Evaluation of Interfering Substances in Routine Chemistry Tests Using Toshiba TBA-C8000 Chemistry Analyzer

  • In clinical chemistry tests, the interfering substances such as hemoglobin, lipid, bilirubin, and drugs, etc. can cause the changes of test results performed by spectrophotometrical methods. We evaluated the effects of interfering substances on the test results by adding interfering substances on the samples in the 19 kinds of clinical chemistry tests such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyltransferase, total protein, albumin, glucose, total cholesterol, total bilirubin, triglyceride, uric acid, calcium, inorganic phosphours, high density lipoprotein cholesterol, low density lipoprotein cholesterol, creatinine, blood urea nitrogen, and C-reactive protein using newly implemented automatic chemical analyzer Toshiba TBA-C8000 under the direction of CLSI EP07-A guideline. Hemolytic samples show increased concentration of total protein, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase and reduced concentration of total bilirubin, alkaline phosphatase by interfering effect. Hyperlipemic samples show increased concentration of total protein and alkaline phosphatase and reduced concentration of low density lipoprotein cholesterol. The samples with conjugated bilirubinemia show increased concentration of inorganic phosphours, otherwise the samples with unconjugated bilirubinemia show no interference or allowable range in the test result.

Effects of Genetic Variants of ${\kappa}$-casein and ${\beta}$-lactoglobulin and Heat Treatment of Milk on Cheese and Whey Compositions

  • Milk samples with different phenotype combination of $\{kappa}$-casein and ${\beta}$-lactoglobulin and different preheating temperatures of 30, 70, 75 and $80^{\circ}C$ were used for cheesemaking under laboratory conditions. For the 853 batches of cheese, mean composition was 59.64% total solids, 30.24% fat and 23.66% protein, and the whey contained 6.93% total solids, 0.30% fat and 0.87% protein. Least squares analysis of the data indicated that heating temperature of the milk and ${\kappa}$-CN/${\beta}$-LG phenotypes had significant effects on cheese and whey compositions. The total solids, fat and protein contents of cheese were negatively correlated with preheating temperatures of milk. Cheese from BB/BB phenotype milk had the highest and those from AA/AA phenotype milk had the lowest concentrations of total solids, fat and protein. Mean recoveries of milk components in the cheese were 53.71% of total solids, 87.15% of fat, and 80.32% of protein. For the 10 different types of milk, maximum recoveries of milk components in cheese occurred with preheating temperature of $70^{\circ}C$ or $75^{\circ}C$ and lowest recoveries occurred at $80^{\circ}C$. The whey averaged 6.94% total solids, 0.30% fat and 0.87% protein. Losses of milk components in the whey were lowest for milk preheated at $80^{\circ}C$ and for milk containing the BB/BB phenotype.

Protein Electrophoresis Fraction in Serum of the Rural Elderly Patient

  • We analyzed total protein (TP), albumin (AL), aspartic aminotransferase (AST), alanine aminotransferase (ALT), urea nitrogen (UN), creatinine (CRE) and serum protein electrophoretic fractions in sera of rural elderly patients to evaluate the health status in rural elderly patients. We observed that the frequencies of patients showed lower level of total protein, albumin, and both total protein and albumin than them of reference range were 20.3%, 22.8% and 19.0%, respectively. The rates of patients showed higher level of AST, ALT, both of AST and ALT, UN, creatinine and both of UN and creatinine than them of reference range were 33.8%, 40.0%, 30.0%, 17.7%, 15.2% and 7.9%, respectively. Comparison of protein fractions of each patient to reference range showed that 77.2% of patients showed normal in all of patterns. Few of patients showed abnormal pattern in albumin (13.6%), ${\alpha}1-globulin (0.0%),\;{\alpha}_2-g10bulin(1.1%),\;{\beta}-globulin(1.9%)\;and\;{\gamma}-globulin(6.2%)$. These data suggest that many of rural elderly patients may suffer from heart, liver and kidney diseases. The serum protein fractions are not typical criterion to evaluate the disease, but production of proteins in rural elderly patients may be affected by liver disease and kidney disease.

Effect of Systemic Fungicide on Total Hemocyte Count and Hemolymph Biochemical Changes in Silkworm, Bombyx mori L., infected with Beau-veria bassiana

  • Silkworm diseases are major constraint in silk cocoon production. Among silkworm diseases, white muscar-dine is highly contagious and most common in winter and rainy seasons. It is suggested that hemocytes involve in defense against invasion of Beauveria bassi-ana and systemic fungicide/chemicals prevent the proliferation of fungi in the hemolymph or preventing the growth of the fungi in the body cavity through enhancing the hemocyte mediated defense response. In the present study the influence of systemic fungicide on hematological changes in silkworms infected with Beauveria bassiana was reported. It is observed that the total hemocyte counts increased in the hemolymph up to 5 days post inoculation in systemic fungicide treated batches while in the inoculated control the increase was up to 3 days indicating the positive hemocyte mediated response in silkworm treated with systemic fungicide. After 2 days in the inoculated control as the multiplication and growth of mycelia increased, defense capacity of the silkworm was decreased. The biochemical changes were also observed in the hemolymph of silkworm infected with B. bassiana. In silkworm infected with Beauveria bassi-ana, the total protein content increased whereas total carbohydrate and total lipids decreased as the infection progresses. In the case of systemic fungicide treated batches the increase in total protein content was comparatively higher and decrease in total carbohydrate and lipids were comparatively lower than the inoculated control.

市販豆腐 成分 및 細菌汚染에 관한 硏究 (A Study on Ingredient and Bacterial Contamination of Bean Curd on the Market)

  • The purpose of this study was to analyze general ingredient of bean cured on the market and to examine its exposure to bacterial contamination. For this study, 17 samples (each 9 samples for general bean curd, soft bean curd, and uncurdled bean curd) were randomly collected from nine areas in Seoul from the beginning of April, 1983 to the beginning of June, 1983. The result of ingredient analysis of moisture, ash, and protein of bean curd was compared with the standard set by the Ministry of Health & Social Affairs. In order to find out exposure of bean curd on the market to bacterial contamination, total biological bacteria and coliform group were examined. Experimental results were shown as follows 1) Results of ingredient analysis of moisture, ash, and protein of general bean curd showed that total samples in both moisture and protein met the standard set by the Ministry of Health & Social Affairs but 44.4% of the samples in ash was below the above standard, indicating average 82.0%, 0.9% and 9.6% in moisture, ash, and protein order. 2) Experimental results of moisture, ash, and protein of soft bean curd demonstrated 90.2%, 0.5% and 4.3% respectively total samples in both moisture and protein satisfied the self-criteria set by the Soft Food Co-operative Association of Seoul City but 11.1% of the samples in ash didn't meet the self-criteria. 3) Total samples of uncurdled bean curd satisfied the self-criteria set by the above association, indicating average 92.0%, 0.4%, and 3.5% in moisture, ash, and protein order. 4) Total biological bacteria and coliform group detected in general bean curd showed that more than 10$^5$/g in total biological bacteria accounted for 88.8% of the samples and that 10$^4$/g or more in coliform group accounted for 77.7% of the samples. The result proves that general bean curd has been exposured to a severe bacterial contamination. 5) Result of total biological bacteria and coliform group detected in such packed bean curd as uncurdled bean curd and soft bean curd showed that 61.6% of the samples exceeded 10$^6$/g in total biological bacteria and 27.7% of the samples exceeded 10$^3$/g in coliform group. 6) According to the change with time and temperatures in total biological bacteria and coliform group of general bean curd, general bean curd began to decay around 72 hours at 4$\circ$C and around 48 hours at 23$\circ$C and around 24 hours at 37$\circ$C and, at that time, total biological bacteria approached 10$^6$/g while coliform group did 10$^6$/g. The result indicates that temperature has a great effect on bacteria counts and decay.

기상요인이 콩 단백질 함량에 미치는 영향 (Climatic Influence on Seed Protein Content in Soybean(Glycine max))

  • This study was carried out to identify how soybean seed protein concentration is influenced by climatic factors. Twelve lines selected for seed protein concentration were studied in 13 environments of North Carolina. Sensitivity of seed protein concentration, total seed protein, and seed yield to climatic variables was investigated using a linear regression model. Best response models were determined using two stepwise selection methods, Maximum R-square and Stepwise Selection. There were wide climatic effects in seed protein concentration, total protein and seed yield. The highest protein concentration environment was characterized by the most high temperature days(HTD) and the smallest variance of average daily temperature range (VADTRg), while the lowest protein concentration environment was distinguished by the fewest HTD and the largest VADTRg. For protein concentration, all lines responded positively to average maximum daily temperature(MxDT), HTD, and average daily temperature range(ADTRg) and negatively to ADRa, while they responded positively or negatively to average daily temperature(ADT), variance of average minimum daily temperature (VMnDT), and VADTRg, indicating that genotypes may greatly differ in degrees of sensitivity to each climatic variable. Eleven lines seemed to have best response models with 2 or 3 variables. Exceptionally, NC106 did not show a significant sensitivity to any climatic variable and thus did not have a best response model. This indicates that it may be considered phenotypically more stable. For total seed protein and seed yield, all the lines responded negatively to both ADTRg and VADRa, suggesting that synthesis of seed components may increase with less daily temperature range and less variation in daily rainfall.

금궤신기탕 및 도홍사물탕(桃紅四物湯)이 Puromycin Aminonucleoside로 유발(誘發)된 백서(白鼠)의 신증(腎症)에 미치는 영향(影響) (The Effects Of Geumguesingitang And Dohongsamultang On Rats With Nephrosis Induced By Puromycin Aminonucleoside)

  • The effects of Geumguesingitang and Dohongsamultang on rats with nephrosis induced by a single tail-intravenous injection of puromycin aminonucleoside(PAN), 2.5mg/l00g of body weight were evaluated in the present study. The effects of Geumguesingitang and Dohongsamultang on PAN nephrosis were evaluated by measuring (1)the concentrations of albumin, total protein, total lipid, cholesterol, triglyceride, creatinine, blood urea nitrogen(BUN) and uric acid in the serum, (2)the concentrations of protein, creatinine, glucose, occult blood and volume of the 24 hours urine and (3)the volume of intake water. The results are summerized as follows; 1. In the Control group as compared to the Normal. the amount of protein of 24 hours urine was significantly increased, the concentrations of albumin and total protein were significantly decreased. Total lipid, cholesterol and triglyceride in the serum were significantly increased. The concentrations of creatinine, BUN, uric acid in the serum, the amount of glucose and occult blood of 24 hours urine were also increased significantly. 2. In the Geumguesingitang group as compared to the Control, the increase in the amount of urinary protein during 24 hours induced by PAN was significantly suppressed, and the concentrations of total protein and albumin in the serum were significantly increased. The concentrations of total lipid, cholesterol and triglyceride in the serum were significantly inhibited. The decrease of the concentrations of creatinine and uric acid in the serum were also observed significantly. 3. In the Dohongsamultang group as compared to the Control, the increase of the amount of protein and glucose of the 24 hours urine induced by PAN were significantly inhibited, and the concentrations of total protein and albumin in the serum were increased significantly. The concentrations of total lipid, cholesterol and triglyceride in the serum were decreased significantly. The decrease of the concentrations of creatinine and uric acid in the serum were observed significantly. It can be inferred that Geumguesingitang has effects on improving proteinuria, hypoproteinemia effectively. It has an effect on hyperlipidemia significantly relieved. And relieving azotemia when nephrotic syndrome is accompanied by the acute renal failure. It can be inferred that Dohongsamultang improves hyperlipidemia effectively. It has effects on proteinuria, hypoproteinemia in nephrotic syndrome significantly relieved. And relieving azotemia when nephrotic syndrome is accompanied by the acute renal failure.

오케타니 유방마사지가 유방불편감, 모유량 및 모유 Total Protein에 미치는 효과 (Effects of Oketani Breast Massage on Breast Discomfort, Breast Milk Amount, and Breast Milk Total Protein)

  • 본 연구의 목적은 오케타니 유방마사지가 산욕초기 수유부의 유방불편감, 모유량 및 모유 Total Protein에 미치는 효과를 검증하는 것이다. 연구설계는 비동등성 대조군 전후시차설계를 이용한 유사실험연구이다. 연구대상은 유방불편감이 있는 산욕초기 산모 60명으로, 실험군과 대조군은 각각 30명이었고, 자료수집은 설문지와 산모가 자가 채취한 모유를 분석하였다. 간호중재로 실험군에게 오케타니 유방마사지를 출산 후 2회 실시하였고, 대조군에게 Self Mamma Care(SMC)을 실시하였다. 수집된 자료는 $x^2-test$, t-test로 분석하였다. 연구결과에서 오케타니 유방마사지를 받은 실험군이 SMC 교육을 받은 대조군보다 유방불편감이 감소하였고, 모유량, 모유 Total Protein이 통계적으로 유의하게 증가하였으므로 오케타니 유방마사지가 유방불편감을 개선하고, 모유량을 증가시키고, 모유 Total Protein을 증가시키는 데 효과적이라 할 수 있다. 그러므로 본 연구는 임상실무에서 산욕초기 수유부에게 유방관련 불편감 감소와 모유량 증가, 모유성분을 개선하는 간호중재로 오케타니 유방마사지가 활용되기를 기대한다.

콩단백질과 고기단백질 식이가 인체내 단백질과 칼슘.인.마그네슘 대사에 미치는 영향 : 중기간 급식 효과 (Effects of Soy Protein and Meat Protein Diets on Protein, Calcium, Phosphorus, and Magnesium Metabolism in Young Korean Adults : Mid-Tern Feeding Study)

  • A 44-day metabolic study was conducted to compare the effects of soy protein and meat protein on N, Ca, P, and Mg metabolism in young Korean males. Meat protein diet(MPD) and soy protin diet(SPD) were provided for 3 weeks each. For MPD, meat protein Supplied 50.3% of total protein and mean daily suplies of Ca, P, and Mg were 33.28mg, 1013.01mg, 226.81mg respectively. For SPD, soy protein supplied 51.8% of total protein and mean contents of Ca, P and Mg were 737.86mg, 1261.76mg, and 451.3mg. Mean daily fecal and urinary excretions of Ca were 350.6mg and 113.3mg for MPD and 515.0mg and 118.3mg for SPD. Apparent absorption rates in MPD and SPD were 4.2mg in MPD and 700.4mg and 475.1mg SPD. Apparent absorption rates were 65.6% in MPD and 44.5% in SPD(p<0.01). For Mg, fecal and urinary excretions were 144.8mg and 81.2mg in MPD and 292.4mg and 65.5mg in SPD. Apparent absorption rates were 37.5% in MPD and 35.2% in SPD. The correlations between urinary Ca, P, and their balance were significant (p<0.001) in MPD, but not in SPD. Urinary Ca was significantly correlated with urinary N, P, Mg(p<0.001) in MPD, but not in SPD. From these results, meat protein seems to increase the excretions of major minerals compared to soy protein.

방사선 방어작용이 있는 인삼 단백분획의 CHO-KI 세포에 대한 세포 독성 (Cytotoxic Effect of Radioprotective Ginseng Protein Fraction on CHO-KI Cells)

  • Radioprotective ginseng protein fraction was isolated from Korean white ginseng and its cytotoxic effect on CHO-K1 cells was studied by the method of measuring the relative cell survival and total cellular protein content (FRAME method). When ginseng protein at the dose of 300, 600, 900, $1200{\mu}g/ml$ was treated to cells for 24 hrs, the relative survival was significantly decreased at the concentration of above $600{\mu}g/ml$, indicating the presence of cytotoxic effect of the protein at certain concentration. When cellular protein content was measured after ginseng protein at the dose of 300, 600, 900, $1200\;{\mu}g/ml$ was treated, the amount of cellular protein was significantly reduced at the concentration above $600{\mu}g/ml$ in the case of 24 hr treatment and at all concentrations including $300{\mu}g/ml$ in the case of 72 hr treatment. The data suggest that the protein may inhibit cell growth, resulting in the reduction of live cells in culture. $ID_{50}$ value which is the concentration of ginseng protein that reduces the total cellular protein content to 50% of the control was calculated as 2276.86 and $1323.32\;{\mu}g/ml$ in groups treated for 24 and 72 hr, respectively. Since $ID_{50}$ value of above $1000{\mu}g/ml$ indicates very weak cytotoxicity, the ginseng protein seems to exert very weak cytotoxic effect on CHO-K1 cells.
