Synergistic anticancer effect of acteoside and temozolomide-based glioblastoma chemotherapy

  • Temozolomide (TMZ) is an alkylating agent commonly used as a first-line treatment for high-grade glioblastoma. However, TMZ has short half-life and frequently induces tumor resistance, which can limit its therapeutic efficiency. In the present study, it was hypothesized that combined treatment with TMZ and acteoside has synergistic effects in glioblastoma therapy. Using cell viability and wound-healing assays, it was determined that this treatment regimen reduced cell viability and migration to a greater extent than either TMZ or acteoside alone. Following previous reports that TMZ affected autophagy in glioma cells, the present study examined the effects of TMZ + acteoside combination treatment on apoptosis and autophagy. The TMZ + acteoside combination treatment increased the cleavage of caspase-3 and levels of B-cell lymphoma 2 (Bcl-2)-associated X protein and phosphorylated p53, and decreased the level of Bcl-2. The combination treatment increased microtubule-associated protein 1 light chain 3 and apoptosis-related gene expression. It was also determined that TMZ + acteoside induced apoptosis and autophagy through the mitogen-activated protein kinase signaling pathway. These findings suggest that acteoside has beneficial effects on TMZ-based glioblastoma therapy.

Gastrodia elata Blume Attenuates 2, 4-Dinitrochlorobenzene-induced Atopic Dermatitis-like Skin Lesions in Balb/c Mice and SD Rats

  • Gastrodia elata Blume is a well-known kind of natural products used as a folk medicine for thousands of years. However, anti-atopic dermatitis-like effects of G. elata Blume had not been evaluated until now. The aim of the present study is to investigate the protective effects of water extract from the roots of G. elata Blume (GE) on 2, 4-dinitrochlorobenzene-induced atopic dermatitis-like skin lesions using balb/c mice. Combination treatment of GE (at a dose of 12.5 mg/kg body weight by administrated per os + 0.5 mg/cm2 as ointment to apply on ear and dorsal skin) was significantly inhibited spleen weight, ear thickness, levels of serum immunoglobulin E and number of mast cells, compared with that of 2, 4-dinitrochlorobenzene-included groups without GE. Furthermore, combination application by oral administration plus by ointment of GE significantly inhibited the histamine release from rat peritoneal mast cells. These results suggest that combination treatment of oral administration plus ointment form of GE could be helpful as a potentially natural pharmaceutical treatment on atopic-like dermatitis.

  • Objectives: This report describes the case of a patient with progressive multifocal leukoencephalopathy (PML) with hemiplegia, who was treated with a combination of traditional Korean medicine and western medicine. Methods: The patient received traditional Korean medical treatments, including acupuncture, during robust antiretroviral therapy with rehabilitative exercise. Results: During 24 months of treatment, the patient showed improvement in hemiplegia on a manual muscle test. Conclusions: The combination of traditional Korean medicine and western medicine was effective in the treatment of PML in a patient with hemiplegia.

The Synergistic Anticancer Effect of Artesunate Combined with Allicin in Osteosarcoma Cell Line in Vitro and in Vivo

  • Background: Artesunate, extracted from Artemisia annua, has been proven to have anti-cancer potential. Allicin, diallyl thiosulfinate, the main biologically active compound derived from garlic, is also of interest in cancer treatment research. This object of this report was to document synergistic effects of artesunate combined with allicin on osteosarcoma cell lines in vitro and in vivo. Methods: After treatment with artesunate and allicin at various concentrations, the viability of osteosarcoma cells was analyzed by MTT method, with assessment of invasion and motility, colony formation and apoptosis. Western Blotting was performed to determine the expression of caspase-3/9, and activity was also detected after drug treatment. Moreover, in a nude mouse model established with orthotopic xenograft tumors, tumor weight and volume were monitored after drug administration via the intraperitoneal (i.p.) route. Results: The viability of osteosarcoma cells in the combination group was significantly decreased in a concentration and time dependent manner; moreover, invasion, motility and colony formation ability were significantly suppressed and the apoptotic rate was significantly increased through caspase-3/9 expression and activity enhancement in the combination group. Furthermore, suppression of tumor growth was evident in vivo. Conclusion: Our results indicated that artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis.

하이드로겔과 하이드로콜로이드 드레싱을 이용한 압박성 궤양의 치험례 (The Treatment of Pressure Sore Using Hydrogel and Hydrocolloid Dressing)

  • Pressure sores are a common complication of hospitalized patients. However, It is often impossible to correct surgically because the general conditions of these patients are poor. It is known that the hydrogel has a powerful autolytic effect by providing moist environments and facilitates wound healing and hydrocolloid dressing is also known to promotes granulation tissue formation and epithelialization. The patients were treated with hydrogel(Purion $gel^{(R)}$, Coloplast A/S, Denmark) and hydrocolloid dressing (Comfeel Plus Transparent $Dressing^{(R)}$, Coloplast A/S, Denmark) after surgical debridement of pressure sores progressed to stage III(n=2) and IV(n=7). This combination treatment could facilitate to debride the nectrotic tissue and promote granulation tissue formation epithelialization simultaneously. We could achieve complete healing of pressure sores using the combination treatment without requiring surgical correction. In conclusion, hydrogel in combination with hydrocolloid dressing is effective in acheiving complete healing of progressed pressure sores.

Sequential administration of camptothecin sensitizes human colon cancer HCT116 cells to paclitaxel via $p21^{Cip1/WAF1}$

  • Colorectal cancer is the third leading cause of cancer-related death in Western countries. Chemotherapeutic agents with different mechanisms of action have shown an increase in cure rates. In the present study, we investigated the effect of a combination of low concentration of paclitaxel (taxol, 5 nM) and topoisomerase 1 inhibitor camptothecin (CPT) on HCT116 colon cancer cells. Although the viability of cells treated with taxol alone was similar to that of control cells, sequential treatment with taxol and CPT exhibited high cytotoxicity. However, the opposite sequence of treatment did not exert cytotoxic effects on HCT116 cells. This enhanced cytotoxicity of the sequential combination therapy was the result of mitotic arrest, which increased the level of $p21^{Cip1/WAF1}$ through the p38 mitogen-activated protein kinase (MAPK) pathway. Knockdown by $p21^{Cip1/WAF1}$ siRNA or treatment with a p38 inhibitor reduced the viability of cells sequentially exposed to taxol and CPT. Taken together, a low taxol concentration in combination with CPT induced mitotic arrest in HCT116 cells, leading to synergistic cell death through enhanced expression of $p21^{Cip1/WAF1}$ and p38 MAPK pathway. Therefore, taxol could playa role as a sensitizer of CPT in colon cancer cells.

흰쥐 적출대동맥의 수축력에 미치는 열과 Nacl의 영향

  • In this study, in order to examine whether salt and heat shock stress would alter or not contraction and relaxation of isolated rat aorta. Under anesthesia with sodium pentobarbital(50 mg Kg$^{-1}$ i.p.), male Sprague Dawley rats weighing 300-330 g were subjected to 0, heat shock combined salt stress, where as the sham group was left at modified Krebs-bicarbonate solution. To measure contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And the strip was checked for expression of heat shock protein(Hsps) by means of western blotting. The combination group of heat and 50 mM NaCl group increased vascular contractility, and the heat and 150 mM NaCl group decreased vascular contractility for 5 hours, and then recovered for 8 hours compared to that of control. Expressin of Hsp 70 of vascular muscle of rat aorta more increased by combination of heat and NaCl treatment than those of single treatment of heat or NaCl treatment, and vascular Hsp 70 showed a little decrease at 8 hours compared at 5 hours. These result indicate that mixed environmental stress either increased or decreased in vascular contractility by combination of heat and NaCl concentration.

한양방 병용 치료로 임신 유지된 조산 고위험군의 조기진통 환자 1례 보고 (A Case Report of Preterm Labor Patient at High Risk for Preterm Delivery Treated by Combination of Herbal Medicine and Conventional Treatment)

  • Objectives: The purpose of this study is to report the effect of combination of herbal medicine and conventional treatment on preterm labor patient at high risk for preterm delivery. Methods: The patient in this case was 33-year-old female with gravid 3, para 2, abortus 1. She experienced 1 preterm birth 5 years ago. She visited our clinic with lower abdominal pain at 14th gestational week. We treated her by herbal medicine. She also take vaginal progesterone from 19th gestational week to 34th gestational week. Results: She maintained her pregnancy until giving a birth by cesarean section at 38th gestational week. No adverse effects were not observed among mother or infant. Conclusions: Combination of herbal medicine and conventional treatment is effective on preterm labor patient at high risk for preterm delivery.

Combined nano-particle drug delivery and physiotherapy in treatment of common injuries in dance-sport

  • Combination of novel technologies with traditional physiotherapy in rehabilitation in injured athletes have shown to provide improved time of recovery. In specific, nanodrugs delivery systems are widely utilized as a counterpart to the physiotherapy in injuries in sports. In the present study, we focus on the common injuries in dance-sports, their recovery and the effect combination of nano-particle drug delivery with the physiotherapy practices. In this regard, a comprehensive review on the common injuries in dance sport is provided. Moreover, the researches on the effectiveness of the nano-particle drug delivery in therapy of such injuries and in similar cases are provided. The possibility of using combination of nano-particle drug delivery and physiotherapy is discussed in detail. Finally, using artificial intelligence methods, predictions on the recovery time and after-treatment side-effects is investigated. Artificial Neural Network (ANN) predictions suggested that using nano-particle drug delivery systems along with physiotherapy practices could provide shortened treatment time to recovery in comparison to conventional drugs. Moreover, the post-recover effects are less than the conventional methods.

  • Objectives : The purpose of this study is to report the effect of Korean-Western medicine combination treatment administered to two patients with acute herpes zoster. Methods : During hospitalization, the two patients were treated with Hyeonggaeyeongyotang-gagam, Hwangryunhaedok-tang wet dressing, pharmacopuncture, acupuncture, and other western medications including antiviral and analgesic agents. Photographs of skin lesions, numeric rating scale(NRS), and visual analog scale(VAS) were used to assess the improvement of symptoms. Results : At the time of discharge, both patients showed remarkable improvement in skin lesions, NRS for pain, and VAS for pruritus and insomnia. Conclusions : The results demonstrated that Korean-Western medicine combination treatment may be effective for treating herpes zoster.
