Effects of Soyoligosaccharides on Blood glucose and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats (대두올리고당이 당뇨 유발 흰쥐의 혈당 및 지질대상에 미치는 영향)

  • This study was conducted to investigate the effect of soyligosaccharides on improvement of diabetes mellitus. Sprague Dawley rats were divided into three groups. Control and one experimental group were fel AIN-93G diet(mixed diet) and the other experimental group was fed a similar diet containing 5% soyligosaccharides(oligosaccharide diet) for 6 weeks. Experimental groups were divided into two sub-groups after streptozotocin(STZ) injection and fed th mixed diet or the oligosaccharide diet for the next 2 weeks. Followings are final experimental and control groups: MM (mixed dietlongrightarrowSTZ injectionlongrightarrowmixed diet). MO(mixed dietlongrightarrowSTZ injectionlongrightarrowoligosaccharide diet), OM( oligosaccharide dietlongrightarrowSTZ injectionlongrightarrowmixed diet), OO(oligosaccharide dietlongrightarrowSTZ injectionlongrightarrowoligosaccharide diet), C(mixed dietlongrightarrowplacebo injectionlongrightarrowmixed diet). Blood glucose was analyzed at d1, d8 and d12 after streptozotocin injection. At the end of the experimental period, fecal bifidobacteria, plasma insulin, total lipid, triglyceride, cholesterol, and leptin concentrations was analyzed, After STZ injection, blood glucose level was significantly elevated in experimental group compared to a control group. Elevated blood glucose level declined faster in OO group compared to the other experimental groups. Meanwhile, insulin concentration decreased in all experimental groups compared to a control group. The number of bifidobacteria was significantly increased in oligosaccharide diet groups after STZ injection (MO and OO) than a mixed diet group(MM). Plasma cholesterol concentration significantly decreased in oligosaccharide diet groups(MO and OO) compared to mixed diet groups (MM and OM) after SZ injection. Plasma triglyceride and liver cholesterol concentrations significantly decreased in MO group compared to other experimental groups. Meanwhile, plasma leptin concentration was significantly lower in oligosaccharide diet groups after STZ injection(MO and OO) compared to a control group. The results in this study suggest that the dietary soyoligosaccharides has strong on reduction of reduction of blood glucose and lipid concentrations in streptozotoicin-induced diabetic rats. Thus, the use of soyoligosaccharides at a diet for IDDM patients may help to improve diabetes mellitus symptoms.(Korean J Nutrition 34(1) : 3-13, 2001)

Anti-Diabetic Effects of Dung Beetle Glycosaminoglycan on db Mice and Gene Expression Profiling

  • Anti-diabetes activity of Catharsius molossus (Ca, a type of dung beetle) glycosaminoglycan (G) was evaluated to reduce glucose, creatinine kinase, triglyceride and free fatty acid levels in db mice. Diabetic mice in six groups were administrated intraperitoneally: Db heterozygous (Normal), Db homozygous (CON), Heuchys sanguinea glycosaminoglycan (HEG, 5 mg/kg), dung beetle glycosaminoglycan (CaG, 5 mg/kg), bumblebee (Bombus ignitus) queen glycosaminoglycan (IQG, 5 mg/kg) and metformin (10 mg/kg), for 1 month. Biochemical analyses in the serum were evaluated to determine their anti-diabetic and anti-inflammatory actions in db mice after 1 month treatment with HEG, CaG or IQG treatments. Blood glucose level was decreased by treatment with CaG. CaG produced significant anti-diabetic actions by inhiting creatinine kinase and alkaline phosphatase levels. As diabetic parameters, serum glucose level, total cholesterol and triglyceride were significantly decreased in CaG5-treated group compared to the controls. Dung beetle glycosaminoglycan, compared to the control, could be a potential therapeutic agent with anti-diabetic activity in diabetic mice. CaG5-treated group, compared to the control, showed the up-regulation of 48 genes including mitochondrial yen coded tRNA lysine (mt-TK), cytochrome P450, family 8/2, subfamily b, polypeptide 1 (Cyp8b1), and down-regulation of 79 genes including S100 calcium binding protein A9 (S100a9) and immunoglobulin kappa chain complex (Igk), and 3-hydroxy-3-methylglutaryl-CoenzymeAsynthase1 (Hmgcs1). Moreover, mitochondrial thymidine kinase (mt-TK), was up-regulated, and calgranulin A (S100a9) were down-regulated by CaG5 treatment, indicating a potential therapeutic use for anti-diabetic agent.

The Effects of Astaxanthin Supplements on Lipid Peroxidation and Antioxidant Status in Postmenopausal Women

  • In postmenopausal women, the incidence of cardiovascular disease(CVD) is common and there is growing evidences that astaxanthin has a strong antioxidant capacity and plays a beneficial role in the prevention of CVD. However, current data are not sufficient to determine the effect of astaxanthin on improving lipid profiles and antioxidant capacity in human. In this study, 15 healthy postmenopausal women were divided into 3 groups and given astaxanthin supplements of 0,2 or 8mg/day for 8 weeks. Blood samples were taken before and after 4 and 8 weeks of astaxanthin supplementation for analysis of serum total choelsterol, LDL-cholesterol, HDL-cholesterol, triglyceride, plasma TBARS, total antioxidant status(TAS) and urinary 8-isoprostanes. HDL-cholesterollevels in 2mg and 8mg group increased significantly after 8 weeks from 50.6$\pm$5.8 to 60.4$\pm$7.1mg/dl, 44.4$\pm$10.7 to 49.4$\pm$2.7$mg/dl$ respectively (p<0.05). In the 2mg group, triglyceride decreased significantly from 171.6$\pm$67.4 mg/$dl$ to 145.8$\pm$5.1$mg/dl$ (p<0.05). Plasma TBARS level in the 2mg group decreased from 1.42$\pm$0.18nM/mg to 1.13$\pm$0.18nM/mg after 8 weeks (p<0.05). In the 8mg group, TBARS level decreased significantly from 1.62$\pm$0.14nM/mg to 1.13$\pm$0.12nM/mg after 8 weeks (p<0.05). TAS, as an indicator of lipid peroxidation, increased significantly from 0.85$\pm$0.42mM/$l$ to 1.90$\pm$0.58mM$l$ after 8 weeks in the 8mg group (p<0.05). Urinary 8-isoprostanes excretion did not decrease significantly with astaxanthin supplementation. In conclusion, it would be helpful for postmenopausal women with common cardiovascular disease to supplement with astaxanthin as an antioxidant.

Effect of Dietary Fibers in Rice and Barley on Lipid and Cadmium Metabolism in the Rat (쌀과 보리의 식이 섬유가 흰쥐의 지방 및 Cadmium 대사에 미치는 영향)

  • This study was performed to investigate effect of dietary fibers in rice and barley on glucose, lipid and cadmium (Cd) metabolism in the rat. Fifty-six male Spague-Dawley rats weighing 244.6$\pm$2.7g were blocked into eight groups according to body weight and raised for four weeks with diets containing 0 or 0.04%(w/w) CdCl2 and four different carbohydrate sources, starch, rice flour, barley flour and mixture of rice and barley flour(7 : 3, w/w). Total dietary fibers and $\beta$-glucan contents of barley were about three times higher than those of rice (10.75% vs. 3.94%, 3.11% vs. 1.06%, respectively). Food intake, weight gain, food efficiency ratio, liver and kidney weights were lower in Cd exposed groups, and barley group among Cd exposed animals showed highest weight gain, food efficiency ratio and organ weights. Fasting serum glucose levels were not significantly different among groups, Serum cholesterol level was lowest in Cd exposed barley group. Serum HDL-cholesterol level was higher in none-Cd exposed starch and barley groups, and HDL-cholesterol : total cholesterol ratios were higher in none-Cd rice and mixed flour groups than other groups. Liver total lipid and triglyceride levels were lowest in barley groups regardless of Cd administration. Fecal total lipid, cholesterol and triglyceride excretions were high in barley and mixed flour groups. Liver Cd concentrations were low in Cd exposed barley and mixed flour groups. In Cd exposed barley group, fecal weight and Cd excretion were highest and Cd retention ratio was lowest among groups. Small intestine metallothionein(MT) concentration was highest in Cd exposed rice group, and kidney MT concentration was highest in Cd exposed barley group. In conclusion, cereals showed different effects on lipid and Cd metabolism that might be mediated by dietary fibers in cereals. Especially $\beta$-glucan-rich barley group showed greatest lipid and Cd lowering effects by increasing fecal lipids and Cd excretions.

  • The purpose of this study is to determine the effect of diabetes education program on Glucose Metabolism(blood sugar, HbA1c) and Lipid Metabolism(total cholesterol, triglyceride, low density lipoprotein, high density lipoprotein), Self-efficacy in non-insulin independent diabetes mellitus. The study design was a non equivalent control group pre-test post-test design. Data for the study were collected from March 12 to June 19, 2001. Sixty-two research subjects were assigned to experimental(36) and control(26) groups. The collected data was analyzed using the Chi-Square test, t-test by spsswin program The results are as follows : 1. Experimental group had higher level of glucose metabolism than control group(FBS ; t=-3.317, p=.002, HbA1c; t=-4.956, p=.000). 2. Level of lipid metabolism were partly a significant different between experimental group and control group(Triglyceride ; t=-2.108, p=.039). 3. Experimental group had higher efficacy score than control group(t=4.651, p=.000). In conclusion, the study supported the effects of diabetes education program to increase metabolism an d self-efficacy. Further study with a longitudinal design is suggested to verify the effect of diabetes education program in NIDDM and standardized diabetes education program.

Effect of Wild Grape Juice on Lipid Metabolism and Antioxidaive Activity in Rat Fed High Oxidized Lipid (산화된 지방식이를 급여한 흰쥐에서의 머루즙 투여가 지질대사와 항산화에 미치는 영향)

  • This study is to find out the antioxidative effect and serum and liver lipid composition of wild grape juice in vivo. Forty 6-week-old white Sprague Dawley rats were divided into 4 groups such as normal lipid group, normal lipid group with wild grape juice, oxidized lipid (basic diet plus 10% of oxidative lipid) group and oxidized lipid group with wild grape juice, and 2ml juice was provided everyday. After 4 weeks of feeding with experimental diet each groups were examined for the antioxidant enzyme activity in blood and liver microsome and their serum and liver lipid composition. Glutanthione peroxidase activity in blood was significantly higher in oxidized lipid group with wild grape juice than in oxidized lipid group. Glutanthione peroxidase activity showed no difference depending on wild grape juice supplementation. Glutanthione peroxidase activity in liver was significantly higher in the groups with wild grape juice than in the groups supplemented only with oxidized lipid. Glutanthione reductase activity showed no difference depending on the supplementation of wild grape juice. Serum triglyceride level in the group supplemented with oxidized lipid diet and wild grape juice showed similar value to the normal lipid group and the normal lipid group with wild grape juiceoxidized fa6. Liver total lipid in the group supplemented with oxidized lipid and wild grape juice showed similar value to the normal lipid group and the group supplemented with normal lipid and wild grape juice. And it was lower than that of oxidative lipid group without juice. The liver triglyceride level in the group supplemented with normal lipid and wild grape juice was lower than that in the oxidative lipid group, but it was as low as in the group supplemented only with normal lipid.

Effects of Dietary Olive Oil on Growth Performance, Carcass Parameters, Serum Characteristics, and Fatty Acid Composition of Breast and Drumstick Meat in Broilers

  • This experiment was conducted to evaluate the effects of dietary olive oil on growth performance, carcass parameters, serum characteristics, and fatty acid composition of breast and drumstick meat in broiler chickens. A total of 480 broilers were randomly allotted into three dietary treatments, including T (basal diet, 5% tallow), O1 (2% olive oil+3% tallow), and O2 (5% olive oil). During d 0 to 21, broilers fed the diet supplemented with 5% olive oil showed lower (p<0.05) body weight gain (BWG) and feed intake (FI) compared with those fed the T diet. Serum triglyceride concentration was reduced (p<0.05), while high density lipoprotein (HDL)-cholesterol concentration was increased (p<0.05) in the O2 treatment group compared with the T and O1 treatment groups. The addition of olive oil to the diets induced a reduction (p<0.05) in the total saturated fatty acid (SFA) contents in breast and drumstick meat, and increased (p<0.05) the total unsaturated fatty acid (USFA) contents and USFA/SFA ratios. In conclusion, a diet with 5% olive oil could decrease BWG and FI of broilers during the starter period (wk 0 to 3), and cause an increase in the serum HDL-cholesterol level, while decreasing the serum triglyceride concentration. Furthermore, USFA level and USFA/SFA ratios in breast and drumstick meat were increased by dietary supplementation of 2 or 5% olive oil.

Effect of Houttuynia cordata Thunb and Herbs Mixture Extract on the Lipid Metabolism in the LPS-induced Hepatotoxicity (어성초 함유 혼합추출물이 LPS 유도 간독성에서 지질대사에 미치는 영향)

  • Lipopolysaccharide (LPS) induces the synthesis of several inflammatory cytokine, chemokine, NO and inflammation in the liver of rats. The purpose of this study was to investigate the preventive effects of Houttuynia cordata Thunb and Oriental herbs mixture extract (HCTM) against the hepatotoxicity-inducing LPS in the lipid metabolism. HCTM of 100 mg/kg concentration was intra-peritoneally administered into rats at dose of 1.5 ml/kg for 20 days. On the day 21, 1.5 ml/kg of LPS was injected 4 hours before anesthetization. The levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), total lipid, triglyceride (TG) and HDL-cholesterol (HDL-C) were measured in serum. LPS-treatment markedly increased the levels of GOT, GPT, total lipid and TG, and decreased the level of HDL-C. However, HCTM pretreatment decreased the levels of GOT, GPT, total lipid and TG by 76.8%, 55.48%, 34.04% and 76.21% respectively, and increased the level of HDL-C by 97.84%. These results showed that HCTM had the protective effect against the hepatotoxicity-inducing LPS in the lipid metabolism, and it suggest that HCTM could be used for functional beverage.

Evaluation the Effects of Short Term Intake Avena sativa Extracts on Blood Glucose and Cholesterol Level (단기간 귀리 추출물 섭취의 혈당 및 콜레스테롤 저하 효과 평가)

  • Recently, studies about various natural extracts to help control blood glucose has been in progress. Avena sativa is well known to have various physilogical effects. Especially, ${\beta}$-glucan has effect about lowering blood glucose level and prevent cardiovascular dz and adult dz related to obesity. In this study we evaluated the effect of Down and control (BM pharmaceutical) which is consist of commercialized Avena sativa fextracts on blood glucose and cholesterol, 6weeks, randomized, double-blind, placebo-controlled trials. The results show not significantly different in all blood index test group from control group, but in glycated albumin decreased 50.33% for test group, decreased 37.91% for control group, in triglyceride decreased 7.51% for test group, increased 3.98 for control group and we can observed Avena sativa has blood glucose and triglyceride lowering effect in some.

The Relationship between the Life Style and the Status of Serum Lipis and Antioxidant Vitamins in University Students (대학생들의 생활습관과 혈청 항산화비타민 및 지질상태의 관련성에 관한 연구)

  • This study was undertaken to investigate the relationship between the life style and the nutritional status of serum antioxidant vitamins and lipids in university male and female students. 48 male and 49 female students attending Andong university, aged between 18 and 25 years, were selected. Questions about the life styles including dietary intakes, food habits, smoking, drinking alcohol, exercise, stress were answered. And serum levels of antioxidant vitamins and lipids were determined. Average serum levels of total cholesterol, LDL C, HDL C, and triglyceride in male and female subjects were 158.6$\pm$32.7, 177.3$\pm$33.8; 86.4$\pm$26.0, 109.0$\pm$31.2; 46.0$\pm$10.7, 49.9$\pm$12.4; 131.2$\pm$22.5, 91.7$\pm$ 38.6mg/dl respectively. Average serum levels of antioxidant vitamin A, E and C in male and female subjects were 42.6$\pm$12.3, 31.4$\pm$9.8 g/dl, 1.11$\pm$0.38, 1.15$\pm$0.29mg/dl and 164.66 $\pm$65.01, 220.06$\pm$80.11 g/dl respectively. There was no significant difference between smoking habits and either serum lipids or antioxidant vitamins level. The serum vitamin C level of drinkers was significantly lower(p=0.038), but serum lipids(total cholesterol, LDL C, and triglyceride) were higher than non alcoholic subjects. The subjects with severe stress had lower in HDL C and higher in atherogenic index than others. This result indicates that oxidative stress may be increased in stressful environment from undesirable life styles and influence the status of serum lipid and antioxidant vitamins.
