Padina arborescens extract protects high glucose-induced apoptosis in pancreatic ${\beta}$ cells by reducing oxidative stress

  • BACKGROUND/OBJECTIVES: This study investigated whether Padina arborescens extract (PAE) protects INS-1 pancreatic ${\beta}$ cells against glucotoxicity-induced apoptosis. MATERIALS/METHODS: Assays, including cell viability, lipid peroxidation, generation of intracellular ROS, NO production, antioxidant enzyme activity and insulin secretion, were conducted. The expressions of Bax, Bcl-2, and caspase-3 proteins in INS-1 cells were evaluated by western blot analysis, and apoptosis/necrosis induced by high glucose was determined by analysis of FITC-Annexin V/PI staining. RESULTS: Treatment with high concentrations of glucose induced INS-1 cell death, but PAE at concentrations of 25, 50 or $100{\mu}g/ml$ significantly increased cell viability. The treatment with PAE dose dependently reduced the lipid peroxidation and increased the activities of antioxidant enzymes reduced by 30 mM glucose, while intracellular ROS levels increased under conditions of 30 mM glucose. PAE treatment improved the secretory responsiveness following stimulation with glucose. The results also demonstrated that glucotoxicity-induced apoptosis is associated with modulation of the Bax/Bcl-2 ratio. When INS-1 cells were stained with Annexin V/PI, we found that PAE reduced apoptosis by glucotoxicity. CONCLUSIONS: In conclusion, the present study indicates that PAE protects against high glucose-induced apoptosis in pancreatic ${\beta}$ cells by reducing oxidative stress.

A Comparative Study on Cognition and Preference of Korean Traditional Food Classified by Age in Busan (부산지역 주민의 한국전통음식에 대한 연령별 인식 및 선호도 조사)

  • This study was carried out to investigate the Korean traditional foods cognition and preference classified by age in Busan. The survey was conducted from January 2 to March 31, 2008, by questionnaires and data analyzed by SPSS program. The results are summarized as follows: Thirty-four point nine percent of the subjects were interested the Korean traditional foods. Over 40's were more interested in the foods than under 30's. Most of the subjects (65.5%) were proud of the Korean traditional foods and the most proud food was kimchi in all of the subjects. The improvement for Korean traditional foods was indicated as cooking method. There was a significant positive correlation between preference of bab, juk, guksu, gug, jeon-juk-sun, gui-jjim, jigae, jorim, bokeum, kimchi, namul, eumcheong and cognition degree of the Korean traditional foods. Therefore, the understanding of the requirements and preference of the subjects according to age and sex is needed to develop our traditional food.

Association between Preferences of Salty Food and Acuity and Preferences of Taste of the Elderly People Living in Rural Area (농촌 노인의 맛 감지능 및 맛 기호도와 짠음식 기호도)

  • The purpose of the study was to investigate the influence of salt (NaCl) recognition threshold and pleasant salt concentrations of Korean rural elderly subjects on preference of salty food as well as food group consumption patterns. The subjects were 213 elderly people (male 71 and female 142) over 65 and under 90 yr of age from Sunchang area. The higher values were found with male subjects in the areas of basic taste recognition threshold and pleasant concentration level of salt. The taste sensitivity scores of male subjects did not decrease with the increasing age, but female subjects exhibited a significant decrease with the age. The major determinant factor of salt preference of the elderly subjects in this study appeared to be personal pleasant concentration of salt rather than salt recognition threshold level and this trend was more evident in males than in females. The subject group of lower salt pleasant concentration i.e. consuming less salt showed the higher number of fruit and fruit juice intakes per week, and higher MNA (define this) scores were implying that they display more desirable nutritional status. Therefore, nutritional education focused on not only a variety of food consumption but also lowering pleasant salt concentration levels is advised to improve the quality of nutrition in the elderly.

High-Level Expression of Pseudomonas sp. LBC505 Endoglucanase Gene in Escherichia coli

  • Endoglucanase gene of Pseudomonas sp. LBC505 was previously cloned in pUC19 to yield plasmid pLCl. The Pseudomonas sp. LBC505 endoglucanase gene was subcloned in a temperature-regulated Es-cherichia coli expression vector, pAS1, containing the leftward promoter $P_L$ of bacteriophage lambda. The level of gene expression was controlled by the thermal inactivation of the heat-sensitive lambda cI857 repressor. Best yield of endoglucanase was obtained by lowering the incubation temperature to $37^{\circ}C$ after induction at $42^{\circ}C$ for 1h. Under these conditions enzyme production continued for about 5h at a gradually decreasing rate. Ecoli harboring recombinant plasmid pASC10 expressed 4.3 times as much CMCase activity as E.coli containing pLCl. To enhance the expression level of endogl, ucanase gene, we have also changed the presumptive Shine-Dalgamo sequence (AGAGGT) of the gene to consensus sequence (AGGAGGT) by site-directed mutagenesis. The genes mutated were subcloned in pASl resulting in the formation of recombinant plasmid pASS50. E.coli harboring the plasmid pASS50 expressed 6.2-fold higher levels of CMCase activity than that of E.coli harboring pLC1.

Analysis on Eating Behavior and Nutritional Status of Nephritic Patients with Mild Kidney Malfunction in Korea (신장염 환자의 식행동 및 영양상태 평가)

  • This study investigated nutritional status and eating behaviors among 59 nephritic patients with mild kidney malfunction in Korea. Nutritional status was measured by blood analysis and 1-3 day dietary recall and records, and eating behaviors were assessed by a questionnaire. Mean Body Mass Index(BMI) was within the normal range, while 21% and 14% of the patients were underweight and overweight, respectively. They received nutrition information mainly from doctors, nurses and mass media, but rarely from nutrition professionals. A quarter of patients skipped breakfast at least 3 times per 12% of the patients, respectively. Energy intake of 83% of the patients was less than the recommended level. Protein intakes of 56% of them were either under or over the recommended levels. Patients had low vitamin B$_2$ and calcium(<75% of the RDA) and excessive phosphorus(138% RDA) and vitamin C(170% RDA). Major food sources to absolute nutrient intakes were similar to those for the average Koreas, except for milk. Milk intake was low in our patients. Dietary quantity and quality were associated positively with BMI, albumin, and HDL-cholesterol and negatively with triglycerides and BUN. Results of this study indicate low nutritional status of the patients and, in turn a need for conducting nutritional education or counselling at regular at regular basis for the nephritic patients with mild kidney malfunction.

Effect of vitamin $B_6$ deficiency on antioxidative status in rats with exercise-induced oxidative stress

  • This study investigated the effect of vitamin $B_6$ deficiency on antioxidant enzyme activities and lipid profile in rats with exercise-induced oxidative stress. Forty eight rats were fed either a vitamin $B_6$ deficient diet (B6-) or a control diet (control) for 4 weeks and then subdivided into 3 groups: pre-exercise (PreE); post-exercise (PostE); recess after exercise (recessE). Compared to those of control group, plasma catalase and hepatic cytosol superoxide dismutase (SOD, EC 1.15.1.1) activities of B6- group were lower regardless of exercise. The ratio of reduced glutathione/oxidized glutathione (GSH/GSSG) of B6 - group was lower in PreE and there was no difference between PostE and recessE. The level of malondialdehyde (MDA) of B6- was significantly higher in PreE and PostE. High-density lipoprotein-cholesterol (HDL-C) level of B6- group was lower regardless of exercise. Atherosclerotic index of $B_6$- group was higher in PreE and there was no difference between PostE and recessE. It is suggested that a reduction in antioxidative status caused by vitamin $B_6$ deficiency may be aggravated under exercise-induced oxidative stress.

Identification of Carotenoid Oxidation Products in Pigment Extracts from Star Ruby Grapefruit Pulp at Different Temperatures with Exposure to Light

  • Pigment extracts obtained from Star Ruby grapefruit pulp were stored at different temperatures (4.5$^{\circ}C$, 23$^{\circ}C$) and exposed to light. many carotenoid oxidation products were formed due to light-exposure during storage periods. They were monitored by using HPLC with photodiode array detection and tentatively identified. Including (all-E)-lycopene and trans-$\beta$-carotene as predominant carotenoids in red grapefruit, 5Z-lycopene, 9Z-lycopene, 13Z-lycopene, and 15-cis-$\beta$-carotene were formed at 4.5$^{\circ}C$, 23$^{\circ}C$. Degradation of all-tarns lycopene was more susceptible to light-exposure and temperature a than that of all-trans $\beta$-carotene. The formation of lycopene cis isomers was favored under lighted condition. Respectively, (5Z)-lycopene was formed in greater amounts than other isomers at 23$^{\circ}C$ storage. The concentration of 15-cis-$\beta$-carotene was significantly increased during storage at 23$^{\circ}C$ storage. The concentration of 15-cis-$\beta$-carotene was significantly increased during storage at 23$^{\circ}C$.

A Study of Effects of Ilex serrata Thunb Extracts (낙상홍 추출물의 효능에 관한 연구)

  • In this study, Ilex serrata Thunb was extracted with 70% methanol and concentrated under reduced pressure to measure its total polyphenol contents; furthermore, we examined the effect of electron donating ability of the various extracts. The extract was fractionated using different solvents such as methylene chloride, ethyl acetate, and methanol to investigate total polyphenol contents and electron donating abilities. The total polyphenol contents were $1,058.59{\pm}85.85{\mu}g\;GAE/mL$ in 70% methanol, $297.29{\pm}29.43{\mu}g\;GAE/mL$ in methylene chloride, $313.28{\pm}9.22{\mu}g\;GAE/mL$ in ethyl acetate, and $837.82{\pm}31.16{\mu}g\;GAE/mL$ in methanol. The total polyphenol contents showed significant differences (p<0.05) between the solvents. The electron donating abilities were $72.93{\pm}0.29%$ for 70% methanol extract, $14.02{\pm}2.01%$ for methylene chloride, $24.05{\pm}1.75%$ for ethyl acetate, and $84.08{\pm}1.95%$ for methanol. The electron donating abilities were significantly different (p<0.05) between the solvents. The two portions of Ilex serrata Thunb extracts obtained from methanol showed significantly relevant results in the total polyphenol contents and electron donating ability, which was higher than the original extract.

Comparing Nutrition Knowledge, Eating Habits, and Neonatal Health Status of Primipara for Pregnant Women of Advanced Maternal Age Compared to Those of Younger Ages (고령 초산모와 비고령 초산모의 임신영양지식, 식습관 및 신생아 건강상태 비교)

  • Purpose: The aim of this study was to examine differences in nutrition knowledge, eating habits during pregnancy, and neonatal health status between primipara for pregnant women of advanced maternal age in comparison to those under the age of 35. Methods: This study used a comparative survey design. Data were collected through self-report questionnaires and patients medical records. A total of 127 participants, mothers after delivery were recruited from metropolitan city B. Results: Primipara in advanced maternal age (n=32) reported significantly higher scores of eating habits (Z=-2.96, p=.003) than younger ages (n=95). There were no significant differences in scores of pregnancy nutrition knowledge (Z=-0.44, p=.660), duration of gestation (Z=-0.28, p=.778), neonatal birth height (Z=-0.10, p=.924), neonatal birth weight (Z=-0.28, p=.777), Apgar score 1 minute (Z=-0.53, p=.599) and 5 minutes (Z=-0.23, p=.816) between two groups. Conclusion: It concludes that age is not the obstacle to the best nutritional status of women and their newborns.

Food Spoilage by Pseudomonas spp. (Pseudomonas spp.에 의한 부패)

  • Pseudomonas spp. are Gram-negative psychrophilic bacteria, which can proliferate at refrigeration temperature. The bacteria produce heat-stable enzymes that can degrade fat and protein in foods. Hence, Pseudomonas spp. are related to the spoilage of milk, dairy products, and meat products under cold storage, causing economic loss. In the food industry, various methods have been used to remove bacteria including Pseudomonas spp. in food-related conditions, but they can be resistant to antimicrobials and sanitizers because they form biofilms regulated by quorum sensing (cell density-dependent cell-to-cell signaling). Since Pseudomonas cells in biofilms can cross-contaminate foods resulting in food spoilage and the survival of food-borne pathogens in food-related conditions, efficient decontamination technology and microbiological criteria should be established to reduce the occurrence of food spoilage by Pseudomonas spp.
