A porous silicate material named as CellCaSi was tested for the removal of microalgae in the water sample from a eutrophic pond. The effects of the CellCaSi on water qualities were investigated on the basis of both the particle size (under 1, 2,and 4 mm) and the added amount (0, 1, 5, and 10 g/l) of the CellCaSi. The removal efficiency of chlorophyll-a was highest at 79% by the addition of 10 g/l of the CellCaSi (under 1 mm) at day 3 after treatment. That is, the removal efficiency of chlorophyll-a by the CellCaSi increased with smaller particle size and more added amount. The dominant species, Chlorella ellipsoidea, was not changed by the addition of the CellCaSi, but the species number and standing crop of the algae diminished. Total nitrogen concentration was not changed much by the addition of the CellCaSi, whereas total phosphorus concentration was reduced. pH and turbidity were not changed by the addition of the CellCaSi, whereas conductivity showed a high correlation with the amount of added CellCaSi ($Y\;=\;29.2 {\cdot}X+306$, $r^2\;=0.984$). Therefore, it seems to be necessary to limit the amount of the CellCaSi under 6.6 g/1 in consideration of a registered maximum conductivity of $500\;{\mu}mhos/cm$ for raw and potable waters.

This study was carried out to develop a fermented juice using persimmon (Diospyros kaki Thunb) and lactic acid bacteria isolated from kimchi, Lactobacillus buchneri BK-1, Pediococcus inopinatus BK-3 and Leuconostoc mesenteroides M-17. The total acidity value was 0.75% and viable cell number reached $1.9{\times}10^8CFU/mL$ when the persimmon and water solution was diluted by 1:3 (w/v) added with rice-syrup ($15^{\circ}Brix$) that was fermented by Lactobacillus buchneri BK-1 for 7 days. Additional levels of rice-syrup increased the total acidity of fermented juice, and the overall acceptability was the highest (4.1 point) for fermented persimmon juice added with rice-syrup $10^{\circ}Brix$. L. buchneri BK-1 and Pediococcus inopinatus BK-3 were selected to ferment the persimmon juice because there total acidity values were 0.83% and 0.80%, respectively, and the final cell concentrations, $5.1{\times}10^8$ and $2.7{\times}10^8CFU/mL$, were more than other treatment, respectively. The total acidity value of persimmon at day 3 of fermented broth were significantly higher than that of day 7 of fermented broth, and the number of viable cell declined from $8.2{\times}10^8$ to $4.3{\times}10^8CFU/mL$. In these results, the suitable period for fermentation was 4~5 days owing to the sourness being strong during fermentation.
The present study was conducted to examine the effect of prunus mume extract on the Kimchi fermentation. The physicochemical and microbiological properties of Kimchi with the addition of various levels(0, 0.1, 0.3 or 0.5%)of prunus mume extract was measured up to 30 days at $4^{\circ}C$. pH was decreased sharply in control after 2 days . A typical decreasing pattern in pH values was noted in all treatments during the initial 18 days of fermentation followed by a gradual decrease afterwards. Total acidity was decreased by the addition of prunus mume extract. Reducing sugar content was reduced to approximately 70% by day 10-12 in prunus mume extract treatments. Total vitamin C content was reached to maximum on 6 days of fermentation and then decreased in all treatments. Reducing sugar and total vitamin C contents were slightly increased by the addition of prunus mume. Total viable cell number increased to reach peak on day 8 and then slowly decreased during the fermentation. Lactobacillus cell number in control treatment continued to increase to reach peak on day 12, while the numbers in prunus mume treatments reached peak on day 8-12 and then gradually decreased throughout the fermentation. The results of this study indicate that prunus mume extract causes to delay the Kimchi fermentation by slowing down pH drop and inhibiting the Lactobacillus cell growth.
In order to find out if staphylococci occur in significant numbers in Korean fermented fish products, a total of 40 different fermented fish products were collected from different markets in Korea and analyzed for their physico-chemical and microbiological states. The pH, salt concentration and water activity of the products were measured and the total viable cell count and the number of Staphylococcus grown on mannitol salt agar were determined. The identification of the strains of Staphylococcus were made by API Staph Strip and MIS identification kits, and the physiological properties of the identified strains were further characterized by different conventional methods. The pH, salt content and water activity of fermented fish samples varied widely from 4.8 to 7.1, 7.4-28.7$%$ and 0.77-0.84, repectively, depending on the type of product. The total viable cell count varied from $10^4-10^9$ cfu/ml, and most of the samples had $10^5-10^6$ cfu/ml No correlation was found between the viable cell count and the pH, NaCl concentration and water activity of the samples. Among the 35 colonies identified as Staphylococcus strains by the identification kits, S. xylosus was the most frequently occurring strain marking 17, and S. warneri was 8, S. epidermidis 4 and S. cohnii 2. S. hominis, S. saprophyticus, S. haemolyticus and S. aureus were also identified once each. In some samples (K-3, P-6, K-8, G-5 and G-10), 2-3 different species of Staphylococcus were found. Considering the region of sampling, among the 10 samples from Kunsan 5 were identified as S. warneri, while in the other regions S. xylosus was predominant. Although the physiological characteristics of the identified strains were generally consistent with those in Bergey's Manual, some discrepances were also observed. All the strains were highly salt tolerant, growing in the media containing over 18$%$ NaCl. All the strains except S. aureus (G-11) showed negative in hemolysis activity, plasma coagulation and DNase tests. All the strains including S. aureus (G-11) showed negative in enterotoxin test.
The objective of this study is to determine developmental pattern and cell allocation to the ICM and TE in haploid and diploid of embryos following parthenogenetic activation and in vitro fertilization. The incidence of development to blastocyst was lower in the combined treatments of ethanol stimulation and cytochalasin B as compared to the control. However, the combined ethanol stimulation and cytochalasin B treatment (diploid) accelerated development to the blastocyst as compared to the ethanol treatment alone (haploid). Significantly reduction in the average number of total cells and ICM was observed in the parthenotes alone as compared to fetilized embryos, but those of combined ethanol stimulation and cytochalasin B treatment embryos were significantly increased as compared to ethanol alone embryos. These results suggested that the ploidy affects preimplantation developmental patters and cell allocation to the ICM and TE in the porcine.

It has been reported that cloning cattle is inefficient. One of the problems was placental abnormality, finally resulting in fetal mortality after transfer of nuclear transfer (NT) bovine embryos. This study was focused on the allocations of embryonic cells to the inner cell mass (ICM) or to the trophectoderm(TE) in NT bovine blastocysts. Somatic cells were derived from a Day 45 fetus of gestation, individually transferred into enucleated oocytes and developed to the blastocyst stage in vitro. Differential staining was used to assess the qualify of blastocysts derived from NT, IVF and in vivo. Development rate of NT embryos to blastocysts (25.0%, 41/164) was similar to that of IVF embryos (28.7%, 49/171). The total cell number of NT blastocysts (101.3$\pm$45.9) was not different compared with that of IVF embryos (107.9$\pm$34.2, P>0.05), but was lower than in vivo embryos (122.5$\pm$21.6, P<0.05). Ratio of ICM/total cells was higher in NT embryos (51.6$\pm$ 18.6%) than in IVF and in vivo embryos (42.3$\pm$ 15.3% and 34.9$\pm$8.9%, respectively) (P<0.05). Most IVF (56.8%, 25/44) and in vivo blastocysts(80.8%, 21/26) was distributed in the proportion of ICM/total cells ranging from 20 to 40% group. However, most NT blastocysts was biased in the 40-60%(34.1%, 15/44) and >60% (31.8%, 14/44) groups. Our findings suggest that placental abnormalities or early fetal losses in the present cloning system may be due to aberrant allocation of NT embryos to the ICM cells.

This study aimed to quality characteristics date for the industrialization of sausage prepared from chicken meat. The sausages were prepared by chicken meat and Jubak powder of CJ1(0%), CJ2(5%), CJ3(10%), CJ4(15%). In the proximate composition of the crude fat content of CJ4 was high and was increased according to Jubak powder addition ratios. Crude protein of CJ1 showed the highest. pH was increased but TBA, WHC, total cell counts were decreased according to Jubak powder addition ratios. WHC was decreased from the initial 46.77% to the final 38.81% according to Jubak powder addition ratios. TBA was increased from initial 0.16-0.19 to 0.2-0.59 at 15 days. Total cell counts in CJ2, CT3 and CJ4 showed the trend to be decreased according to Jubak powder addition ratios. Number of total cell counts of CJ1, CJ4 were increased from initially 2.2 to 8.9, 6.2 at 15 days. Elasticity, hardness, gumminess and cohesiveness of CT4 showed high value. L, a, b were increased according to Jubak powder addition ratios. In the sensory evaluation, Preference for color, overall of CJ1 showed high value. Preference for appearance flavor, taste of CJ2 showed high value. This result can be applied in storage property of chicken meat sausage added with Jubak powder.

The clarification of spatial-temporal patterns of phytoplankton from southern coastal waters during the period of March to November in 2004 was carried out. Total cell numbers were shown in 5,286 cells $ml^{-1}$ on March and reached to encounter a peak of 27,775 cells $ml^{-1}$ on July. Mean cell number was also shown in maximum of 1,587 cells $ml^{-1}$ on July, which recorded approximately two times higher than on June. The cell number of phytoplankton from southmiddle waters attained an abundance of ${\geq}35%$ regardless of months, which was the highest the abundance of phytoplankton in 2004 than any other waters in this study. Southwestern waters were lower the cell number of 2-5 times than those of southmiddle and southeastern waters. In particular, Prorocentrum occurred in southeastern waters on June and the highest cell number of 8,200 cells $ml^{-1}$ around Tongyeong region on July, which was recorded to occupy the value of 60.9% in southeast waters. The abundance of Skeletonema costatum as a dominant taxa in southwest was shown in ${\geq}60%$ on March, July, September, and October, whereas was also recorded to achieve the abundance of above 80% in southmiddle waters on March, July, and September. The majority of the taxa in southeastern waters was diatom: Eucampia zoodiacus, and Chaetoceros spp.. They occupied above 45%. On November, most of southern waters were abundant to Chaetoceros spp. On the basis of cluster analysis using SPSS ver 10.0, phytoplankton occurring on March showed somewhat no correlation with all of southern waters. In contrast to on March, the relationship between southwestern and southmiddle waters was shown on August and November, indicating a distinction from southeastern waters. However, the distance between southwestern/middle and southeastern waters appeared to be less than 5. Consequently, the abundance of phytoplankton in southern waters showed much fluctuations in temporal and spatial assays. In particular, southwestern and southmiddle waters during the periods of summer and winter appeared to be a similar to environmental characteristics.
To save cost and time in Kimchi manufacture, the use of raw red-pepper paste and device-mashed vice materials, in place of dried red-pepper powder, was examined. Two kinds of Kimchi were prepared: One with dried red pepper powder and device-not mashed vice materials and the other with raw red pepper paste and device-mashed vice materials. Then pH, total acidity, total viable cell counts, total lactic acid bacteria and sensory characteristics were evaluated. Comparisons of the two Kimchis, indicated that changes in pH, total acidity, the total number of viable cells and total lactic acid bacteria were similar between the two groups. The acceptability score of the Kimchi made using the raw red pepper paste and device-mashed vice materials was slightly lower than that of Kimchi made using the dried red-pepper powder. This color was indistinctly changed since the vice materials were mashed and mixed. In conclusion, the results indicate that when manufacturing Kimchi using device-mashed vice materials, Kimchi of better quality can be made by adding dried red-pepper powder.
This investigation is to determine whether the surface complexation of iron influence acute pulmonary responses induced by silica. For this study, three varieties of cation complexed silica were used: $silica-H^+,\;-Zn^{2+},\;and\;-Fe^{3+},$ since the first two are not active in the transport of electrons and generate little free radicals relative to the dust with the surface iron. Rats (270 to 280 g) were intratracheally (IT) instilled with saline, $silica-H^+,\;-Zn^{2+},\;or\;-Fe^{3+}$(5 mg in 0.5 ml saline). After 4 h, cell number, type, and differentiation were analysed in the bronchoalveolar lavage cells, and the levels of lactate dehydrogenase (LDH) and total protein were determined in the lavage fluid. In addition, bronchoalveolar lavage cells were cultured, and nitric oxide production was measured using nitrate assay. Inducible nitric oxide synthase (iNOS) mRNA in the bronchoalveolar lavage cells was also determined by northern blot analysis. Differential counts of the lavage cells showed that red blood cells were increased by 9-, 8-, and 13-fold and total leukocytes (lymphocytes plus polymorphonuclear neutrophils) by 48-, 36-, and 33-fold, following IT $silica-H^+,\;-Zn^{2+},\;and\;-Fe^{3+},$ respectively compared with the saline group. Meanwhile, there were no significant differences in red blood cells and total leukocytes among any of the cation complexed silica groups. The levels of LDH and total protein in the lavage fluid were significantly increased by 3- to 4-fold. However, compared among these silica groups, $Fe^{3+}$? complexation did not significantly change the LDH activity and total protein. NO production in cultured bronchoalveolar lavage cells was elevated by 2-fold, following IT any of the silica treatments compared with the saline group. Furthermore, the steady-state levels of iNOS mRNA in the lavage cells were greatly increased. There were any differences in iNOS mRNA expression among the silica-treated groups as with NO production. These findings suggest that surface complexed iron may not influence the acute pulmonary responses resulted from 4h exposure to silica.
