Cyclin-dependent Kinase저해 단백질 p16^{INK4A}의 인체 암세포에서의 세포사멸 유도 활성 (A Cyclin-Dependent Kinase Inhibitor, p16^{INK4A}, Induces Apoptosis in The Human Cancer Cells.)

  • 본 연구진은 토양미생물의 배양액으로부터 cyclin-dependent kinase 저해활성의 Toyocamycin을 분리하였으며 〔16〕, 화학적 전합성을 통하여 활성이 개선된 유도체인 신물질 MCS-5A를 합성하였다〔3〕. 이 MCS-5A를 이용한 항암 기전규명을 위한 연구를 통하여 , human promyelocytic leukemia cell(HL-60)에서 MCS-5A에 의해 cyclin-dependent kinase inhibitor p16$^{INK4A}$ 단백질의 발현증가가 암세포의 세포주기 억제와 동시에 HL-60 cell희 세포사멸을 유도하는 것을 확인하였다(data not shown). 그러나 HL-60 cell의 경우와는 달리 non small cell lung cancer cell(NSCLC)인 A549 cell(p16$^{INK4A}$ 결핍 세포주)에 MCS-5A를 처리할 경우에는 전혀 세포사멸이 유도되지 않았다. 따라서 MCS-5A에 의한 HL-60 cell에서의 세포사멸 유도는 발암억제 유전자인 P16$^{INK4A}$의 세포 내 발현 및 존재 여부에 의해 좌우되는 것으로 판단되었다. 이러한 배경에서 본 연구는 p16$^{INK4A}$.의 기존에 알려진 세포주기 억제를 유발하는 cyclin-dependent kinase inhibitor(CKI)로서의 역할 뿐 아니라, p16$^{INK4A}$ 유전자가 세포사멸을 유도할 수 있다는 새로운 기능을 규명하기 위하여 다음의 연구를 시도하였다. 즉 $p^{INK4A}$ 결핍 세포주인 A549(-p16/+p53)와 H1299(-pl6/-p53) 그리고 p16$^{INK4A}$ 함유 세포주인 HeLa(+p16/+p53)세포에 외부로부터 p16$^{INK4A}$ 유전자를 도입시켜, 각 세포주에서의 세포사멸 유도 여부를 비교하고자 하였다. 우선 wild-type p16$^{INK4A}$ 유전자를 가진 HeLa cell에서 총 RNA를 추출하여, 역전사 반응으로 cDNA를 만들고, PCR을 통해 p16$^{INK4A}$ 유전자를 증폭하였다. pcDNA3.1/His is A vector에 p16$^{INK4A}$ 유전자를 끼워 넣고 competent cell (XL1-Blue)에 형질 전환하여 cloning한 후, p16$^{INK4A}$ clone을 다량으로 추출하였다. 위에 언급한 각각의 cell line에 p16$^{INK4A}$유전자를 농도(0, 1, 5, 10$\mu\textrm{g}$)별로 transfection 시킨 후, p16 단백질을 일정 시간 동안(12시간) 발현시킨 뒤, TUNEL등의 분석을 통해 세포사멸이 유도되는지를 확인하였으며, 또한 Western blot 분석을 통하여 p16단백질과 세포사멸 유도 인자인 caspase 3의 발+현 양상을 확인하였다. 연구 결과, Western blot을 통해 transfection시킨 p16/INK4A/유전자의 농도에 따라 각각의 cell line에서 Pro-caspase 3의 감소함을 관찰할 수 있었고, TUNEL분석을 통해 A549및 HeLa cell에서 세포사멸이 유도됨을 확인할 수 있었다 특히 A549(-p16/+p53)와 HeLa cell(+p16/+p53)에서는 TUNEL 분석 및 Western blot을 통한 pro-caspase 3의 caspase 3로의 전환 등을 통해 세포사멸이 발생하였음을 확연하게 확인할 수 있었으나, 반면 H1299(-pl6/-p53) cell에서는 단지 Western blot을 통한 pro-caspase 3의 활성화만을 통해 간접적으로 세포사멸을 확인 할 수 있었다. 또한 p53이 결핍된 H1299(-pl6/-p53)세포주에서의 $^{INK4A}$ 에 의한 세포사멸 유도는 p53 비의존적으로 작용한다는 사실을 확인할 수 있었다. 결론적으로 발암억제 유전자인 $^{INK4A}$ 는 CKI로서의 기능뿐 아니라, 세포사별 유도와도 밀접하게 관련되어 있으며, 이 기능은 발암 억제 유전자인 p53과는 독립적으로 작용한다는 사실을 확인하였다. 세포사멸 유도 기전연구에서 $p16^{INK4A}$ 가 세포사멸을 유도하는 기전에 대해서는 아직 명확하게 밝혀진 바는 없으며, 현재 본 연구실에서 다양한 실험을 통해 연구가 진행 중이다.

Obtainment and Characterization of Brain Tumor Cell Using Vasopressin-SV40 T Ag Transgenic Mouse

  • In previous reports, pVPSV.IGR2.1 transgenic mouse were described that brain tumor and lymphoma by reason of Vasopressin-SV40 T antigen. In this study, we produced pVPSV.IGR3.6 transgenic mouse that used pVPSV.IGR3.6 vector. Expression of transgene was vary different in transgenic mouse. We obtained 6 transgenic mouse line, moreover they had died at the age of 2-6 weeks without transmitting the transgene to their offspring, and had tumorigenesis on same location with pVPSV.IGR2.1 transgenic mouse. Only a founder mouse was investigated for expression of fusion gene. Here we extended this transgenic approach to the study of tumor progression. From the mouse, we confirmed brain tumor cell, after then cultured for investigate characterization. In this report, we demonstrate that reduction of survival rate in transgenic mouse fused vasopressin gene length, acquisition of brain tumor cell, composition with astrocyte cells and neuronal cells. Finally, cells had no change with increase of passage.

BAG5 regulates PTEN stability in MCF-7 cell line

  • The phosphatase and tensin homolog deleted on chromosome 10 (PTEN) is a tumor-suppressing lipid phosphatase that is frequently absent in breast tumors. Thus, the stability of PTEN is essential for tumor prevention and therapy. The ubiquitin-proteasome pathway has an important role in regulating the functions of PTEN. Specifically, carboxyl terminus Hsp70-interacting protein (CHIP), the E3 ubiquitin ligase of PTEN, can regulate PTEN levels. In this study, we report that BCL-2-associated athanogene 5 (BAG5), a known inhibitor of CHIP activity, reduces the degradation of PTEN and maintains its levels via an ubiquitylation-dependent pathway. BAG5 is identified as an antagonist of cell tumorigenicity.

Radiosensitizing and Topoisomerase I Inhibitory Effects of Aloe vera, Formitella fraxinea, and Ulmus davidiana Extracts

  • Ulmus davidiana, Formitella fraxinea, and Aloe vera extracts were detected to have inhibitory effects against topoisomerase I at treatment of $5{\mu}g$. Ulmus davidiana and Aloe vera extracts were found to show inhibitory effect similar to camptothecin, Formitella fraxinea extract was found to have weak activity. We also found the potential use of those extracts as a radiation sensitizer. Radiosensitizing effect at combination treatment was increased more than 2 times at single treatment of radiation, Ulmus davidiana or Formitella fraxinea extracts. Ulmus davidiana and Formitella fraxinea extracts were found to have significant radiosensitizing effect on test tumor cell line. But, Aloe vera extract was not detected to have activity as a radiosensitizer. Ulmus davidiana and Formitella fraxinea extracts are potent radiosensitizers on tumor cell lines and should be considered for further study of active compounds.

Sulforaphane Inhibits the Proliferation of the BIU87 Bladder Cancer Cell Line via IGFBP-3 Elevation

  • Aim: To investigate effects of sulforaphane on the BIU87 cell line and underlying mechanisms involving IGFBP-3. Methods: Both BIU87 and IGFBP-3-silenced BIU87 cells were treated with sulforaphane. Cell proliferation was detected by MTT assay. Cell cycle and apoptosis were determined via flow cytometry. Quantitative polymerase chain reaction and Western blotting were applied to analyze the expression of IGFBP-3 and NF-${\kappa}B$ at both mRNA and protein levels. Results: Sulforaphane (80 ${\mu}M$) treatment could inhibit cell proliferation, inducing apoptosis and cell cycle arrest at G2/M phase. All these effects could be antagonized by IGFBP-3 silencing. Furthermore, sulforaphane (80 ${\mu}M$) could down-regulate NF-${\kappa}B$ expression while elevating that of IGFBP-3. Conclusions: Sulforaphane could suppress the proliferation of BIU87 cells via enhancing IGFBP-3 expression, which negatively regulating the NF-${\kappa}B$ signaling pathway.

Biphasic Activity of Chloroquine in Human Colorectal Cancer Cells

  • Autophagy is a homeostatic degradation process that is involved in tumor development and normal development. Autophagy is induced in cancer cells in response to chemotherapeutic agents, and inhibition of autophagy results in enhanced cancer cell death or survival. Chloroquine (CQ), an anti-malarial drug, is a lysosomotropic agent and is currently used as a potential anticancer agent as well as an autophagy inhibitor. Here, we evaluate the characteristics of these dual activities of CQ using human colorectal cancer cell line HCT15. The results show that CQ inhibited cell viability in dose- and time-dependent manner in the range between 20 to 80 uM, while CQ did not show any antiproliferative activity at 5 and 10 uM. Cotreatment of CQ with antitumor agent NVP-BEZ235, a dual inhibitor of PI3K/mTOR, rescued the cell viability at low concentrations meaning that CQ acted as an autophagy inhibitor, but CQ induced the lethal effect at high concentrations. Acridine orange staining revealed that CQ at high doses induced lysosomal membrane permeabilization (LMP). High doses of CQ produced cellular reactive oxygen species (ROS) and cotreatment of antioxidants, such as NAC and trolox, with high doses of CQ rescued the cell viability. These results suggest that CQ may exert its dual activities, as autophagy inhibitor or LMP inducer, in concentration-dependent manner.

  • Evaluation of potentials of chemicals to alter expression of genes that are involved in carcinogenesis may serve useful tools in toxicological research. In this investigation, we developed reporter cell lines that expressed luciferase in response to transactivation of hypoxia inducible factor-1, P53 tumor suppressor and Nur77 of which roles have been well established in cancer development and progression. Whereas these reporter cell lines displayed low constitutive backgrounds, the reporter activities were significantly enhanced in response to $desferriosamine/CoCl_2$, adriamycin or 6-mercaptopurine, which are hypoxia mimicking chemicals, P53 activator or Nur77 inducer, respectively. The activation of the reporter was time- and dose-dependent. Known tumor initiators and promoters, such as phorbol 12-myristate 13-acetate and phorbol 12, 13-dicaprinate induced the reporter activity at as low as 10nM in these stable cell lines. Further, known anti-tumor promoters, such as ascorbic acid and ${\beta}-carotene$ repressed the reporter activities. These results indicate that our stable reporter cell lines could serve as a useful system for rapid assessment of carcinogenicity of toxic chemicals.

Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitor Versus Placebo as Maintenance Therapy for Advanced Non-small-cell Lung Cancer: A Meta-analysis of Randomized Controlled Trials

  • Background: Use of epidermal growth factor receptor inhibitors (EGFR-TKIs ) is now standard for non-small-cell lung cancer (NSCLC). However, the effects of EGFR-TKIs in maintenance therapy for advanced NSCLC patients are still unclear. The preent meta-analysis was performed to examine pooled data of randomized control trials (RCT) where EGFR-TKIs were compared against placebo in maintenance regimens for patients with advanced NCSLC to quantify potential benefits and determine safety. Methods: Several data bases were searched, including PubMed, EMBASE and CENTRAL, and we performed an internet search of conference literature. The endpoints were objective response rates (ORR), progression-free survival (PFS) and overall survival (OS). We performed a meta-analysis of the published data, using Comprehensive Meta Analysis software (Version 2.0). with a fixed effects model and an additional random effects model, when applicable. The results of the meta-analysis are expressed as hazard ratios (HRs) or risk ratios (RRs), with their corresponding 95% confidence intervals (95%CIs). Results: The final analysis included six trials, covering 3,758 patients. Compared with placebo, EGFR-TKIs maintenance therapy improved ORR and PFS for patients with advanced NSCLC, the difference being statistically significant (P<0.05), but proved unable to prolong patients' OS. The main adverse reactions were diarrhea and rashes. Conclusion: EGFR-TKIs demonstrated encouraging efficacy, safety and survival when delivered as maintenance therapy for patients with advanced NSCLC after first-line chemotherapy, especially for the patients who had adenocarcinomas, were female, non-smokers and patients with EGFR gene mutations.

LINE-1 and Alu Methylation Patterns in Lymph Node Metastases of Head and Neck Cancers

  • Background: The potential use of hypomethylation of Long INterspersed Element 1 (LINE-1) and Alu elements (Alu) as a biomarker has been comprehensively assessed in several cancers, including head and neck squamous cell carcinoma (HNSCC). Failure to detect occult metastatic head and neck tumors on radical neck lymph node dissection can affect the therapeutic measures taken. Objective: The aim of this study was to investigate the LINE-1 and Alu methylation status and determine whether it can be applied for detection of occult metastatic tumors in HNSCC cases. Methods: We used the Combine Bisulfite Restriction Analysis (COBRA) technique to analyse LINE-1 and Alu methylation status. In addition to the methylation level, LINE-1 and Alu loci were classified based on the methylation statuses of two CpG dinucleotides in each allele as follows: hypermethylation ($^mC^mC$), hypomethylation ($^uC^uC$), and 2 forms of partial methylation ($^mC^uC$ and $^uC^mC$). Sixty-one lymph nodes were divided into 3 groups: 1) non-metastatic head and neck cancer (NM), 2) histologically negative for tumor cells of cases with metastatic head and neck cancer (LN), and 3) histologically positive for tumor cells (LP). Results: Alu methylation change was not significant. However, LINE-1 methylation of both LN and LP was altered, as demonstrated by the lower LINE-1 methylation levels (p<0.001), higher percentage of $^mC^uC$ (p<0.01), lower percentage of $^uC^mC$ (p<0.001) and higher percentage of $^uC^uC$ (p<0.001). Using receiver operating characteristic (ROC) curve analysis, $%^uC^mC$ and $%^mC^uC$ values revealed a high level of AUC at 0.806 and 0.716, respectively, in distinguishing LN from NM. Conclusion: The LINE-1 methylation changes in LN have the same pattern as that in LP. This epigenomic change may be due to the presence of occult metastatic tumor in LN cases.
