A practical approach was proposed to produce transgenic chimeric chickens using blastodermal cells (BCs). The chicken BCs were mechanically dissociated and transferred into the recipient eggs that had been exposed to 500 rads irradiation of$^{60}Co$ and windowed on the equatorial plane. Chimeric chickens were generated using two models: the crosses (MXL) from Black Minors (ii,EE,b/b) ♂${\times}$Barred Leghorns (ii,ee,B/-) ♀ as donors and White Leghorns (WL, II) as acceptors (Model 1), or the Black Heifengs (BH, ii,EE,bb) as donors and Hua-xing white (HW, II) as recipients (Model 2). The treated eggs were incubated in their original shells in normal conditions until hatching. Green fluorescent protein (GFP) gene was transferred into the BCs derived from MXL and BH via lipofectamine and the pEGFP-C1, and transfection efficiency into the BCs was examined under a fluorescent microscope. Potential transgenic chimeras were selected based on the proposed methods in this study. Using the fresh BCs, the best rate of phenotypic chimeras was 6.7% and 26.0% in model-1 groups, and model-2 groups, respectively. We also described the optimized conditions for transfection. Although 30% of the BCs transfected in vitro emitted green light under an inverted fluorescent microscope, no embryos injected with the transfected BCs expressed foreign GFP gene at 3-4 days.
The quail (Coturnix japonica) has been used as a model animal in many research fields and its application is still expanding in other fields. Compared to the chicken, the quail is quicker to reach sexually maturity, has short generation intervals, is easy to handle, requires less space and feed, and is sturdy. In addition, it produces many eggs and the research tools developed for chicken can be applied directly to quail or with some modifications. Due to recent advances in next-generation sequencing, abundant sequence data for the quail genome and transcripts have been generated. These sequence data are valuable sources for studying functional genomics using quail, which is one of the model animal used to investigate gene function and networks. Although there are some obstacles to be removed, the quail is the best optimized model to study the functional genomics of poultry. In many research fields, functional genomics study using the quail model will provide the best opportunity to understand the phenomena and principles of life. We review why, among many other birds, the quail is the best model for studying poultry functional genomics.
본 연구는 오브알부민 프로모터의 최적 크기를 결정하기 위해 수행하였고, 이를 위해 2.8, 5.5, 그리고 Mut-4.4 kb의 오브알부민 프로모터를 리포터 벡터에 클로닝하여 다양한 세포들에서 프로모터의 활성을 검증하였다. Mut-4.4kb-pOV는 2.8와 5.5kb-pOV에 비해 HeLa, MES-SA, 그리고 LMH/2A에서 높은 수준의 프로모터 활성을 유도하였으나, cEF 세포에서는 낮은 활성을 보였다. 한편, Mut-4.4kb-pOV/pGL4.11 벡터가 도입된 HeLa, MES-SA, LMH/2A, 그리고 cEF 세포에서 에스트로겐 처리에 의한 반응성을 검증한 결과, cEF 세포를 제외한 나머지 세포들은 에스트로겐을 처리했을 때 리포터 유전자의 발현량이 증가하였다. 또한 LMH/2A 세포에 500 nM 에스트로겐 처리 결과, Mut-4.4kb-pOV는 에스트로겐 처리 후에도 2.8와 5.5kb-pOV에 비해 높은 수준의 프로모터 활성을 유도하였다. 더불어 ERE 영역이 없는 2.8kb-pOV는 LMH/2A 세포에서 500 nM 에스트로겐 처리 후 프로모터의 활성에 효과가 없었으나, ERE 영역을 포함하는 5.5 그리고 Mut-4.4kb-pOV는 에스트로겐 처리에 의해 프로모터의 활성이 증가한 결과를 보였다. 이상의 결과는 Mut-4.4kb-pOV가 형질전환 암탉을 생산하기 위한 재조합바이러스 벡터의 프로모터 영역으로 2.8와 5.5kb-pOV에 비해 보다 효율적인 크기임을 보여주는 결과이다.
As a bioreactor, bird has proved to be most efficient system for producing useful therapeutic proteins. More than half of the egg white protein content derives from the ovalbumin gene with four other proteins(lysozyme, ovomucoid, ovomucin and conalbumin) present at levels of 50 milligrams or greater. And the naturally sterile egg also contains egg white protein at high concentration allowing for a long shelf life of recombinant protein without loss in activity. In spite of these advantages, transgenic procedures for the bird have lagged far behind because of its complex process of fertilized egg and developmental differences. Recently, a system to transplant mouse testis cells from a fertile donor male to the seminiferous tubules of an infertile recipient male has been developed. Spermatogenesis is generated from transplanted cells, and recipients are capable of transmitting the donor haplotype to progeny. After transplantation, primitive donor spermatogonia migrate to the basement membrane of recipient seminiferous tubules and begin proliferating. Eventually, these cells establish stable colonies with a characteristic appearance, which expands and produces differentiating germ cells, including mature spermatozoa. Thus, the transplanted cells self-renew and produce progeny that differentiate into fully functional spermatozoa. In this study, to develop an alternative system of germline chimera production that operates via the testes rather than through developing embryos, the spermatogonial stem cell techniques were applied. This system consisted of isolation and in vitro-culture of chicken testicular cells, transfer of in vitro-maintained cells into heterologous testes, production of germline chimeras and confirmation of germline transmission for evaluating production of heterologous, functional spermatozoa.
미세주입법에 의한 형질전환 마우스 제작에는 대량의 전핵기란을 필요로 한다. 본 실험에서는 심플한 형질전환 마우스 제작방법을 확립하기 위해 초급속 동결한 전핵기란을 공시했다. 초급속 동결법으로 동결한 전핵기 체외수정란 139개를 융해 후 공시하고, ${\beta}-actin/luc^+$ 융합유전자를 미세주입하였다. 주입 조작 후, 형태학적으로 정상적인 수정란 101개(72.6%)를 5마리의 수란암컷 마우스에 이식하였다. 그 결과, 이식한 모든 마우스가 임신하고, 최종적으로 15마리(14.8%)의 산자가 태어났다. 한편, 450개의 체외수정란에 대해 동일한 배아조작 후에 338개(75.1%)가 생존하고 14마리의 수란암컷 마우스에 이식 하였다. 그 중에 78%의 수정암컷 마우스가 임신하고, 54마리(19.1%)의 산자가 태어났다. 태어난 산자에 대해서는 southern blot 법에 의해 염색체 내의 도입유전자의 도입을 확인한 결과, 동결수정란 처리구와 체외수정란구에서 각각 6.6%(1/15), 5.5% (3/54)의 마우스에서 도입유전자의 도입이 확인되었다. 더욱이 두 처리구 전부의 형질전환 마우스의 미부조직에서 도입유전자인 루시페라제 유전자의 발현이 관찰되었다. 이상의 결과에 의해 체외수정란 초급속 동결보존법을 사용한 Tg 마우스 제작방법의 확립을 확인하였고, 이러한 결과들로부터 실험기간의 단축과 작업의 간소화에 크게 이바지할 것으로 사료된다.
Gene and cell transfer technique will serve as a powerful tool for the genetic improvement of the poultry and to yield useful products. For avian transgenesis, Japanese quail may serve as an excellent animal model because of its small body size and fast growth rate. Recent progress was described on the manipulation of quail embryos such as the introduction of foreign genes and cells, and the subsequent culturing of the manipulated embryos yielding hatchlings. Intraspecific donor-derived offspring have been available in quail, however, further investigation will be required to obtain interspecific offspring with the aim of rescuing endangered species. Trans genesis will also be useful for improving the profitability and quality of poultry stocks and for developing stocks with novel uses. Considerable progress should soon be made toward the production of transgenic poultry. The key feature of the procedure described here is that embryos are initially taken out from the shell for ease of manipulation and then placed back in culture in addition to various operations midway during culture.
Genetic modification enables modification of target genes or genome structure in livestock and experimental animals. These technologies have not only advanced bioscience but also improved agricultural productivity. To introduce a foreign transgene, the piggyBac transposon element/transposase system could be used for production of transgenic animals and specific target protein-expressing animal cells. In addition, the clustered regularly interspaced short palindromic repeat-CRISPR associated protein 9 (CRISPR-Cas9) system have been utilized to generate chickens with knockout of G0/G1 switch gene 2 (G0S2) and myostatin, which are related to lipid deposition and muscle growth, respectively. These experimental chickens could be the invaluable genetic resources to investigate the regulatory pathways and mechanisms of improvement of economic traits such as fat quantity and growth. The gene-edited animals could also be applicable to the livestock industry.
배자 생식세포 발달에 관련된 메카니즘을 밝혀내기 위해서, 닭 배자 생식기에서 추출한 원시 생식세포의 단백질체 지도를 만들었다. 총 500 배자를 6일간 배양하여 배자 생식기를 획득했고, 7-10일 배양 후, 배양된 원시생식세포는 2차원 젤 전기 영동법에 의해 분할되어 졌다. 유의적 발현 수준을 나타낸 많은 단백질 스팟 들은 MALDI-TOP 와 LC-MS/MS에 의해 확인되었으며, 89개의 단백질 스팟 중에 50개의 mass spectra 들이 데이터베이스에서 조류 단백질과 일치함을 확인하였다. 본 실험에서 행한 단백질체 지도는 형질전환 연구와 생식세포 생물학 분야에 중요한 참고 문헌으로 가치를 가질수 있을 것이다.
