능이버섯의 향기특성 (Aroma Characteristics of Neungee(Sarcodon aspratus))

  • 능이버섯의 향기성분을 조사하기 위하여 SDE, SFE, headspace 로 추출하고 GC, GC-MS로 동정하였으며, 능이버섯의 향기특성을 조사하기 위하여 GC-olfactometry 방법중 AEDA법을 이용하였다. 그 결과 SDE 추출물에서는 33개, SFE추출물에서는 26개, headspace 추출물에서는 17개의 화합물이 동정되었다. 확인된 화합물의 주성분은 1-octen-3-ol, 1-octen-3-one, 3-octanone, 2-octen-1-ol, 3-octanol, 1-octanol 등의 $C_8$ 화합물과 benzeneacetaldehyde등이었다. AEDA 결과 능이버섯의 주된 향기특성으로는 생능이버섯내(unknown), 생버섯내(1-octen-3-ol), 곰팡이내(1-octen-3-one), 흙내(3-octanol), 풀내(1-octanol), 강한 풀내(2-octenal), 단 꽃내(benzeneacetaldehyde), 목재내(L-linalool), 치즈내(3-hydroxy-2-butanone), 상큼한 단내(2-decanone), 약간의 불쾌한 고기내(3-methyl thiopropanol), 시큼한 우유내(propanoic acid) 등으로 나타났다.

Biosynthesis of $\beta$-Lactam Antibiotics by Cell-free Extract from Lysobacter lactamgenus

  • Using cell-free extract of Lysobacter lactamgenus, enzymatic conversion of $\delta$-L-($\alpha$-aminoadiphyl)-L-cysteinyl-D-valine (ACV) the first substrate of $\beta$-lactam biosynthesis, into antibiotic compounds was attempted. In high performance liquid chromatographic (HPLC) analysis, the biosynthetic intermediates for cephalosporin antibiotics including isopenicillin N, deacetoxycephalosporin C, deacetylcephalosporin C and unknown cephem compound were detected in reaction mixtures. It implies that cephabacin compounds from L lactamgenus could be produced by biosynthetic routes through penicillin ring formation and its expansion to cephalosporin ring, likely as cephalosporin C from Cephalosporium or cephamycin C from Streptomyces. Among biosynthetic enzyme in cell-free extract, the ring formation activity (isopenicillin N synthetase activity) was separated in 50-60% of ammonium sulfate fraction, and ring expansion activity (deacetoxycephalosporin C synthetase activity) was found to be in 40-50% fraction. The partially purified isopenicillin N synthetase could convert as much as 90% ACV to isopenicillin N during 6-hour reaction.

Constitutive flavonoids of the flowers of Tamarix tetragyna

  • A phytochemical investigation of the aqueous ethanolic flower extract of Tamarix tetragyna led to the isolation and characterization of the hitherto unknown conjugates, kaempheride 3,7-dipotassium sulphate and kaempferol 3,4'-dipotassium sulphate as well. Twelve known flavonol compounds, including kaempheride 3-potassium sulphate and kaempheride $3-O-{\beta}-glucuronide$ were also isolated and identified. $^1H-\;and\;^{13}C-NMR$ spectra for the known kaempheride derivatives have been recorded and assigned for the first time. Structures of all compounds were established by conventional methods of analysis and confirmed by $^1H-,\;^{13}C-NMR$ and mass spectral analysis.

Multi-Target Cytotoxic Actions of Flavonoids in Blood Cancer Cells

  • To date, cytotoxic effects of flavonoids in various cancer cells are well accepted. However, the intracellular signaling cascades triggered by these natural compounds remain largely unknown and elusive. In this mini-review, the multiplicity of molecular targets of flavonoids in blood cancer cells is discussed by demonstrating the involvement of various signaling pathways in induction of apoptotic responses. Although these data reveal a great potential of flavonoids for the development of novel agents against different types of hematological malignancies, the pleiotropic nature of these compounds in modulation of cellular processes and their interactions certainly need unraveling and further investigation.

Volatile Flavor Components in Pumpking Vinegar

  • Pumpkin venegar was produced using autoclaved pumpkin juice by fermenting with cetobacter starter and ethanol at the ratio of 4 % and 10% to the volume of pumpkin juice, respectivley. Fermentation was carried out at 2$0^{\circ}C$ for 14 days followed by aging at 1$0^{\circ}C$ for 14 days. Flavor components of pumpkin vinegar was determined using GC/MS. Identified components, were 2 aldehydes (4.74%), 5 acohols (30.06%), 4 ketones (8.99%), 4 acids (16.39%), 5 alkanes (11.10%), 11 miscellaneous compounds (27.01%) and 9 unknown compounds (1.71%). Pumpkin vinegar showed very similar flavor characteristics to those of conventional wien vinegar and sherry wine vinegar in particular , acetioin, methyl acetate, and butanoic acid were typical volatile components of these three kinds of vinegar. Pumpkin vinegar showed possiblity to compete with European wine vinegar.

도시환경의 총부유먼지 중 미지성분의 분포 특성에 대한 연구 (A study of distribution characteristics of unidentified particulate components in an urban area)

  • 대기 중의 총부유먼지(total suspended particulate, TSP)를 구성하는 성분들 중 organic carbon (OC), element carbon (EC), 금속성분은 양적으로 중요한 구성인자에 해당한다. 이들을 제외한 나머지 부분은 아직까지 정성 및 정량분석이 취약한 미지 물질(${\Sigma}X$)에 해당한다. 본 연구에서는 서울시 강서 지역에서 2009년 2-12월 기간동안 관측한 자료를 이용하여, TSP의 주요 성분들 간의 관계에 대한 분석을 시도하였다. 이를 통해, TSP의 거동을 다양한 각도에서 해석하기 위한 기초 자료로 활용하고자 하였다. 전체 연구기간 동안, TSP를 이루는 주요인자로서 미지의 영역이 평균적으로 48.6%를 차지할 정도로 더 지배적이란 것을 확인하였다. 그리고 TSP에 대한 그 구성 성분들간의 관계를 검토하였을 때, 미지 물질들은 이온성분들과 밀접한 연관성을 보일뿐 아니라 TSP의 함량과 정비례 관계임을 확인하였다. 반면, 양자를 TSP의 농도로 표준화(normalization)를 하였을 경우, 뚜렷한 반비례 관계에(강한 역상관성)이 두드러지게 나타났다. 이러한 현상은 ${\Sigma}X$가 이온성분과는 달리 수용성이 떨어지는 특성을 지닌 것을 반영한 것으로 사료된다. TSP의 거동을 보다 체계적으로 이해하고 이를 도시 대기질 관리에 적용하기 위해서는, ${\Sigma}X$의 조성이나 거동을 명확하게 규정하기 위한 여러 가지 연구가 필요하다.

석유계 UVCB 물질의 산업안전보건법 규제 적용을 위한 일부 휘발성 유기화합물 분석 (An analysis of Volatile Organic Compounds for the Application of Petroleum-based UVCB Substances to the Occupational Safety and Health Act)

  • Objectives: To ensure that employers handling petroleum compounds understand whether petroleum-based UVCB (unknown or variable composition, complex reaction products, or biological materials) substances contain hazardous substances and comply with the Occupational Safety and Health Act, petroleum-based UVCB substances were analyzed and the results from samples were compared with MSDS (Material Safety Data Sheet) contents. Methods: Twenty-one petroleum samples were analyzed using GC-MS (Gas Chromatography-Mass Spectrometry), targeting ten volatile organic compounds regulated by the Occupational Safety and Health Act. Results: The target chemicals were detected in 13 out of 21 samples. All 13 samples were in the naphtha (low boiling point naphthas (gasolines)) group. There were also naphtha group samples containing 2% benzene. Some naphtha samples used as solvents contained about 1% benzene. Conclusions: This study shows that naphtha group petroleum substances contain hazardous chemicals in many cases. In particular, if benzene, n-hexane, and toluene with low occupational exposure limits are contained above the limit concentration. Such information should be delivered in the article on MSDS legal regulations.

가압조건의 마이크로웨이브 추출에서 Ginsenosides의 추출특성 모니터링과 추출효율 비교 (Monitoring of Extraction Characteristics and Comparison of Extraction Efficiencies for Ginsenosides in the Microwave-Assisted Process Under Pressure)

  • 가압형 마이크로웨이브 추출장치를 이용하여 추출조건(에탄올농도, 추출온도 및 추출시간)에 따른 인삼 ginsenosid서 추출특성을 반응표면분석에 의해 모니터링하고, 마이크로파 추출법의 추출효율을 확인하였다. 조건별 추출물의 ginsenoside함량에 대한 회귀식의 $R^2$는 대부분 0.830이상으로 $10\%$이내의 수준에서 유의성이 인정되었다. $Ginsenoside-Rb_2$ -Rc, -Re 및 $-Rg_1$은 추출온도가 높을수록 증가하여 $140^{\circ}C$에서 가장 높게 나타났으며, 에탄을 농도가 $50\~75\%$에서 가장 높은 추출율을 보였다. Ginsenoside의 HPLC 패턴은 MAP 추출온도 $120^{\circ}C$에서 unknown peak가 확인되었으며, $150^{\circ}C$추출에서는 unhown peak가 증가되었다. $Ginsenoside-Rb_2$와 -Re 함량에 미지화합물(unknown peak)을 포함하였을 때 는 최대의 추출온도가 $129^{\circ}C$에서 $147^{\circ}C$로 증가하였고, 동시에 추출시간과 $R^2$가 증가하였다. MAP 추출에서 unknown화합물의 함량이 최소가 되는 조건은 에탄올 농도 $67.33\%$, 추출온도 $99.34^{\circ}C$, 추출시간 3.65분이었다. 추출효율의 확인시험에서 8분간의 MAP추출법은 현행 추출법(40시간) 매우 유사한 수준의 ginsenoside 추출효율을 나타내었다.

십전대보탕 발효물의 성분 분석 및 뇌신경 세포 보호 활성 (The Study on Compounds of the Fermented Sipjundaebo-tang and its Neuroprotective Activity)

  • Sipjundaebo-tang was a well-known restorative traditional herbal prescription that used to treat anemia, anorexia, fatigue and inflammation. In this study, we examined the bioconversion of compounds in the Sipjundaebo-tang (SJ) and fermented Sipjundaebo-tang with Lactobacillus fermentum KFRI 164 (FSJ) using established HPLC-DAD method. The chromatogram of FSJ has shown that the contents of six bioactive compounds 5-HMF, paeoniflorin, ferulic acid, cinnam aldehyde, decursin, glycyrrhizin in SJ has decreased. And the contents of unknown compounds (1), (2), (3), (4) and (5) in FSJ were higher than each contents of SJ. The antioxidant activities of SJ and FSJ were conducted by DPPH free radical and Hydrogen peroxide ($H_2O_2$) scavenging assay. Electron donating activity (EDA, %) value of FSJ has shown higher than 21.9% and 14.5% at a concentration of 0.5 mg/ml for DPPH radical scavenging activity and $H_2O_2$ scavenging activity, respectively. Also, the neuroprotective activities of SJ and FSJ against glutamate-induced oxidative stress in a mouse hippocampal cell line (HT22) were evaluated by MTT assay. As a result, FSJ has shown higher neuroprotective activity than 56.5% comparing with SJ. In conclusion, the fermented SJ using microorganism could convert compounds in SJ and enhance antioxidant activity and neuroprotective activity.
