This study was carried out to examine the proportion of fatty acids and contents of trans fatty acid in commercial processed foods in Jeon-Buk area. Contents of trans fatty acid in bakery products ranged from 0.2% to 1.0% per 100 g food. Especially garlic bread and pastry contained the highest amount. Popcorns contained trans fatty acid from 0.0% to 13.4%. Among them, instant popcorns for microwave contained the highest amount, while trans fatty acid was hardly detected in popcorns for theater. And trans fatty acid detected in popcorns was mostly in the form of 18 : 1 trans. The content of trans fatty acid in pizza was nearly 0.2%. Amounts of saturated fatty acid and unsaturated fatty acid were similar between each type of pizza and frozen pizza. Fritters such as fried chickens and vegetables contained trans fatty acid less than 0.2% per 100 g food.
Cis-trans forms of fatty acids in cigarette smoke condensate were separated and characterized using SP 2340 glass capillary gas chromatography. The transforms of fatty acids, palmitoeladic , 1-vaccenic, eladic and linoeladic acid were identified. These components were not present only 3% of the total fatty acid fraction, but they showed a very low transfer rate.
In this study, we determined lipid content, total fatty acid composition, trans fatty acid(tFA) content, and acid value of twenty black-bean-sauce noodles collected in Seoul city area. Total lipid contents of twenty samples were determined to be 3.33~9.23% on wet base. Total unsaturated fatty acids were from 47.83% to 83.18%(mainly oleic and linoleic acid). Total saturated fatty acids ranged from 16.06% to 51.48%. Besides, tFA contents in total lipid of samples were ranged from 0.38% to 1.39%, equivalent to 20 to 80mg per 100g of black-bean-sauce noodles. Acid values of lipids extracted from samples ranged from 0.56 to 2.88.

The consumption of foods containing trans fatty acids (TFAs) is a matter of concern at present. According to many studies, trans fatty acids (TFAs) may cause illnesses such as the coronary heart disease, diabetes mellitus, large intestine cancer, and breast cancer. They can also raise low density lipoprotein (LDL) cholesterol and reduce high density lipoprotein (HDL) cholesterol. TFAs can also inhibit the synthesis of phospholipids containing polyunsaturated fatty acids in arterial cells. As a consequence the Food and Drug Administration has deemed that saturated fatty acid, cholesterol and trans fatty acid levels be listed on food labels as of 2006. The Korea Food and Drug Administration also has required the listing of trans fatty acid content on food labels since 2007. The aim of this study was to determine the total lipid and trans fatty acid (TFA) contents in retort food, powdered milk, biscuit and pizza products. The number of samples examined were 2 retort food, 6 powdered milk, 7 biscuit and 3 pizza products. The extraction of total lipids in retort food and powdered milk followed the chloroform methanol method. The extraction of total lipids in biscuit and pizza was by the acid digestion method. All samples were analyzed by gas chromatography (GC) using a SP-2560 capillary column and a flame ionization detector. The TFA contents per 100g of sample were 1-2.8% (1.9%) in retort foods, 0.4-2.4% (1.37%) in powdered milk products, 0-2.9% (1.23%) in biscuits, and 2.8-3.45% (3.03%) in pizzas.
Objective: Caseins and fatty acids of milk are synthesized and secreted by the epithelial cells of the mammary gland. All-trans retinoic acid (ATRA), an active metabolite of vitamin A, has been shown to promote mammary development. This study was conducted to determine the effect of ATRA on casein synthesis and fatty acid composition in MAC-T cells. Methods: MAC-T cells were allowed to differentiate for 4 d, treated with ATRA (0, 1.0, 1.5, and 2.0 μM), and incubated for 3 d. We analyzed the fatty acid composition, the mRNA expression of casein and fatty acid synthesis-related genes, and the phosphorylation of casein synthesis-related proteins of MAC-T cells by gas chromatography, quantitative polymerase chain reaction, and western blotting, respectively. Results: In MAC-T cells, ATRA increased the mRNA levels of αS1-casein and β-casein, janus kinase 2 (JAK2) and E74-like factor 5 of the signal transducer and activator of transcription 5 β (STAT5-β) pathway, ribosomal protein S6 kinase beta-1 (S6K1) and eukaryotic translation initiation factor 4E binding protein 1 of the mammalian target of rapamycin (mTOR) pathway, inhibited the mRNA expression of phosphoinositide 3-kinase and eukaryotic initiation factor 4E of the mTOR pathway, and promoted the phosphorylation of STAT5-β and S6K1 proteins. Additionally, ATRA increased the de novo synthesis of fatty acids, reduced the content of long-chain fatty acids, the ratio of monounsaturated fatty acids to saturated fatty acids (SFA), the ratio of polyunsaturated fatty acids (PUFA) to SFA, and the ratio of ω-6 to ω-3 PUFA. The mRNA levels of acetyl-CoA carboxylase 1, fatty acid synthase, lipoprotein lipase, stearoyl-CoA desaturase, peroxisome proliferator-activated receptor gamma, and sterol regulatory element-binding protein 1 (SREBP1) were enhanced by ATRA. Conclusion: ATRA promotes the synthesis of casein by regulating JAK2/STAT5 pathway and downstream mTOR signaling pathway, and it improves the fatty acid composition of MAC-T cells by regulating SREBP1-related genes.
An in vitro study was conducted to determine the effect of C18-polyunsaturated fatty acid on direct incorporation into the rumen bacteria, bio-hydrogenation and production of CLA in vitro. Sixty milligrams of linoleic acid ($C_{18:2}$) or linolenic acid ($C_{18:3}$) were absorbed into the 0.5 g cellulose powder was added to the 150 ml culture solution consisting of 120 ml McDougall's buffer and 30 ml strained rumen fluid. Four uCi of 1-$^{14}C_{18:2}$ or 1-$^{14}C_{18:3}$ (1 uCi/15 mg each fatty acid) were also added to the corresponding fatty acids to estimate the direct incorporation into the bacterial lipids. The culture solution was then incubated anaerobically in a culture jar with stirrer at 39$^{\circ}C$ for 12 h. Ammonia concentration and pH of the culture solution were slightly influenced by the fatty acids. Amount of fatty acid incorporated into the bacteria was 1.20 mg and 0.43 mg/30 ml rumen fluid for $C_{18:2}$ and $C_{18:3}$, respectively during 12 h incubation. Slightly increased CLA (sum of cis-9, trans-11 and cis-10, trans-12 $C_{18:2}$) was obtained from the $C_{18:3}$ addition compared to that from $C_{18:2}$ after 12 h incubation in vitro.
Although negative evidences of trans fatty acids(tFAs) are gradually increasing, the hypercholesterolemic effect of tFAs are controversial and its biological significances are still not known. The objective of this study was to examine the effect of dietary tFAs on cholesterol metabolism and membrane fluidity in rats. Animals were fed diets containing 0.5% cholesterol and 20% test fats(margarine, butter, corn oil) for 8 weeks. Each test fats(margarine, butter, corn oil) contained 25%, 8.5% and no tFAs, respectively, Plasma total triglyceride(TG) were not different among diet treatments. Feeding trans fat diets (margarine and butter) lowered plasma cholesterol. Specially, butter diet elevated LDL-cholesterol and decreased HDL-cholesterol levels, resulting in the highest atherogenic index among diet treatments. Hepatic cholesterol concentration and HMG CoA reductase activity were also decreased, whereas fecal excretion of cholesterol was increased in trans fat-fed animals. trans fat containing diets also decreased hepatic membrane fluidity. From these results, it can be concluded that hypercholesterolemic effect of tFAs can be modulated by the fatty acid composition of diets, but its spatial cofiguration may help behave like saturated fatty acid in membrane fluidity.
In this study, the degree of rancidity and trans fatty acids formation was assessed in Soybean oil(SBO), Corn germ oil (CGO), Canola oil(CNO) and Olive oil(OLO). All samples treated under various conditions were analyzed in order to determine their physicochemical characteristics(RI: Refractive index, Tocopherol, AV: Acid Value, IV: Iodine Value) and total trans fatty acid contents via GC. The results were as follows: The AV of corn germ oil was the highest (0.49$\pm$0.01 and 0.72$\pm$0.04 respectively) among the 4 kinds of oils at $170{\pm}2^{\circ}C$ and $210{\pm}2^{\circ}C$. The IV of olive oil was the lowest(88.7$\pm$0.6 and 89.2$\pm$0.5) among the 4 kinds of oils at $170{\pm}2^{\circ}C$ and $210{\pm}2^{\circ}C$. The trans fatty acid contents of the soybean oil, corn germ oil, canola oil and olive oil, respectively(in g/100) increased from 0.41, 0.60, 0.44 and 0.11 prior to heating to 0.84, 1.36, 0.94 and 0.81 after 7 hours. Catechin and BHT reduced trans fatty acid formation by 0.5-15.5% under all treatment conditions. In particular, Catechin exerted a more profound inhibitory effect on trans fatty acids formation than that did BHT.
In this study, several standard fatty acids were analyzed by three analysis instruments. And also, for the two kinds of soybean oils, fatty acids compositions were determined by three instruments. The results were obtained as follows: 1. In the case of Gas Chromatography (GC), standard fatty acids (Myristic, Stearic, Linoleic, Linolenic, Arachidonic acid) were determined with high reproducibility, but oleic acid/elaidic acid were not seperated. By Capillary Gas Chromatography (CGC), most of standard fatty acids were determined with very high reproducibility than saturated fatty acids, and palmitic acid/oleic acid were not seperated. 2. In the analytical ability of cis-trans fatty acids isomer (oleic acid/elaidic acid), CGC was shown better analytical ability of geometrical isomer than HPLC. Oleic acid/elaidic acid were not seperated by packed column (15% DEGS). The rquire time for standard fatty acids analysis was as follows; GC, 7.21 min., CGC, 9.84 min., HPLC, 24.48 min. 3. The major compositions of fatty acids of each soybean oil (CSOY; refined, DSOY; unrefined) by GC and CGC were linoleic acid, oleic acid, palmitic acid, linolenic acid and stearic acid. But in the case of HPLC, palmitic acid/oleic acid were not seperated. Analytical ability of three instruments on fatty acids composition in each soybean oil was same trend as in the standard fatty acids mixture.
