changed into oblique form in middle portion of roof and their numbers were increased gradually. 4) Pyronin-philic stain of fibroblast was gradually deepened in 7 days fetuses and this finding also suggested to the depreciation of collagen synthesis in this specimen. 5) PAS positive line was observed continuousely at the portion of cervical to the middle root surface.

  • Background: The histomorphological properties of the left anterior desecending artery (LAD), the left internal thoracic arteries (LITA), the radial arteries (RA) and the intercostal arteries (ICA) were studied for their use as a conduit for coronary artery bypass grafting (CABG), and we compared them with each other. Material and Method: All the vessels were harvested from nineteen cadavers (17 males and 2 females). The mid-portion of the LAD, the mid-portion of the LITA, the distal RA and the mid-portion of the 5th ICA were obtained. All of them were stained with hematoxylin-eosin and with Van Gieson's elastin stain. The morphological characteristics were examined and the thicknesses of the intima and media (I/M ratio: the intima to media ratio) were compared using one-way ANOVA tests. Result: The mean age of the cadavers was $61.5{\pm}9.6$ years. The LITA and ICA were elastic arteries, and the LAD and RA were muscular arteries. The I/M ratio showed statistically significant differences: $0.07{\pm}0.03$ in the LITA, $0.16{\pm}0.11$ in the ICA, $0.45{\pm}0.29$ in the RA and $0.93{\pm}0.52$ in the LAD, respectively. Conclusion: This study showed that the degrees of intimal hyperplasia of the CA and the various conduits for CABG were different significantly. The ICA was found to have relatively favorable characteristics as a coronary by pass conduit, but its suitability for clinical use is a challenging issue.

  • The purpose of this study is to evaluate the tissue response in applying of various bone substitutes included toothash-plaster mixture, resorbable hydroxylapatite (HA) and demineralized freeze-dried bone and to show the clinical usefulness of toothash-plaster mixture for the repair of craniomaxillofacial bone defect. For this experiment, 100 Sprague-Dawley rats weighing 200gm or more were used. There were four experimental groups: group I, toothash-plaster mixture; group II, demineralized freeze-dried bone; group III, resorbable HA; and group IV, control group. A full thickness, round bone defect measuring 10mm in diameter was created in the midcranium, and the substitutes cited above were embedded in the experimental rats based on their group assignment. Blood clot was filled in the rats assigned to the control group. Experimental rats were sacrificed on the 1st, 3rd, 5th, 8th, 12th and 24th week after implantation and stained with the hematoxylineosin, Masson's Trichrome, using Van Gieson's stain method, and were examined under light microscope. The results were as follows: 1. In all the groups, prominent inflammatory reaction and the infiltration of multinucleated giant cells were noted during the early stage. Gradual healing decreased this reaction. 2. Among the rats in the experimental group II, which were given demineralized freeze-dried bone implants, active formation of new bone traveculae manifested. Chondroid tissues appeared, and it was suggested that the defect was filled with newly formed bone by virtue of osteoinductive activity. On the 12th week after the experiments, most of the defect was filled with newly formed bone trabeculae. 3. In experimental groups I and III, it was noted that HA manifested a healing process similar to that characterized by the toothash-plaster mixture, but inflammatory reaction was more prominent in experimental group I. Active osteoblasts were observed along the periphery of osteoid tissues, while newly formed bone trabeculae appeared adjacent to the implanted materials three weeks later. Formation increased to the extent that newly formed bone trabeculae fused directly with the host bone. Increase in new bone ingrowth into the filling materials was revealed by both experimental groups. 4. In the control group, new bone formation adjacent to the host bone was observed, but most of the defect was filled with mature connective tissue 24 weeks after the experiments.
