Thalictrum rochebrunianum var. grandisepalum (TRG) is a Korean endemic plant, and it is widely used for edible, medicinal, landscape materials. In this study, we examined the protein and mRNA expression levels of MITF, tyrosinase, TRP-1 and TRP-2 by TRG extract (TRGE) in IBMX-treated melanocytes to evaluate the possibility of using TRG as a whitening material. IBMX were reported as melanin synthesis enhancers. It could increase intracellular melanin synthesis by activation of the microphthalmia-associated transcription factor (MITF) signaling pathway. TRGE did not show cytotoxicity at concentrations below $100{\mu}g/ml$ in B16F10 cells. TREG dose-dependently inhibited protein and mRNA levels of MITF, tyrosinase, TRP-1 and TRP-2. Therefore, we suggest that TRGE is an important natural resource for cosmetic raw materials for whitening function.

Melanogenesis is a well-known physiological response of human skin that may occur because of exposure to ultraviolet light, for genetic reasons, or due to other causes. In our effectors to find new skin lightening agents, acanthoic acid (AA) was investigated for its ability to inhibit melanogenesis. The effects of AA isolated from A.koreanumun the expression of $\alpha$-MSH-induced melanogenic factors (tyrosinase, tyrosinase related protein (TRP)-1, TRP-2 and MITF (microphthalmla-associated transcriptional factor)) were investigated in murine B16F10 melanoma cells. The results indicate that AA was an effective inhibitor of melanogenesis in B16F10 cells. To elucidate the mechanism of the effect of AA on melanogenesis, we performed Western blotting for melanogenic proteins. AA inhibited melanogenic factors (tyrosinase, TRP-1, TRP-2) expressions. In this study, we also confirmed that AA decreased the protein level of MITF proteins, which would lead to a decrease of tyrosinase and related genes in B16F10 melanoma cells. In order to apply AA to the human skin, the cytotoxic effects of the AA were determined by MTT assays using human keratinocyte HaCaT cells. Based on these results, we suggest that AA be considered possible anti-melanogenic agent and might be effective against hyperpigmentation disorders for the topical application.
Objectives : The various grape extracts derived from grape pulp, seed and skin, containing various types of polyphenols and flavonoids, have been known to have anti-inflammatory, antioxidant and improve cardiovascular condition as well as sun's damaging effects. However, there have been rare reports of various beneficial effects of grape fruit stem extract (GFSE), one of the waste products of grapes. We investigated anti-inflammatory and melanogenesis inhibitory effects of GFSE. Methods : One-hundred gram of grape fruit stem was extracted with 80% ethanol at room temperature for 3 days. After filtration, the ethanol was removed using vacuum evaporator, then lyophilized to obtain the dry extract which was stored at $-20^{\circ}C$ until used. NO levels were measured by using Greiss reagent. Prostaglandin $E_2$ ($PGE_2$) production was measured by ELISA assay. The expression levels of iNOS, COX-2, TRP-1 and TRP-2 were evaluated by western blot analysis. Results : GFSE reduced the level of nitric oxide and prostaglandin $E_2$ ($PGE_2$) production in a dose-dependent manner, compared to control. Expressions of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) protein were also effectively inhibited by the GFSE. In a tyrosinase inhibitory activity, GFSE significantly reduced the tyrosinase activity and melanin content in a dose dependent manner, compared to control. GFSE also decreased the expression of tyrosinase related protein-1 (TRP-1) and tyrosinase related protein-2 (TRP-2), known as a melanocyte-specific gene product involved in melanin synthesis. Conclusions : Therefore, these results indicated that GFSE had powerful anti-inflammatory and anti-melanogenic effects.
본 연구에서 루꼴라(E. sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 루꼴라 추출물 처리 시 항산화 활성($ID_{50}$, 17.60 mg/L)이 매우 우수하게 나타났으며 루꼴라 추출물 처리시 높은 tyrosinase 활성저해($ID_{50}$, 132.54 mg/L) 및 멜라닌 함량 저하($ID_{50}$, 158.90 mg/L)를 보여주었다. 또한, 루꼴라 추출물 농도 의존적으로 tyrosinase 발현이 억제되었으며, 이는 루꼴라 추출물이 직접적인 tyrosinase 활성저해 및 세포 내 tyrosinase 발현을 억제시킴으로서 멜라닌 합성을 저해하는 것으로 판단된다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부미백 소재 등 피부개선 효과를 지닌 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단되며 향후 주름개선 효과 등 추가적인 실험을 통하여 화장품 소재 개발 응용 가능성을 타진할 것이다.
Objectives : Under normal condition melanin protects the skin from extracellular stimuli including ultraviolet (UV)-induced oxidative skin damages, but excess production and accumulation of melanin can induce hyperpigmentation causing esthetic problems. Therefore, in this study we tried to search for natural skin whitening materials from marine natural resources. Methods : Water and ethanol extracts of marine natural resources were prepared from Porphyra thalli (PT), Laminariae thallus (LT), Ostreae concha (OC), Sargassum thallus (ST), Undaria thallus (UT), Codium thalli (CT), Enteromorpha thalli (ET), Syngnathoides biaculeatus (SB), and Hippocampus coronatus (Hc). Their effects against UVB and ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis were investigated based on melanin formation in B16 mouse melanoma cells. The mRNA and protein expression of enzymes involved in the melanogenic process were further examined by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, respectively. Results : Water extract of Ostreae concha (OCW/E) effectively inhibited UVB and ${\alpha}$-MSH-induced melanin production in B16 melanocytes, which seemed to be mediated by inhibition of mRNA expression of tyrosinase and tyrosinase-related protein 1 (TRP-1). In another experiment, ethanol extracts from Porphyra thalli (PTE/E), Laminariae thallus (LTE/E), Sargassum thallus (STE/E), Undaria thallus (UTE/E), Codium thalli (CTE/E), Syngnathoides biaculeatus (SBE/E), and Hippocampus coronatus (HcE/E) significantly suppressed UVB and ${\alpha}$-MSH-induced melanin formation. Furthermore, ethylacetate fraction isolated form LTE/E (LTE/EEt) decreased UVB and ${\alpha}$-MSH-elevated extracellular melanin levels via inhibition of tyrosinase protein expression. Conclutions : These results suggest that marine natural resources such as Porphyra thalli, Laminariae thallus, Ostreae concha, Sargassum thallus, Undaria thallus, Codium thalli, Syngnathoides biaculeatus and Hippocampus coronatus have anti-melanogenic effects, thereby exhibiting high potentials to be utilized as one of the ingredients for the development of new whitening functional cosmetics.
본 연구에서는 화장품 천연소재로서 해죽순 열수 추출물의 이용 가능성을 확인하였다. tyrosinase 저해활성을 측정한 결과 1,000 ㎍/mL에서 52.0%의 활성을 나타내었다. 세포 생존율을 MTT 분석법으로 확인한 결과 멜라노마 세포(B16F10)의 농도 구간이 100 ㎍/mL 일 때 84.8%의 생존율을 보였다. Western blot을 통한 단백질 발현 억제 효과를 측정하기 위해 25, 50, 100 ㎍/mL 농도의 해죽순 열수 추출물과 β-actin을 사용하였다. 그 결과, MITF, TRP-1, TRP-2, tyrosinase의 단백질 발현양이 100 ㎍/mL에서 70.7%, 83.3%, 45.7%, 45.9%로 억제됨을 확인하였다. 따라서 해죽순 열수 추출물의 미백효과가 우수함을 확인하였고, 해죽순 열수 추출물의 화장품 소재로서의 가능성을 확인하였다.
본 연구는 말오줌나무 70% 에탄올 추출물의 항산화 효과와 미백 효과를 검증하였다. 말오줌나무 추출물의 전자공여능 측정실험은 $1,000{\mu}g/ml$에서 86.21%의 효과를 나타내었고, ABTS 소거능을 측정한 결과 $1,000{\mu}g/ml$ 농도에서 97.9%의 소거능을 보였다. Tyrosinase 저해활성 측정은 $1,000{\mu}g/ml$ 농도에서 37%의 효과를 보였다. 말오줌나무 추출물의 세포 생존율을 melanoma cell (B16F10)에서 확인하기 위해 MTT assay를 진행하였는데 세포 생존율을 측정한 결과, $100{\mu}g/ml$ 농도에서 90% 이상의 생존율을 보였다. 말오줌나무 추출물의 MITF, TRP-1, TRP-2 및 tyrosinase의 단백질 발현 효과를 25, 50, $100{\mu}g/ml$ 농도에서 측정하였으며 양성 대조군으로 ${\beta}-actin$을 사용하였다. 그 결과, MITF, TRP-1, TRP-2 및 tyrosinase의 각각 $100{\mu}g/ml$ 농도에서 34.5%, 45.6%, 58.4%, 79.6%의 단백질 발현 억제 효과를 보였다. MITF, TRP-1, TRP-2 및 tyrosinase의 mRNA발현을 RT-PCR로 25, 50, $100{\mu}g/ml$ 농도에서 측정하였고, 양성 대조군으로 GAPDH를 사용하였다. 그 결과, 말오줌나무 추출물의 $100{\mu}g/ml$ 농도에서 각각 85.4%, 67.5%, 85.2%, 67.1%의 mRNA 발현이 감소함을 확인 할 수 있었다. 따라서, 말오줌나무 추출물이 미백 소재로서의 가능성을 확인할 수 있었다.

Abnormal melanin synthesis results in several hyperpigmentary disorders such as freckles, melanoderma, age spots, and other related conditions. In this study, we investigated the anti-melanogenic effects of an extract from the microalgae Chlamydomonas reinhardtii (CE) and potential mechanisms responsible for its inhibitory effect in B16F10, normal human epidermal melanocyte cells, and human skin-equivalent models. The CE extract showed significant dose-dependent inhibitory effects on ${\alpha}$-melanocyte-stimulating, hormone-induced melanin synthesis in cells. Additionally, the CE extract exhibited suppressive effects on the mRNA and protein expression of microphthalmia-associated transcription factor, tyrosinase, tyrosinase-related protein-1, and tyrosinase-related protein-2. The CE extract also inhibited the phosphorylation of protein kinase A and extracellular signal-related kinase, which function as upstream regulators of melanogenesis. Using a three-dimensional, reconstructed pigmented epidermis model, the CE-mediated, anti-pigmentation effects were confirmed by Fontana-Masson staining and melanin content assays. Taken together, CE extract can be used as an anti-pigmentation agent.
본 연구는 B16F10 melanoma 세포에서 산딸기 잎 추출물(Rubus crataefifolius Leaf Extract, RCLE)의 멜라닌 생성 억제 효과를 확인하고자 수행되었다. α-MSH로 자극한 B16F10 melanoma 세포에서 멜라닌 함량과 tyrosinase 활성, 멜라닌 생성관련 효소들인 TRP-1, TRP-2 및 MITF의 단백질 발현 수준을 조사하였고 이에 대한 RCLE의 효과를 검증하였다. RCLE는 tyrosinase 활성과 멜라닌 생성을 효과적으로 억제하였고 멜라닌 생성 경로에 관여하는 PKA와 CREB의 인산화와 MITF의 발현을 억제하였으며 멜라닌 생성 관련 단백질의 발현을 하향 조절하였다. 이러한 결과는 RCLE가 MITF 발현을 억제하여 α-MSH로 자극된 멜라닌 합성을 억제한다는 것을 보여준다. 따라서 이러한 연구결과는 RCLE가 과도한 멜라닌 생성으로 인한 색소 침착을 완화시킬 수 있는 기능성 화장품 소재로 활용될 수 있음을 시사한다.
