Effect of $N_2$-back-flushing Time and TMP in Lake Water Treatment Using Multichannel Ceramic Microfiltration Membranes (다채널 세라믹 정밀여과막으로 호소수 처리시 질소 역세척 시간 및 막간 압력차의 영향)

  • In this study, we treated lake water by 2 kinds of multichannel ceramic micro filtration membranes. We could investigate effects of $N_2-back-flushing$ time (BT) and transmembrane pressure (TMP), and find optimal operating conditions. The BT were changed in $10{\sim}60$ sec, TMP in $0.6{\sim}2.0$ bar at fixed filtration time (FT) 8 min, flow rate 2.0 L/min and back-flushing pressure 2.0 bar. Also, the optimal conditions were discussed in the viewpoints of resistance of membrane fouling $(R_f)$, dimensionless permeate flux $(J/J_o)$, permeate flux (J) and total permeate volume $(V_T)$. As result, optimal back-flushing conditions for HC04 ($0.4{\mu}m$ pore size) and HC10 membrane $(1.0{\mu}m)$ were BT=10 sec and BT=20 sec, respectively. Then, higher TMP should increase the driving force, and could produce more VT. Average rejection rates of pollutants were higher than 95.4% for turbidity, $12.7{\sim}20.1%\;for\;COD_{Mn},\;0.0{\sim}6.4%\;for\;NH_3-N,\;1.9{\sim}4.6%$ for T-N and $34.9{\sim}88.4%$ for T-P.

Nitrogen and Phosphorus Removal in Long Term Pilot Plant Operation Using Submerged Hollow Fiber Membrane and Ferric Chloride (침지형 중공사막과 철염을 이용한 Pilot MBR 공정의 장기운전에 따른 질소, 인 제거 특성)

  • Pilot scale vertical-type membrane bioreactor was operated to examine the effect of $FeCl_3$ injection on the removal of organics, nitrogen and phosphorous, and additionally trans-membrane pressure (TMP) was observed. The membrane type was hollow fiber membrane with pore size of $0.25\;{\mu}m$, and the material was polytetrafluoroethylene (PTFE). The membrane permeate was continuously removed by a pump under a constant flux ($25\;L/m^2/h$). Air back-flushing technique were adopted to reduce fouling. As a result, TMP was increased more slowly than that of the operation without air back-flushing, During long-term operation, approximately 310 days, the injection of $FeCl_3$ was effective not only in removing phosphorous chemically but also in reducing TMP increase. Furthermore, while the average COD and T-N concentration of the effluent without $FeCl_3$ injection was 14.3 mg/L and 6.0 mg/L respectively, that of effluent with $FeCl_3$ was 11.3 mg/L and 6.0 mg/L respectively, which confirmed the effects of $FeCl_3$.

Characterization of Root Transcriptome among Korean Ginseng Cultivars and American Ginseng using Next Generation Sequencing (차세대염기서열 분석을 이용한 고려인삼과 미국삼의 전사체 분석)

  • The transcriptomes of four ginseng accessions such as Cheonryang (Korean ginseng cultivar), Yunpoong (Korean ginseng cultivar), G03080 (breeding line of Korean ginseng), and P. quinquefolius (American ginseng) was characterized. As a result of sequencing, total lengths of the reads in each sample were 156.42 Mb (Cheonryang cultivar), 161.95 Mb (Yunpoong cultivar), 165.07 Mb (G03080 breeding line), and 166.48 Mb (P. quinquefolius). Using a BLAST search against the Phytozome databases with an arbitrary expectation value of 1E-10, over 20,000 unigenes were functionally annotated and classified using DAVID software, and were found in response to external stress in the G03080 breeding line, as well as in the Cheonryang cultivar, which was associated with the ion binding term. Finally, unigenes related to transmembrane transporter activity were observed in Cheonryang and P. quinquefolius, which involves controlling osmotic pressure and turgor pressure within the cell. The expression patterns were analyzed to identify dehydrin family genes that were abundantly detected in the Cheonryang cultivar and the G03080 breeding line. In addition, the Yunpoong cultivar and P. quinquefolius accession had higher expression of heat shock proteins expressed in Ricinus communis. These results will be a valuable resource for understanding the structure and function of the ginseng transcriptomes.

Use of Response Surface Methodology for Optimization of Clarified Mixed Apple and Carrot Juice Production (반응표면 분석을 이용한 사과.당근 혼합주스의 청징공정 최적화)

  • Response surface methodology was used to investigate the quality of clarified mixed apple and carrot juices using ultrafitration. Apple and carrot juices were blended at the ratio of 1:3, 1:1, and 3:1. A three-variable, three-level central composite design was employed where the independent variables were the blend ratio, temperature and average transmembrane pressure (ATP). With increasing temperature and pressure, flux linearly increased regardless of blending ratio. Blend juice with 75% apple showed the highest soluble sugar and total sugar content in apple and carrot blend juices. Soluble solid contents were more affected by blending ratio than temperature and ATP. Total sugar contents were greatly affected by temperature; increasing temperature led to higher total sugar content up to $25^{\circ}C$. Higher carrot ratio led to higher vitamin C content. In general, higher acidity was achieved by higher apple content and acidity was increased with increasing temperature. Turbidity increased for all samples as APT increased, with the blending ratio of 1:1 (apple:carrot) showing the highest turbidity. Viscosity was greatly changed in the blending ratio of 3:1 (apple:carrot) juice. The polynomial models developed by RSM were satisfactory to describe the relationships between the studied factors and the responses. Analytical optimization gave $flux=0.216\;L/m^2.h$, soluble $solids=10.39^{\circ}Brix$, total sugar=71.32 mg/mL, vitamin C=315.18 mg%, acidity=7.78 mL, turbidity=0.017, and viscosity=1.44 cp, when using a $temperature=44.97^{\circ}C$, ATP=113.57 kPa, and blend ratio=28.50%.

A Study on Operating Condition of Test-Bed Plant using Membrane filtration of D Water Treatment Plant in Gwang-Ju (D정수장 정밀여과막 실증플랜트의 최적 운전조건 연구)

  • Membrane filtration has become more popular in drinking water treatment recently, since the filtration can control not only particulate matters but also pathogenic microorganisms such as giardia and cryptosporidium very effectively. Pilot-scale ($120m^3/d$ of treatment capacity) and test-bed ($25,000m^3/d$ of treatment capacity) microfiltration experiments were conducted to find optimum operating mode and the critical flux. Optimum operating mode of pilot-test was assessed as inflow 1.0 min, filtration 36.5 min, air backwash 0.9 min, backwash 1.0 min and outflow 1.0 min with 50 LMH ($L/min{\cdot}m3^$) of critical flux. Critical Flux was calculated to be $50L/m^2-h$ (within TMP 0.5 bar) based on the increase formula of the transmembrane pressure difference according to the change of time at Flux 20, 40, 56 and 62 LMH in pilot operation. Chemical cleaning was first acid washed twice, and alkali washing was performed secondarily, and a recovery rate of 95% was obtained in the test-bed plant. The results of operating under these appropriate conditions are as follows. Turbidity of treated water were 0.028, 0.024, 0.026 and 0.028 NTU in spring, summer, autumn and winter time, respectively. Microfiltration has superior treatment capability and performance characteristics in removing suspended solids and colloidal materials, which are the main cause of turbidity and important carrier of metal elements, and it has shown great potential in being an economically substitute to traditional processes (sand filtration).

Debittering of Citrus Products Using ${\beta}-Cyclodextrin$ Polymer and Ultrafiltration Process (${\beta}-Cyclodextrin$ 중합체와 한외여과 공정을 이용한 감귤류의 쓴맛 성분 제거)

  • ${\beta}-Cyclodextrin\;({\beta}-CD)$ polymers were prepared using epichlorohydrin as a cross linking agent. The polymers were separated into ${\beta}-CD$ soluble polymer $({\beta}-CD\;SP)$ and ${\beta}-CD$ insoluble polymer $({\beta}-CD\;ISP)$ on a 10,000 molecular weight cut-off membrane (YM 10). Optimum separation conditions in the YM 10 were: transmembrane pressure 51.7 kPa, separation temperature $35^{\circ}C$, and volume concentration ratio 10. The flux was $0.025\;mL/cm^{2}/min$ under the optimum conditions. Gel permeation chromatography indicated that ${\beta}-CD\;SP\;and\;{\beta}-CD\;ISP$ had a degree of polymerization of $2{\sim}8$ and over 10, respectively. The formation of an inclusion complex with hydrophobic compounds such as 4-dimethylaminoazobenzene, methyl red, and naringin was compared among ${\beta}-CD,\;{\beta}-CD\;SP\;and\;{\beta}-CD\;ISP$. The molar absorptivity for the two chromatic compounds was increased and the absorption peak was shifted in the presence of ${\beta}-CD$ polymers. Naringin, the principal flavonoid bitter tasting component of citrus fruit, had a low water solubility. The solubility of naringin was increased through the formation of an inclusion complex with ${\beta}-CD$ polymers. There was no significant difference in the formation of an inclusion complex between ${\beta}-CD\;SP\;and\;{\beta}-CD\;ISP$. Reduction of the bitter components from citrus products was shown to be possible when employing ${\beta}-CD\;SP$, while the usage of ${\beta}-CD$ monomer has been limited due to the low water solubility.

Advanced Treatment of Liquid Fertilizer from Livestock Night Soil Treatment Facility by Membrane Separation Processes (분리막 공정을 이용한 축산분뇨 처리장 액비의 고도처리)

  • This paper is to develop the process suitable for the advanced treatment of liquid fertilizer from the livestock night soil treatment facility (biogas plant). Nanofiltration (NF) and reverse osmosis (RO) process was used, respectively, for the advanced treatment of liquid fertilizer. And membrane bioreactor (MBR) with and without biomedia were tested, respectively, for the pretreatment. It was found that almost T-N of the liquid fertilizer was composed of ammoniacal nitrogen. Transmembrane pressure of MBR with biomedia increased slowly during the operation time, while that of MBR without biomedia increased rapidly at the initial time. But there was no difference observed in the removal efficiencies of COD, T-N, and T-P irrespective of the dosage of biomedia. When the liquid fertilizer was pretreated by MBR with biomedia, the removal efficiencies of COD, T-N, and T-P were 99.8, 86.5%, and 99.8% by NF, and 99.9, 86.8%, and 99.8% by RO, respectively. Compared with the effluent quality standards of the livestock night soil treatment facility, the water quality treated by MBR and NF/RO process met the standard for COD and T-P, but exceeded the permitted standard for T-N. In order to meet the effluent quality standard for T-N, it is necessary to change the MBR operation cycle or to add the secondary treatment by NF/RO.

Ultrafiltration and Separation Process Optimization of Hen Egg White Lysozyme as Natural Antimicrobial Enzyme (천연 항균 효소제 난백 lysozyme의 한외여과 조건 최적화)

  • Hen egg white lysozyme (HEWL) is very valuable as a natural preservative in food processing due to its selective bactericidal activity. HEWL which traditionally isolated by crystallization or freeze drying was simply separated from 13 different hen egg white (HEW) proteins by a single-step ultrafiltration. Freeze dried HEW (0.25%, w/v) dissolved in a citrate-phosphate buffer (pH 4.6) was ultrafiltered with a PM30 membrane under various operating conditions, by changing concentration, temperature, transmembrane pressure $({\triangle}P_T)$, and stirring speed. Optimum separation conditions were decided when maximal flux was obtained. Under the optimum separation conditions, the effect of membrane material and fouling on flux as time passed as well as lysozyme concentration, protein concentration, specific activity (SA) in the permeate were measured. Best separation conditions of HEWL with PM30 membrane were sample concentration 0.25%, temperature $35^{\circ}C$, ${\Delta}P_T\;30\;psi$, and stirring speed 300 rpm. During the first 12 min, the flux of YM30 was higher, but at the steady-state it was lower than that of PM30. The SA of the PM30 permeate was over 2 times higher in spite of the lysozyme and protein concentration being lower than that of YM30 permeate. The flux of 5 times used PM30 decreased 30% compared to a new PM30, but both had the same tendency in flux decrease when time passed. Both of them reached a steady-state after 35 min and remained at 70% of the initial flux. In the PM30 permeate, the lysozyme concentration and SA were 110 units/mL and 2,821 units/mg protein, respectively. Therefore, PM30 membrane separation was very effective for separation of antimicrobial lysozyme.

Control of Membrane Fouling in Submerged Membrane Bioreactor(MBR) using Air Scouring (침지형 생물 반응기 공정에서 플럭스 향상을 위한 공기 세척 효과에 관한 연구)

  • Membrane bioreactor(MBR) processes have been widely applied to wastewater treatment for last decades due to its excellent capability of solid-liquid separation. However, membrane fouling was considered as a limiting factor in wide application of the MBR process. Excess aeration into membrane surface is a common way to control membrane fouling in most MBR. However, the excessively supplied air is easily dissipated in the reactor, which results in consuming energy and thus, it should be modified for effective control of membrane fouling. In this study, cylindrical tube was introduced to MBR in order to use the supplied air effectively. Membrane fibers were immersed into the cylindrical tube. This makes the supplied air non-dissipated in the reactor so that membrane fouling could be controlled economically. Two different air supplying method was employed and compared each other; nozzle and porous diffuser which were located just beneath the membrane module. Transmembrane pressure(TMP) was monitored as a function of airflow rate, flux, and ratio of the tube area and cross-sectioned area of membrane fibers(A$_m$/A$_t$). Flow rate of air and liquid was regulated to obtain slug flow in the cylindrical tube. With the same flow of air supply, nozzle was more effective for controlling membrane fouling than porous diffuser. Accumulation of sludge was observed in the tube with the nozzle, if the air was not suppled sufficiently. Reduction of membrane fouling was dependent upon the ratio, A$_m$/A$_t$. For diffuser, membrane fouling was minimized when A$_m$/A$_t$ was 0.27, but 0.55 for nozzle.

Ultrafiltration of Humic and Natural Water: Comparison of Contaminants Removal, Membrane Fouling, and Cleaning (휴믹산 용액 및 자연수의 한외여과: 제거율, 막오염 및 세척특성 비교)

  • NOM and fine particles are the main target materials in water treatment using membranes. Particularly, humic substances extracted from soils are frequently used in many fundamental studies representing natural organic matter in raw water for drinking water treatment. In this study, ultrafiltration (UF) of artificial humic water and natural river water was conducted and the characteristics of removal efficiency and permeability were compared. In the UF of river water, the transmembrane pressure increased in the same pattern with that of 5 mg/L humic water. For the removal of organic matter and fine particles, however, two types of feed water had shown different trends. Kaolin particles and humic acids added to artificial water were better removed, while colloids and organics in natural water were relatively poorly removed. From the $UV_{254}$ and GPC analyses, it seemed that the hydrophobicity and size of humic substances contributed to the greater removal of organic matter. The UF membrane applied for humic water also showed a higher flux recovery by caustic chemical cleaning than that for river water.
