Expression of Cancer-Testis Antigens in Pediatric Cancers

  • Cancer-testis antigens (CTAs) are a group of tumor-associated antigens with more than 140 members whose expression has been shown to be limited to gametogenic tissues and placenta among normal tissues. However, malignant tissues of different origins have shown aberrant and elevated expression of these antigens. Such a pattern of expression endows beneficial properties for use as cancer biomarkers as well as immunotherapeutic targets as a result of the immune-privileged status of the testes. CTAs have been shown to be expressed in pediatric brain tumors, different types of sarcomas, leukemias, and lymphomas as well as neuroblastomas. Although data regarding their expression pattern in childhood tumors are not as comprehensive as for adult tumors, it is supposed that CTA-based immunotherapeutic approaches can also be used for pediatric cancers. However, there are limited data about the objective clinical responses following immunotherapy in such patients. Here we try to review the available information.

  • Odontogenic carcinosarcoma is an extremely rare malignant odontogenic tumor with only a few reported cases. It is characterized by a true mixed tumor showing malignant cytology of both epithelial and mesenchymal components. It has been assumed to arise from pre-existing lesions such as ameloblastoma, ameloblastic fibroma, and ameloblastic fibrosarcoma. To date, the reported cases have exhibited considerably aggressive clinical behavior. The case of an odontogenic carcinosarcoma in the mandible of a 61-year-old male is described herein. The tumor destroyed the cortex of the mandible and invaded the adjacent tissues. Treatment was performed by surgical resection and reconstruction. The purposes of this article are to introduce odontogenic carcinosarcoma through this case study, to distinguish it from related diseases and to discuss features of the tumor in the existing literature.

흉폐부위에 악성소세포종 -증례 보고- (Malignant Small Cell Tumor of the Thoracopulmonary Region - Report of One Case and a Review of the Literature -)

  • 흉폐부위에서 발생하는 악성소세포종양은 1979년 Askin이 보고했고 이를 Askin종양이라고 부른다. Askin종양은 상당히 희귀하며 흉벽에 연조직이나 폐가장자리에서 잘 생기며 특히 젊은 층에 호발한다. 임상적, 병리학적으로 Ewing's sarcoma와 Rhabdomyosarcoma와 상당히 흡사하나 정밀한 전자현미경 관찰시 병리학적으로 별개에 종양으로 간주되었다. Askin종양은 국소재발이 많고 전신으로 잘 터지지 않는 경향이 있으나 생존율은 상당히 나쁜 것으로 되어 있다. 임상적으로나 병리학칙으로 Askin종양에 해당되는 24계 환자를 치료결과와 더불어 보고한다.

  • Osteosarcoma is a common malignant tumor of bone, but mechanisms underlying its development are still unclear. At present, it is believed that the inhibition of normal apoptotic mechanisms is one of the reasons for the development of tumors, so specific stimulation of tumor cell apoptosis can be considered as an important therapeutic method. Livin, as a member of the newly discovered inhibitor of apoptosis proteins (IAPs) family, has specifically high expression in tumor tissues and can inhibit tumor cell apoptosis through multiple ways, which can become a new target for malignant tumor treatment (including osteosarcoma) and might of great significance in the clinical diagnosis of tumors and the screening of anti-tumor agents and carcinoma treatment.

생쥐 종양모델에서 57Co-Vitamin B12의 종양 친화성에 관한 연구 (Affinity for 57Co-Vitamin B12 by a Wide Histologic Variety of Tumor Types in Mice)

  • 종양영상과 치료에 이용할 수 있는 새로운 종양 친화성 물질을 발견하는 것은 핵의학 분야에서 중요하다. Vitamin $B_{12}$는 thymidylate 생합성과정에서 DNA 합성에 필수적인 조효소로서 분열속도가 빠른 세포와 조직 즉 태생기 세포와 일부 종양에 많은 양이 축적된다. 본 연구의 목적은 마우스에서 다양한 조직학적 형태의 종양에 의한 방사표지 vitamine $B_{12}$의 종양 친화성을 결정하여 $^{57}Co$-vitamine $B_{12}$가 종양영상 물질로서 가능한지를 규명하려고 하였다. 17 종류의 종양을 Balb/C와 Balb nu/nu 누드마우스에 피하주사하였다. 종양이 직경 약 $1.0cm^3$ 되었을 때 $^{57}Co$-methylcobalamin을 복강 내로 주사하였다. 24시간 후에 마우스를 희생하여 신장, 간, 종양, 비장, 뼈(우측 대퇴골), 장, 지방조직(복강내), 근육, 혈액(100 ul), 췌장을 제거하여 무게를 챈 후 gamma countcr로 방사능을 측정하였고 cpm/mg tissue로 표시하였다. 이때 마우스 문제에 따른 차이를 없게하기 위해 20g 몸무게로 표준화 하였다. 다양한 종류의 종양에서 $^{57}Co$-vitamine $B_{12}$가 의의 있게 축적되었고 종양대 정상조직의 섭취비도 대부분의 조직에서 높았으며 특히 배후 방사능에 중요한 영향을 미치는 뼈, 근육, 혈액에서 뚜렷하게 높았다. 본 연구의 결과 vitamin $B_{12}$는 종양친화성 물질로서 종양영상에 사용이 가능한 물질로 생각되며 앞으로 종양영상면에서 임상적 이용에 대한 더 많은 연구가 이루어져야겠다.

Tumor-Suppression Mechanisms of Protein Tyrosine Phosphatase O and Clinical Applications

  • Tyrosine phosphorylation plays an important role in regulating human physiological and pathological processes. Functional stabilization of tyrosine phosphorylation largely contributes to the balanced, coordinated regulation of protein tyrosine kinases (PTKs) and protein tyrosine phosphatases (PTPs). Research has revealed PTPs play an important suppressive role in carcinogenesis and progression by reversing oncoprotein functions. Receptor-type protein tyrosine phosphatase O (PTPRO) as one member of the PTPs family has also been identified to have some roles in tumor development. Some reports have shown PTPRO over-expression in tumors can not only inhibit the frequency of tumor cell division and induce tumor cell death, but also suppress migration. However, the tumor-suppression mechanisms are very complex and understanding is incomplete, which in some degree blocks the further development of PTPRO. Hence, in order to resolve this problem, we here have summarized research findings to draw meaningful conclusions. We found tumor-suppression mechanisms of PTPRO to be diverse, such as controlling G0/G1 of the tumor cell proliferation cycle, inhibiting substrate phosphorylation, down-regulating transcription activators and other activities. In clinical anticancer efforts, expression level of PTPRO in tumors can not only serve as a biomarker to monitor the prognosis of patients, but act as an epigenetic biomarker for noninvasive diagnosis. In addition, the re-activation of PTPRO in tumor tissues, not only can induce tumor volume reduction, but also enhance the susceptibility to chemotherapy drugs. So, we can propose that these research findings of PTPRO will not only support new study ideas and directions for other tumor-suppressors, importantly, but also supply a theoretical basis for researching new molecular targeting agents in the future.

FZD6 expression is negatively regulated by miR-199a-5p in human colorectal cancer

  • Colorectal cancer (CRC), the third most common cancer worldwide, also has the highest rate of cancer-related morbidity and mortality. WNT signaling is initiated by binding of WNT to various receptors, including frizzleds (FZDs), and plays a critical role in CRC and other tumor development by regulating proliferation, differentiation, migration, apoptosis, and polarity. Among the members of the FZD family, FZD6 is broadly expressed in various tissues, and its overexpression has been reported in several cancers, suggesting an important role in cancer development. In this study, we investigated the expression of FZD6 in patients with CRC and found it to be increased in tumors, as compared to paired adjacent non-tumor tissues. Additionally, we found that FZD6 expression was negatively regulated by miR199a5p in CRC cells. These results suggest that overexpression of FZD6, mediated by reduced expression of miR-199a-5p, may play an important role in the development of CRC. [BMB Reports 2015; 48(6): 360-366]

  • Undifferentiated pleomorphic sarcoma (UPS), previously known as malignant fibrous histiocytoma, occurs commonly in the soft tissues in adult, but is rare in the maxillofacial region. It consists of undifferentiated mesenchymal tumor cells resembling histiocytes and fibroblasts. The purpose of this article is to report a case of UPS in the mandible. A 44-year-old patient presented with a painful growing mass in the mandible of two months' duration. Computed tomography and positron emission tomography-computed tomography revealed an ill-defined heterogenous, hypermetabolic mass about 4 cm in size in the left mandible invading adjacent soft tissues. A left mandiblulectomy and reconstruction with a fibular free flap were performed. Immunohistochemical study gave a diagnosis of UPS. The patient was referred for adjuvant chemotherapy after surgical removal of the tumor.

Comparison of Two Methods to Extract DNA from Formalin-Fixed, Paraffin-Embedded Tissues and their Impact on EGFR Mutation Detection in Non-small Cell Lung Carcinoma

  • Objective: Molecular pathology tests are often carried for clinicopathological diagnosis and pathologists have established large collections of formalin-fixed, paraffin-embedded tissue (FFPE) banks. However, extraction of DNA from FFPE is a laborious and challenging for researchers in clinical laboratories. The aim of this study was to compare two widely used DNA extraction methods: using a QIAamp DNA FFPE kit from Qiagen and a Cobas Sample Preparation Kit from Roche, and evaluated the effect of the DNA quality on molecular diagnostics. Methods: DNA from FFPE non-small cell lung carcinoma tissues including biopsy and surgical specimens was extracted with both QIAamp DNA FFPE and Cobas Sample Preparation Kits and EGFR mutations of non-small cell lung carcinomas were detected by real-time quantitative PCR using the extracted DNA. Results and Conclusion: Our results showed that DNA extracted by QIAamp and Cobas methods were both suitable to detect downstream EGFR mutation in surgical specimens. Howover, Cobas method could yield more DNA from biopsy specimens, and gain much better EGFR mutation results.

Analysis of FHIT Gene Methylation in Egyptian Breast Cancer Women: Association with Clinicopathological Features

  • Background: Fragile histidine triad (FHIT) gene is a tumor suppressor gene which involved in breast cancer pathogenesis. Epigenetics alterations in FHIT contributes to tumorigenesis of breast cancer. Objective: Our objective was to study FHIT promoter region hypermethylation in Egyptian breast cancer patients and its association with clinicopathological features. Materials and Methods: Methylation-specific polymerase chain reaction was performed to study the hypermethylation of FHIT promoter region in 20 benign breast tissues and 30 breast cancer tissues. Results: The frequency of hypermethylation of FHIT promoter region was significantly increased in breast cancer patients compared to bengin breast disease patients. The Odd's ratio (95%CI) of development of breast cancer in individuals with FHIT promoter hypermethylation (MM) was 11.0 (1.22-250.8). There were also significant associations between FHIT promoter hypermethylation and estrogen, progesterone receptors negativity, tumor stage and nodal involvment in breast cancer pateints. Conclusions: Our results support an association between FHIT promotor hypermethylation and development of breast cancer in Egyptian breast cancer patients. FHIT promoter hypermethylation is associated with some poor prognostic features of breast cancer.
