Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) has no effect on normal cells, but selectively can induce apoptosis in tumor cells. Gartanin, a xanthone compound in mangosteen, has been shown to inhibit cancer cell growth by arresting the cell cycle and inducing autophage. In this study, we revealed that gartanin can sensitize TRAIL-induced human liver cancer cell death. We also found that gartanin enhances DR5 expression, a death receptor for TRAIL. This effect appears to be related to CHOP activation associated with the response of endoplasmic reticulum stress. Gartanin treatment also inhibited p62 protein expression and cleaved LC3 to activate autophagy flux, which is related with TRAIL-induced cell death. Pretreatment with autophagy flux inhibitor, LY294002, inhibited gartanin-induced DR5 expression. In summary, our results reveal that the combined treatment of gartanin and TRAIL can be a valuable tool for cancer treatment.
The purposes of this study are developing a stress index for each individual major trail in the national parks and contribute to establishing a park management policy based on scientific analyses using the index. We collected data from 144 sections of trails in national parks, conducted the Delphi survey and factor analysis, and generated the stress index by standardizing scores of factors contributing to the stress for each trail. As aresult, total eight factors contributing to the stress were considered and four of them, i.e. the amount of visits, the effect of visits, the degree of trail destruction, and species diversity, were selected to generate the stress index. How to weigh factors was determined by their importance obtained in the second Delphi survey and the average from a 5 point Likert scale. Then, the cumulative probabilities from the standardized scores were graded into five groups. In conclusion, there were twenty trails from the six national parks including Jirisan, Bukhansan, Deogyusan, Naejangsan, Seoraksan in the first grade which gained over 80 as the stress index.
Interest in the development of urban trails is growing in South Korea. River front trails have been developed in Seoul, Daegu and other major cities. Such trails, and the corridors in which they are developed, often provide a myriad of benefits to the community, including open space, exercise, and stress release. This trend mirrors development patterns found in the United States and European nations. This study examined differences in trail users perceptions of trail attributes, activity participation, willingness to pay for trail access, and perceptions of trail benefits between trail users in the United States and those in South Korea. Perceptions of trail users at three trail sites in the metropolitan area of Daegu were examined, and then were compared to results reported in several studies conducted in the U. S. While this research was exploratory, it did uncover many interesting differences in perceptions of trail attributes between trail users in the two countries. This information may provide insight into the development of greenway systems in South Korea.
This study deals with the proper conditions of the trail rolling method (TRM) for the prevention of the buckling welding distortion at the AA5083 GMA butt weldment. For it, the effect of the working conditions of TRM including the rolling depth and the rolling distance between the welding torch and the roller on the longitudinal welding shrinkage force of the weldment was evaluated by using 3 dimensional thermo-mechanical FE analyses. The longitudinal welding shrinkage force inducing the buckling welding distortion at the GMA butt weldment was mitigated with an increase in the rolling depth and the rolling distance between the welding torch and the roller. Based on the results, the proper conditions of trail rolling method were established to reduce the longitudinal welding shrinkage force of the GMA butt weldment to below the critical value corresponding to the bucking distortion.
The resistance of cancer cells to anti-cancer drugs is the leading cause of chemotherapy failure. The clinical use of nonsteroidal anti-inflammatory drugs (NSAIDs) has been gradually extended to cancer treatment through combination with anti-cancer drugs. In the current study, we investigated whether NSAIDs including celecoxib (CCB), 2,5-dimethyl celecoxib (DMC), and ibuprofen (IBU) could enhance the cytotoxic effects of imatinib and TNF-related apoptosis inducing ligand (TRAIL) on human cancer cells. We found that the NSAIDs potentiated TRAIL and imatinib cytotoxicity against human hepatocellular carcinoma (HCC) cell lines SNU-354, SNU-423, SNU-449, and SNU-475/TR and against leukemic K562 cells with high level of CD44 (CD44highK562), respectively. More specifically, CCB induced endoplasmic reticulum stress via up-regulation of ATF4/CHOP which is associated with the induction of autophagy against HCC and CD44high K562 cells. NSAID-induced autophagic activity accelerated TRAIL cytotoxicity of HCC cells through up- and down-regulation of DR5 and c-FLIP, respectively. The NSAIDs also potentiated imatinib-induced cytotoxicity and apoptosis through down-regulation of markers in CD44highK562 cells that express a stemness phenotype. Our results suggest that the ability of NSAIDs to induce autophagy could enhance the cytotoxicity of TRAIL and imatinib, leading to a reverse resistance to these drugs in the cancer cells. In conclusion, NSAIDs in combination with low-dose TRAIL or imatinib may constitute a novel clinical strategy that maximizes therapeutic efficacy of each drug and effectively reduces the toxic side effects.
Purpose: The purpose of this study was to identify the relationship between depression, perceived stress, fatigue and anger in clinical nurses. Method: A descriptive survey was conducted using a convenient sample. Data was collected by questionnaires from four hundred clinical nurses who worked at a university hospital. Radloff's CES-D for depression, Cohen, Kamarck & Mermelstein's Perceived Stress Scale, VAS for Fatigue, and Spielberger's STAXI for anger were used. The data was analyzed using the pearson correlation coefficient, students' t-test, ANOVA, and stepwise multiple regression with SPSS/WIN 12.0. Result: The depression of clinical nurses showed a significantly positive correlation to perceived stress(r=.360, p=.000), mental fatigue(r=.471, p=.000), physical fatigue(r=.350, p=.000), trait anger(r=.370, p=.000), anger-in expression(r=.231, p=.000), and anger-control expression(r=.120, p=.016). There was a negative correlation between depression and age(r=-.146, p=.003). The mean score of depression of nurses, 26, was a very high score and 40.8% of clinical nurses were included in a depression group. The main significant predictors influencing depression of clinical nurses were mental fatigue, trail anger, perceived stress, anger-in expression, and state anger, which explained about 32.7%. Conclusion: These results indicate that clinical nurses with a high degree of perceived stress, mental fatigue and anger-in expression are likely to be depressed.
Objective of this study is to provide basic information for developing more differentiated and targeted forest healing policy and Korean medical healing programs grounded on consumer preference for forest based Korean medical healing tourism products. The internet survey(CAWI) by percentage quota sampling with 400 Seoulite ages over 30 by the age, area, and gender was conducted, and 317 samples were used for a final analysis. 61.5% of the Seoulite associated 'forest bath/walking in the woods/tree' with an image of a forest based Korean medical healing tourism product, and preference for the product and the intention to use were positive at the percentages of 72.9% and 67.5%, respectively. Preferred areas were Seoul/Gyeonggi-do(53.5%) and Gangwon-do(38.8%). 'Stress solving and refreshment', 'taking a forest bath and a walk', and 'maintaining and promoting health' were the main purposes of the use. As for a therapy, 'walking therapy' was most preferred, and 'ergotherapy' was the next. First priority as for a use facility was 'healing trail', and 'professional medical facility' ranked second. Although important decision attributes were ' cost of use', 'food', and 'friendliness of medical staff', all the other sets of attributes related to use convenience, quality of medical service and tourism activities also recorded high, which forecasts higher consumer expectation for the product. As the result showing differences in consumer preference by the demographic segmentation, differentiated and segmented consumer needs should be considered when planing and managing a product. The scope of the study is limited to a demographic segmentation which is a basic stage of understanding consumer preference, therefore more detailed future researches on complicated and multi-dimensional consumer needs are required.

Background: Phytochemical compounds are emerging as a new generation of anticancer agents with limited toxicity in cancer patients. The purpose of this study was to investigate the potential effcts of thymoquinone, caffeic acid phenylester (CAPE) and resveratrol on inflammatory markers, oxidative stress parameters, mRNA expression levels of proteins and survival of lung cancer cells in Vitro. Materials and Methods: The A549 cell line was treated with benzo(a)pyrene, benzo(a)pyrene plus caffeic acid phenylester (CAPE), benzo(a)pyrene plus resveratrol (RES), and benzo(a)pyrene plus thymoquinone (TQ). Inflammatory markers, oxidative stress parameters, mRNA expression levels of apoptotic and anti-apoptotic proteins and cell viability were assessed and results were compared among study groups. Results: TQ treatment up-regulated Bax and down-regulated Bcl2 proteins and increased the Bax/Bcl2 ratio. CAPE and TQ also up-regulated Bax expression. RES and TQ down-regulated the expression of Bcl-2. All three agents decreased the expression of cyclin D and increased the expression of p21. However, the most significant up-regulation of p21 expression was observed in TQ treated cells. CAPE, RES and TQ up-regulated TRAIL receptor 1 and 2 expression. RES and TQ down-regulated the expression of NF-kappa B and IKK1. Viability of CAPE, RES and TQ treated cells was found to be significantly decreased when compared with the control group (p=0.004). Conclusions: Our results revealed up-regulation of the key upstream signaling factors, which ultimately cause increase in their regulatory p53 levels affecting the induction of G2/M cell cycle arrest and apoptosis. Overall these results provide mechanistic insights for understanding the molecular basis and utility of the anti-tumor activity of TQ, RES and CAPE.
Withaferin A is a steroidal lactone purified from the Indian medicinal plant Withania somnifera. It exhibits a wide variety of activities, including anti-tumor, anti-inflammation, and immunomodulation properties. In this review, we focused on the anti-cancer effects of withaferin A. Withaferin A inhibits cell proliferation, metastasis, invasion, and angiogenesis in cancer cells. Furthermore, it sensitized irradiation, tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-, and doxorubicin-mediated apoptosis. The results showed that multiple mechanisms were involved in withaferin A-mediated anti-cancer effects. First, withaferin A increased intracellular reactive oxygen species (ROS) production and induced ER stress- and mitochondria-mediated apoptosis. Second, withaferin A inhibited the signaling pathways (Jak/STAT, Akt, Notch, and c-Met), which are important in cell survival, proliferation, and metastasis. Third, it induced apoptosis and inhibited cancer cell migration through the up-regulation of prostate apoptosis protein-4 (Par-4). Finally, withaferin A up-regulated pro-apoptotic protein expression levels through the inhibition of proteasome activity. Our findings suggested that withaferin A is a potential, potent therapeutic agent.
