돼지감자를 이용한 고농도 알코올발효 균주의 탐색 (Screenig and Indentification of Wild Strains for the Production of High Concentration of Alcohol from Jerusalem artichoke Tubers)

  • Yeast screening for effective production of alcohol from Jerusalem artichoke tubers as an alternative energy source was performed. Inulin assimilative strains with high alcohol tolera- nce were isolated from wild sources and cultured in the liquid media of Jerusalem artichoke powder varying its concentraion from 15 to 30%. As a result, four strains of 2,445 isolates showing the inulin assimilation were selected as alcohol fermentative and alcohol tolerant yeasts. These strains were assignated to be Kluyveromyces marxianus F043 and Kluyveromyces sp. F173, E040, and F334, respectively, by their cultural and physiological characteristics. The F043 strain produced ethanol of 98.1 g/l in the 25% Jerusalem artichoke medium for 3 days.

광선의 영향에 따른 감자의 Chlorophyll과 Glycoalkaloid의 함량 변화 (The Changes of Chlorophyll and Glycoalkaloid Contents in Potato Tubers after Exposure of Fluorescent and UV Light)

  • To determine changes of chlorophyll and potato glycoalkaloid(PGA) contents in potato tubers after lightening, Dejima potatoes cultivated in Korea were used and PGA was analyzed by HPLC. Potatoes were stored under fluorescent and UV light exposure or darkness for 7 days at $5^{\circ}C$. The contents of chlorophyll and glycoalkaloid in the peel of potatoes exposed to fluorescent light were increased to $84\%,\;69.5\%$ respectively compared to potatoes stored without lightening. While a marked effect of lightening on the peel of potatoes exposed to fluorescent light, UV light lead to no change of chlorophyll and less increase in glycoalkaloid than fluorescent did. The increase of chlorophyll was provoked by only fluorescent light. Cortex part did not accumulate any chlorophyll or glycoalkaloid.

Expression Patterns of CaMV 35S Promoter-GUS in Transgenic Poatoes and Their Clonal Progenies

  • Two potato (Solanum tuberosum L.) cultivars were transformed by Agrobacterium tumefaciens harboring cauliflower mosaic virus (CaMV) 35S promoter and $\beta$-glucuronidase (GUS) gene. Expression patterns of the CaMV 35S promoter according to tissue types and developmental stages, and genetic stability of GUS gene were investigated in the clonal progenies of transgenic potatoes. Kanamycin-resistant shoot emerged from tuber disc after 4 weeks of culture, and root was induced 6 weeks after culture on the selection medium. Shooting frequency of cvs. Superior and Dejima were 43% and 27%, respectively. Mature transformants and their clonal progenies showed no phenotypical abnormality. GUS activity was expressed primarily at parenchymatous cells of phloem tissue around the vascular cambium in the stem and root, and higher activity was found at the apical meristem of shoot, root and adventious shoot bud. GUS activity was higher at tubers of young explants than at stored tubers. These facts indicate that expression level of the CaMV 35S promoter differed according to tissue types and developmental stages of the organs. The GUS gene was stably inherited to each clonal progeny and normally expressed.

  • The bioassay-guided fractionation of the methylene chloride soluble portion of a methanol extract of Gastrodia elata tubers led to the isolation of a new furfural, 5-(4-hydroxy-benzyloxymethyl)-furan-2-carbaldehyde (2), together with four known compounds (1, 3-5), which exhibited potent inhibitory activity at the concentration of 25 $\mu\textrm{g}$/ml on melanin biosynthesis in cultured B-16 mouse melanoma cells.

Fusarium oxysporum Causes Root Rot on Gastrodia elata in Korea: Morphological, Phylogenetic, and Pathogenicity Analyses

  • Gastrodia elata infected with root rot disease was collected from cultivated G. elata fields in Gimcheon, Korea, in 2018. G. elata tuber surfaces exhibited root rot disease symptoms of dark-grey lesions and white fungal mycelial growth. The fungus was isolated from symptomatic tubers and cultured. Based on morphological characteristics and molecular analysis of the internal transcribed spacer region of ribosomal DNA and translation elongation factor 1-alpha, the isolated fungus was Fusarium oxysporum. This is the first report of root rot caused by F. oxysporum on G. elata tubers in Korea.

Changes of Endogenous Gibberellins in Tubers of Chinese Yam(Dioscorea opposita) during Storage Period

  • For storage periods of tubers in Chinese yam, the levels of $\textrm{GA}_{44}$ and $\textrm{GA}_{20}$ was constant, meanwhile both $\textrm{GA}_{53}$ and $\textrm{GA}_{19}$ level were always higher than that of $\textrm{GA}_{44}$ and $\textrm{GA}_{20}$$\textrm{GA}_9$ content as precursor of $\textrm{GA}_4$ was not changed during storage. $\textrm{GA}_{24}$ content was low to below 0.2 ng for 90 days after storage, $\textrm{GA}_{36}$ content as precursor of $\textrm{GA}_4$ like $\textrm{GA}_9$ was about 6-8 fold higher than that of $\textrm{GA}_9$ during storage. GA contents of the two gibberellin biosynthetic pathways were gradually increased when storage periods were progressed. Bioactive GA$_1$ content as the GA members of an early C-13 hydroxylation was always constant, and its content was very low as below 0.1ng per dry weight, meanwhile, bioactive $\textrm{GA}_4$ content as the GA members of non C-13 hydroxylation was drastically increased, also, its content was highest at 90 days after storage, and then decreased at 120 days after storage. Consequently, we suggest that $\textrm{GA}_4$ may be involved in controlling tuber sprouting in Chinese yam.

Mass Propagation of Plug Seedling using Stem Cutting and Their Tuber Yield in Potato

  • For the mass production of plug seedlings in cultivar ‘Dejima’ potato (Solanum tuberosum L.) the optimal apical cutting diameter for rooting and rapid multiplication of stem cuttings in hydroponics were determined. In addition, the best planting date was predicted to increase tuber yield of plug seedlings at fall cropping in Cheju-Do, Korea. Days to initial rooting decreased as the cutting diameter was reduced. Plant height, leaf number, root length and root weight per plant were favorable as the cutting diameter was small. The ideal cutting diameter was 1-2 mm in this experiment. In the hydroponic cultures, the Japanese standard (JS) nutrient solution was the most effective for multiplication of stem cuttings. It was able to propagate more than 20 times a month from a single mother plant. Viability of plants, which were derived from plug seedlings using stem cuttings, was excellent when transplanted to the field. The number of tubers and tuber yield in both of the plug seedlings and seed potato planting plots were high when planted on 25 August. The number and yield were reduced when planted on 15 August, 5 September and 15 September. The degree of decrease of tuber yield in the plug seedling planting plot however, was lower than that of seed potatoes when the planting date was late. In the case of small tubers (under 30 g), the number of tubers and tuber yield were evidently increased in the seed potato tuber planting plot; the yield of large tuber (over 80g) in the plug seedling planting plot was higher than that of the seed potato. The total tuber yield per plant in the plug seedling planting plot was less than that of the seed potato; therefore, in order to increase tuber yield it was necessary to increase field plant density.

한국산 마의 지질 성분의 분리 및 분석 (Fractionation and Quantitative Analysis of Lipid Components in Korean Yam (Dioscorea) Tubers)

  • 한국산 마의 주요 품종인 Dioscorea batatas, D. aimadoimo and D. japonica 3가지 품종의 시료들을 동결건조하여 분말로 만든 다음 지질의 추출, silicic acid column chromatography에 의한 지질의 분획, thin layer chromatography에 의한 지질의 분리 및 gas chromatography에 의한 지방산 조성 등을 행하였다. 총지질의 함량은 D. japonica가 다른 마에 비해 적은 편이었으며, 지질을 분획한 결과 3가지 품종 모두 중성지질이 가장 많았고 그 다음 당지질, 인지질 순으로 나타났다. 분획한 각 지방의 구성 지질로는 중성지질이 sterolester, triglyceride, 1,3-diglyceride, 1,2-diglyceride 및 monoglycerid였고, 당지질에서 acylsterylglycoside, sterylglycoside, digalactosyldiglyceride 및 sulfolipid였고, 인지질에서 phosphatidylethanolamine, phosphatidylcholine 및 phosphatidylinositol 이었다. GC에 의한 총지질의 지방산 조성은 팔미트산과 리놀레산이 가장 많은 부분을 차지하였고 불포화지방산 함량이 포화지방산 함량보다 많았다. 또한 중성지질, 당지질 및 인지질의 지방산 조성도 총 지질을 구성하는 지방산 조성과 같은 양상을 보여 주었다.
