Effect of bark on the adsorption of heavy metal ions (2) - Effect of Pinus densiflora and Quercus mongolica barks on the adsorption of Cu and Cd ions - (수피(樹皮)에 의(衣)한 중금속(重金屬) 흡착효과(吸着效果)(2) - 소나무와 신갈나무 수피(樹皮)에 의한 Cu와 Cd의 흡착효과(吸着效果) -)

  • This investigation involves a study of the physical and chemical factors of Pinus densiflora SIEB. et ZUCC. and Quercus mongolica Fisher barks affecting on the adsorption of heavy mteal ions. The results obtained can be summarized as follows. 1. The capacity of the untreated bark to remove the Cu and Cd from solution was similar to or 5% higher than that of formaldehyde treated bark in both species. Considering that untreated bark lead to color-leaching problem, bark treated with formaldehyde are economical. 2. With decreasing particle size of bark(20-80), the adsorption ratio of the Cu and Cd from solution was increased. Quercus bark adsorbed more Cu and Cd at smaller particle size compared to Pinus bark. 3. The heavy metal eqilibrium adsorption of the bark from Cu and Cd solution was attained within 10 min. Pinus bark removed about 48% of the Cu and 41% of the Cd from solution in 10 min while Quercus bark removed about 50% during that period. 4. As the initial metal concentration increased. the absolute metal uptake was increased while percentage removal was decreased. At the lower metal concentration (10 ppm). Pinus and Quercus barks removed 77-94% of the Cu and 72-84% of the Cd. At high metal concentration (200 ppm), the adsorption ratio was 40% Cu and 25% Cd, respectivelty. 5. The maximum adsorption of the Cu and Cd from solution was obtained at pH 5-6 in filtrate. 6. With increased bark weight per given metal concentration, absolute removal of metal ion from solution was increased, but the percentage removal was decreased. The amount of adsorption was 4.2 mg Cu and 4.2 mg Cd per gr. Pinus, bark and 5.4 mg Cu and 4.3 mg Cd per gr. Quercus bark, respectively.

Heavy Metal Adsorption of Untreated Barks by Treatment Conditions of Aqueous Solution (용액의 처리조건에 따른 미처리 수피에 의한 중금속 흡착)

  • This study was designed to investigate the adsorption of heavy metal ions by untreated bark according to the treatment conditions of aqueous solution. The effect of temperature and pH of aqueous solution, particle size of bark, addition of light metal ions on the adsorption was examined, and the competition in adsorption among heavy metal ions was also evaluated. te The adsorption ratio of $Cu^{2+}$ and $Zn^{2+}$ increased with increasing themperature of solution from $-5^{\circ}C$ to $10^{\circ}C$ however, it was relatively constant at temperatures between $10^{\circ}C$ and $55^{\circ}C$. The adsorption ratio of $Cr^{6+}$ increased continuously with increasing the temperature of solution. The maximum adsorption ratio of $Cu^{2+}$, $Zn^{2+}$, and $Pb^{2+}$ was noted at pHs ranged 6 to 7; however, the adsorption ratio declined sharply on either sides of the optimum. The adsorption ratio of $Cr^{6+}$ decreased continuously with increasing the pH of solution. The adsorption ratio increased as decreasing the particle size of bark, and there was little differences in adsorption tendency between pine and oak bark. By the addition of $Ca^{2+}$ or $Mg^{2+}$(10~25 ppm), the adsorption ratio of $Cu^{2+}$ and $Zn^{2+}$ increased. An increase of the adsorption ratio was higher in oak bark than in pine bark. However, the adsorption ratio of $Pb^{2+}$ and $Cr^{6+}$ was not affected by the addition of light metal ions. As the mixed solution of 2 or 3 kinds of heavy metal ions($Cu^{2+}$, $Zn^{2+}$, $Pb^{2+}$) was treated with the untreated bark, the adsorption of $Zn^{2+}$ decreased considerably because of the competitive adsorption among heavy metal ions. Also the adsorption of $Cu^{2+}$ was more and less reduced. However the adsorption of $Pb^{2+}$ was not affected by the presence of other heavy metal ions.

Antidiabetic activity of Diospyros malabarica Kostel bark: a preliminary investigation for possible mode of action

  • The defatted methanol extract of bark of Diospyros malabarica (DM) at doses of 200 and 400 mg/kg, p.o. showed significant hypoglycemic activity on normal rats. The extract also exerted significant antihyperglycemic effect in alloxan-induced hyperglycemia and resulted in increase in plasma protein content and decrease in alkaline phosphatase, cholesterol and triglyceride levels when compared with those in the diabetic control group. However there were no significant changes in body and kidney weights of the DM extract-treated animals, compared to those of the untreated diabetic rats as a control. However, the DM extract showed a potential antioxidant activity by increasing catalase activity and reducing lipid peroxidation in liver. The results demonstrate antidiabetic activity of the defatted methanol extract of DM bark.

  • The objective of this study was to investigate the physical and chemical factors of Pinus densiflora SIEB. et ZUCC. and Quercus mongolica Fisher barks affecting on the adsorption of heavy metals. The results obtained can be summarized as follows: 1. With decreasing the particle size of bark, the adsorption rate of two heavy metal ions were increased. In case of using same particle size, the adsorption of $Fe^{++}$, and $Ni^{++}$ by Quercus bark showed higher than by Pinus bark. 2. The effect of untreated bark on the adsorption of heavy metal was more or less 5% higher than that of HCHO-treated bark in both species. But the color absorbances of the filtrates from HCHO-treated Pinus and Quercus barks were 5.8 and 11.8 times smaller than those of the filtrate from untreated Pinus and Quercus barks, respectively. 3. The maximum adsorption of $Fe^{++}$, and $Ni^{++}$ by bark was shown after 30 min. of the reaction. 4. With increasing the concentration of heavy metal, the amount of adsorption by bark was increased, but the adsorption ratio were decreased. 5. The maximum adsorption of $Fe^{++}$, and $Ni^{++}$ appeared at final pH of $4{\sim}5$, and pH of $3.6{\sim}4.0$ in filtrate, respectively. 6. With increasing the bark weight per a given heavy metal solution, the adsorption ratio were increased, but the amount of adsorption per gram of bark was the highest on the reaction with 2g of bark in a economical sense showing the amount of adsorption of 21mg $Fe^{++}$/g and 7mg $Ni^{++}$/g of Pinus bark, 36mg $Fe^{++}$/g and 9mg $Ni^{++}$/g of Quercus bark, respectively.

Efficacy of Cu(II) Adsorption by Chemical Modification of Pine Bark (소나무 수피의 화학적 처리에 의한 Cu(II) 흡착 효과)

  • Korean pine(Pinus densiflora) bark was evaluated for its adsorption capacity of Cu(II) ions from aqueous solution by running a series of batch experiments. Prior to the tests, the milled barks were treated with 1 N NaOH or 1 N HCl to examine the effect of surface modification. For comparison, untreated bark was tested under same condition. Within the tested pH range between 3 and 6, NaOH treatment increased Cu(II) adsorption capacity by $139\sim184%$, while HCl treatment decreased it by $37\sim42%$. Maximum copper ion uptake by bark was observed at pH $5\sim6$, but pH of solution was not a potent influence. A pseudo second-order kinetic model fitted well for the sorption of copper ion onto bark. For NaOH-treated bark, the calculated sorption capacity$(q_e)$ increased from 6.58 to 12.77 mg/g, while the equilibrium rate constant$(k_2)$ decreased from 0.284 to 0.014 g/mg/min as initial Cu(II) concentration doubled from 100 mg/L. A batch isotherm test using NaOH-treated bark showed that equilibrium sorption data were represented by both the Langmuir model and the Freundlich model. It was confirmed that carboxylic acid of bark was involved in the Cu(II) adsorption. For NaOH-treated bark, in particular, carboxylate ion produced by hydrolysis or saponification appeared to be a major functional roup responsible for the enhanced Cu(II) sorption.

Effects of CEPA on the Secretion of Lacquer and Bark Physiology of Lactree(Rhus verniciflua Stokes) (CEPA 처리(處理)가 옻나무의 칠액분비(漆液分泌) 및 수피생리(樹皮生理)에 미치는 영향(影響))

  • This study was conducted to investigate the effects of CEPA(2-chloroethyl phosphonic acid) on bark physiology and anatomy of lactree(Rhus verniciflua Stokes). Sample trees of similar size and growth rate were selected from 7-year-old lactree plantation located in Hyengsung-kun, Kangwon-do. Lanolin pastes containing 0.1, 1, or 10% CEPA were put into the bark-removed hole made by corer(${\phi}1cm$) on the main stem at 1.2m above the ground on June 16, 1995. Five weeks after application of CEPA, bark thickness was markedly increased as a result of the increase in the amount of phloem and intercellular spaces, and correlated with the increased production of urushiol. By the application of 10% CEPA, bark thickness was increased approximately 2.5 times, and the urushiol content within bark was increased 2.8 times compared to that of untreated trees because CEPA stimulated the accumulation of urushiol within bark. Treatment of 10% CEPA also increased the size and the total number of secretory canals, and induced an increase in ray width. The phloem parenchyma cells of CEPA-treated trees were well-developed and closely packed with little intercellular space.

The effects of biomaterials in growing medium on the response of Zelkova serrata in a containerized production system

  • Changes in the physical and chemical properties of soil materials during the nursing process have a great influence on the quality of containerized seedlings and on growth and survival after planting. In this study, the effect of biomaterials and their mixed ratios on the growth of Zelkova serrata seedlings in a containerized seedling production system was investigated. Mushroom sawdust, pine bark, and carbonized rice husk were used as biomaterials. The mixed ratios were 10% and 20% volume ratio of the growing medium volume, including the untreated controls. There was no significant difference in the height growth of the Zelkova serrata seedlings according to the biomaterials. The root collar diameter was the highest with the 20% carbonized rice husk and the lowest with the mushroom sawdust. The difference between the highest quality index and the lowest quality index was 30% in the order of the carbonized rice husk, pine bark, control, and mushroom sawdust, but there was no statistical significance. In this study, if the growing medium mixed with biomaterials does not reduce the seedling growth compared with the control, it is considered that the biomaterial can replace a part of the growing media. Therefore, the results show that some of the growing media can be replaced with carbonized rice husk or pine bark when producing Zelkova serrata seedlings.

Isolation and Identification of Adventitious Root Formation Inducing Substances from Cortex of cinnamomum cassia J.Presl (육계(Cortex of Cinnamomum cassia J.Presl) 추출물로부터 부정근 형성 유도물질 분리 및 동정)

  • In this study, as a result of exploring the physiological activity of plants useful for agriculture on various plant resources, it was possible to confirm an activity similar to auxin that promotes plant rooting in methanol extract of Cinnamon Bark (cortex of Cinnamomum cassia J.Presl). After separating the active body by applying column chromatography and HPLC to the CHCl3 active fraction obtained by solvent extraction for each polarity from the methanol extract of cinnamon bark, cinnamyl alcohol was identified through GC/MS analysis. By bioassay using cinnamyl alcohol standard and the active fraction separated and purified from the methanol extract of cinnamon bark, the rooting rate of mung bean seedlings of the cinnamyl alcohol standard was 290% compared with the untreated control at 134.2 ㎍/mL concentration, and the adventitious root formation activity similar to the rooting rate (268.6%; 100 ㎍/mL) of the active fraction was shown. In conclusion, it is believed that cinnamyl alcohol contained in methanol extract of Cinnamon Bark is the main compound that induces adventitious root formation in mung bean.

LC/MS-based Analysis of Bioactive Compounds from the Bark of Betula platyphylla var. japonica and Their Effects on Regulation of Adipocyte and Osteoblast Differentiation

  • Betula platyphylla var. japonica (Betulaceae), also known as Asian white birch, is an endemic medicinal tree, the bark of which has been used in Chinese traditional medicine for the treatment of various inflammatory diseases. In our continuing search for bioactive compounds from Korean natural resources, a phytochemical investigation of the bark of B. platyphylla var. japonica led to the isolation of 7-oxo-${\beta}$-sitosterol (1) and soyacerebroside I (2) from its ethanol extract as main components by liquid chromatography (LC)/mass spectrometry (MS)-based analysis. The structures of isolates were identified by comparison of $^1H$ and $^{13}C$ nuclear magnetic resonance spectroscopic data and physical data with the previously reported values and LC/MS analyses. To the best of our knowledge, this is the first study to demonstrate that the isolated compounds, 7-oxo-${\beta}$-sitosterol and soyacerebroside I, were isolated in B. platyphylla var. japonica. We examined the effects of the isolates on the regulation of adipocytes and osteoblast differentiation. These isolates (1 and 2) produced fewer lipid droplets compared to the untreated negative control in Oil Red O staining of the mouse mesenchymal stem cell line without altering the amount of alkaline phosphatase staining. The results demonstrated that both compounds showed marginal inhibitory effects on adipocyte differentiation but did not affect osteoblast differentiation.

Improvement of Lacquer Collection Method by CEPA Application in Lactree(Rhus verniciflua Stokes) (CEPA 처리(處理)에 의한 옻나무 칠액(漆液) 채취법(採取法) 개량(改良)에 관(關)한 연구(硏究))

  • In order to improve 'Salso' method (conventional tapping method) which was very inefficient in the aspects of collection time and labor, this study was carried out to investigate the effect of the application date and the distance from treatment point in the lactree(Rhus verniciflua) treated with 10% CEPA and to decide the possibility of application of the technique in the field. Bark thickness was significantly increased to the part 40cm above and below the treatment point, but urushiol content was increased to the part 20cm above and 10cm below the zone treated with 10% CEPA. The urushiol content of the bark was highest at 5cm above the treated zone and decreased in the order of 10, 20, and 40cm. And the urushiol contents of the bark of the upper part were higher than that of the low part. The effects of CEPA on bark thickness were similar to urushiol contents. Urushiol production of lactree is highly dependent on climatic conditions and particularly on the precipitation, and duration of sunshine. Ten percent of CEPA-lanolin pastes which was treated on June 16 affected bark anatomy and urushiol contents, while the treatment on August 24 did not affect. In the Rhus verniciflua treated with 10% CEPA, the urushiol contents was initially increased from 7 days after treatment, continued for the 4 weeks, and then slight decrease occurred at 5 weeks after the treatment. We measured a total sap yield by Salso method in lactree treated with 10% CEPA. By applying 10% CEPA, the sap yield was increased 3-4 times compared to that of untreated trees in the first tapping. But the relative ratio was gradually decreased from the second tapping to sixth, and after seventh tapping, the untreated trees secreted more sap than the treated trees. We discussed about the causes.
