To evaluate the antioxidative effect of maengjong-juk (Phyllostachys pubescens) extract coated rice in vivo system, maengjong-juk extract coated rice diets were fed to C57BL/6 mice for 16 weeks. Plasma total antioxidative capacity, hepatic lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content were measured. Plasma total antioxidative capacity was elevated significantly in maengjong-juk extract diets supplemented group in a dose dependant manner. Hepatic TBARS contents were significantly decreased in maengjong-juk extract diets supplemented group compared to high cholesterol group. Maengjong-juk extract coated rice diets suppressed the protein oxidation significantly in liver. Activities of hepatic antioxidative enzymes such as total SOD, CuㆍZn-SOD, Mn-SOD, GSH-Px and catalase activities of maengjong-juk extract coated rice diets were significantly higher than those of high cholesterol diet. Total hepatic glutathione content was significantly increased by maengjong-juk extract coated rice diets administration. According to this study, numerous antioxidative materials and phytochemicals containing in maengjong-juk extracts appear to protect antioxidative systems in C57BL/6 mice fed bamboo extract coated rice diet. The oxidative stress level and antioxidant activities in two green algae (Ulva pertusa and Ulva linza), two brown algae (Agarum cribrosum and Dictyota dichotoma), and three red algae (Grateloupia lanceolata, Carpopeltis affinis, and Gracilaria verrucosa) collected from intertidal regions of Korea were assessed. In the two green algae, although the total glutathione content was not as high as that of the brown algae, the glutathione pool was extremely reduced, and the glutathione reductase (GRd)/glutathione peroxidase (GPx) activity ratio was high, which apparently plays an important role for protection against oxidative damage, as manifested by low lipid peroxidation. In the brown algae, which exhibited a low lipid peroxidation level that was comparable to the green algal species, the highest glutathione content, together with high GPx activity, appears to be the most important factor in their antioxidant protection. The red algal species exhibited extremely high lipid peroxidation levels. They also contained the lowest and most oxidized glutathione among the species, as well as the lowest GRd activity. In spite of the marked difference in the glutathione content, the significant difference in the activity of ${\gamma}$-glutamylcysteine ligase, the rate limiting enzyme for glutathione synthesis, among the species was not exhibited. Our results suggest that there is a significant difference in the levels of oxidative stress and antioxidant capacity among the algal species, and that the glutathione system, especially the efficiency of glutathione recycling, plays a vital role in antioxidative protection in algal species.
Objective Trichotillomania is a relatively common illness whose neurobiology is poorly understood. One treatment for adult trichotillomania, n-acetyl cysteine (NAC), has antioxidative properties, as well as effects on central glutamatergic transmission. Preclinical models suggest that excessive oxidative stress may be involved in its pathophysiology. Methods Adults with trichotillomania provided a blood sample for analysis of compounds that may be influenced by oxidative stress [glutathione, angiotensin II, ferritin, iron, glucose, insulin and insulin growth factor 1 (IGF1), and hepcidin]. Participants were examined on symptom severity, disability, and impulsivity. The number of participants with out-of-reference range oxidative stress measures were compared against the null distribution. Correlations between oxidative stress markers and clinical measures were examined. Results Of 14 participants (mean age 31.2 years; 92.9% female), 35.7% (n=5) had total glutathione levels below the reference range (p=0.041). Other oxidative stress measures did not have significant proportions outside the reference ranges. Lower levels of glutathione correlated significantly with higher motor impulsiveness (Barratt Impulsiveness Scale sub-score) (r=0.97, p=0.001). Conclusion A third of patients with trichotillomania had low levels of glutathione, and lower levels of glutathione correlated significantly with higher motor impulsiveness. Because NAC is a precursor for cysteine, and cysteine is a rate limiting step for glutathione production, these results may shed light on the mechanisms through which NAC can have beneficial effects for impulsive symptoms. Confirmation of these results requires a suitable larger follow-up study, including an internal normative control group.
The purpose of this study was to investigate the effect of dietary mushroom powder on blood glucose levels, seam lipid levels, glucose 6-phosphtase (G6Pase), thiobarbituric arid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ). Four groups of rats (Sprague-Dawley male rats, 180-200 g) were fed as follows: normal rats were fed a control diet (C), diabetic rats were file a control diet (CD), normal fats were fed a mushroom powder diet (M), and diabetic rals were find mushroom powder diet (MD). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). The animals were fed ad libium each of the experimental diets for five weeks. Food and water intake was determined every day. Blood glucose and serum total cholesterol levels were determined every week. After five weeks, the rats were sacrificed and blood glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. HDL-cholesterol levels were analyzed and LDL-cholesterol concentrations were calculated by equation. There was body weight loss in the diabetic rats, but the MD group showed less body weight loss than the CD group. Blood glucose and serum total cholesterol level of the MD group were lower than those of the CD group (p < 0.05). Also, serum total cholesterol of the M group was lower than that of the C group (p < 0.05). But the serum triglyceride level of the diabetic rats (CD and MD) was higher than that of the normal rats (C and M). However, there was no significant difference between the control diet group and the mushroom diet group. Serum HDL-cholesterol levels of the C group and CD group were higher than that of the M group (p < 0.05), and the MD group was not significantly different. But the serum LDL-cholesterol levels of the M group were lower than those of the C group (p < 0.05). Activity of hepatic microsomal G6Pase significantly increased in the CD and MD, reaching levels higher than those of the C and M groups. Hepateic gutathione S-transferase (GST, glutathione reductase (GR) and glutathione peroxidase (GPX) activity was not significant. But renal GST, GR and GPX activity in the MD group was lower than that of the CD group (p < 0.05). These results suggest that dietary mushroom reduces renal disorders such as oxidation and aging of tissue. In conclusion, dietary mushroom groups reduced blood glucose and cholesterol levels in STZ-induced diabetic rats and renal glutathione enzymes activity was averted in diabetic rats.
In this study, coffee waste was extracted with different solvents such as ethyl acetate, methylene chloride and methanol to investigate the total polyphenol contents, electron donating ability and the inhibitory effect on glutathione S-transferase. The total polyphenol contents were $3,060.61{\pm}357.12{\mu}g\;GAE/mL$ in ethyl acetate, $909.09{\pm}35.71{\mu}g\;GAE/mL$ in methylene chloride, and $1,602.27{\pm}30.36{\mu}g\;GAE/mL$ in methanol. The total polyphenol contents showed a significant difference (p<0.05) between the solvents. The electron donating ability was $80.20{\pm}1.45%$ for ethyl acetate, $81.94{\pm}0.45%$ for methylene chloride, and $85.14{\pm}1.53%$ for methanol. The electron donating abilities were significantly different (p<0.05) between the solvents. The inhibitory effect of the various extracts on glutathione S-transferase (% inhibition) was $92.12{\pm}0.56%$, $88.48{\pm}0245%$ with methylene chloride extract, and $90.85{\pm}0.14%$ with methanol extract. These too were significant different (p<0.05) between the solvents. The two portions of coffee waste extracts obtained from ethyl acetate and methanol showed meaningful results on the total polyphenol contents, and the inhibition effects on glutathione S-transferase. Therefore, they can be utilized to develop health care foods and can be applied as antioxidants for cosmeceuticals.
The total phenolic and flavonoid contents of Lentinus edodes extracts were determinated by spectrophotometrical .method, and antihepatotoxic activity was detected on glutathione S-Transferases(GST). The total phenolic contents was highest water extract than solvent(ethanol, methanol) extracts, but flavonoid content was appear on opposite. GST activity was the highest appears in water extract. This fact verified of anticancer effect indirectly of Lentinus edodes.
Tomato (Lycopersicon esculentum Mill) seedlings exposed to various concentrations of $CdCl_2(0{\sim}100{\mu}M)$ in a nutrient solution for up to 9 days were analyzed with respect to the thiol changes and oxidative stress. The Cd exposure increased total non-protein thiols (NPT) and cysteine in both leaves and roots, total glutathione in leaves, and the ratios of oxidized glutathione (GSSG)/reduced glutathione (GSH) in both leaves and roots, but decreased the ratio of dehydroascorbate (DASA)/ascorbate(ASA) in leaves. Our results suggest that the Cd-induced GSH depletion due to thiol synthesis and oxidation alters the antioxidant activity of seedlings for $H_2O_2$, and the subsequent $H_2O_2$ accumulationand oxidative stress result in phytotoxicity.
We investigated the levels of glutathione (GSH) and its oxidized form (GSSG) in 24 cell lines derived from various plant species to understand the antioxidative mechanism in plant cell cultures. The total glutathione content was 98$\pm$27 $\mu$g/g cell fresh wt, showing a slight difference in plant species. The average contort of GSH and GSSG was 72$\pm$20 and 26$\pm$10 $\mu$g/g cell fresh wt, respectively. The average GSH content in plant cell lines occupies approximately 73% in total glutathione. During the suspension cultures of Scutellaria baicalensis, one of the plant species we tested, the GSH content decreased in proportion to the cell growth during the exponential growth stage, showing the low level at the stationary growth stage (84 $\mu$g/g cell fresh wt), whereas the GSSG content increased to the stationary growth stage (31 $\mu$g/g cell fresh wt). The results suggested that the ratio of GSH and GSSG should be involved in the cell growth and antioxidative mechanism in cultured cells.
Livers isolated from 18 hours fasted rats were subjected to N$_{2}$ hypoxia (for 45 min) followed by reoxygenation (for 45 min). The perfusion medium used was Krebs-Henseleit bicarbonate buffer (KHBB, pH 7.4). Lactate and alanine were added as gluconeogenic and ureagenic substrates and Trolox C was also added to perfusate. Oxygen consumption, lactate dehydrogenase (LDH), alanine transaminase (ALT), total glutathione, oxidized glutathione, bile flow, glucose and urea were measured. After hypoxia oxygen consumption significantly dropped but Trolox C had no influence on this decrease. ALT and LDH were significantly increased by hypoxia/reoxygenation. This increase was markedly attenuated in the presence of Trolox C. The total glutathione and oxidized glutathione efflux increased following hypoxia, which were prevented by the treatment of Trolox C. Bile flow rate decreased following hypoxia/reoxygenation but did not continue to decrease in the reoxygenation phase by Trolox C. Following hypoxia/reoxygenation glucose and urea releases decreased. Trolox C had no influence on inhibition of glucose and urea production. These results suggest that Trolox C protected the liver cells against hypoxia/reoxygenation injury, yielding further evidence for a causative role of oxidative stress in this model.
This study was conducted to investigate the effect of dietary grape skin on lipid peroxidation and antioxidant defense system in rats fed high fat diet. The Sprague-Dawley rats were fed either control (5% fat) diet or high fat (25% fat) diet which was based on AIN-93 diet for 2 weeks, and then they were grouped as control group (C), control + 5% grape skin group (CS), high-fat group (HF), high fat + 5% grape skin group (HFS) with 10 rats each and fed corresponding diets for 4 weeks. The hepatic thiobarbituric acid reacting substances (TBARS) were increased in high fat group as compared with control group, but reduced by grape skin. The serum total antioxidant status, and activities of hepatic catalase and superoxide dismutase, xanthine oxidase and glucose-6-phosphatase were increased by supplementation of grape skin. Glutathione peroxidase activity was significantly higher in CS group than in C group. Grape skin feeding tended to increase the concentration of total glutathione, especially in control group. The ratio of reduced glutathione to oxidized glutathione was lower in high fat groups than in control groups. The ratio was increased by dietary supplementation of grape skin in control group. These results suggest that dietary supplementation of grape skin would be effective on protection of oxidative damage by lipid peroxidation through improvement of antioxidant defense system in rats fed high fat diet as well as rats with low fat diet.
