The influences of Phaseolus radiatus L.'s Ethanol Extracts and Fractions on Skin Whitening and Anti-inflammatory Effects (녹두(綠豆) 에탄올 추출물 및 분획물이 피부의 미백, 항염증 효과에 미치는 영향)

  • Objectives : This study is designed to clarify whitening, anti-inflammatory effect of fractions extracted from the mixture of Phaseolus radiatus L. and ethanol. Methods : In this experiment, we were intended to reveal whitening, anti-inflammatory effect of fractions extracted from the mixture of Phaseolus radiatus L. and ethanol. The whitening activity was confirmed by UV blocking activity, tyrosinase inhibiting activity, and melanin formation inhibiting activity. Anti-inflammatory activity is confirmed by measurement of cytotoxicity level by MTT assay and measurement of Cytokine expression, which is the main mediator of inflammation reaction. Results : As a results, overall activity was high in the ethyl acetate fraction. Tyrosinase inhibitory activity was less than 20% at all concentrations, but the activity to inhibit melanin self-production was higher than that of ethyl acetate fraction at $32.19{\pm}2.79%$ at $100{\mu}g/m{\ell}$. And ethyl acetate fraction had a relatively high UV blocking activity. In the anti-inflammatory test, the concentration-dependent activity was shown, and the chloroform and ethyl acetate fractions showed significant NO production inhibitory activity. Cytokine expression was superior to that of the final stage of B cell differentiation, and cell viability was over 80% except for the chloroform fraction at the concentration of $200{\mu}g/m{\ell}$. Conclusions : The results of this experiment confirmed the whitening effect and anti-inflammatory effect of Phaseolus radiatus L.'s extracts and fractions and report the possibility of application as external medicine.

  • Objectives : The cosmeceutical activity of Cuacuta japonica Choisy were investigated for cosmetics industrial application. Methods : Cosmeceutical activities include antioxidant, tyrosinase inhibition effects and nitrite-scavenging ability were examined through the Cuacuta japonica Choisy Results : The result were obtained as follows : The electron donating ability of water and ethanol extract was 69.5%, 78.3% at 1,000ppm. In SOD-like activity, water extract showed low effect, but ethanol extract scored 63.4% at 1,000ppm. In inhibition activity of xanthine oxidase, water extract showed low effect, which is 27.2% at 500ppm, in contrast to ethanol extract high effect: 99.5% at 500ppm, in nitrite-scavenging ability test, 500ppm of C. japonica Choisy water extract showed the effect of 51.3%, while 500ppm of C. japonica Choisy ethanol extract showed the effect of 72.7%. From the test of inhibitory effect of tyrosinase, both water and ethanol extract showed insufficient inhibitory rate. Conclusion : According to all the test results that have been given, it is possible that the extract of C. japonica Choisy can be used as a new natural material of the cosmeceutical industry.

Effects of Inoculation of Bacillus subtilis Cells on the Fermentation of Korean Traditional Soy Paste(Doenjang) (메주 제조 시 Bacillus subtilis의 첨가가 재래식 된장의 발효에 미치는 영향)

  • Korean traditional soy paste(Doenjang) was fermented for 90 days using, as a starter, a Meju prepared by inoculation of Bacillus subtilis cells. Changes in physiochemical and functional properties were investigated during fermentation. Amylase and protease activities increased and showed maximal levels(4.11 and 7.75 units/mL, respectively) after 60 days of fermentation. Both enzyme activities then fell. Inhibitory activities of the soy paste against tyrosinase and angiotensin converting enzyme(ACE) increased during the fermentation period. Antimutagenic activities during fermentation were determined using the S. enterica serovar Typhimurium TA100 and TA98 analysis system. No significant differences in activity were observed in soy pastes prepared with or without B. subtilis. Antimutagenic activities against the activities of N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitro-O-phenylenediamine (NPD) increased and reached 70.33% and 60.22%, respectively, after 90 days of fermentation, as assessed using the tester strain TA100. Against the actions of NPD and 4-nitroquinoline-1-oxide(NQO), antimutagenic activities also increased during fermentation and reached 50.84% and 47.01%, respectively, as assessed using the tester strain TA98. The genistin content was much higher(187.48 g/g) in soy paste prepared using B. subtilis inoculation than the level(31.30 g/g) seen in soy paste prepared without bacterial inoculation, although the contents of daidzein and genistein were slightly reduced under such circumstances.

A Study of Anti-wrinkle Activities as a Functional Cosmetic Ingredient of Rhododendron brachycarpum Extracts (만병초(Rhododendron brachycarpum) 추출물의 기능성 화장품 소재로써의 주름개선 활성에 관한 연구)

  • The purpose of this study was to investigate activities as functional cosmetic materials, focusing on Rhododendron brachycarpum (RB) and Rhododendron fortunei (RF). The tyrosinase inhibitory effect, related to skin-whitening, was 32.6% and 39.3% respectively at the concentration of 1,000 ㎍/ml. The elastase inhibitory effect, related to skin anti-wrinkling activity, was 30% and 36.2% at 1,000 ㎍/ml concentration. Collagenase inhibitory activity showed 77.7%, and 80.2% respectively at 1,000 ㎍/ml concentration, which demonstrated excellent inhibitory activity. For a cell viability test, that measured on fibroblast cells by RB and RF extracts. Furthermore, the cell viability test showed 100.9% and 98.9% with cell viability at 100 ㎍/ml concentration in CCD-986Sk. The protein expression inhibitory effect of RB and RF extracts was measured by western blot at 25, 50, and 100 ㎍/ml concentrations, and the β-actin was used as a positive control. As a result, western blot of RB and RF extracts was measured by the expression inhibition rate of the MMP-1, MMP-2, MMP-3 protein, and was decreased by 67.2%, 65.5%, 13.6%, and 89.1%, 85.0%, and 62.7% at 100 ㎍/ml concentration. The mRNA expression inhibitory effect of RB and RF extracts was measured by RT-PCR at 25, 50, and 100 ㎍/ml concentrations, and the GAPDH was used as a positive control. According to the results of RT-PCR of RB and RF extracts, the expression inhibition rate of the MMP-1, MMP-2, and MMP-3 mRNA was decreased by 70.1%, 9.1%, 37.9%, and 38.2%, 8.3%, 57.3% at 100 ㎍/ml concentrations. So, RB and RF extracts showed the anti-wrinkle effectiveness as a functional cosmetic material.

Enzyme Inhibitory and Anti-Proliferation Effects of Peanut Skin Extracts Depending on Cultivar (품종별 땅콩 종피 추출물의 효소저해활성 및 암세포주 증식억제 효과)

  • The purpose of this study was to investigate the inhibitory effect of enzyme activity and anti-proliferation of human cancer cell lines (HCT 116, NCI-H460 and MCF-7) of peanut skin depending on cultivars (Arachis hypogaea L. cv. K-Ol, cv. Sinpalkwang, cv. Daan, cv. Heuksaeng) and extraction solvent. Peanut skin was extracted with 80% ethanol, 80% methanol, 80% acetone, and distilled water, followed by analysis of the enzyme inhibitory activity and anticancer activity. Methanol extract of Daan cultivar most effectively inhibited ${\alpha}$-gluosidase (65.08%, 0.025 mg/mL), tyrosinase (82.49%, 2 mg/mL) and ACE (73.61%, 10 mg/mL). The inhibitory effect of peanut skin extracts on colon cancer cell (HCT-116), lung cancer cell (NCI-H460) and breast cancer cell (MCF-7) growth were investigate using MTT assay. The highest anti-proliferation of cancer cell line of peanut skin extracts was observed in the methanol extract of Daan cultivar. The cell viability on HCT 116, NCI-H460 and MCF-7 cell lines of methanol extracts from peanut skin of Daan cultivar was 48.13%, 41.03%, and 36.02% at $200{\mu}g/mL$, respectively. These results suggest that peanut skin extracts may mediate physiological activity, and provide valuable information for the use of peanut byproduct as a functional food material.

Antioxidative Effects and Tyrosinase Inhibitory Activities of Mate (Ilex paraguariensis) Extract/Fractions (마테(Ilex paraguariensis) 추출물/분획물의 항산화능 및 타이로시네이즈 저해 활성)

  • In the present study, 50% ethanol extract, the ethyl acetate and aglycone fraction were prepared from mate (Ilex paraguariensis) and their antioxidative ability was evaluated. The yields of extract and fractions were 32.0, 4.48 and 0.82% per dried powder, respectively. Free radical scavenging activities were performed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay and total antioxidant capacity was estimated using luminol-dependent chemiluminescence assay. Free radical scavenging activities ($FSC_{50}$) of 50 % ethanol extract, ethyl acetate fraction and aglycone fraction were 8.83, 5.84 and $6.05{\mu}g/mL$, respectively. Their total antioxidant capacities ($OSC_{50}$) were similar to that of L-ascorbic acid ($1.72{\mu}g/mL$), known as a prominent water soluble antioxidant, in all extracts and 50% ethanol extract ($1.03{\mu}g/mL$) was the most effective. The cellular protective effects on the $^1O_2$-induced cellular damage of erythrocytes were evaluated and the results showed that all extracts were significantly higher than (+)-${\alpha}$-tocopherol at $10{\mu}g/mL$. Especially, the ${\tau}_{50}$ value of aglycone fraction was 5 times higher than (+)-${\alpha}$-tocopherol at $10{\mu}g/mL$ and $50{\mu}g/mL$. The inhibitory effects of the ethyl acetate and aglycone fractions on tyrosinase were similar to arbutin, known as the whitening agent in cosmetics. These results suggest that the extracts of mate have the applicability as antioxidant and anti-aging cosmeceutical ingredients.

Hesperidin Suppresses Melanosome Transport by Blocking the Interaction of Rab27A-Melanophilin

  • We investigated the inhibitory effects of hesperidin on melanogenesis. To find melanosome transport inhibitor from natural products, we collected the structural information of natural products from Korea Food and Drug Administration (KFDA) and performed pharmacophore-based in silico screening for Rab27A and melanophilin (MLPH). Hesperidin did not inhibit melanin production in B16F10 murine melanoma cells stimulated with ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH), and also did not affect the catalytic activity of tyrosinase. But, hesperidin inhibited melanosome transport in melanocyte and showed skin lightening effect in pigmented reconstructed epidermis model. Therefore, we suggest that hesperidin is a useful inhibitor of melanosome transport and it might be applied to whitening agent.

Oleoresin Content and Physiological Activities of Fresh Red pepper by microwave-Assiated Extraction (마이크로웨이브 추출공정에 의한 홍고추 올레오레진의 함량 및 기능적 특성)

  • Water and ethanol extracts of fresh red peppers were obtained by two methods, conventional extraction (CE) and microwave-assiated extraction (MAE), under different extraction conditions. Red pepper oleoresins extracted by CE and MAE were examined in oleoresin yield and physiological activities. The proper extraction time of MAE was about 5 minutes, whereas that of CE was 2 hours. therefore extraction time was decreased drastically by MAE but there was no significance in oleolesin yields. the electron donating abilities also showed negligible difference between two extracts obtained by CE and MAE, and 80% level in all extracts . the nitrite scavenging effect was reduced by increased of ph , and showed a high elimination effect over 85% at ph 1.2. All extracts had a high tyrosinase inhibitory effects of 100%. The angiotensin I-conventing enzyme effect showed higher activity with over 80% in MAE than 70% level in CE. the capsanthin was extracted with ethanol and was 11.4 and 12.9 ${\mu}$moles per 1 g of fresh red pepper by CE and MAE, respectively.

The Study on Pharmacological Activation as Cosmetic Material of Rosa rugosa Thunb. Flowers Extract (해당화꽃 추출물의 화장품소재로서 약리활성에 관한 연구)

  • Objectives : In this study, we investigated the effect of ethanol extract from Rosa rugosa Thunb. flowers(ERF) on the activities of antioxidant, antiwrinkle and whitening. Methods : We measured antioxidant efficacy of ERF by using 1,1-diphenyl-2-picrylhydrazyl(DPPH) assay. Also we confirmed the inhibitory effect of ERF on collagenase activity and melanin synthesis by using collagenase assay kit and dihydroxiphenylalanine staining, respectively. To evaluate the anti-inflammatory effects of ERF, we examined the inflammatory mediator IL-6. Results : ERF showed highly efficacy in DPPH radical scavenging activity. ERF dose-dependently suppressed collagenase activity. Ultraviolet-induced production of IL-6 decreased by ERF treatment. In B16F10 cell, ERF significantly reduced tyrosinase activity and melanin synthesis. Conclusions : From the above results, it was indicated that ERF could be utilized as anti-aging and whitening cosmetic ingredients.

Effects of Three Korean White Ginseng Extracts on Atopic Dermatitis-Related Cytokines, and Antioxidant and Anti-aging Activities (백삼 추출물이 아토피 피부염 관련 사이토카인 분비 및 항산화, 항노화에 미치는 영향)

  • Inflammation that is considered to be mainly related to pathogenesis of atopic dermatitis (AD) is the biological response of a host to stimuli, such as cellular injury or infection. In this study, we investigated the anti-inflammatory and anti-oxidative activities of white ginseng roots by ultra high pressure extraction (Gin-UHP), fermentation followed by ultra high pressure extraction (Gin-UHPF), and polyol extraction (Gin-POL). As a result, ginseng extracts were able to decrease the secretion of pro-inflammatory cytokines (interleukin-8 and tumor necrosis factor-alpha) and immunoglobulin E. Also, Gin-POL had the highest DPPH radical scavenging activity and when we compared the SOD-like activity, Gin-UHP had the highest. Moreover, we looked into the effect of these ginseng extracts on anti-aging to show the possible usefulness as a raw material of cosmetics. As a result, ginseng extracts were able to reduce the production of melanin, and inhibit the tyrosinase and elastase activities in a dose-dependent manner. The extracts also decreased the expression of MMP-1 and had a significant hyaluronidase inhibitory activity. Taken together, these results demonstrate that ginseng extracts may have an improvement effect on AD by using its anti-inflammatory and antioxidant properties.
