  • Welding is one of the most important joining processes and the effect of welding residual stresses in the structure has a great deal of influence on its quality. In this paper, recent development in computational welding mechanics, particularly calculation of welding residual stresses, is introduced. The hypoelastic formulation of finite element analysis for thermoelastic-plastic deformation is applied to welding processes to find residual deformations and stresses. Leblond's phase evolution equation coupled with the energy equation is employed to calculate the phase volume fraction; this plays an important role as a kinetics parameter affecting phase fraction effects in the mechanical constitutive equation of welded materials. Furthermore, transformation plasticity is taken into account for an accurate evaluation of stress. The influence of the phase transformation and the transformation plasticity on residual stress is investigated by means of numerical analyses using metallurgical parameters in Leblond's phase evolution equation that are adjusted with respect to various cooling rates in a CCT-diagram. Coding implementation is conducted by way of the ABAQUS user subroutines, UMAT.

  • Purpose: The Malaysia Education Blueprint is primarily concerned with the transformation of students' minds through the curriculum offered at the school level (2013-2025). Diversity in the application of teaching and learning methods is one means of achieving the transformation of students' minds through the Secondary School Standard Curriculum. Consequently, the production of ExSheetLink's Module for Accounting Education is the primary outcome of this study, which had three objectives: the need for ExSheetLink's Module in the process of producing financial statements for Accounting Students in secondary school to the Accounting Teacher; and the design of ExSheetLink's Module that meets the entire process in the production of financial statements for Accounting Students in secondary school based on the Documents Curriculum and the Accounting Students' needs. Research design, data and methodology: This study outlines the research framework for module development in accordance with the Design and Development Research Method, which combines multiple research techniques (Mixed Method). Results: The development of ExSheetLink's Module is completed and can be used for the level of effectiveness purposes. Conclusion: The transformation of Accounting Students' minds is a success thanks to the ExSheetLink Module. Researchers also suggested that all Malaysian Secondary School accounting students test the ExSheetLink Module.

A Study on Color Management of Input and Output Device in Electronic Publishing (I) (전자출판에서 입.출력 장치의 컬러 관리에 관한 연구 (I))

  • In this paper, an experiment was done where the input device used the linear multiple regression and the sRGB color space to perform a color transformation. The output device used the GOG, GOGO and sRGB for the color transformation. After the input device underwent a color transformation, a $3\;{\times}\;20\;size$ matrix was used in a linear multiple regression and the scanner's color representation of scanner was better than a digital still camera's color representation. When using the sRGB color space, the original copy and the output copy had a color difference of 11. Therefore it was more efficient to use the linear multiple regression method than using the sRGB color space. After the input device underwent a color transformation, the additivity of the LCD monitor's R, G and B signal value improved and therefore the error in the linear formula transformation decreased. From this change, the LCD monitor with the GOG model applied to the color transformation became better than LCD monitors with other models applied to the color transformation. Also, the color difference varied more than 11 from the original target in CRT and LCD monitors when a sRGB color transformation was done in restricted conditions.

Transformation of an Alkalin Protease Overproducer, Vibrio metschnikovii Strain RH530, and Improvement of Plasmid Stability by the par Locus

  • Vibrio metschnikovii strain RH530 is a non-pathogenic, industrially-important alkaline protease producer which has been isolated from wastewater. In this paper, we report on the transformation of this strain by using the method of electroporation. A field strength of $7.5\;kVcm^{-1}$ and $25\;{\mu}F$, and using a 0.2-cm cuvette, appeared to be the optimal conditions for electroporation of the cells with the recombinant pSBCm plasmid carrying the vapK alkaline protease gene and the ColE1 replicon. Cells were subjected to osmotic shock in order to remove extracelluar DNase, and adding 200 mM of sucrose to electroporation buffer cells showed an increased transformation efficiency. Maximum efficiency of transformation was obtained at an early exponential growth phase. Using all of the conditions mentioned above, we routinely obtained a transformation efficiency of more than $10^4{({\mu}g\;plasmid\;DNA)}^{-1}$. The stability of the plasmid pSBCm in V. metschnikovii RH530 was 25% after 18h of growth (27 generations) in the medium without antibiotic selection. The insertion of the par locus to the pSBCm increased the stability of the plasmid up to 42% without selective pressure. The increase in plasmid stability was accompanied by the increase in the productivity of alkaline protease in the recombinant V. metschnikovii strain RH530. Determining optimal conditions for the transformation of the industrially-important, nonpathogenic Vibrio strain, and the improvement of plasmid stability by introducing the par locus into the high copy number plasmid vector, will allow the development of procedures involved in the genetic manipulation of this strain, particularly for its use in the production of industrial enzymes such as alkaline protease.

  • Wheat is one of the world's top three crops and is an important staple crop, accounting for 20% of the nutrient calories consumed by the world's population. However, due to its complex heterogeneous hexaploid chromosomes and vast genome of approximately 16 Gb, compared to those of other crops, molecular biology and biotechnology studies on wheat are lacking. In recent years, wheat genome analysis has been performed using the latest next-generation sequencing technology so that useful genes can be easily obtained, and wheat biotechnology research is accelerating in various fields. In this review, wheat transformation, an indispensable technique for developing new functional biotech wheat by revealing the function of wheat genes, is described in detail. In addition, the latest research results for overcoming plant diseases, abiotic stresses, and wheat-related diseases that are difficult to solve by classical breeding through wheat transformation and biotechnology are described.

  • Integration is an important intellectual ability to reconfigure several separated elements into one unified form and to concisely and categorically express them. In this paper, we classify the types of integration in C language, and propose a systematic and incremental method to transform to better code based on the integration types. This transformation method could also be used as a means to improve integrated thinking and efficiently learn C language, and will improve development ability in C programming or other language programming.

Effect of Reverse Transformation on the Microstructure and Retained Austenite Formation of 0.14C-6.SMn Alloy Steel (0.14C-6.5Mn 합금강의 미세조직과 잔류오스테나이트 형성에 미치는 역변태처리의 영향)

  • The present study aimed to develop the TRIP(transformation induced plasticity) aided high strength low carbon steel sheets using reverse transformation process. The cold-rolled 0.14C-6.5Mn steel was reverse-transformed by slow heating to intercritical temperature region and air cooling to room temperature. An excellant combination of tensile strength and elongation of $98.3kgf/mm^2$ and 44.4% appears. This combination comes from TRIP phenomena of retained austenite during deformation. The stability of retained austenite Is very Important for the good ductility and it depends on diffusion of carbon and manganese during reverse transformation. The air cooling after holding at intercritical temperature retards the formation of pearlite and provides the carbon enrichment in retained austenite, resulting the increase of elongation in cold-roiled TRIP steel.

Matrix Attachment Regions (MARs) as a Transformation Booster in Recalcitrant Plant Species

  • For genetic engineering to be commercially viable, an efficient transformation system is needed to produce transgenic plane from diverse genotypes ("generalized protocol"). Development of such a system requires optimization of a number of components such as gene transfer agent, plant tissues competent for both regeneration and transformation, and control of transgene expression. Although several novel gene transfer methods have been developed for plane, a majority of stably transformed plane express the introduced genes at low levels. Moreover, silencing of selectable marker genes shortly after their incorporation into plant chromosomes may result in low recovery of transgenic tissues from selection. Matrix attachment regions (MARs) are DNA sequences that bind to the cell's proteinaceous nuclear matrix to form DNA loop domains. MARs have been shown to increase transgene expression in tobacco cells, and reduce position in mature transgenic plants. Flanking an antibiotic resistance transgene with MARs should therefore lead to improved rates of transformation in a diversity of species, and may permit recalcitrant species and genotypes to be successfully transformed. Literature review and recent data from my laboratory suggest that MARs can serve as a transformation booster in recalcitrant plant species.

Development of Transgenic Orchardgrass (Dactylis glomerata L.) Using Agrobacterium tumefaciens

  • To produce of transgenic orchardgrass, the seed-derived calli of orchardgrass (Dactylis glomerata L.) co-cultivated with Agrobacterium turnefaciens EHAlOl harboring binary vector pIG121-Hm were selected with hygromycin and then transferred onto N6 regeneration medium containing 1 rngl l of NAA, 5 rngl l of kinetin, 250 rngl l of carbenicillin and 50 mg/ l of hygromycin. The efficiency of transformation was differed on cultivars, that is, 'Potomac' appeared 12% of transformation efficiency while 'Amba' did 5.5%. The addition of acetosyringone during co-cultivation was a key to successhl transformation of orchardgrass. Transgene fragments were identified by PCR analysis and the constitutive expression of GUS gene was confirmed by Northern blot analysis.   • Purpose: The purpose of this study is to examine the characteristics and problems of existing research through various literature investigations on the existing R&D process. Implications are identified through the investigation of existing studies and the characteristics of the digital transformation era are analyzed. We research and propose a new R&D process suitable for the new era of digital transformation. Methods: The research method investigates various literature precedent studies on the existing R&D process and the methods used by companies. Research new R&D processes that reflect the characteristics of the digital transformation era and develop new related models. Results: In the era of digital transformation, the parallel R&D process is being reorganized to respond quickly to the demands of rapid market changes. In order to respond quickly to various customers, the R&D process can respond flexibly in a parallel structure. Conclusion: Along with the development of the R&D process, additional research on indicators and evaluation models that can evaluate them fairly and flexibly is needed. Further research is needed to determine whether the R&D process suitable for the digital transformation era is operating properly.
