Optimization of Polyethylene Glycol-Mediated Transformation of the Pepper Anthracnose Pathogen Colletotrichum scovillei to Develop an Applied Genomics Approach

  • Colletotrichum acutatum is a species complex responsible for anthracnose disease in a wide range of host plants. Strain C. acutatum KC05, which was previously isolated from an infected pepper in Gangwon Province of South Korea, was reidentified as C. scovillei using combined sequence analyses of multiple genes. As a prerequisite for understanding the pathogenic development of the pepper anthracnose pathogen, we optimized the transformation system of C. scovillei KC05. Protoplast generation from young hyphae of KC05 was optimal in an enzymatic digestion using a combined treatment of 2% lysing enzyme and 0.8% driselase in 1 M NH4Cl for 3 h incubation. Prolonged incubation for more than 3 h decreased protoplast yields. Protoplast growth of KC05 was completely inhibited for 4 days on regeneration media containing 200 ㎍/ml hygromycin B, indicating the viability of this antibiotic as a selection marker. To evaluate transformation efficiency, we tested polyethylene glycol-mediated protoplast transformation of KC05 using 19 different loci found throughout 10 (of 27) scaffolds, covering approximately 84.1% of the entire genome. PCR screening showed that the average transformation efficiency was about 17.1% per 100 colonies. Southern blot analyses revealed that at least one transformant per locus had single copy integration of PCR-screened positive transformants. Our results provide valuable information for a functional genomics approach to the pepper anthracnose pathogen C. scovillei.

Development of Transient Expression System Using Transformed Seedlings of Brassica napus var. napus (유채유묘의 형질전환을 통한 일시발현시스템의 개발)

  • For molecular breeding purpose, genetic transformation of Brassica napus cultivars has been extensively performed using Agrobacterium method. B. napus cv. napus, one of major oil crops, can be transformed via Agrobacterium-based method. We demonstrated that Agrobacterium-mediated transformation via vacuum infiltration slightly worked for the seedlings of B. napus cv. napus according to fluorometric GUS enzyme analysis. In contrast, transformation efficiency was highly enhanced when the seedlings, prior to agroinfiltration, were treated with sodium hydrosulfite solution as a chemical wounding agent. GUS gene expression in transformed seedlings that was confirmed by RT-PCR suggests their usefulness for the development of transient expression system.

  • This paper presents an automated API translator for embedded software development based on a model-driven approach. Since MDA(Model Driven Architecture) provides little support for the development of embedded software, we propose a new approach containing its advantages. First, we define #generic APIs# which do not depend on any RTOS#s but provide most of typical RTOS services. We can describe RTOS-related behaviors of target application using these generic APIs in a CIC(Common Intermediate Code). Then, we propose a transformation tool for translating between a CIC using generic APIs and a C-code for specific RTOS. The proposed API translator converts them using XML transformation rule which is defined outside. It indicates that an API translator extends to other RTOS#s by modifying or adding the transformation rule. From the experiment. we validate the proposed method.

  • The paradigm of rural development projects has shifted and expanded from agricultural production based on the way of improving the residential environments in order to achieve the quality of life staying in rural areas. The purpose of this study is to evaluate policy directions of rural development projects by investigating government projects conducted successively starting from the Participatory Government of President Roh in 2003 and to the present Government of President Moon. In consideration of administrative affairs, the projects include the Balanced National Development Project, Quality of Life Improvement and Regional Development for Rural Villages Projects. The study examined changes in visions and key agendas proposed in national affairs as well as those in the visions and strategies exhibited by local development project plans through various governments. The core contents examined are the project characteristics, transformation process through government changes, and the law of status pertaining to rural development projects. The direction of implementation for the rural development projects implemented by each government provide an opportunity of understanding the transformation process in each project that has been still effective currently. This study is expected to provide a basic set of data to be used for a proposal for new rural development projects while still being useful to be referred or integrated to other national and regional projects.

Assessment of Factors Influencing Agrobacterium Mediated Transformation in Cucumber (Cucumis sativus L)

  • Five day old cotyledon explants of Cucumber (Cucumis sativus L) cv Poinsett 76 were cocultivated with two Agrobacterium strains (EHA105 and LBA 4404) each carrying GUS as the reporter gene and npt-II as the selection marker gene in the T-DNA region of the vector. Transformed shoots were selected at 150 mg/L kanamycin. A two day cocultivation coupled with $20\;{\mu}M$ acetosyringone increased the frequency (8.2 and 15.4 shoots) of GUS expression in the shoots of transformed plant. Among the two Agrobacterium strains, EHA 105 performed better than LBA 4404 in bringing two-fold increase in transformation efficiency (14%) than LBA 4404 (7.4%). PCR analysis was done to confirm the integration of T-DNA into cucumber genome.

  • Creeping bentgrass (Agrostis stolonifera L.) is economically important as the principal turfgrass species for golf course greens and fairways in temperate climates around the world. As the utilization area of the turfgrass species increases recently, the demand for new and improved cultivars increases. Thus, substantial progress has been made in applying modern biotechnology to develop genetically engineered (i.e., biotech) creeping bentgrass with new traits that eluded the breeders. This review article addresses the advances made in developing biotech creeping bentgrass, which are categorized in the following topics: (i) genetic transformation of creeping bentgrass, (ii) development of various biotech creeping bentgrasses by genetic engineering, and (iii) progresses in the deregulation of herbicideresistant creeping bentgrass.

  • The term color management system design, an ensemble of algorithm that provides a framework in which color information can be processed consistently throughout a digital imaging system. This is most commonly achieved through the use of special color transformation, known as device independent color transformation based on ICC device profiles. The purpose of this paper is to present some of the scientific principles of color management, and the original color management algorithms and solutions for digital soft color proofing system development.

  • The development of human civilization has been accompanied by a corresponding development of storage facilities. In particular, the storage of residential spaces not only performs the storage management function for personal property but also is considered an important prerequisite for a comfortable life. While examining the history of the transformation of built-in furniture, this study emphasizes the social and cultural implications of Korean residences. Therefore, We attempt to uncover the Korean characteristics that can be discovered in the transformation of storage furniture according to the changes that occurred in Korean residential forms following the modern era and to question the problematic issues of the built-in furniture in Korea.

Practical Algorithms on Lunar Reference Frame Transformations for Korea Pathfinder Lunar Orbiter Flight Operation

  • This technical paper deals the practical transformation algorithms between several lunar reference frames which will be used for Korea pathfinder lunar orbiter (KPLO) flight operation. Despite of various lunar reference frame definitions already exist, use of a common transformation algorithm while establishing lunar reference frame is very important for all members related to KPLO mission. This is because use of slight different parameters during frame transformation may result significant misleading while reprocessing data based on KPLO flight dynamics. Therefore, details of practical transformation algorithms for the KPLO mission specific lunar reference frames is presented with step by step implementation procedures. Examples of transformation results are also presented to support KPLO flight dynamics data user community which is expected to give practical guidelines while post processing the data as their needs. With this technical paper, common understandings of reference frames that will be used throughout not only the KPLO flight operation but also science data reprocessing can be established. It is expected to eliminate, or at least minimize, unnecessary confusion among all of the KPLO mission members including: Korea Aerospace Research Institute (KARI), National Aeronautics and Space Administration (NASA) as well as other organizations participating in KPLO payload development and operation, or further lunar science community world-wide who are interested in KPLO science data post processing.
