This study is to evaluate the effect of different levels of ensiling density on the fermentation quality of guineagrass silages during the early stage of ensiling. Guineagrass at the milky ripe stage was chopped and ensiled into a small-scale laboratory silo at two ensiling density levels (high density at 95 g/silo and low density at 75 g/silo). Three silos per level were opened after six ensiling periods (0.5, 1, 1.5, 2, 3 and 7 days of ensiling) and the fermentation qualities were analyzed. Within the initial 1.5 days of ensiling there were not significant (p>0.05) differences in the fermentation qualities between two density levels silages, and an almost constant pH and no or only small amounts of lactic acid, acetic acid and total volatile fatty acids were detected. However, the high density silage significantly (p<0.05) increased the rate and extent of fermentation after 1.5 days of ensiling, which was well reflected in significantly (p<0.05) faster and larger pH decline and lactic acid production at each elapsed time as compared with the low density silage. This resulted in significantly (p<0.05) lower finial pH and significantly (p<0.05) higher lactic acid content at the end of the experiment. Moreover, there was higher AA content relative to LA in both the H-D and L-D silages during the full fermentation course, and resulted in the AA-type silage. There were generally somewhat or significantly (p<0.05) higher acetic acid, volatile fatty acids and ammonia-N/total nitrogen in the high density silage than in the low density silage during the initial 3 days of ensiling. However, there were higher (p>0.05) ammonia-N/total nitrogen and significantly (p<0.05) higher butyric acid content in the low density silage at day 7 of ensiling. The silages of two density levels showed an initial increase in glucose between 0.5 and 1 day for the high density silage and between 1 and 1.5 days for the low density silage, respectively, thereafter showed a large decrease until the end of the experiment. There were not large differences (p>0.05) in ethanol content between the low density and high density silages that showed small amounts within initial 3 days of ensiling. However, the low density silage had a significantly (p<0.05) higher ethanol content than the high density silage at the end of experiment. From the above results it was suggested that the increase in ensiling density was an effective method to improve the fermentation quality, especially for tropical grasses.
Since 2005, food waste has been separately collected and recycled to animal feed or aerobic compost in South Korea. However, the conventional recycling methods discharge process wastewater, which contain pollutant equivalent to more than 50% of food waste. Therefore, anaerobic digestion is considered as an alternative recycling method of food waste to reduce pollutant and recover renewable energy. Recent studies showed that hydrogen can be produced at acidogenic stage in two-stage anaerobic digestion. In this study, the authors investigated the effects of pretreatment time and pH low set value on continuous mesophilic hydrogen fermentation of food waste. Food waste was successfully converted to $H_2$ when heat-treated at $70^{\circ}C$ for 60 min, which was milder than previous studies using pH 12 for 1 day or $90^{\circ}C$. Organic acid production dropped operational pH below 5.0 and caused a metabolic shift from $H_2/butyrate$ fermentation to lactate fermentation. Therefore, alkaline addition for operational pH at or over 5.0 was necessary. At pH 5.3, the result showed that the maximum hydrogen productivity and yield of 1.32 $m^3/m^3$.d and 0.71 mol/mol $carbohydrate_{added}$. Hydrogen production from food waste would be an effective technology for resource recovery as well as waste treatment.
Development of an effective method for the preparation of dongchimi juice was investigated by addition of NaCl, sucrose and hydrolytic enzymes before fermentation and addition of dongchimi juice during fermentation. The Chinese radish was ground and suspended in water (1:1, w/v) with addition of spices of garlic, green onion and ginger followed by fermentation at $25^{\circ}C$. Increase in NaCl concentration of brinning solution from 1.0 to 5.0% resulted in a significant decrease in the rates of pH decrease and acidity increase. The sugar addition resulted in a faster changes of them, particulary after 24 hours at $25^{\circ}C$. The fermentation rate was also greatly improved by enzymatic hydrolysis with using viscozyme, a commercial polysaccharides hydrolyzing enzyme, before fermentation. When the fermented juices of two stage (pH 5.4 and pH 4.4) were added up to 15% before (pH 5.4 juice) and during (pH 4.4 juice) fermentation, the initial and second stage of fermentation were significantly improved. Therefore a method of addition of sugar, hydrolytic enzymes and dongchimi juice before or during fermentation was suggested for dongchimi juice preparation.

This study investigated the manufacturing conditions of apple vinegar with high acetic acid content following fermentation according to alcohol concentration without any nutrients. We compared and analyzed the quality characteristics of high acetic acid fermentation by varying the initial alcohol content (6%, 7%, 8%, and 9%). In the results, it was possible to manufacture high acetic acid vinegar with 12% titratable acidity and an alcohol content of 6% and 7%. Lower initial alcohol content was associated with higher yield due to a shorter lag phase. For quality characteristics of the high acetic acid apple vinegar, pH was 2.91~3.20, titratable acidity was 12.0%, and organic acid consisted of acetic acid, malic acid, critic acid and oxalic acids. Based on the results, high acetic acid apple vinegar was produced using a two stage fermentation process after alcohol fermentation but the further research is needed to reduce the time of fermentation in fed-batch culture for industrial use.

The steam explosion-chemical pretreatment is a more effective wood pretreatment technique than the conventional physical pretreatment by accelerating reactions during the pretreatment process. In this paper, two-stage pretreatment processes of hardwood were investigated for its enzymatic hydrolysis and the succinic acid yield from the pretreated solid. The first stage pretreatment was performed under conditions of low severity to optimize the amount of solid recovery. In the second stage pretreatment washed solid material from the first stage pretreatment step was impregnated again with chemical (alkaline or chlorine-based chemicals) to remove a portion of the lignin, and to make the cellulose more accessible to enzymatic attack. The effects of pretreatment were assessed by enzymatic hydrolysis and fermentation, after the two stage pretreatments. Maximum succinic acid yield (16.1 g $L^{-1}$ and 77.5%) was obtained when the two stage pretreatments were performed at steam explosion -3% KOH.
Two stage fermentations of glucose/xylose mixture which is similar composition with rice straw hemicellulose hydrolysate were performed by Candida mogii ATCC 18364. In first stage, glucose was consumed rapidly for cell growth in aerobic condition (2 vvm, 300 rpm), then D-xylose was used for xylitol production in semi-aerobic condition (1 vvm, 300 rpm). After 4 days of fermentation, about $24\;g/{\ell}$ xylitol was produced with a yield of 0.58 g/g and volumetric productivity of $0.25\;g/{\ell}{\cdot}h$. To improve the xylitol yield by reduction of xylose consumption for cell growth and maintenance, D-glucose was continuously supplemented during the second stage of fermentation. By D-glucose feeding of $6.8\;g/{\ell}{\cdot}$ day, xylitol was produced up to $29\;g/{\ell}$ with a yield of 0.8 g/g and volumetric productivity $0.30\;g/{\ell}{\cdot}h$ which are 1.2-1.3 times higher than those obtained without D-glucose feeding.
The changes in yields and nutritive composition of whole crop wheat (Triticum aestivum L.) during maturation and effects of maturity stage and lactic acid bacteria (LAB) inoculants on the fermentation quality and aerobic stability were investigated under laboratory conditions. Whole crop wheat harvested at three maturation stages: flowering stage, milk stage and dough stage. Two strains of LAB (Lactobacillus plantarum: LAB1, Lactobacillus parafarraqinis: LAB2) were inoculated for wheat ensiling at $1.0{\times}10^5$ colony forming units per gram of fresh forage. The results indicated that wheat had higher dry matter yields at the milk and dough stages. The highest water-soluble carbohydrates content, crude protein yields and relative feed value of wheat were obtained at the milk stage, while contents of crude fiber, neutral detergent fiber and acid detergent fiber were the lowest, compared to the flowering and dough stages. Lactic acid contents of wheat silage significantly decreased with maturity. Inoculating homofermentative LAB1 markedly reduced pH values and ammonia-nitrogen ($NH_3$-N) content (p<0.05) of silages at three maturity stages compared with their corresponding controls. Inoculating heterofermentative LAB2 did not significantly influence pH values, whereas it notably lowered lactic acid and $NH_3$-N content (p<0.05) and effectively improved the aerobic stability of silages. In conclusion, considering both yields and nutritive value, whole crop wheat as forage should be harvested at the milk stage. Inoculating LAB1 improved the fermentation quality, while inoculating LAB2 enhanced the aerobic stability of wheat silages at different maturity stages.
A two-stage fed-batch fermentation was carried out to increase xylitol productivity by Candida tropicalis. The first stage for cell growth was performed in the pH-stat and continuous fed-batch modes. The higher cell growth and lower ethanol production obtained in the fed-batch mode where the growth medium was fed when pH of culture broth increased over 5.7. And also the effect of oxygen transfer on xylitol production was investigated by changing agitation speed under 0.5 vvm of aeration. The maximum xylitol productivity and yield were obtained at 500 rpm of agitation.

In situ extraction by organic solvent was studied in order to improve the recovery yield of hydrocarbon from the culture of Botryococcus braunii, a green colonial microalga. When the solvent mixture of octanol as an extractive solvent and n-octane as a biocompatible solvent was added to a two-phase column, the algal growth was seriously inhibited, even at a low concentration of polar octanol. Therefore, a two-stage cell-recycle extraction process was proposed to improve the contact area between the organic phase and the aqueous phase. The hydrocarbon recovery with in situ cell-recycle extraction showed a three-fold increase (57% of cell) in yield over that with two-phase extraction. In addition, over 60% of the hydrocarbon could be recovered without serious cell damage by downstream separation when this process was applied to the culture broth after batch fermentation.
