Growth hormone (GH) treatment has been increasingly widely used for children with GH deficiencies as the survival rate of pediatric patients with malignancies has increased. Both GH and insulin-like growth factor-I have mitogenic and antiapoptotic activity, prompting concern that GH treatment may be associated with tumor development. In this review, the authors examined the relationship between GH treatment and cancer risk in terms of de novo malignancy, recurrence, and secondary neoplasm. Although the results from numerous studies were not entirely consistent, this review of various clinical and epidemiological studies demonstrated that there is no clear evidence of a causal relationship between GH treatment and tumor development. Nonetheless, a small number of studies reported that childhood cancer survivors who receive GH treatment have a small increased risk of developing de novo cancer and secondary malignant neoplasm. Therefore, regular follow-ups and careful examination for development of cancer should be required in children who receive GH treatment. Continued surveillance for an extended period is essential for monitoring long-term safety.

In the era of immunotherapeutic control of cancers, many advances in biotechnology, especially in Ab engineering, have provided multiple new candidates as therapeutic immuno-oncology modalities. Bispecific Abs (BsAbs) that recognize 2 different antigens in one molecule are promising drug candidates and have inspired an upsurge in research in both academia and the pharmaceutical industry. Among several BsAbs, T cell engaging BsAb (TCEB), a new class of therapeutic agents designed to simultaneously bind to T cells and tumor cells via tumor cell specific antigens in immunotherapy, is the most promising BsAb. Herein, we are providing an overview of the current status of the development of TCEBs. The diverse formats and characteristics of TCEBs, in addition to the functional mechanisms of BsAbs are discussed. Several aspects of a new TCEB-Blinatumomab-are reviewed, including the current clinical data, challenges of patient treatment, drawbacks regarding toxicities, and resistance of TCEB therapy. Development of the next generation of TCEBs is also discussed in addition to the comparison of TCEB with current chimeric antigen receptor-T therapy.
연구배경: HSVtk/GCV를 이용한 유전자치료에서 면역반응은 1) adenovirus 혹은 retrovirus와 같이 벡타로 사용된 virus의 단백질, 2) 치료목적으로 이입된 HSVtk 유전자의 생성물, 3) 암세포에 대해서 일어날 수 있다. 그리고 이러한 면역반응은 cytokines의 생성 혹은 cytotoxic tumor-specific T-cell의 생성을 초래하여 bystander effect에 의한 살상효과를 증가시키거나, anti-tumor immunity를 유도하여 tumor vaccine의 효과를 나타낼 수 있다. 한편 이와는 대조적으로 면역반응용 HSVtk 유전자를 발현하는 세포들을 파괴하여 이입된 HSVtk 유전자의 발현기간을 제한함으로서 유전자치료의 효과를 감소시킬 수도 있다. 본 연구는 retrovirus 벡타로 이입한 HSVtk 유전자치료에서 면역체계가 bystander effect에 의한 살상효과에 미치는 영향을 규명하고 면역체계가 이입한 유전자의 발현에 미치는 영향을 조사하고자 하였다. 방 법: Immunocompetent mice인 Balb/c mouse와 immunodeficient mouse인 Balb/c-nude 및 SCID mouse에서 retrovirus 벡타를 사용하여 HSVtk 유전자를 이입하고 치료효과를 조사하였다. 그리고 Balb/c mouse에 면역억제제인 cyclosporin을 투여하여 면역억제제가 bystander effect 및 유전자치료 효과와 유전자의 발현기간에 미치는 영향을 조사하였다. 결 과: Balb/c mouse에 HSVtk 유전자를 이입하고 GCV를 투여한 군은 GCV를 투여하지 않은 대조군에 비해 종양의 성장이 유의하게 억제되었으나 Balb/c-nude mouse와 SCID mouse의 경우 GCV를 투여한 군과 대조군 사이에 유의한 차이가 없었다. 면역억제제인 cyclosporin을 투여한 군에서 유전자 치료 효과가 cyclosporin을 투여하지 않은 정상 mouse에 비해 치료효과가 유의하게 작았다. Cyclosporin 투여에 따른 유전자의 발현기간에는 유의한 차이가 없었다. 결 론: Retrovirus 벡타를 사용한 HSVtk 유전자치료에는 면역증강이 치료효과를 증가시킬 것으로 생각된다.
Background: Concurrent chemo-radiotherapy is the recommended standard treatment modality for patients with locally advanced lung cancer. The purpose of three-dimensional conformal radiotherapy (3DCRT) is to minimize normal tissue damage while a high dose can be delivered to the tumor. The most common dose limiting side effect of thoracic RT is radiation pneumonia (RP). In this study we evaluated the relationship between dose-volume histogram parameters and radiation pneumonitis. This study targeted prediction of the possible development of RP and evaluation of the relationship between dose-volume histogram (DVH) parameters and RP in patients undergoing 3DCRT. Materials and Methods: DVHs of 41 lung cancer patients treated with 3DCRT were evaluated with respect to the development of grade ${\geq}2$ RP by excluding gross tumor volume (GTV) and planned target volume (PTV) from total (TL) and ipsilateral (IPSI) lung volume. Results: Were admitted statistically significant for p<0.05. Conclusions: The cut-off values for V5, V13, V20, V30, V45 and the mean dose of TL-GTV; and V13, V20,V30 and the mean dose of TL-PTV were statistically significant for the development of Grade ${\geq}2$ RP. No statistically significant results related to the development of Grade ${\geq}2$ RP were observed for the ipsilateral lung and the evaluation of PTV volume. A controlled and careful evaluation of the dose-volume histograms is important to assess Grade ${\geq}2$ RP development of the lung cancer patients treated with concurrent chemo-radiotherapy. In the light of the obtained data it can be said that RP development may be avoided by the proper analysis of the dose volume histograms and the application of optimal treatment plans.
꿀벌에 의해 생산되는 프로폴리스는 이미 항균, 항산화, 항염증, 항바이러스 그리고 항종양 등의 기능성으로 중요성이 증가하고 있으며, 인공적으로 생성된 화합물이 아닌 자연에서 얻어진 천연물이라는 점에서 특히 가치가 증가하고 있다. 그 중에서도 인류에게 대표적 난치병 질병 중의 하나인 암에 대한 항종양 특징은 화학적 예방 요법이라는 측면에서 중요성과 활용도가 주목받고 있다. 그러나 프로폴리스가 가지는 항종양 같은 현상적인 효과라던가 프로폴리스 구성 성분의 특징에 대해서만 연구가 이루어지고 있고, 암세포에서 나타나는 구체적인 분자 기전에 대해서는 아직까지도 연구가 미비한 상황이다. 본 연구에서는 암세포에 대해서 프로폴리스가 나타내는 구체적인 분자기전을 제시함으로써 어떤 메커니즘으로 의해 프로폴리스가 항암에 대해 효과를 나타내는지 확인하고자 하였다. 암세포에 대해서 확실한 항암 효과를 보이기 위해서는 프로폴리스의 세포 사멸 농도를 확인하고 그 안에서 이루어지는 단백질 분자의 발현 양상 또는 분자 기능의 활성화 등을 기본적으로 확인해야 한다. 수많은 단백질들이 암세포의 성장 및 분화에 관여하고 있지만 현실적으로 세포 내부의 모든 단백질의 기능을 확인하기엔 불가능하기 때문에 세포의 분열과 사멸에 관련된 기본적인 단백질과 더불어 암에 관련된 단백질의 활성화 등은 반드시 확인해야 한다. 따라서 본 연구에서 제시된 결과를 보면 프로폴리스가 암세포에 대해서 항암 효과를 보이기 위해서는 100 ㎍/mL의 농도에서 비로소 세포 사멸 현상을 나타낼 수가 있으며, 그에 따라 암세포 내부의 수많은 단백질의 변화가 나타남을 알 수 있다. 특히 주목할 점은 국내의 지역에 따라 수집된 프로폴리스의 항암 효과가 암세포 내부에서 작용할 때는 큰 차이를 보인다는 것이다. 이것은 매우 중요한 결과로서 각 지역에 식재된 밀원수에 따라 프로폴리스의 구성성분이 달라지는 것이며 그에 따라 프로폴리스의 기능성에 큰 차이를 나타낸다는 의미이다. 이는 프로폴리스가 가지는 기능에서 항암뿐만이 아니라 항염증, 항산화 그리고 항균 작용에서도 지역별로 구성된 식물상에 따라 기능성의 차이가 결정된다는 것이다. 역으로 추적해서 각 지역에서 나타난 프로폴리스의 기능성 차이를 확인하고 해당 지역의 식물상을 분석한다면 질병 또는 특정 기능성을 가진 프로폴리스를 생산할 수 있다는 의미가 될 것이다. 자궁경부암 세포에 대한 프로폴리스의 성장저해 효과는 확인했지만 아직도 확인해야 할 기전은 매우 다양하다. 세포는 내부의 신호 체계가 매우 복잡하고 단일 신호 체계가 아닌 다중 신호 체계를 가지고 있기 때문에, 프로폴리스에서 항암 효과가 나타난다고 하더라도 암세포는 다른 방향으로 anti-apoptitic signal을 생성할 수 있다. 따라서 다양한 신호 체계에 대한 암세포 내 단백질 발현에 대한 변화를 확인해야 하며, 추가로 단백질의 phosphorylation, acetylation, protein folding 그리고 mutation과 같은 기능성을 검증해야 한다. 이런 분석을 통해 프로폴리스의 구체적인 세포 신호 경로를 확인해야만 항암과 관련하여 약물을 제조할 때 손쉽게 표적 특이적인 신약 제조가 이루어질 수 있을 것이다. 특히 암은 조직별로 그 특성을 다르게 가질 수 있기 때문에 조직 특이적인 맞춤형 약제의 생산이 가능할 것이다. 따라서 본 연구에서 제시된 지역별 프로폴리스의 항암 분자 기전 분석에 더하여 해당 지역의 밀원수 분석 및 세포 내 단백질 변화추적을 기반으로 앞으로 더 많은 종류의 암세포 저해 기전을 밝힌다면 대표적 양봉 산물인 프로폴리스를 이용한 특정 암에 대한 효과적인 항암제의 개발이 가능할 것으로 보여진다.
목적 네 가지 방법으로 토끼의 뇌에 VX2 세포를 이식하여 뇌종양의 생성 유무와 위치, 체적을 분석하고자 한다. 대상과 방법 공여 토끼를 희생하여 얻은 VX2 세포 현탁액을 1) 당일 이식, 2) 냉장 보관 7일 후 이식, 3) 초저온 냉동보관 7일 후 이식, 그리고 4) 공여 토끼를 희생하지 않고 조직생검 기구를 이용하여 VX2 세포현탁액을 얻은 후 당일 이식하였다. 뇌 이식에는 정위 기구를 사용하였으며 하방 레버를 이용하여 5 mm 진입하여 VX2 세포 현탁액을 주입하였다. 10일 경과 후 MR imaging을 실시하였으며 다음 날 뇌 조직을 얻어 조직검사를 실시하여 MR 영상과 현미경 검사상 뇌종양의 생성 유무와 위치, 체적 등을 살펴보았다. 결과 VX2 세포 현탁액을 당일 이식한 토끼(n = 18)에서는 17마리(94.4%)에서 뇌종양이 생겼다(평균 체적은 106.32 mm3). 냉장 보관 후 7일에 세포 현탁액을 만들어 뇌 이식한 토끼(n = 5)에서는 1마리(20%)에서 뇌종양이 생겼으나 현미경으로만 확인되었다. 초저온 냉동 보관 후 7일에 세포 현탁액을 만들어 뇌 이식한 토끼는 5마리에서는 3마리(60%)에서 뇌종양이 생겼고 현미경으로만 확인되었다. 생검 횟수를 10회 실시한 후 이식한 3마리에서는 뇌종양이 생기지 않았고, 20회 실시한 후 이식한 2마리에서는 모두 뇌종양이 생겼으며 MR 영상과 조직검사에서 확인되었다(평균 체적 38.71 mm3). 네 가지 방법으로 발생한 종양의 위치는 superficial cortex가 대부분이었다. 결론 VX2 세포를 이식하여 토끼의 뇌종양 모델을 제작하는 기법은 쉽고 재현성은 다양했다. 이 모델은 뇌종양에 대한 영상, 치료 방법, 중재 시술 및 약물 전달 등 다양한 연구에 활용될 수 있을 것이다. 연구자의 처한 다양한 환경에 따라 다양한 방법으로 VX2 세포를 이식할 수 있을 것으로 생각된다.
The aim of this study was to ana1yze T-cell lymphoma invasion and metastasis-inducing factor 1 (Tiam1) expression in 1ung cancer patients. A total of 204 patients with lung cancer tissue lesions were enrolled in the present study, along with 40 cases of normal lung tissue and 40 of normal fetal lung tissue. Tiam1 protein expression level was determined using intensity quantitative analysis, for comparison in lung cancer, metastatic, normal lung, and fetal lung tissue. The positive unit (PU) of Tiam1 was $13.5{\pm}5.42$ in lung cancer,$5.67{\pm}1.56$ in norma1 epithelial cells, and $5.89{\pm}1.45$ in fetal lung epithelial cells. The value in the lung cancer tissue was significantly higher than that in the normal lung tissue and the fetal lung tissue (P<0.01). The Tiam1 PU values with lymph node metastasis and without 1ymph node metastasis were $15.2{\pm}4.34$ and $12.5{\pm}4.23$, respectively, and the difference was statistically significant (P<0.05). The Tiam1 PU values in different tumor, nodes, metastasis (TNM) stages, III-IV period, and I-II phase were $14.7{\pm}4.14$ and $11.0{\pm}5.34$ (P<0.05). A correlation was found between Tiam1 expression and the age of patient, tumor size, tumor type, and tumor differentiation. Tiam1 protein expression in the lung tumor tissue is significantly higher than that in the normal lung tissue and fetal lung tissue. Tiam1 expression may be closely related to lung cancer development and metastasis.

In the present study, we expected the anti-tumor effect by combined treatment of arsenic trioxide and interferon (IFN)-${\alpha}$ on murine Lewis lung carcinoma (LL2) cells through in vivo study. As a experimental model, LL2 cells ($1{\times}10^{6}$/mouse) were injected subcutaneously into the back region of mice. When the tumor volume reached $100mm^3$, mice were treated with 1 mg/kg arsenic trioxide, 50000 IU IFN-${\alpha}$, or arsenic trioxide and IFN-${\alpha}$. The development of tumor cells was significantly inhibited by combined treatment with arsenic trioxide and IFN-${\alpha}$. In arsenic trioxide and IFN-${\alpha}$ treated group, apoptotic index was reached a peak valve at 48 hr after the treatment and it was restored to approximately the control level at 8 days. Also, positive signals of Bax and Bad were increased at 48 to 96 hr and decreased at 8 day. Whereas, positive cells of Bcl-2 were steadily decreased at 12 to 48 hr and restored to the background level at 8 days. Our data showed that immunoreactivity of Bcl-2 was decreased at 12 to 48 hr, while positive signals of Bax and Bad were increased in accordance with apoptotic index at these times. In conclusion, our results suggest that the combined treatment with arsenic trioxide and IFN-${\alpha}$ significantly inhibited the growth of LL2 tumor cells and induced apoptosis through the up and down-regulation of Bcl-2 gene family.

$Foxp3^+$$CD25^+CD4^+$ regulatory T (Treg) cells are crucial for the maintenance of immunological self-tolerance and are abundant in tumors. Most of these cells are chemo-attracted to tumor tissues and suppress anti-tumor responses inside the tumor. Currently, several cancer immunotherapies targeting Treg cells are being clinically tested. Cisplatin is one of the most potent chemotherapy drugs widely used for cancer treatment. While cisplatin is a powerful drug for the treatment of multiple cancers, there are obstacles that limit its use, such as renal dysfunction and the development of cisplatin-resistant cancer cells after its use. To minimize these barriers, combinatorial therapies of cisplatin with other drugs have been developed and have proven to be more effective to treat cancer. In the present study, we evaluated the effect of the combination therapy using methyl gallate with cisplatin in EL4 murine lymphoma bearing C57BL/6 mice. The combinatorial therapy of methyl gallate and cisplatin showed stronger anti-cancer effects than methyl gallate or cisplatin as single treatments. In Treg cell-depleted mice, however, the effect of methyl gallate vanished. It was found that methyl gallate treatment inhibited Treg cell migration into the tumor regardless of cisplatin treatment. Additionally, in both the normal and cisplatin-treated tumor-bearing mice, there was no renal toxicity attributed to methyl gallate treatment. These findings suggest that methyl gallate treatment could be useful as an adjuvant method accompanied with cisplatin therapy.
Background and Aims: Advances in the treatment of cervical cancer over the last decade have predominantly involved the development of genes directed at molecular targets. Gene therapy is recognized to be a novel method for the treatment of cervical cancer. Genes can be administered into target cells via nanocarriers. This study aimed to develop systemically administrable nano-vectors. Floate (Fa) containing gene loaded nanoparticles (NPs) could target HeLa human cervical cancer cells through combination with receptors on the cells to increase the nuclear uptake of genetic materials. Methods: Fa was linked onto Poly (ethylene glycol)-b-poly (D, L-lactide) (PEG-PLA) to form Fa-PEG-PLA, and the resulting material was used to load plasmids of enhanced green fluorescence protein (pEGFP) to obtain gene loaded nanoparticles (Fa-NPs/DNA). Physical-chemical characteristics, in vitro release and cytotoxicity of Fa-NPs/DNA were evaluated. The in vitro transfection efficiency of Fa-NPs/DNA was evaluated in HeLa cells and human umbilical vein endothelial cells (HUVEC). PEG-PLA without Fa was used to load pEGFP from NPs/DNA as a control. Results: Fa-NPs/DNA has a particle size of 183 nm and a gene loading quantity of 92%. After 72h of transfection, Fa-NPs/DNA displayed over 20% higher transfection efficiency than NPs/DNA and 40% higher than naked DNA in HeLa cells. However, in HUVECs, no significant difference appeared between Fa-NPs/DNA and NPs/DNA. Conclusions: Fa-PEG-PLA NPs could function as excellent materials for gene loading. This nano-approach could be used as tumor cell targeted medicine for the treatment of cervical cancer.

서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
