Inflammatory Polyps in 2 Cats (두 마리 고양이에서의 염증성 용종 예)

  • Inflammatory polyps in feline ear are nonneoplastic, inflammatory growths that arise from the middle ear or the eustachian tube and extended into the pharynx or external ear canal. Two 2-year-old female Russian blue cats showed 2-3 weeks history of aural discharge, crust formation in external ear, and head or ear shaking. Two masses were surgically excised from ear canal, and submitted for diagnosis. Histopathologically, these masses were covered with hyperplastic ciliated epithelium or nonkeratinizing squamous epithelium with partial erosion and ulceration. The core of masses was consisted of proliferated connective tissue and massive infiltration of mononuclear cells. Immunohistochemically, about 90% of infiltrated mononuclear cells demonstrated CD3 positive T cell. According to both polymerase chain reaction (PCR) and reverse transcriptase-PCR, tissues samples were negative for feline viral pathogens. Based on the clinical, gross, histopathologic findings, these two cases were diagnosed as inflammatory polyps originated from the middle ear in cats.

The Cross Ability and the Phenotypic Characteristics of F1 Hybrid in the Interspecific Crosses between Brassica napus and B. campestris, B. rapa (Brassica 속 식물 내 종간교잡에 따른 교잡효율 및 F1잡종의 표현형질)

  • Interspecific crosses between rapeseed (Brassica napus L.) and chinese cabbage (B. campestris var. pekinensis Makino) and turnip (B. rapa L.) were made in order to examine the cross possibility and morphological phenotype of $F_1$ hybrid. The growth of pollen tube of cross between rapeseed and chinese cabbage was more rapid than cross between rapeseed and turnip. Silique formation and seed setting in silique were relatively high in the cross between rapeseed and chinese cabbage. The percentage of silique set from interspecific cross between rapeseed and chinese cabbage was 90.6% and higher 23.3% than the percentage of silique from interspecific cross rapeseed and turnip. The average number of seed per silique obtained from the cross rapeseed and chinese cabbage, and rapeseed and turnip reached 15.5 and 11.6, respectively. The morphological phenotypes of $F_1$ hybrid plants obtained from seeds in the cross between rapeseed and chinese cabbage resembled rapeseed mainly, but leaf length and leaf width were increased. The size, shape and lobation of leaves of $F_1$ hybrid plants from interspecific cross between rapeseed and turnip were intermediated between their parent species, but color of leaves was dark-green.

Concerning the Formation of the Acquired Cholesteatoma (상고실 진주종의 형성에 관하여)

  • Concerning the pathogenesis of acquired cholesteatoma in attic, there has been postulated theories by immigration from the Shrapnell's portion of the tympanic membrane, posterosuperior quardrant of the deep meatal skin and invagination of the margin of the central perforation. Otherwise, squamous metaplasia of the epithelium lining the middle ear cleft has been supported as a possible cause of cholesteatoma. Clinically, there has been known of the facts that cholesteatoma is formed in the attic but the pathogenesis concerning the acquired cholesteatoma is not still exactly reported. Recently, authors analyzed 170 cases of cholesteatomatous middle ear performed the operation to the middle ear cleft. On the operation finding, when the primary focus of the cholesteatoma was in the attic, we observed two types of perforation, marginal and central perforation in the Shrapnell's portion, and retraction to the Prussak's space, bony defect on the Rivinus notch. Among 36 cases of the cholesteatoma, the perforation of the Shrapnell's portion are 5 cases. Bony defect on the Rivinus notch and marginal perforation on the posterosuperior quadrant of the Shrapnell's portion are 21 cases. Among these cases, 3 cases are combined with central perforation of the Shrapnell's portion. Conclusively, the reasons that cholesteatoma is favorable site in the attic: 1) Excretion of the inflammatory discharge in the attic is difficult because of the distance of the E-tube. 2) The Shrapnell's portion has less collagen fiber than the pars tensa and it is thin because the elastic fibers are rich in it. It is easy to retract within the Prussak's space to the cases of keratinizing hyperplasia. 3) The epidermis attached at the Rivinus notch of the superior portion on the Shrapnell's portion is invaginated through the destructed bony wall of the Rivinus notch and the margin of the tympanic membrane in the response to the keratinizing hyperplasia.

The Comparison of Volatile Organic Compounds (VOCs) Analysis and Tradescantia Micronucleus (Trad-MCN) Bioassay for Evaluation of Hazardous Materials in Chemical Workplace Field (화학공장 실내 작업장에서의 유해물질 평가를 위한 VOC 분석법과 자주달개비 미세핵 분석법의 비교)

  • This research examined the presence of hazardous materials in chemical workplace field using an integrated chemical/biological monitoring. Chemical workplace field air for volatile organic compounds (VOCs) analysis was collected using a collection tube packed with Tena.x TA adsorbent 400 mg. Workplace field air samples were analyzed by gas chromatography/mass spectrometry (GC/MS). Simultaneously, Tradescantia BNL 4430 clone was exposed in situ to monitor hazardous materials in chemical workplace field. GC/MS analysis showed the presence of various VOCs such as trichloroethylene, toluene, ethylbenzene, (m,p,o)-xylenes, styrene, 1,3,5-trimethylbenzene, and 1,2,4-trimethylbenzene. The results showed that in situ monitoring of VOCs with the Tradescantia-micronucleus (Trad-MCN) assay gave positive results in chemical workplace field and negative response at outdoor air. In conclusion, inhalation of these field air by workers may affect chronic demage to their health by inducing micronuclei formation in Tradescantia pollen mother cells. The combination of chemical/biological monitoring is very effective to evaluate hazardous materials in workplace field and can be alternatively used for screening hazardous materials.

A Study on the Comparison of Brazed Joint of Zircaloy-4 with PVD-Be and Zr-Be Amorphous alloys as Filler Metals (PVD-Be와 비정질 Zr-Be 합금을 용가재로 사용한 Zircaloy-4의 브레이징 접합부의 비교 연구)

  • Brazing is an important manufacturing process in the fabrication of Heavy Water Reactor fuel rods, in which bearing and spacer pads are joined to Zircaloy-4 cladding tubes. The physical vapor deposition(PVD) technique is currently used to deposit metallic Be on the surfaces of pads as a filler metal. Amorphous Zr-Be binary alloys which are manufactured by rapid solidification process are under developing to substitute the conventional PVD-Be coating. In the present study, brazed joint with PVD and amorphous alloys of $Zr_{1-x}Be_{x}(0.3{\le}x{\le}0.5)$ as filler metals are compared by mechanism, microstructure and hardness. The thickness of brazed joint with amorphous alloys became much smaller than that of PVD-Be. The erosion of base metal did not occur in the brazed joint with amorphous alloys. The brazing mechanism for PVD-Be seems to be Be diffusion into Zr-4 with capillary action resulting from eutectic reaction while that for amorphous alloys are associated with the liquid phase formation in the brazed joint. The brazed joint microstructure with PVD-Be consists of dendrite while that with amorphous alloys is globular. The $Zr_{0.7}Be_{0.3}$ alloy shows the smooth interface with little erosion in the base metal and is recommended a most suitable brazing filler metal for Zircaloy-4.

Strain-Relaxed SiGe Layer on Si Formed by PIII&D Technology

  • Strain-relaxed SiGe layer on Si substrate has numerous potential applications for electronic and opto- electronic devices. SiGe layer must have a high degree of strain relaxation and a low dislocation density. Conventionally, strain-relaxed SiGe on Si has been manufactured using compositionally graded buffers, in which very thick SiGe buffers of several micrometers are grown on a Si substrate with Ge composition increasing from the Si substrate to the surface. In this study, a new plasma process, i.e., the combination of PIII&D and HiPIMS, was adopted to implant Ge ions into Si wafer for direct formation of SiGe layer on Si substrate. Due to the high peak power density applied the Ge sputtering target during HiPIMS operation, a large fraction of sputtered Ge atoms is ionized. If the negative high voltage pulse applied to the sample stage in PIII&D system is synchronized with the pulsed Ge plasma, the ion implantation of Ge ions can be successfully accomplished. The PIII&D system for Ge ion implantation on Si (100) substrate was equipped with 3'-magnetron sputtering guns with Ge and Si target, which were operated with a HiPIMS pulsed-DC power supply. The sample stage with Si substrate was pulse-biased using a separate hard-tube pulser. During the implantation operation, HiPIMS pulse and substrate's negative bias pulse were synchronized at the same frequency of 50 Hz. The pulse voltage applied to the Ge sputtering target was -1200 V and the pulse width was 80 usec. While operating the Ge sputtering gun in HiPIMS mode, a pulse bias of -50 kV was applied to the Si substrate. The pulse width was 50 usec with a 30 usec delay time with respect to the HiPIMS pulse. Ge ion implantation process was performed for 30 min. to achieve approximately 20 % of Ge concentration in Si substrate. Right after Ge ion implantation, ~50 nm thick Si capping layer was deposited to prevent oxidation during subsequent RTA process at $1000^{\circ}C$ in N2 environment. The Ge-implanted Si samples were analyzed using Auger electron spectroscopy, High-resolution X-ray diffractometer, Raman spectroscopy, and Transmission electron microscopy to investigate the depth distribution, the degree of strain relaxation, and the crystalline structure, respectively. The analysis results showed that a strain-relaxed SiGe layer of ~100 nm thickness could be effectively formed on Si substrate by direct Ge ion implantation using the newly-developed PIII&D process for non-gaseous elements.

Paired Ig-Like Type 2 Receptor-Derived Agonist Ligands Ameliorate Inflammatory Reactions by Downregulating β1 Integrin Activity

  • The paired immunoglobulin-like type 2 receptor (PILR) family consists of two functionally opposite members, inhibitory $PILR{\alpha}$ and activating $PILR{\beta}$ receptors. PILRs are widely expressed in various immune cells and interact with their ligands, especially CD99 expressed on activated T cells, to participate in immune responses. Here we investigated whether PILR-derived agonists inhibit ${\beta}1$ integrin activity as ligands for CD99. PILR-derived peptides as well as PILR-Fc fusion proteins prevented cell adhesion to fibronectin through the regulation of ${\beta}1$ integrin activity. Especially, PILRpep3, a representative 3-mer peptide covering the conserved motifs of the PILR extracellular domain, prevented the clustering and activation of ${\beta}1$ integrin by dephosphorylating FAK and vinculin, which are major components of focal adhesion. In addition, PILRpep3 inhibited transendothelial migration of monocytes as well as endothelial cell tube formation. Furthermore, upon intraperitoneal injection of PILRpep3 into mice with collagen-induced arthritis, the inflammatory response of rheumatoid arthritis was strongly suppressed. Taken together, these results suggest that PILR-derived agonist ligands may prevent the inflammatory reactions of rheumatoid arthritis by activating CD99.

Angiogenic effects of wood-cultivated ginseng extract and ginsenoside Rg5 in human umbilical vein endothelial cells (혈관내피세포에서 산양삼 추출물과 진세노사이드 Rg5의 혈관신생 효과)

  • Ginsenoside Rg5, one of the protopanaxadiol ginsenosides of wood-cultivated ginseng, has been implicated in various diseases, such as diabetes, cancer, and hypertension; however, its angiogenic activity and molecular mechanisms have not yet been elucidated. Here, we found that wood-cultivated ginseng extract and ginsenoside Rg5 increase in vitro proliferation, migration, and tube-like structure formation, which are typical phenomena associated with angiogenesis, in cultured human umbilical vein endothelial cells (HUVECs). Moreover, Ginsenoside Rg5 stimulated the phosphorylation of Akt, endothelial nitric oxide (NO) synthase (eNOS), and extracellular-regulated kinase (ERK)1/2, which are well-known signal mediators of the angiogenic pathway. Furthermore, Ginsenoside Rg5 did not accelerate the activation of ICAM-1 and VCAM-1 which are inflammatory response mediators. These results suggest that wood-cultivated ginseng extract and ginsenoside Rg5 stimulated in vitro angiogenesis by activating the Akt/eNOS- and ERK1/2-dependent signal pathways without inducing vascular inflammation.

Arg-Leu-Tyr-Glu Suppresses Retinal Endothelial Permeability and Choroidal Neovascularization by Inhibiting the VEGF Receptor 2 Signaling Pathway

  • Vascular endothelial growth factor (VEGF) plays a pivotal role in pathologic ocular neovascularization and vascular leakage via activation of VEGF receptor 2 (VEGFR2). This study was undertaken to evaluate the therapeutic mechanisms and effects of the tetrapeptide Arg-Leu-Tyr-Glu (RLYE), a VEGFR2 inhibitor, in the development of vascular permeability and choroidal neovascularization (CNV). In cultured human retinal microvascular endothelial cells (HRMECs), treatment with RLYE blocked VEGF-A-induced phosphorylation of VEGFR2, Akt, ERK, and endothelial nitric oxide synthase (eNOS), leading to suppression of VEGF-A-mediated hyper-production of NO. Treatment with RLYE also inhibited VEGF-A-stimulated angiogenic processes (migration, proliferation, and tube formation) and the hyperpermeability of HRMECs, in addition to attenuating VEGF-A-induced angiogenesis and vascular permeability in mice. The anti-vascular permeability activity of RLYE was correlated with enhanced stability and positioning of the junction proteins VE-cadherin, ${\beta}$-catenin, claudin-5, and ZO-1, critical components of the cortical actin ring structure and retinal endothelial barrier, at the boundary between HRMECs stimulated with VEGF-A. Furthermore, intravitreally injected RLYE bound to retinal microvascular endothelium and inhibited laser-induced CNV in mice. These findings suggest that RLYE has potential as a therapeutic drug for the treatment of CNV by preventing VEGFR2-mediated vascular leakage and angiogenesis.

Antiangiogenic activity of non-aqueous fraction from Sparassis crispa extract in human umbilical vein endothelial cells (혈관내피세포에서 꽃송이버섯(Sparassis crispa) 소수성 추출물의 항혈관신생 활성)

  • Sparassis crispa is an edible mushroom that is distributed in Korea, Japan, Europe, and North America. It exerts various biological activities such as immunopotentiation, anti-diabetic, anti-cancer, and anti-inflammatory effects. Recently, we separated the health functional non-aqueous fraction from the chloroform extract of S. crispa (SCF4). In this study, we evaluated the antiangiogenic activity of SCF4 in human umbilical vein endothelial cells (HUVECs). SCF4 effectively inhibited vascular endothelial growth factor (VEGF)-induced cell growth at concentrations ($5-25{\mu}g/mL$) showing no cytotoxic effects. SCF4 inhibited VEGF-induced invasiveness and tube formation ability, which are in vitro angiogenic features of HUVECs, in a dose-dependent manner. In addition, SCF4 markedly suppressed in vivo angiogenesis of chorioallantoic membrane from growing chick embryos without cytotoxicity. Furthermore, SCF4 downregulated the phosphorylation of VEGFR2, AKT, and ERK1/2, which are major angiogenic signal mediators. These results suggest that SCF4 inhibited angiogenesis by suppressing the VEGFR2 signaling pathways without cytotoxicity.
