Busan New Port, under construction aiming for the hub of Northeast Asia and Partly in operation, had damaged up to 48 billion Won due to Typhoon 'maemi' in 2003. The present criteria of domestic harbor design only describes about the critical wave height with respect to the size of vessel for harbor tranquility. The berth operation ratio which represents the annual available berthing days is depending on the efficiency of cargo handling work and this depends on the motion of the moored vessel due to the wave action and the characteristics of cargo gears. The motion of moored vessel might be related not only to the wave height but also to wave period. Furthermore, the berth operation ratio relies on external forces such as currents and winds, including the characteristics of mooring system and the specification of the moored vessel. In this study we only deal with berth operation ratio in normal sea state, considering wave and current by measured data and numerical calculation. Especially we tried to evaluate the berth operation ratio for each berth adopting the variation of dredging and reclamation plan and the change of wave environment during the process of the new port construction. For better understanding and analysis of wave transformation process, we applied the steady state spectral wave model and extended mild-slope wave model to the related site. This study summarizes comparisons of harbor responses predicted by two numerical predictions obtained at Busan New port site. Field and numerical model analysis was conducted for the original port plan and the final corrected plan.

Hybrid ceramic membrane (HCM) processes that combined ozonation with a ceramic membrane (CM) or a reactive ceramic membrane (RM), an iron oxide nanoparticles (IONs) incorporated-CM were investigated for membrane fouling control. Alumina disc type microfiltration and ultrafiltration membranes doped with IONs by sintering method were tested under varying mass fraction of IONs. Scanning electron microscope (SEM) images showed that IONs were well-doped on the CM surface and doped IONs were approximately 50 nm in size. Change in the pure water permeability of RM was negligible compared to that of CM. These results indicate that IONs incorporation onto CM had little effect on CM performance in terms of the flux. Natural organic matter (NOM) fouling and fouling recovery patterns during HCM processes confirmed that the RM-ozonation process enhanced the destruction of NOM and reduced the extent of fouling more than the CM-ozonation process by hydroxyl radical formation in the presence of IONs on RM. In addition, analyses of NOM in the feed water and the permeate showed that the efficiency of membrane fouling control results from the NOM degradation during HCM processes; leading to removal and transformation of relatively high contents of aromatic, high molecular weight and hydrophobic NOM fractions.
Ti plasmid of A. tumefaciens was labeled with $^3H-thymidine$, purified and encapsulated into phosphatidylserine (PS) and PS-cholesterol (Chol; 1 : 1 molar ratio) liposomes by lyophilization-rehydration method. PS was supplemented with 1 mole percent octadecyl rhodamine B for fluorometric measurement of PS. Liposomes entrapping $^3H-Ti plasmid$ were fused with Nicotiana sanderae protoplasts by treating with 5 mM $CaCl_2$ and 10% PEG. The fusion was evidenced by fluorescence microscopic technique. The amounts of Ti plasmid and PS associated with protoplasts were assayed by the radioactivity of $^3H-Ti plasmid$ and by the fluorescence of rhodamine B. About 7.9% of the PS liposome and 7.2% of PS-Chol liposome were fused with protoplasts. During the fusion process, about 30% of the liposomal contents of PS-Chol liposome was leaked, in contrast to about 60% leakage of its contents in PS liposome. Accounting the number of liposomes fused with protoplasts together with the encapsulation efficiency and the leakage of liposomal contents, it was calculated that ca. 1,700 Ti plasmid was transfered into one protoplast by the present method. This result may indicates that the present method transfers enough Ti plasmid into plant protoplast to elicit genetic transformation of plants.

With the development of economy and improving of people's living standards, people dietary needs will be achieved from subsistence to high nutrition and from high nutrition to healthy transformation. Mushroom as a kind of highly nutritious, low fat, rich vitamin food has a great interest among the people. This makes the mushroom into a new sunrise industry and it gradually from pure manual cultivation develops toward the fully automatic factory. In the process of mushroom factory production, regulation of environmental factors directly affects the yield and quality of mushroom. In related to the methods of mushroom cultivation, the recent technologies apply the new technology such as sensors and IT convergence services. And then cultivating mushroom is managed effectively. This paper in order to solve the above problems and construct an effective mushroom growth environment using technology such as humidity sensor construct an environment that can automatically adjust the humidity. This environment has important significance to improve the level of automation mushroom production, increase yield per unit area and quality of mushroom, increase economic efficiency of mushroom production, and enhance the competitiveness of mushroom production.
In this study we tried to transform an antimicrobial peptide gene (Shiva) under the promoter of tomato phenylalanine ammonia-lyase (tPAL5) into tobacco and potato plants. Antimicrobial peptide gene was isolated originally from giant silk moth (Hyalophora cecropia) and modified ie nucleotide sequence to increase antimicrobial activity. Transgenic tobacco plants were regenerated and their seeds were tested on the media containing kanamycin (500 mg/L). The results of PCR amplification and genomic Southern blot hybridization confirmed the integration of construct (tPAL5 promoter-Shiva-NOS-GUS-NOS) into chromosome. We observed that one of the transgenic tobacco plants showed chromosome rearrangement when integrated. In case of potato transformation, the efficiency of regeneration was maximized at the medium containing Zeatin 2mg/L, NAA 0.01mg/L, GA$_3$ 0.1mg/L. We also observed the high expression of GUS (${\beta}$-glucuronidase) enzyme which was located next to the terminator sequence of nopaline synthase gene (NOS) in the vascular tissue of stem, leaves of transgenic potatoes. This result suggested that a short sequence of Shiva gene (120 bp) and NOS terminator sequence might be served as a leader sequence of transcript when translated.
This study was carried out to investigate activity difference in the superoxide dismutase (SOD) and peroxidase (POD) of tomato callus transformed with Agrobacterium containing the GUS gene. Than those of other two tomato cultivars, the hypocotyl explant of JA101 was shown to have higher POD and SOD specific activity of 23 unit/mg protein and 2,156 unit/mg protein, respectively. Relatively high frequency of callus formation was obtained from the hypocotyl explant on MS medium containing 1 mg/L 2,4-D for 30 days and its POD(47 unit/mg protein) and SOD (95,786 unit/mg protein) specific activities were higher than other 2,4-D concentration. The hypocotyl explant and callus cocultivated with Agrobacterium for 72 hours were transferred to MS medium supplemented with 1 mg/L 2,4-D, 30 mg/L kanamycin, 30 g/L sucrose and 4 g/L Gelrite. The hypocotyl explants transferred to the medium formed callus with 45.5% effeciency after 8 weeks. The transformation efficiency confirmed by GUS assay was 21.6%. POD specific activity of the transformed callus (54 unit/mg protein) were somewhat lower than the non-transformed callus (64 unit/mgg protein) and SOD specific activity of the transformed callus (30,300 unit/mg protein) were also lower than the non-transformed callus (37,077 unit/mg protein). However there was no significant difference in POD and SOD isozyme patterns between the transformed and the non-transformed calluses. From these results, it revealed that there was no difference of antioxidant enzyme activities between the transformed callus and the non-transformed callus in tomato.
The bialaphos is a potent inhibitor of glutamine synthease in higher plants and is used as a non-selective herbicide. We have used the bialaphos resistant gene(Bar) encoding for an acetyltransferase isolated from Streptomyces hygroscopicus SF1293. Callus derived from mature seeds of rice(Oryza sativa L. cv. Dong Jin) were co-cultivated with Agrobacterium tumefaciens EHA101 carring a plasmid pGPTV-HB containing genes for hygromycin resistance (HygR) and Bar. Transgenic plants showing in vitro resistance to 50 mg/L hygromycin and 10 mg/L bialaphos were obtained by using a two-step selection/regeneration procedure. Transformation efficiency of rice was about 30% which was as high as reported in other dicotyledons. Progenies ($\textrm{T}_{1}$ generation) derived from primary transformant of 17 lines were segregated with a 3 resistant : 1 sensitive ratio in medium containing hygromycin and bialaphos. Stable integration of Bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from $\textrm{T}_{2}$ progenies. Transgenic plants ($\textrm{T}_{3}$) grown in the field were resistant to bialaphos (Basta) at a dosage lethal to wild type plants.
This study is kinematics for the manipulator development of cucumber harvesting. A theory value was verified by repeated error measurement after the forward kinematics or inverse kinematics analysis of manipulator. Manipulator is consisted of one perpendicular link and two revolution link. The transformation of manipulator can be valued by kinematics using Denavit-Hartenberg parameter. The value of inverse kinematics which is solved by three angles faction shows two types. Repeated errors refered maximum 2.60 mm, 2.05mm and 1.55 mm according to X, Y, Z axis. In this study, the actual coordinates of maximum point and minimum point were agreement in the forward kinematics or inverse kinematics. The results of repeated error measurement were reflect to be smaller compared to a diameter of cucumber. measurement errors were determined by experimented errors during the test. For reducing errors of manipulator and improving work efficiency, the number of link should be reduced and breeding and cultural environment should be considered to reduce the weight and use the hard stuff. The velocity of motor for working should be considered, too.

In this study, for conducting a static load test of an external fuel tank used for an aircraft, the flight load acting on the external fuel tank was converted to the test load and the suitability of the converted test loads was confirmed. In order to calculate the test load from the flight load, the external fuel tank was divided into several sections. Shear load, moment by unit shear load, and unit moment were calculated for each section. Test loads for each section were then calculated by computing the shear load, the moment of each section, and flight load condition. In actual static load tests, it might not be possible to impose the test load in the calculated position due to physical constraints. Therefore, after determining positions in which the load could be imposed in the actual test, the test load calculated for each section was redistributed to selected positions. Finally, a test load plan was established by applying a whiffle tree to enhance the efficiency of the test performance while making it easier to operate the actuator. The reliability of the test load plan was verified by comparing it with flight load conditions.

As the 4th industrial revolution is in full swing and next-generation ICT(Information & Communications Technology) convergence technology is being developed, various smart construction technologies are being rapidly introduced in the construction field to respond to technological changes. In particular, since the earth-volume calculation process for site design accounts for a large part of the design cost at the construction site, related researches are being actively conducted to improve the efficiency of the process and accurately calculate the earth-volume. The purpose of this study is to present a method for quickly constructing the topography of a construction site in 3D and efficiently calculating earth-volume using the results. For this purpose, the construction site was constructed as a 3D realistic model using large-scale aerial photos obtained from UAV(Unmanned Aerial Vehicle). At this time, since the constructed 3D realistic model has a surface model structure in which volume calculation is impossible, the structure was converted into a 3D solid model to enable volume calculation. And we devised a methodology to calculate earth-volume based on CAD(Computer-Aided Design and Drafting) using the converted solid model. Automatically calculating earth-volume from the solid model by applying the method. As a result, It was possible to confirm a relative deviation of 1.52% from the calculated earth-volume from the existing survey results. In addition, as a result of comparative analysis of the process time required for each method, it was confirmed that the time required is reduced of 60%. The technique presented in this study is expected to be utilized as a technology for smart construction management, such as periodic site monitoring throughout the entire construction process, as well as cost reduction for earth-volume calculation.
