Chloramphenicol acetyl CoA transferase (CAT) and angiotensin- converting enzyme inhibitory peptide (ACEI) were fused to C-terminal region of rice ubiquitin to examine the level of transcripts or enzyme activities in yeast. When two chimeric genes under an inducible Gall promoter control were transformed into Saccharomyces cerevisaie, both CAT and ACE inhibitory activities were enhanced by three to four-fold as compared to those containing no ubiquitin gene. However, the levels of transcripts of ubiquitin fused and un fused genes were not significantly different each other. Therefore, it was suggested that the expression of foreign genes was post-transcriptionally enhanced by fusion of plant ubiquitin in heterologous organisms such as yeast.

Major symptoms of al1ergic rhinitis are nasal obstruction, sneezing and watery rhinorrhea. Onpyetang has been used to treat for nasal obstruction, which is one of the symptoms of allergic rhinitis. This Experimental study was done to research effects of Onpyetang and transformed Onpyetang(Allii Radix is deducted from Onpyetang) on the anti-allergic effects. We have studied the vascular permeability response induced by serotonin and histamine, the contact dermatitis response induced by picryl chloride, the delayed type hypersensitivity response to SRC, the mice paw edema induced by carrageenin, the writhing syndrome induced by $0.7\%$ acetic acid, and the rectal temperature in febrile rats induced by yeast. The results were as follows : 1. In the vascular permeability response to intradermal serotonin and histamine. Onpyetang proved significant inhibitory effect(P<0.05, p<0.001) But transformed Onpyetang proved significant inhibitory effect only to histamine(P<0.05) 2. In the contact dermatitis response induced by picryl chloride, Onpyetang and transformed Onpyetang proved significant inhibitory effect.(P<0.05, P<0.05) 3. In the delayed type hypersensitivity response induced by SRC, Onpyetang and transformed Onpyetang proved significant inhibitory effect on mice paw edema.. (P<0.05, P<0.05) 4. In the delayed type hypersensitivity response induced by SRC, Onpyetang proved significant inhibitory effect to serum IgE.(P<0.01) But transformed Onpyetang proved insignificant inhibitory effect on serum Ig E. 5. In the mice paw edema induced by carrageenin, Onpyetang and transformed Onpyetang proved significant anti-inflammatory effect. (P<0.01, P<0.01) 6. In the writhing syndrome induced $0.7\%$ acetic acid, Onpyetang and transformed Onpyetang proved significant analgestic effect. (P<0.01, P<0.01) 7. In the rectal temperature in febrile rats induced by yeast, Onpyetang and transformed Onpyetang proved significant anti-pyretic effect.(P<0.001, P<0.01) according to this result Onpyetang was conclude to be effective on anti-allergic, anti-pyretic, anti-inflammatory and analgestic action. but transformed Onpyetang(Allii Radix is deducted from Onpyetang) was not effective on the vascular permeability response to intradermal serotonin and increasing Ig E of delayed type hypersensitivity response induced by SRC. In addition, transformed Onpyetang is not effective as Onpyetang. More study should be done about the role of Allii Radix.

A genomic DNA library of the fungus Trametes versicolor was constructed in a yeast integration vector which contains the URA3 gene of the budding yeast Saccharomyces cerevisiae and the gene responsible for hygromycin B resistance, and fragments acting as autonomously replicating sequences (ARSes) in the budding yeast were identified from the genomic DNA library. Sixteen recombinant plasmids from the library transformed the budding yeast Saccharomyces cerevisiae to Ura+ at high frequencies. They were maintained stably under selective conditions, but were gradually lost from yeast cells at different rates under nonselective conditions, indicating that they contain eukaryotic origins of DNA replication and exist as extrachromosomal plasmids. Base sequences of four ARS DNAs among the 16 cloned fragments revealed that all or the four contain at least one 11 bp [(A/T)TTTA(T/C)(A/G)TTT(A/T)]consensus sequence of the budding yeast ARS.
The effect of red ginseng saponins (total saponins, Rbl- and Rgl- fraction of saponins) on the induction of $\beta$-galactosidase in yeast, hccharomyces cereuisiae, was investigated to see that ginseng saponins would penetrate the cell membrane and have a function in a nucleus as steroid hormones do. To attain such a kind of purpose, a DNA fragment (685bp) containing GALI promoter was inserted into the sites of EcoRl and BamHl of polylinker region, upstream of lace gene of the plasmid YEp356 (7.966 Kb), and thus the resulting plasmid pGALl-lacZ is supposed to express $\beta$- galactosidase only in the presence of galactose. The plasmid pGALl -lacZ was introduced into yeast, Ky106 (a leu2 ura3 his3 trp 1 Iys2), and the growth of the transformed cells was much slower in the presence of galactose than glucose. The effects of saponins on the specific activity of P-galactosidase from transformed yeast cells were detected. No significant increase was observed in case of total saponins, but the Rbl- or Rgl- fraction of saponins gave much higher increase in the activity. Maximum increase was observed as 35% in 10-3% of Rbl and as 75% in 10-1% of Rgl. These data suggest that ginseng saponins might be able to enter the nucleus and stimulate transcription. However, further studies to find out the putative saponin receptor are needed to confirm this possibility. Key words : Red ginseng saponin, $\beta$-galactosidase induction, Saccharomyces cerevisiae.
A stuructural gene (ycl) encoding novel yeast cell wall hydrolase, YCL, was cloned from alkalophilic Bacillus alcalophilus subsp. YB380 by PCR, and transformed into E. coli JM83. Based on the N-terminal and internal amino acid sequences of the enzyme, primers were designed for PCr. The positive clone that harbors 1.8 kb of the yeast cell wall hydrolase gene was selected by the colony hybridization method with a PCR fragment as a probe. According to the computer analysis, this gene contained a 400-base-paired N-terminal domain of the enzyme. Based on nucletide homology of the cloned gene, a 850 bp fragment was amplified and the C-terminal domain of the enzyme was sequenced. With a combination of the two sequences, a full nucleotide sequence for YCL was obtained. This gene, ycl, consisted of 1,297 nucleotides with 27 nucleotides with 27 amino acids of signal sequence, 83 redundant amino acids of prosequence, and 265 amino acids of the mature protein. This gene was then cloned into the pJH27 shuttle vector and transformed into the Bacillus subtilis DB104 to express the enzyme. It was confirmed that the expressed cell wall hydrolase that was produced by Bacillus subtilis DB104 was the same as that of the donor strain, by Western blot using polyclonal antibody (IgY) prepared from White Leghorn hen. Purified yeast cell wall hydrolase and expressed recombinant protein showed a single band at the same position in the Western blot analysis.
Using yeast, Saccharomyces cerevisiae, and the integrate vector system, we have isolated and characterized an autonomously replicating sequence (ARS) from Aspergillus nidulans. The DNA fragment, designated ANR1, is 5.0 kb in size and maintained free from the chromosome in S. cerevisiae. The YIplac211-ANR1 recombinant plasmid, which consists of sequences derived from the yeast integrative vector YIplac211 and 5.0 kb ANR1 fragment, showed a 104-fold enhancement in transformation efficiency over that found for YIplac211, and was easily recovered from the transformed yeast. Genetic analysis of transformants showed that YIplac21-ANR1 could be over 96% cured when cultured over 20 generations in complete medium and thus suggests that this sequence is mitotically unstable. In A. nidulans, recombinant plasmid PILJ16-4.5 which carries the 4.5 kb EcoRI fragment of ANR1 showed a 170-fold enhancement in transformation efficiency compared to that of the integrative vector PILJ16.
Using a modified yeast secretory expression vector, $\alpha$-amylase of Schwanniomyces occidentalis was produced from Saccharomyces cerevisiae. The expression vector contains the a-amylase gene (AMY) harboring its own promoter without the regulatory region and the adenine base at the -3 position from the ATG start codon, its own signal sequence, CYC1 transcription terminator, and SV40 enhancer. The expressed $\alpha$-amylase activity from cells carrying the plasmid was approximately 26 times higher than that from the cells harboring an unmodified plasmid. When Saccharomyces diastaticus was transformed with this modified vector, a 2.5 times higher level of amylolytic activity than that from Sch. occidentalis was observed.
The cuclodextrin glucanotransferase (CGTase) gene of Bacillus macerans was subcloned at the downstream of yeast ADH1 promoter, and then the resulting plasmid pVT-CGTM(9.15 kb) was introduced into the yeast host strain, Saccharomyces cerevisias 2805. The transformed yeast, S. cerevisiae 2805/pVT-CGTM, showed the starch-hydrolyzing activity on the starch-azure plate. The optimal conditions for the CGTase expression were found to be 2% dextrose, initial pH5.5, 3$0^{\circ}C$, and 48hr cultivation. Under this condition, the extracellular CGTase activity reached at 0.53 U/mL, whereas the intracellular activity was about 0.03U/mL. This result indicates that the signal peptide of Bacillus CGTase functioned well in S. cerevisiae.
We have previously reported recombinant productions of bacteriocins using yeast expression plasmid pAUR123, which contains the alcohol dehydrogenase (ADH) promoter, in Saccharomyces cerevisiae cells and their antibacterial activities. In order to improve the antibacterial activities of bacteriocidal yeast cells, a strong glyceraldehyde phosphate dehydrogenase (GPD) promoter gene of S. cerevisiae was amplified and inserted upstream into bacteriocin genes such as the OR-7, Subpeptin JM4-A or JM4-B gene in the corresponding recombinant yeast plasmid. Yeast cells transformed by the recombinant plasmid containing the GPD promoter represented higher antibacterial activities against both Gram positive B. subtilis and Gram negative E. coli cells compared to those transformed by the corresponding recombinant plasmid containing the ADH promoter. Thus, yeast cells harboring the recombinant plasmid containing the GPD promoter constructed in this study could be applied in the food preservative or animal feed industries.

Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic reaction follows a ping-pong mechanism in which the enzyme is transiently phosphorylated on a histidine residue conserved in all nucleoside diphosphate kinases. Beside their role in nucleotide synthesis, these enzymes present additional functions, possibly independent of catalysis, in processes such as differentiation, cell growth, tumor progression, metastasis and development. To clone murine nm23-M5, several expressed sequence tags (ESTs) of the GenBank data base, selected according to their homology to nm23-H5 cDNA, reconstituted a complete open reading frame (GenBank AF222750). To test whether murine NDPKs (1, 2, 3, 4, 5, and 6) can inhibit Bax-mediated toxicity in yeast, co-transformation was performed respectively. The yeast S.cerevisiae was transformed with a copy expression plasmid containing the histidine selection marker and expressing murine Bax under the control of a galactose-inducible promoter. Several clones were selected and found to be growth inhibited when Bax expression was induced with galactose. A representative clone was transformed again with a copy expression plasmid containing the tryptophane selection marker and expressing either murine Bcl-xL or NDPK under the control of a galactose-inducible promoter. Several subclones of the double-transformants were selected and characterized. The ability of Bcl-xL and NDPKs to suppress Bax-mediated toxicity was determined by growing yeast cells overnight in galactose media and spot-testing on galactose plates starting with an equal number of yeast cells as determined by taking the OD$_{600}$. Ten-fold serial dilutions were used in the spot-test. Plates were grown at 3$0^{\circ}C$ for 2-3 days. All murine NDPKs suppressed Bax dependent apoptosis. Futher study will be peformed whether Bax-toxicity inhibition was caused by NDP kinase activity or additional function.n.
